Effects of Jagamcho-tang on the C6 Glial Cell Injured by LPS Combined PMA (자감초탕(炙甘草湯)이 LPS와 PMA에 의해 손상된 C6 glial 세포에 미치는 영향)

  • The water extracts of Jagamcho-tang has been used for treatment of arrhythmia and palpitation in oriental traditional medicine. Brain is provided with blood flow by heart. Jagamcho-tang has been studied on ischemia and infarction in heart. However, little is known about the mechanism by which the water extracts of Jagamcho-tang rescues brain cells from ischemic damages. To elucidate the protective mechanism on ischemic induced cytotoxicity, the effects of Jagamcho-tang on ischemia induced cytotoxicity and generation of nitric oxide(NO) are investigated in C6 glioma cells. Jagamcho-tang induce NO in a dose dependent manner up to 2.5mg/ml in C6 glioma cells. The pretreatment of Jagamcho-tang protect sodium nitroprusside(SNP) (2mM) induced cytotoxicity. This effect of Jagamcho-tang is mimicked by treatment by pretreatment of SNP($100{\mu}M$), an exogenous NO donor. NG-monomethyl-L-arginine($N^{G}MMA$), a specific inhibitor of nitric oxide synthase (NOS), significantly blocks the protective effects of Jagamcho-tang on cell toxicity by ischemia. In addition, lipopolysaccharide(LPS) and phorhol 12 myristate 13-acetate(PMA) treatment for 72h in C6 glial cells markedly induce NO, but treatment of the cells with the water extracts of Jagamcho-tang decrease nitrite formation in a dose dependent manner. In addition, LPS and PMA treatment for 72h induce severe cell death and LDH release into medium in C6 glial cells. However treatment of the cells with the water extracts of Jagamcho-tang dose not induce significant changes compare to control cells. Furthermore, the protective effects of the water extracts of Jagamcho-tang is mimicked by treatment of $N^{G}MMA$. Taken together, I suggest that the protective effects of the water extracts of Jagamcho-tang against ischemic brain damages may be mediated by regulation of iNOS during ischemic condition.

Tetrahydrobiopterin Inhibits PDGF-stimulated Migration and Proliferation in Rat Aortic Smooth Muscle Cells via the Nitric Oxide Synthase-independent Pathway

  • Tetrahydrobiopterin (BH4), an essential cofactor for nitric oxide synthase (NOS) activity, is known to play important roles in modulating both NO and superoxide production during vascular diseases such as atherosclerosis. However, the role of BH4 in functions of vascular smooth muscle cells is not fully known. In this study, we tested the effects of BH4 and dihydrobiopterin (BH2), a BH4 precursor, on migration and proliferation in response to platelet-derived growth factor-BB (PDGF-BB) in rat aortic smooth muscle cells (RASMCs). Cell migration and proliferation were measured using a Boyden chamber and a 5-bromo-2'-deoxyuridine incorporation assay, respectively, and these results were confirmed with an ex vivo aortic sprout assay. Cell viability was examined by 2,3-bis [2-methoxy-4-nitro-5-sulfophenyl]-2H-tetrazolium-5-carboxanilide assays. BH4 and BH2 decreased PDGF-BBinduced cell migration and proliferation in a dose-dependent manner. The inhibition of cell migration and proliferation by BH4 and BH2 was not affected by pretreatment with $N^G$-nitro-L-arginine methyl ester, a NOS inhibitor. Moreover, the sprout outgrowth formation of aortic rings induced by PDGF-BB was inhibited by BH4 and BH2. Cell viability was not inhibited by BH4 and BH2 treatment. The present results suggest that BH4 and BH2 may inhibit PDGF-stimulated RASMC migration and proliferation via the NOS-independent pathway. Therefore, BH4 and its derivative could be useful for the development of a candidate molecule with an NO-independent anti-atherosclerotic function.

Anti-inflammatory effect of aged black garlic on 12-O-tetradecanoylphorbol-13-acetate-induced dermatitis in mice

  • BACKGROUND/OBJECTIVES: Although aged black garlic has various biological activities such as anti-allergy, anti-inflammation and neuroprotection, effect of aged black garlic on chemically contact dermatitis is unclarified. MATERIALS/METHODS: To evaluate anti-dermatitic activity of aged black garlic extract, we investigated effects of a fraction of aged black garlic extract (BG10) on both in vivo and in vitro. RESULTS: BG10 almost inhibited formation of nitric monoxide and interleukin-6 (IL-6; $IC_{50}$, $7.07{\mu}g/mL$) at $25{\mu}g/mL$, and dose-dependently reduced production of tumor necrosis $factor-{\alpha}$ ($TNF-{\alpha}$; $IC_{50}$, $52.07{\mu}g/mL$) and prostaglandin $E_2$ ($IC_{50}$, $38.46{\mu}g/mL$) in lipopolysaccharide-stimulated RAW264.7 cells. In addition, BG10 significantly inhibited the expression of inducible nitric oxide synthase, cyclooxygenase-2 and nuclear $NF-{\kappa}B$, and improved that of cytosolic levels of $NF-{\kappa}B$ and $I{\kappa}B{\alpha}$ in the cells. Consistent with in vitro studies, BG10 (0.5 mg/mL) not only reduced ear edema but also suppressed the formation of IL-6 and $TNF-{\alpha}$ induced by 12-O-tetradecanoylphorbol-13-acetate in ear tissues of mice. CONCLUSIONS: These findings suggest BG10 has anti-dermatitic activity through inhibiting activation of macrophages. Therefore, such effects of BG10 may provide information for the application of aged black garlic for prevention and therapy of contact dermatitis.

