Effects of Chitosan with Different Molecular Weight and Nitrite Addition on the Residual Nitrite Contents and Self-life of Emulsified Sausage during Cold Storage (분자량이 다른 키토산과 아질산염 첨가가 유화형 소시지의 냉장 저장 중 아질산염잔존량 및 저장성에 미치는 영향)

  • The objective of this study was to determine the effects of residual nitrite contents, chitosan with different molecular weight and nitrite addition on emulsified sausage during cold storage. Six types of sausages were evaluated: control, 0.5% 50 kDa chitosan (T1), 0.5% 200 kDa chitosan (T2), 150 ppm nitrite (T3), 0.5% 50 kDa chitosan+150 ppm nitrite (T4), and 0.5% 200 kDa chitosan+150 ppm nitrite (T5). Each type of sausage was tested in triplicate and assigned to one of four storage periods: 0, 10, 20 and 30 days. As the storage time increased, the presence of chitosan and nitrite resulted in decreased residual nitrite value and increased pH (in control and T2), TBARS (thiobarbituric acid reactive substance) values, and total plate counts (TPC). Values for pH, TBARS, residual nitrite and total plate counts decreased significantly in response to the addition of chitosan and nitrite relative to the control (p<0.05). T5 was redder than the control (higher CIE$a^*$) at 30 d; however, no difference in the CIE $L^*$ and $b^*$ values was observed. T5 was significantly (p<0.05) more effective at delaying lipid oxidation when compared to the other treatment groups. T5 presented TPC that was significantly lower (p<0.05) than the other groups after three days of storage. In addition, the use of chitosan and nitrite in combination had much better antioxidant and antimicrobial effectiveness than other treatment groups. In conclusion, this study demonstrates that the addition of 0.5% 200 kDa chitosan and 150 ppm nitrite in combination with emulsified sausages tended to improve antioxidative and antimicrobial effects during storage when compared to other treatment groups.

Characteristics of Denitrification from Municipal Wastewater Treatment using a Combined Fixed Film Reactor (CFFR) Process (복합생물막 반응기를 이용한 하수처리시 탈질화 특성)

  • A new biological nutrient removal system combining $A^2/O$ process with fixed film was developed in this work and the characteristics of denitrification were especially investigated in the combined fixed film reactor(CFFR). Media was added in the anaerobic, anoxic and aerobic reactors, respectively. Tests were made to establish the effluent level of $NO_x-N$, COD, DO and nitrite effects on $NO_x-N$ removal in the CFFR by decreasing hydraulic retention time (HRT) from 10.0 to 3.5 hours and by increasing internal recycle ratio form 0% to 200%. The influent was synthesized to levels similar to the average influent of municipal wastewater treatment plants in Korea. SARAN media with a porosity of 96.3% was packed 40% / 130% / 25% based on its reactor volume, respectively. It was found that COD rarely limited dentrification in the anoxic reactor because of high $C/NO_x/-N$ ratio in the anoxic reactor, while DO concentration in the anoxic reactor and $NO_2-N/NO_x/-N$ from the aerobic effluent inhibited denitrification in the anoxic reactor. It was proved that the critical points of DO concentration in the anoxic reactor and $NO_2-N/NO_x/-N$ from the aerobic effluent were 0.15mg/L and 10%, respectively. As the internal recycle ratio increased, DO concentration in the anoxic reactor and $NO_2-N/NO_x/-N$ from the aerobic effluent increased. Especially, at the condition of internal recycle ratio, 200%, DO concentration in the anoxic reactor and $NO_2-N/NO_x/-N$ from the aerobic effluent exceeded the critical points of 0.15mg/L and 10%, respectively. Then, denitrification efficiency considerably decreased. Consequently, it was represented that the control of DO concentration in the anoxic reactor and $NO_2-N/NO_x/-N$ from the aerobic effluent can assure effective denitrification.

  • Nitrite, one of the precursors of dimethylnitrosamine distributes widely in natural foods and also used as a color fixative in meat products. In this paper Nitrites and Nitrates contents in Korean-Kimchi and Human Saliva were as follows. (1) pH: $pH:\;3.8{\sim}5.8$(in Korean-Kimchi soup) (2) NaCl: $1.5{\sim}3.1%$(in Korean-Kimchi soup) (3) Acidity: $0.38{\sim}0.62%$(in Korean-Kimchi soup) (4) $NO_2-$ $0.25{\sim}0.68ppm$ (in Korean-Kimchi) $1.9{\sim}5.0ppm$ (in Human Saliva) (5) $NO_3-$ $35.0{\sim}92.0ppm$ (in Korean-Kimchi) $7.6{\sim}28.0ppm$ (in Human Saliva) These results showed low contents in Korean-Kimchi and Human Saliva.

