Evaluation of Anti-Oxidant and Anti-Inflammatory Activities of Ganoderma lucidum Cultured on Hulled Barley (겉보리에서 배양한 영지버섯 추출물의 항산화 및 항염증 효능 평가)

  • Background: Ganoderma lucidum cultured on hulled barley was investigated as a potential natural source of antioxidants and anti-inflammatory agents. Methods and Results: The yields from Ganoderma lucidum cultured on hulled barley water and ethanol extract were 17.69% and 25.77%, respectively. The antioxidant activity of Ganoderma lucidum cultured on hulled barley extracts was confirmed by various methods including assayss of 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2-azino-bis-(3-ethylbenzo thiazoline-6-sulfonic acid) (ABTS), nitrite radical scavenging, and $Fe^{3+}$ to $Fe^{2+}$ reducing power activity. The ethanol extract of Ganoderma lucidum cultured on hulled barley showed improved DPPH, ABTS and nitrite radical scavenging activity compared with the water extract. After treatment of RAW264.7 cells with Ganoderma lucidum cultured on hulled barley ethanol extracts, the cell viability compared with the control was 92.82%, even at a concentration of $3,000{\mu}g/m{\ell}$. The ethanol extract inhibited reactive oxygen species (ROS) generation in RAW264.7 cells stimulated with $H_2O_2$, even at low concentrations. In addition, the ethanol extract showed an inhibitory effects on the production of lipopolysaccharide-induced nitric oxide (NO) in RAW264.7 cells. Conclusions: This study suggests that the extract of Ganoderma lucidum cultured on hulled barley is a potential source of natural antioxidants and anti-inflammatory agents.

Effects of added chlorides and their concentrations on growth and nitrate content in leaf lettuce(Lactuca sativa L.) in hydroponics.

  • Nitrate in edible vegetables is converted to nitrite by nitrate reductase(NR) and/or bacteria in intestines. Nitrite and amino, in the intestine of some animals and human, bind to form nitrosamine, which is toxic and known as carcinogen. This study was carried out to examine the effect of added chlorides and their concentrations on growth, yield and nitrate content in leaf lettuce plants in hydroponics. Seeds of lettuce cv, "Samsunjokchukmyon" were planted on April 29, and seedlings were planted on June 2, and were cultured until July 5 in 1998. KCI and CaCl₂ were used as chloride source and their concentrations were 1, 2 and 4 me/L, respectively, in the lettuce standard nutrient solution for National Horticultural Research Institute(NHRI). Completely randomized design with 3 replications was used. Nitrate content and NR activity were measured 2 and 5 weeks after planting(WAP). The obtained results were summarized as the follows : Leaf weight per plant was difference from harvest dates and treatments, but total leaf weight was not significantly different among treatments. Number of leaves was higher in KCI 2 me/L, CaCl₂1 me/L and control at 2 WAP than the others, and was higher in KCI 1 me/L, and control at 3 WAP than others, and was higher in control at 5 WAP. Total number of harvested leaves was the highest in control with 14, which followed by KCI 2 me/L and CaCl₂1 me/L. Nitrate content was decreased by addition of chloride in nutrient solution. Nitrate content in the 3rd and 9th leaves was significantly decreased. NR activity was higher in control and CaCl₂ addition treatments, while KCI addition treatments reduced NR activity. However, no direct relationship with nitrate was observed. Growth characteristics such leaf length and leaf width were not significantly influenced by chloride addition.

Production of Nitric Oxide and Erythropoietin in Serum of Newborn (신생아의 혈청내 Nitric Oxide와 Erythropoietin의 생성)

  • Nitric oxide(NO) is a potent and selective pulmonary artery vasodilator. Erythropoietin(EPO) is produced in the kidney in response to reduced oxygen availability. In this study, blood samples were collected for determination of NO and EPO concentrations in 18 normal newborns and 16 newborns with respiratory distress syndrome(RDS). Serum was measured by Ding's method for NO concentration and by enzyme-linked immunosorbant assay for EPO concentration. Nitric ion concentration in serum was 14.9$\pm$3.2 $\mu$M in normal control group and 12.8$\pm$3.3 $\mu$M in RDS group. EPO concentration in serum was 16.2$\pm$3.4 mU/ml in normal control group and 21.2$\pm$5.4 mU/ml in RDS group. These results show the decrease of NO and increase of EPO in RDS newborn patients in comparison with normal newborns. Such imbalances may contribute to the development of several clinical symptoms.

