Inhibitory Effect of Panax notoginseng on Nitric Oxide Synthase, Cyclo-oxygenase-2 and Neutrophil Functions

  • Our preliminary aim is to elucidate the pharmacokinetic features of the PNS(Panax notoginseng Buck F.H. Chen. (Arialiaceae) root). First, we assessed the prevention of neurtrophil functions. A Panax notoginseng inhibited neutrophil functions, including degranulation, superoxide generation, and leukotriene B4 production, without any effect on 5-lipoxygenase activity. This Panax notoginseng reduced nitric oxide (NO) and prostaglandin E2 production in mouse peritoneal macrophages stimulated with lipopolysaccharide, whereas no influence on the activity of inducible NO synthase, cyclo-oxygenase-2 or cyclo-oxygenase-1 was observed. Panax notoginseng significantly reduced mouse paw oedema induced by carrageenan. The results indicate that Panax notoginseng exerts anti-inflammatory effects related to the inhibition of neutrophil functions and of NO and prostaglandin E2 production, which could be due to a decreased expression of inducible NO synthase and cyclo-oxygenase-2.

  • Aim of the present study was to investigate whether methanol extract from the leaves of Staphylea bumalda could be used to suppress lipopolysaccharide (LPS)-induced inflammatory responses in murine macrophage cell lines, Raw 264.7 cells. The extract reduced nitric oxide (NO), cyclooxigenase-2 (COX-2) and pro-inflammatory cytokines production from LPS-stimulated Raw 264.7 cells. These inhibitory effects were associated with decreases in the phosphorylation of MAP kinases and the activity of $NF{\kappa}B$ signal pathways. Our results indicate that Staphylea bumalda significantly inhibits the inflammatory activity of activated macrophages, suggesting that Staphylea bumalda could be a potential candidate for the treatment of inflammatory disease.

  • Pueraria flos and Adzuki Beans contains several bioactive compounds, such as saponin, oleanolic acid, and flavone. Pueraria flos and Adzuki Beans has traditionally been used to treat disorders of antioxidant activity, diabetes and liver detoxication, and it has antinociceptive and anti-inflammatory properties. However, complex saponin were validity of the anti-inflammatory activity has not been scientifically investigated. In this study, to determine anti-inflammatory activity of complex saponin separated from Pueraria flos and Adzuki Beans mixture extract on nitric oxide and prostaglandinE2 assay. The anti-inflammatory activities of complex saponin separated from Pueraria flos and Adzuki Beans mixture extract were evaluated for inhibitory activities against lipopolysacchride induced nitric oxide and prostaglandinE2 production protein expressions in RAW264.7 cell lines. The complex saponin separated from Pueraria flos and Adzuki Beans mixture extract inhibitory activity for both tests with protein high depressions(%) values showed in the ranges of 50~100 ㎍/ml. Overall, prostaglandinE2 tests had a higher inhibitory effect on inflammation than nitricoxide tests. Theseis result suggest a potential role of complex saponin separated from Pueraria flos and Adzuki Beans mixture extract as source of anti-inflammation agent.

  • Nitric oxide (NO) has been suggested to play an important role in endotoxin-mediated shock and inflammation. In this study, we investigated the effect of Rosa laevigata Michx. (Rosaceae) on the production of NO and the molecular mechanism of its action. Rosa laevigata inhibited NO generation and iNOS expression in LPS-stimulated murine macrophages. Activity of nuclear $factor{-\kappa}B\;(NF{-\kappa}B)$ and the degradation of $I{\kappa}B-{\alpha}$ were suppressed by Rosa laevigata. Furthermore, extracellular signal-stimulated kinase (ERK), which is known to be involved in $NF{-\kappa}B$ activation, is inhibited by Rosa laevigata. These results suggest that Rosa laevigata could exert its anti-inflammatory actions by suppressing the synthesis of NO through inhibition of $NF{-\kappa}B$ activity.