Electrodeposition of Gold on Fluorine-Doped Tin Oxide: Characterization and Application for Catalytic Oxidation of Nitrite

  • Sub-micrometer size gold particles were electrodeposited on a transparent fluorine-doped tin oxide (FTO) from acetonitrile solution containing $AuCl_4{^-}$ and tetramethylammonium tetraflouroborate (TMATFB) for detecting $NO_2{^-}$. A series of two-electron ($2e^-$) and one-electron ($1e^-$) reductions of the $AuCl_4{^-}-AuCl_2{^-}-Au$ redox systems were observed at FTO and a highly stable and homogeneous distribution of Au on FTO (Au/FTO) was obtained by stepping the potential from 0 to -0.55 V (vs. Ag/$Ag^+$). The Au/FTO electrode exhibited sufficiently high catalytic activity toward the oxidation of $NO_2{^-}$ with a detection limit (S/N = 3) and sensitivity of 2.95 ${\mu}M$ and 223.4 ${\mu}A{\cdot}cm^{-2}{\cdot}mM^{-1}$, respectively, under optimal conditions. It exhibited an interference-free signal for $NO_2{^-}$ detection with excellent recoveries from real samples.

Spectroscopic, Thermal and Biological Studies of Zn(II), Cd(II) and Hg(II) Complexes Derived from 3-Aminopyridine and Nitrite Ion

  • Microwave assisted syntheses of Zn(II), Cd(II) and Hg(II) complexes with 3-aminopyridine (3AP) and nitrite ($NO_2{^-}$) ions have been reported. The metal complexes were characterized by elemental analyses, molar conductance, IR, Far-IR, electronic, NMR ($^1H$, $^{13}C$), thermal and electron impact mass spectral studies. The spectroscopic studies reveal the composition, the nature of nitrite ligand in the complexes, electronic transitions, chemical environments of C and H atoms thermal degradation of the complexes. On the basis of characterization data, distorted tetrahedral geometry is suggested for Zn(II), Cd(II) and Hg(II) complexes. The organic ligand (3AP) and their metal complexes were screened against gram negative pathogenic bacteria and fungi in vitro. The results are compared with our previous report J. Korean Chem. Soc. 2013, 57, 341 on 4-aminopyridine and nitrite ion complexes of the same metal ions.

Histopathological study of chronic nitrite toxicity on the japanese eel, Anguilla japonica (아질산의 만성중독증에 의한 참장어(뱀장어)의 병리조직학적 연구)

  • The research was carried out to examine the chronic toxic effects of nitrite on the Japanese eel, Anguilla japonica by neans of histological observations. Young eel, 10.8g mean body weight. were exposed to 6 different concentrations of nitrite(1, 5, 10, 20, 30 and 40ppm) for 10 weeks. Each concentration was treated under 5 different levels of pH(5.5, 6.0, 6.5, 7.0 and 7.5) and each of these treatment was tested at 2 different temperature regimes($25^{\circ}C$ and $30^{\circ}C$). Proper concentration of nitrite was made by $NaNO_2$ and proper pH levels were made by the combinations of 0.1M $KH_2PO_4$ and 0.1M $NaHCO_3$. Histopathological test of gill tissues were made along with the test of the formation of thrombocystes and chloride cells on the gill filaments. At the lower pH levels, mucus secretion from the gill was incrased as the nitrite concentration increased. As the level of nitrite increased the number of chloride cells on the gill filament were decreased. Most of the remained chloride cells were observed only at the terminar part of the gill filament at 40ppm of nitrite. Degeneration of gill tissues were observed when nitrite levels were over 10ppm along with detachement and sweption of the epithelial cells of the gill lamellae. Shrunken gill lamellae and formation of thrombosis in the capillaries of gill lamellae were also observed. When temperature goes higher at the higher level of nitrite, necroses in the gill lamellae was increased. At the lower than 10ppm of nitrite, degeneration of gill lamellae was occured at the beginning of the test period but regenerated later. Negative effects of nitrite on the growth of young eel was started between 5~10ppm at the pH level of 7.0 and 7.5. Thrombosis formation were also started at this level. The safety concentration of nitrite at the pH levels of 7.0 and 7.5 on the small eel seems to be 1ppm. Thrombosis and gill lamella detachment and necrosis in the gill capillaries were not observed at this level. Chloride cells were appeared the whole part of the gill filament.