A Study on Intakes of Some Food Additives by Korean (한국인의 식품첨가물 섭취량 조사)

  • A survey for BHA (butyl hydroxy anisol), BHT(dibutyl hydroxy toluene), DHA (dehydroxy acetic acid), BA (benzoic acid), Sor. A. (Sorbic acid), POBA-Esters (P-hydroxy benzoic acid esters of ethyl, propyl, isopropyl, butyl and isobutyl), propionic acid, nitrite (as $N0_2$) and sulfites (as $S0_2$) of food additives which have much interesting to food consumers was carried out to assess the intake amount by Korean through the years of 1985-1987. Additive intakes were estimated by applying the determined a.dditive contents in the specific food to the specific daily food-intake amount obtained from the report of Korean national nutrition survey, and the level in cities were compared higher in general to that in rural communities, and the average level of the nation showed as follows; the estimated value of butyl hydroxy anisol and dibutyl hydroxy toluene in total was 0.017 mg, and 0.030 mg for dehydroxy acetic acid 0.064 mg for benzoic acid, 6.011 mg for sorbic acid, 0.247 mg for p-hydroxy benzoic acid eaters in total, 0.1438 mg for propionic acid 0.016 mg for nitrite ($N0_2$) and 0.022 mg for sulfite ($S0_2$) respectively, and those values were far below to that of FAO/WHO's ADls.

  • As the concentration of nitrogen in the sewage flowing into the sewage treatment plant increases due to urbanization and industrialization, the degree of adverse effects such as eutrophication and toxicity to the aquatic ecosystem is also increasing. In order to treat sewage containing high concentration of nitrogen, various studies on the biological nitrogen removal process are being conducted. Existing biological nitrogen removal processes require significant costs for supplying oxygen and supplementing external carbon sources. In this respect, as a high-level nitrogen removal process with economic improvement is required, an anaerobic ammonium oxidation process (ANAMMOX), which is more efficient and economical than the existing nitrification and denitrification processes, has been proposed. The purpose of this study is to confirm the stability of the ANAMMOX process in the water treatment process and to derive the ratio of ammonia nitrogen (NH4+) to nitrite nitrogen (NO2-) for the implementation of the mainstream ANAMMOX process. A laboratory-scale Mainstream ANAMMOX reactor was operated by applying the ratio calculated based on the substrate ratio suggested in the previous study. In the initial range, the removal efficiency of NH4+ was 58~86%, and the average removal efficiency was 70%. In the advanced range, the removal efficiency of NH4+ was 94~99%, and the average removal efficiency was 95%. As a result of the study, as the NH4+/NO2- ratio increased, the stability of the mainstream ANAMMOX process was secured, and it was confirmed that the NH4+ removal efficiency and the total nitrogen (TN) removal efficiency increased. As a result, the results of this study are expected to be used as basic data in the application of the ANAMMOX process in the mainstream.

Electrode Kinetics for Mixed Ligand Complexes of Cobalt (III) with Bis (ethylenediamine) and Monodendate Ligands (한자리 배위자와 에틸렌디아민의 코발트 (III) 착물에 대한 전극반응 속도론)

  • The heterogeneous rate constants for the electrochemical reduction by $trans-[Co(en)_2X_2](ClO_4)_n$(where X is cyanide, nitrite, ammonia, and isothiocyanate) at mercury and glassy carbon electrode were investigated by cyclic voltammetry, DC polarography, and by using rotating disk electrode. The good linear relationship was obtained between the activation energy of reduction and absorption wave number of complexes on glassy carbon electrode. At mercury electrode, $NO_2^-$ ligated complex showed the large deviation from the linear relationship. The difference in the value of rate constants for $NO_2^-$ ligated complex between mercury and glassy carbon electrode was about three order of magnitude which was much larger than the other complexes. It was suggested that $NO_^-$ ligated complex was reduced by inner-sphere mechanism on mercury electrode from the larger value of activation energy and entropy on mercury than carbon electrode.

Studies on Physiological and Functional Properties of Methanol Extract from Chicken Bile (닭 쓸개 methanol 추출물의 생리기능적 특성에 관한 연구)

  • Methanol extracts and powder of chicken bile were evaluated to determine antimicrobial, electron-donating, nitrite-scavenging, and inhibitory abilities against angiotensin I-converting enzyme (ACE). HPLC revealed taurochonodeoxycholic acid (TCDOA) and taurocholic acid (TCA) were major bile salts, at 5,893 and 395 mg/100g, respectively. Methanol extracts showed inhibitory effect on growth of Bacillus cereus, Salmonella typhimurium, Staphylococcus aureus, and Vibrio parahaemolyticus, whereas no effect on Escherichia coli. Electrondonating and nitrite-scavenging abilities increased significantly with increasing amount of bile samples. Electrondonating activity of dried powder was higher than that of methanol extracts, whereas nitrite-scavenging activity showed opposite trend. Both samples showed positive inhibitory activity of ACE. Methanol extracts showed higher activity than that of freeze-dried powder at high level of bile sample (5 and 10%).