Anti-Oxidative and Nitric Oxide Production Inhibitory Activities of Phenolic Compounds from the Fruits of Actinidia arguta

  • Phytochemical examination of the fruits of Actinidia arguta has led to the isolation three organic acids, one coumarin, and three flavonoids. Structures of these compounds were elucidated as protocatechuic acid (1), caffeic acid (2), $caffeoyl-\beta-D-glucopyranoside$ (3), esculetin (4), quercetin (5), quercetin $3-O-\beta-D-galactopyranoside$ (6), and quercetin $3-O-\alpha-L-rhamnopyranosyl$ $(1\rightarrow6)-O-\beta-D-glucopyranoside$ (7) by comparisons with previously reported spectral data. To investigate the anti-inflammatory and anti-oxidative effects of these compounds, nitric oxide production inhibitory activity in LPS-stimulated RAW 264.7 cells and DPPH radical scavenging activities were examined. Nitric oxide productions were reduced significantly by the addition of compounds $[1(IC_{50}=59.27{\mu}g/ml),\;2(IC_{50}=27.95{\mu}g/ml),\;3(IC_{50}=73.09{\mu}g/ml),\;4(IC_{50}=67.44{\mu}g/ml),\;5(IC_{50}=17.40{\mu}g/ml),\;6(IC_{50}=41.99{\mu}g/ml),\;7(IC_{50}=54.46{\mu}g/ml)]$, and extracts $(IC_{50}=56.21{\mu}g/ml)$ compared with positive control, L-NMMA $(IC_{50}=14.48{\mu}g/ml)$. The phenolic compounds also showed anti-oxidative activities. Especially, Compounds $1(IC_{50}=8.87{\mu}g/ml),\;4(IC_{50}=3.41{\mu}g/ml),\;and\;5(IC_{50}=6.06{\mu}g/ml),\;$, showed potent anti-oxidative activities similar to L-ascorbic acid $(IC_{50}=5.89{\mu}g/ml)$.

청동풍탱이(Anomala albopilosa)추출물의 항산화성 및 생리기능 (Antioxidant Activity and Physiological Function of the Anomala albopilosa Extracts)

  • 산화적인 스트레스(oxidative stress)는 신경염증의 발병 요인 중의 하나로 알려져 있다. 이에 본 연구는 약용곤충으로 알려진 청동풍뎅이(Anomala albopilosa) 성충과 3령 유충(궁벵이) 에탄올 추출물을 대상으로 항산화활성을 비교 측정하였으며, 또한 대식세포인 RAW 264.7 세포에서의 NO 의 생성억제 효과 및 세포독성 그리고 분자 염증관련 인자인 iNOS, COX-2와 $PGE_2$ 생성 및 활성 억제를 조사하여 항염 증효과 등의 생리활성을 측정 비교하였다. 우선, 분자염증은 활성산소 관련의 물질과 밀접한 관련이 있으므로 향산화 실험과 관련하여 청동풍탱이 성충과 3령 유충 에탄올 추출물 의 DPPH(1,1-diphenyl-2-picrylhydrazyl) radical 소거능, superoxide radical 소거능, xanthine oxidase 저해활성 그리고 nitric oxide 소거능에 대한 항산화활성 등의 assay를 실시하였다. 그 결과, 청동풍뎅이 에탄올 추출물은 항산화능을 갖고 있었으며, 3령 유충 추출물에서보다는 성충 추출물에서 다소 높은 항산화활성을 보여주었다. 또한 RAW 264.7 세포에 LPS로 자극을 주고 청동풍뎅이 성충과 3령 유충 에탄올 추출물을 처리하여 항염증관련 활성을 확인해본 결과, 고농도에서는 다소 세포독성을 나타냈지만 농도의존적으로 NO, iNOS, COX-2 그리고 $PGE_2$의 생성 억제효과가 나타났다. 이러한 결과는 유용곤충자원을 이용한 유효성분 추출을 통한 항산화 및 항염증 물질의 연구 또는 예방하거나 치료할수 있는 염증 억제 성분의 분리 및 그 작용기전 연구에 중요한 기초 자료가 될 것이라 사료된다.

Di- and Sesqui-Terpenoids Isolated from the Pods of Sindora sumatrana and Their Potential to Inhibit Lipopolysaccharide-Induced Nitric Oxide Production