Antioxidant Effects and Anti-inflammation Effects of Lophatheri Herba Water Extracts Via Reducing iNOS Synthesis Induced by LPS in RAW 264.7 Cell (담죽엽의 항산화 효과와 RAW 264.7 세포에서 LPS로 유도된 iNOS 발현에 미치는 영향)

  • We studied to know the anti-inflammation effect on water extracts of Lophatheri Herba which was growing in every places in our country. We objected free radical scanvenger effect and nitrite eliminate effect of the Lophatheri Herba water extracts, and the cell viabillity, the effects of Lophatheri Herba water extracts on NO production, iNOS synthesis induced by LPS. Free radical scavenger effects were $27.91{\pm}0.12%$, $38.96{\pm}0.10%$, $46.22{\pm}0.15%$ depend on 0.5, 1.0, 2.0 mg/ml each dose of Lophatheri Herba water extracts. Nitrite eliminate effects were $9.86{\pm}0.3%$, $80.61{\pm}0.23%$, $97.62{\pm}0.56%$ in 0.1, 1.0, 2.0 mg/ml Lophatheri Herba water extracts on pH 1.2. NO production and iNOS synthesis induced by LPS were reduced in RAW 264.7 cell by Lophatheri Herba water extracts. As the above results, Lophatheri Herba water extracts have anti-inflammation effects via NO production decrease, iNOS synthesis decrease mechanism. So Lophatheri Herba water extracts will be used as the protection or treatment in chronic inflammation desease like a asthma, stomatitis etc.

Immune Cells Activity, Nitrite Scanvenging and ABTS Radical Scavenging Activities of Codonopsis lanceolata Ethanol Extracts from Districts in Korea

  • This study was executed to evaluate the immune activity, nitrite scavenging activity and ABTS radical scavenging activity against extracts of various concentration of ethanol solvent from Codonopsis lanceolata cultured at 6 local regions. The immune responses from both human T and B cell line was significantly enhanced in the cell growth compared to control while the cell growth was influenced at a certain period of culture. The results revealed that the cell growth of both human T and B cell was altered in a time dependent manner. The nitrite scavenging activity of ethanol extracts from various solvent concentration of C. lanceolata were affected by pH. At a pH of 1.2, the nitrite scavenging effect of all of the extracts tested observed higher than that of the other two pH ranges. There was no distinct detection of nitrite scavenging effects of the pH range 6.0. The ABTS radical scavenging activity was progressively increased in a dose-dependent manner, and the activity was the highest in 100% ethanol extract. The result from this investigation suggests that the extract of Codonopsis lanceolata could be an addition to basic medicine for immune modulation and natural food additives.

Effect of Temperature and FA Concentration on the Conversion of Ammonium to Nitrite (온도와 FA 농도가 암모늄 이온의 아질산 전환에 미치는 영향)

  • The effects of free ammonia (FA) concentration and temperature on nitrite accumulation were studied. To estimate the most effective ammonium oxidation and nitrite build-up condition, nitrification tests were conducted in batch conditions at various FA concentrations, and at different ammonium concentration and temperature. The activation energies of ammonium oxidizer were 81.7 KJ/mol below $20^{\circ}C$, and 32.5 KJ/mol over $20^{\circ}C$, while that of nitrite oxidizer was 35.5 KJ/mol irrespective of temperature variations. The results of nitrification tests conducted at different FA concentrations and temperatures showed that temperature strongly affects nitrite accumulation, while effects due to FA concentrations were found negligible.

New nirS-Harboring Denitrifying Bacteria Isolated from Activated Sludge and Their Denitrifying Functions in Various Cultures

  • By using PCR with nirS gene primers, three nirSharboring denitrifying bacteria (strain N6, strain N23, and strain R13) were newly isolated from activated sludge of a weak municipal wastewater treatment plant. Small-subunit rRNA gene-based analysis indicated that strain N6, strain N23, and strain R13 were closely related to Arthrobacter sp.,Staphylococcus sp., and Bacillus sp., respectively. In an attempt to identify their roles in biological nitrate and nitrite removal from sewage, we investigated their specific denitrification rates (SDNRs) for $NO_-^3$ - and $NO_-^2$ - in various cultures. All purecultures of each isolated nirS-harboring bacterial strain could remove $NO_-^3$ - and $NO_-^2$ - simultaneously in high efficiency, and the carbon requirements for $NO_-^3$ - removal of strain N6 and strain R13 were effectively low at 3.1 and 4.1 g COD/g $NO_3N$, respectively. In the case of mix-cultures of the strains (N6+N23, N6+R13, N23+R13, and N6+N23+R13), their SDNRs for $NO_-^3$ - were also effective, and their carbon requirements for $NO_-^3$ - removal were also effective at 3.0- 3.8 g COD/g NO3N. However, all tested mix-cultures accumulated $NO_-^2$ - in their culture media. On the other hand, the continuous culture of activated sludge mixed with strain N6 showed no significant increase of $NO_-^3$ - removal in comparison with strain N6's pure culture. These results suggest that nitrate and nitrite removal in biological wastewater treatment might be dependent on complicated bacterial interactions, including several effective denitrifying bacteria isolated in this study, rather than the specific bacterial types present and the number of bacterial types in activated sludge.
