Abies nephrolepis(Pinaceae) extracts were tested for determined immune system regulating activity based on antiinflammatory activity, antioxidant activity and anti-proliferative effect on HeLa cell line. The A. nephrolepis extracts increased dose-dependently anti-proliferation of HeLa cell line. The DM fraction of the extracts having anti-proliferatative effects of HeLa cell line was fractionalized four subfractions($D1{\sim}D4$). Inflammation-induced NO production was inhibited by D2 and D4 in LPS-activated RAW264.7 macrophages. And also, this fractions showed antioxidant activity examined by DPPH radical scavenging effects. These results suggest that the potential use of DM fraction of A. nephrolepis in chemoprevention and regulation overproduction of NO on pathogenic conditions. The mechanism of the inflammatory effects, however, must be evaluated through various parameters in the induction cascade of NO production.
This study was undertaken to define the effect of ChungSangSaWhaTang on the norepinephrine-induced arterial contraction and the mechanism of ChungSangSaWhaTang-induced relaxation. In order to investigate the effect of ChungSangSaWhaTang on contracted rabbit carotid arterial strips, transverse strips with intact or damaged endothelium were used for the experiment using organ bath. To analyze the mechanism of ChungSangSaWhaTang-induced relaxation, ChungSangSaWhaTang extract infused into contracted arterial strips induced by norepinephrine after treatment of indomethacin, $N_{\omega}$-nitro-L-arginine, or tetraethylammonium chloride. ChungSangSaWhaTang relax arterial strip with endothelium contracted by norepinephrine, but in the strips without endothelium, ChungSangSaWhaTang-induced relaxation was significantly inhibited. The endothelium-dependent relaxation induced by ChungSangSaWhaTang was decreased by the pretreatment of $N_{\omega}$-nitro-L-arginine, but it was not observed in the strips pretreated with indomethacin or tetraethylammonium chloride. We suggest that ChungSangSaWhaTang may inhibit agonist-induced contraction through the formation of nitric oxide in the vascular endothelial cells.
With the progress of chemical researches in ginseng studies, efforts to elucidate the pharmacological actions of ginseng have been greatly increased. The majority of ginseng reaserches in the past has been performed with crude extracts from ginseng roots to verify scientifically the empirical application of ginseng in men and animals recently. Ginseng reaserches have been done mostly with pure ginsenosides and there has been a shift in focus to the various biochemical pathways. It was demonstvated that ginseng had diverse effects by modulating the second-messenger system, such as cyclic nucleotides. calcium The demonstration in 1987 of the formation of nitric oxide(NO, endothelium-derlled rectating factor) by an enzyme in vascular endothelial cells opened up a new area of biological reaserches of ginseng. It was shown that vascular relaxations induced by glnsenosides are mediated by release of nitric oxide from endothelial cells. According to the literature search from'hledline". There have been 737 original and review articles during the last 30 years. In these review articles, an attempt has been made to summalize some results from some of these published papers. Ginseng has a wide range of phal.macologtcal and therapeutical actions. It acts on the centralral nervous system and cardiovascular system, promotes immune function and metabolism. In the present study, we assayed a number of compounds isolated from Panax ginseng C. A. Meyer (Araliaceae) for an ability to protect rat cortical cell cultures from the deleterious effects of the neurotoxicant, glutamate. We found that ginsenosides Rbl and Rg3 significantly attenuated glutamate-induced neurotoxicity. Brief exposure of cultures to excess glutamate caused extensive neuronal death. Glutamate-induced neuronal cell damage was significantly reduced by pretreatment with Rbl and Rgl. Ginsenosides Rbl and Rg3 inhibited the overproduction of nitric oxide which routinely follows glutamate neurotoxicity and preserved the level of superoxide dismutase in glutamate-treated cells. Furthermore, in cultures treated with glutamate, these ginsenosides inhibited the formation of malondialdehyde, a compound produced during lipid peroxidation, and diminished the influx of calcium. These results show that ginsenosides Rbl and Rg1 exerted significant neuroprotective effects on cultured cortical cells. As such, these compounds may be efficacious in protecting neurons from oxidative damage produced by exposure to excess glutamate.

Nitric oxide (NO) is synthesized by various cells involved in inflammatory reactions and may then act on mast cells. In the present work, we attempted to clarify the role of this molecule on the proliferation of mouse bone marrow derived-mast cells (BMMC). Swiss 3T3 fibroblastsproduced nitrite ($NO_{2}$) and nitrate ($NO_{3}$) upon treatment with interferon ${\gamma}$(IFN-${\gamma}$). This formation was dependent of L-arginine and could be inhibited by the -L-arginine analogue $N^{G}$-monomethyl-L-arginine ($N^{G}$MMA). The effect of IFN-g was drastically invreased by cotreatment with tumor necrosis factor g(IFN-g). BMMC were maintained in vitro for as long as 30 days when cocultured with Swiss 3T3 fibroblasts. coculture with $N^{G}$MMA, significantly increased the number of BMMC. These results indicate that NO involves the inhibition of proliferation of BMMC when cocultured with Swiss 3T3 fibroblasts.
In this study, powder of Orostachys japonicus A. Berger (O. japonicus) was extracted with 95% ethyl alcohol and fractionated using a series of organic solvents, including n-hexane (hexane), dichloromethane (DCM), ethylacetate (EtOAc), n-butanol (BuOH), and water ($H_2O$). We investigated the anti-inflammatory effects of these O. japonicus extracts on lipopolysaccharide (LPS)-stimulated RAW 264.7 cells. Their effects on the expression of inflammatory mediators and transcription factors were analyzed by Western blotting. DCM fraction significantly inhibited formation of reactive oxygen species (ROS) as well as nitric oxide (NO) in LPS-stimulated RAW 264.7 cells. Phosphorylation of the pro-inflammatory transcription factor complex nuclear factor-kappa B (NF-${\kappa}$B) p65 and expression of inducible nitric oxide synthase (iNOS), one of its downstream proteins, were also suppressed by DCM fraction. These effects were regulated by upsteam proteins in the mitogen-activated protein kinase (MAPK) and phosphoinositide 3-kinase/Akt (PI3K/Akt) signaling pathways. Taken together, our data suggest that O. japonicus could be used as a potential source for anti-inflammatory agents.