Oxidation-treated of Oxidized Carbons and its Electrochemical Performances for Electric Double Layer Capacitor (산화처리 탄소 및 이를 이용한 EDLC 특성)

  • The oxidation treatment of several carbon materials with a sodium chlorate and 70 wt.% of nitric acid, combined with heat treatment, were attempted to achieve an electrochemical active material with a larger capacitance. Among pitch, needle coke, calcinated needle coke and natural graphite, the structure of needle coke and calacinated needle coke were changed to the graphite oxide structure with the expansion of the inter-layer. On the other hand, the calcinated needle coke after oxidation and heating at $200^{\circ}C$ has exhibited largest capacitance per weight and volume of 29.5 F/g and 24.5 F/ml at the two-electrode system in the potential range of 0 to 2.5 V. The electrochemical performance of the calcinated needle coke was discussed with the phenomenon of the electric field activation and the formation of new pores between the expanded inter-layer at first charge.

Characteristics of NOB Formation in a Coaxial Multi-Air Staged LPG Flame (동축 공기다단 LPG화염의 NOx 생성특성에 관한 연구)

  • Experimental and numerical studies have been done to examine the effects of excess air ratio and tertiary air swirl number on the formation characteristics of NOx in a pilot scale combustor adopting a multi-air staged burner. In numerical calculation the mathematical models for turbulence, radiation and nitric oxide chemistry were taken into account. The radiative transfer equation was solved using the discrete ordinates method with the weighted sum of gray gases model. In the NOx chemistry model, the chemical reaction rates for thermal and prompt NOx were statistically averaged using a probability density function. The results were validated by comparison with measurements. For the experiment, a 0.2 MW pilot multi-staged air burner has been designed and fabricated. Using the numerical simulation developed here, a variation of thermal and prompt NOx formation was predicted by changing the excess air ratio and tertiary air swirl number. As the excess air ratio increased up to 1.9, the formation of the total as well as thermal NOx at exit increased while the prompt NOx decreased. The formation of thermal NOx was more affected by concentration of $O_2$ and $N_2$ than gas temperature. When the tertiary air swirl number increased, the formation of the total as well as the prompt NOx slightly decreased.

A Study on the Visualization of NO Concentration Distributions in $CH_4/O_2N_2$ Premixed Flames by PLIF (평면 레이저 유도 형광법(PLIF)을 이용한 $CH_4/O_2N_2$ 예혼합화염의 NO 농도 분포 가시화에 관한 연구)

  • In this study, quantitative measurement of nitric oxide concentration distributions visualization were investigated in the laminar $CH_4/O_2N_2$ nixed flame by Planar laser-induced fluorescence(PLIF). The NO A-X (0,0) vibrational band around 226nm was excited using a XeCl excimer-pumped dye laser. Selecting an appropriate NO transition minimizes interference from Rayleigh scattering and $O_2$ fluorescence. The measurements were taken in $CH_4/O_2N_2$ premixed flame with equivalence ratios varying from $1.0{\sim}1.6$, and a fixed flow rate of 3slpm. NO was found to produce primarily between an inner premixed and an outer nonpremixed flame front, and total NO concentration is raised when equivalence ratios increase. These results suggest that prompt NO is likely to contribute to MO formation in $CH_4/O_2N_2$ premixed flame. Furthermore, this trend was well matched with previous works.

Multidimensional Engine Modeling: NO and Soot Emissions in a Diesel Engine with Exhaust Gas Recirculation

  • The effects of EGR(Exhaust Gas Recirculation) on heavy-duty diesel engine performance, NO and soot emissions were numerically investigated using the modified KIVA-3V code. For the fuel spray, the atomization model based on the linear stability analysis and spray wall impingement model were developed for the KIVA-3V code. The Zeldovich mechanism for the formation of nitric oxide and the soot model suggested by Hiroyasu et al. were used to predict the diesel emissions. In this paper, the computational results of fuel spray, cylinder pressure, and emissions were compared with experimental data, and the optimum EGR rates were sought from the NO and soot emissions trade-off. The results showed that the EGR is effective in suppressing NO but the soot emission was increased considerably by EGR. Using cooled EGR, soot emission could be enhanced without worsening of NO.

Protection of Peroxynitrite-Induced DNA Damage by Dietary Antioxidants

  • The present study was undertaken to test the hypothesis that dietary antioxidants protect DNA damage induced by peroxynitrite, a potent physiological inorganic toxin. The present study showed that dietary antioxidants such as (-)-epigallocatechin gallate, quercerin, rutin, resveratrol, and ursolic acid inhibit single strand breaks in supercoiled plasmid DNA induced by 3-morpholinosydnomine N-ethylcarbamide (SIN-1), a generator of peroxynitrite through the reaction between nitric oxide and superoxide anion. The formation of 8-hydroxy-2'-deoxyguanosine (8-OH-dG) in calf thymus DNA by SIN-1 was also inhibited by dietary antioxidants. When U937 cells were incubated with 1 mM SIN-1 bolus, a significant increase of 8-OH-dG level was observed. However, oxidative DNA damage was significantly lower in the cells pre-treated with dietary antioxidants when cells were exposed to SIN-1.

RNAi-mediated reduction of xanthine dehydrogenase results in increased biomass of Arabidopsis seedlings

  • Xanthine dehydrogenase (XDH), a classic enzyme involved in purine catabolism, can catalyze the formation of redox-signaling reactive oxygen and nitrogen species such as superoxide and nitric oxide. We generated transgenic plants of Arabidopsis in which XDH was knocked out by introduction of hairpin RNA-expression vector. Expression analysis by reverse transcription-PCR and in-gel staining of XDH activity revealed that transgenic lines efficiently suppressedXDH expression at the transcriptional level, demonstrating that RNA interference was successfully induced. XDH-suppressed transgenic lines exhibitedincreased biomass production during the growth of seedlings.