Effects of Addition of Precursor and Inhibitor on Formation of N-nitrosamines During Kimchi Fermentation (니트로스아민의 전구 및 억제물질 첨가가 김치 숙성중 니트로스아민의 생성에 미치는 영향)

  • The study was focused to investigate the effect of nitrosodimethylamine (NDMA) formation due to nitrosamine (NA) precursor and/or inhibitor addition to Kimchi during the fermentation of Kimchi at room temperature ($16{\pm}2^{\circ}C$). The addition of nitrite and dimethylamine (DMA) to Kimchi showed the average 32-fold ($21.3{\sim}113.9\;\mu\textrm{g}/kg$) and 9-fold ($1.3{\sim}40.3\;\mu\textrm{g}/kg$) higher NDMA formation than control ($O{\sim}5.6\;\mu\textrm{g}/kg$), respectively. Also, the addition of both nitrite and DMA resulted in the increase of NDMA formation to average 42-fold ($39.4{\sim}155.7\;\mu\textrm{g}/kg$) higher than control. On the other hand, the addition of 4 ruM ascorbic acid to Kimchi with nitrite and DMA inhibited the formation of NDMA down to 71.3% of control. However, the addition of cysteine and erythorbic acid affected no inhibition to NDMA formation during the test. The addition of sodium sulfite increased NDMA formation. Accordingly, this showed that the amount of ascorbic acid generated during Kimchi fermentation played an important inhibition role for NA formation.

Effect of Drone Pupa Meal Added as Replacement of Sodium Nitrite and Vitamin C on Physico-chemical Quality Characteristics of Emulsion-type Sausage (아질산나트륨 및 비타민 C 대체로 첨가한 수벌번데기 분말이 유화형 소시지의 이화학적 품질 특성에 미치는 영향)

  • This study estimated the effect of drone pupa meal (DPM) added as replacement of sodium nitrite (SN) and vitamin C (VC) on physico-chemical quality characteristics of emulsion-type sausages. Samples were prepared either with 150 ppm SN+200 ppm VC (control); 75 ppm SN+100 ppm VC+6.015% DPM (T1); or 12.03% DPM (T2) and then stored at $4^{\circ}C$ for 30 days. The pH value decreased (p<0.05) with increase in the levels of DPM. Moisture and protein content decreased (p<0.05) but fat and ash content increased (p<0.05) with higher levels of DPM. T1 and T2 had higher (p<0.05) saturated fatty acids content and lower (p<0.05) unsaturated and polyunsaturated fatty acids content compared to the control. Lower (p<0.05) $L^*$ and $a^*$ values and higher (p<0.05) $b^*$ and $h^{\circ}$ values were exhibited in the T1 and T2 than in the control; and $C^*$ value was the lowest (p<0.05) in T2. The TBARS content was the highest (p<0.05) in T2, especially, 2 times higher (p<0.05) than in the control. T1 and T2 had harder (p<0.05) texture compared to the control. These findings suggest that the DPM has no replacement effects against SN and VC in emulsion-type sausage, but it has negative effects on color, lipid oxidation stability, and texture.

Anti-inflammatory effect of various solvent extract from Atractylodes japonica on Lipopolysaccharide-induced Inflammation in BV2 cells. (창출 추출물의 BV2 cell 소염작용에 관한 실험적 연구)

  • Objective : In this study, the effect of Atractylodes japonica against LPS induced inflammation in mouse microglia BV2 cells was investigated. Method : Microglia BV2 Cells viability was determined using the MTT assay. We used water, ethanol extract from Atractylodes japonica and studied on the anti-inflammatory effect of lipopolysaccharide-induced inflammation using reverse transcription polymerase chain reaction (RT-PCR), western blot, and nitric oxide detection on mouse microglia BV2 cells. Result : The MTT assay revealed that it's extract has no significant cytotoxicity in the microglia BV2 cell. Various solvent extract from Atractylodes japonica inhibited nitrite production, iNOS protein and mRNA expression levels. And also it's extracts significantly reduced lipopolysaccharide-induced COX-2 activation in RT-PCR and western blot in lipopolysaccharide-induced microglia BV2 cells Conclusion : In this study, it's extracts was shown to suppress NO production by inhibiting iNOS expression and COX-2 activity. With this effects of anti-inflammation, we suggests that, it's extracts may be a useful candidate for the development of a drug on the related inflammatory diseases in brain.