The present study was carried out to clarify whether isoflavonoids isolated from Belamcandae Rhizoma (Iridaceae) inhibit inflammation and angiogenesis by the experimental methods in vitro and in vivo. Among the isolated isoflavonoids, such as irigenin, irisflorentine, and iristectorene B inhibited nitric oxide (NO) production, as measured by nitrite formation at 3-30 ${\mu}M$. Also these compounds reduced cyclooxygenase-2 (COX-2) and inducible nitric oxide synthase (iNOS) enzyme expression in a concentration dependent manner, when measured by western blotting, at 3-30 ${\mu}M$. Irigenin, irisflorentine and iristectoren B decreased angiogenesis of chick embryos in the chorioallantoic membrane assay. These compounds also reduced the proliferation of calf pulmonary arterial endothelial (CPAE) cells and found to possess relatively weak gelatinase/collagenase inhibitory activity in vitro. These compounds, when administered subcataneously at the dose of 30mg/kg for 20 days to mice implanted with murine Lewis lung carcinoma (LLC), caused a significant inhibition of tumor volume. Therefore, antiangiogenic activities of isoflavonoids from Belamcandae Rhizoma might be due to antiproliferative activities under inhibition the induction of COX-2 and iNOS enzyme.
Research and development studies in internal combustion engines are set on a turning point due to requirements mostly purify the polluted environments. Naturally, basic studies concerned about engines are objected to elucidate formation mechanism of harmful matters, such as nitric oxide $(NO_x)$ and particulate matters. And for diesel engines, phenomenon in combustion chambers are analyzed in several approach ways in order to obtain detail understandings in closed and hardly observing space. In this article. it is discussed that the formation mechanism of diesel fuel sprays, mostly non-evaporating free diesel sprays. From that it would be promoted some new innovations in internal combustion engines of next generation.

Diesel engine is a major source of the air pollution. In general the concentrations of these pollu-tants in diesel engine exhaust differ from values calculated assuming chemical equibrium. Thus the detailed chemical mechanisms by which these pollutions form and the kinetic of these process-es are important in determining emission levels. In this study the computer program has been developed to calculate the required thermodynam-ic properties of combustion products(10 spacies) for both equilibrium and non-equilibrium in cylin-der for diesel engines. Nitric oxide emissions are calculated by using the extended Zeldovich Kinet-ic mechanism with a steady state assumption for the N concentration and equilibrium values used for H, O, $O_2$ and OH concentrations. By the results it is confirmed that developed simulations program with the NO prediction model is validated against residual mass fraction combustion index of Wiebe's functions pre-mixed com-bustion ration fuel injection timing.
영지균사체로부터 기능적인 면역활성 물질을 얻고자, 여러 조건으로 추출된 ${\beta}-glucan$성 다당류(GAM)의 Raw 264.7 대식세포 활성 정도를 알아보았다. GAN으로 자극된 Raw 264.7 대식세포는 대조군보다 약 $2{\sim}5$배의 nitric oxide 생성을 촉진하였다. 대식세포의 활성화 정도는 GAM의 구조와 분자량 그리고 화학적 조성에 따라 NO생성에 차이를 보였다. 서당을 탄소원으로 배양된 균사체로부터 추출된 알칼리 가용성이며 수용성인 G-BUC로 활성화된 Raw 264.7세포는 $22{\mu}M$의 NO생성을 유도하였다. G-SUC(WS)는 87%의 탄수화물과 3.4%의 단백질로된 평균분자량 30 kD의 다당류이다. 탄수화물의 구성당은 48%의 포도당과 22%의 mannose등으로 구성된 glucomannan성 GAM이다. 세포분획성 GAM가운데 세포벽으로부터 추출된 알칼리 가용성(CW-AB-WS)인 ${\beta}-glucan$성 다당류가 가장 많은 NO생성을 촉진시켜 영지의 대식세포 활성 다당류로 가장 잠재력이 있는 다당류인 것으로 나타났다. NO생성에 GAN과 대식세포 활성물질(LPS 및 $IFN-{\gamma}$)의 혼합 효과는 독자적인 자극보다는 혼합하여 자극했을 때 NO의 생성이 증폭되었다. GAM으로 활성화된 대식세포의 유사분열은 대조군에 비해 현저히 억제되었으며, 활성화된 대식세포의 배양액은 in vitro에서 L1210 암세포 주에 세포독성을 나타내었다. 이와 같은 결과로 보아 영지의 ${\beta}-glucan$성 다당류(GAN)는 ${\beta}-anomeric$ glucan receptor를 갖고 있는 대식세포를 자극하여 NO를 생성하며, 이때 NO생성의 촉진하는 다당류의 조건은 어느정도의 수용성을 갖고 있는 소량의 단백질이 포함된 고분자성 ${\beta}$glucan다당류이다.