  • Activity-guided fractionation of the n-hexane and ${CHCl_3}-soluble$ fractions of Sindora sumatrana using a bioassay based on the inhibition of lipopolysaccharide (LPS)-induced nitric oxide (NO) production by inducible nitric oxide synthase (iNOS) in murine macrophage RAW 264.7 cells led to the isolation of the known compound, $(+)-7{\beta}-acetoxy-15,16-epoxy-3$, 13(16), 14-clero-datriene-18-oic acid (2) as an active constituent. In addition, a new trans-clerodane diterpenoid, (+)-2-oxokolavenic acid (1), together with six known compounds, (+)-3, 13-clerodadiene-16,15-olide-18-oic acid (3), $(+)-7{\beta}-acetoxy-3$,13-clerodadiene-16,15-olide-18-oic acid (4), $(+)-7{\beta}-acetoxy-16-hydroxy-3$,13-clerodadiene-16, 15-olide-18-oic acid (5), ${\beta}-caryophyllene$ oxide (6), $clovane-2{\beta},9{\beta}-diol (7),{\;}and{\;}caryolane-1,9{\beta}-diol$ (8) were isolated and found to be inactive. The structure of compound 1 was determined using physical and spectroscopic methods such as 1D and 2D-NMR experiments. The known compounds 2-8 were identified by the spectroscopic data and by comparison with the published values. Of eight isolates (1-8), only compound 2 exhibited an iNOS inhibitory activity with $IC_{50}$/ value of $51.6{\;}\mu\textrm{m}M$.

  • Prostaglandins biosynthesis and nitric oxide production have been implicated in the process of inflammation. In this study we investigated on the effects of ethyl acetate extract of Angelica Koreana Radix (EAKR) on the activities of prostaglandin $E_2\;(PGE_2)$, proteoglycan (PG) degradation and nitric oxide synthase (NO) in inflammation cytokines-activated rabbit articular chondrocytes. EAKR exhibited inhibitory activities on NO production and $PGE_2$ production as $73.08\%\;and\;89.49\%$, respectively at $20{\mu}g/ml$ and inhibited the degradation of PG in a concentration-dependent manner Zelatin zymography analysis demonstrated that EAKR significantly inhibited MMP-2, 9 expression in chondrocytes. In vivo, EAKR was shown to have inhibitory effects on acetic acid-induced pain. This study suggests that modulation of $PGE_2$, NO, PG degradation and MMP-2, 9 by EARK may be important in the prevention of inflammation and osteroarthritis.

Bioassay-coupled LC-QTOF MS/MS to Characterize Constituents Inhibiting Nitric Oxide Production of Thuja orientalis

  • The ethyl acetate fractions prepared from the leaves of Thuja orientalis significantly inhibited nitric oxide (NO) production in lipopolysaccharide-stimulated BV2 microglial cells. According to bioassay-coupled LC-QTOF MS/MS, the components near 22 and 25 mins in the mass chromatogram highly inhibited NO production and were expected to be labdane diterpenes, and the active components were characterized via further isolation. The results of the NO production inhibitory assay of the isolated compounds correlated well with the results of bioassay-coupled LC-QTOF MS/MS. Among the identified constituents, NO production inhibitory activities of 16-hydroxy-labda-8(17),13-diene-15,19-dioic acid butenolide (2) and 15-hydroxypinusolidic acid (3) were newly reported. Taken together, these results demonstrated that LC-QTOF MS/MS coupled with NO production inhibition assay was a powerful tool for accurately predicting new anti-inflammatory constituents in the extracts from natural products. Moreover, it provided a potential basis for broadening the application of bioassay-coupled LC-QTOF MS/MS in natural product research.

Anti-oxidative, Nitric Oxide Inhibitory Activities and Irritation Test of the Fermented Opuntia humifusa Cladodes

  • Opuntia humifusa is a member of the Cactaceae family. In the present study, the antioxidant, nitric oxide(NO) inhibitory activities and potential irritation response of the fermented Opuntia humifusa cladodes(FOH) were investigated for cosmetic use. Antioxidant activities were tested using 1,1-diphenyl-2-picrylhydrazyl(DPPH) and xanthine oxidase assay, we found that FOH could scavenge DPPH free radicals and inhibit xanthine oxidase activity in a dose dependent manner, with $IC_{50}$ of 2599.46${\mu}g/ml$ and 721.38${\mu}g/ml$, respectively. To investigate the possible anti-inflammatory effects of FOH, RAW 264.7 macrophages were pretreated with FOH($0{\sim}400{\mu}g/ml$) for 30 min and then treated with LPS for 24 h. We found that cell number did not vary significantly with the treatment of FOH, and FOH did not show any inhibitory effects on LPS-induced NO production. After application of FOH to rabbits for skin and eye irritation test, the experimental sites did not show any response compared to the control. FOH were considered to be a non-irritant to the skin and eye. Based on the above information, we suggest that FOH can be considered to be a non-irritant base cosmetic material for safely use.
