Flow of Soluble Non-ammonia Nitrogen in the Liquid Phase of Digesta Entering the Omasum of Dairy Cows Given Grass Silage Based Diets

  • An experiment was conducted to quantify the flow of soluble non-ammonia nitrogen (SNAN) in the liquid phase of ruminal (RD) and omasal digesta (OD), and to investigate diurnal pattern in SNAN flow in OD. Five ruminally cannulated Finnish-Ayrshire dairy cows in a $5{\times}5$ Latin square design consumed a basal diet of grass silage and barley grain, and that supplemented with four protein feeds (kg/d DM basis) as follows: skimmed milk powder (2.1), wet distiller' solubles (3.0), untreated rapeseed meal (2.1) and treated rapeseed meal (2.1). Ruminal digesta was sampled using a vacuum pump, whereas OD was collected using an omasal sampling system at 1.0 h interval during a 12 h feeding cycle. Both RD and OD were acidified, centrifuged to remove microbes and precipitated with trichloroacetic acid followed by centrifugation. The SNAN fractions (free amino acid (AA), peptide and soluble protein) in RD and OD were assessed using ninhydrin assay. Free AA, peptide and soluble protein averaged 60.0, 89.4 and 2.1 g/d, respectively, for RD, and 81.8, 121.5 and 2.5 g/d, respectively, for OD. Although free AA flow was relatively high, mean peptide flow was quantitatively the most important fraction of SNAN, indicating that degradation of peptide to AA rather than hydrolysis of soluble protein to peptide or deamination may be the most limiting step in rumen proteolysis. Diurnal pattern in flow of peptide including free AA in OD during a 12 h feeding cycle peaked 1 h post-feeding, decreased by 3 h post-feeding and was relatively constant thereafter. Protein supplementation showed higher flow of peptide including free AA immediately after feeding compared with no supplemented diet. There were no differences among protein supplements in diurnal pattern in flow of peptide including free AA in OD.

Soluble Non-ammonia Nitrogen in Ruminal and Omasal Digesta of Korean Native Steers Supplemented with Soluble Proteins

  • An experiment was conducted to study the effect of soluble protein supplements on concentration of soluble non-ammonia nitrogen (SNAN) in the liquid phase of ruminal (RD) and omasal digesta (OD) of Korean native steers, and to investigate diurnal pattern in SNAN concentration in RD and OD. Three ruminally cannulated Korean native steers in a $3{\times}3$ Latin square design consumed a basal diet of rice straw and corn-based concentrate (control), and that supplemented (kg/d DM basis) with intact casein (0.24; IC) or acid hydrolyzed casein (0.46; AHC). Ruminal digesta was sampled using a vacuum pump, whereas OD was collected using an omasal sampling system at 2.0 h intervals after a morning feeding. The SNAN fractions (free amino acid (AA), peptide and soluble protein) in RD and OD were assessed using the ninhydrin assay. Concentrations of free AA and total SNAN in RD were significantly (p<0.05) lower than those in OD. Although free AA concentration was relatively high, mean peptide was quantitatively the most important fraction of total SNAN in both RD and OD, indicating that degradation of peptide to AA rather than hydrolysis of soluble protein to peptide or deamination may be the most limiting step in rumen proteolysis of Korean native steers. Diurnal variation in peptide concentration in OD for the soluble protein supplemented diets during the feeding cycle peaked 2 h post-feeding and decreased thereafter whereas that for the control was relatively constant during the entire feeding cycle. Diurnal variation in peptide concentration was rather similar between RD and OD.

재래식 메주 및 된장중의 항산화성 물질에 관한 연구 3. 수용성 갈변물질의 항산화력 (Antioxidative Materials in Domestic Meju and Doengjang 3. Separation of Hydrophilic Brown Pigment and Their Antioxidant Activity)

  • Hydrophilic brown pigments(HBPs) produced during fermentation and aging the domestic Meju and Doenjang were separated by dialysis and chromatography , and their antioxidant activity were measured . The chemical properties of HBP were determined by UV and IR spectrophotometry. HBPs contents were found to be 93.1 mg/g and 183.2mg/g in Meju fermentated for 30 days and 80 days , respectively. The ratio of dialysate to diffusate of the HBPs were appeared to be 70 : 30 and 87 :13 in the Meju fermented for 30 days and 80 days, respectively. and the rtio in the Doenjang aged for 60 days was 91 :9 , indicating that dialysate slowly days, respectively, and the ratio in the Doenjang aged for 60 days was 91 : 9, incidatin that dialysate slowly increased by the fermentation . Both portion exhibited strong antioxidant activity, but more stronger antioxidant activity was found in the dialysate. DEAE-celluose column chromatography showed that dialysate contained more materials eluted by 0.01-0.03M HCI solution than 0.01M acetate buffer, but diffusate showed opposite results. The degree of browning reaction and antioxdiant activity found in the fraction eluted by HCI solution was stronger than that of the fraction eluted by acetate buffer. The fraction eluted from DEAE-cellulose column chormatogrphy was further fractionaged by TLC and found that strong antioxidant activity was present in the fractions which did not possess fluorescenece and showed a negative ninhydrin reaction. TLC fractions of HCI eluant in Meju exhibited a strong absorbance at 260-280nm, but most of other fractions did not show any absorbance at UV region. TLC fractions from dialysate and diffusate showed fairly identical IR spectrum with absorbance at 3400cm-1, 2800cm-3000cm -1, 1600cm, -1 1400cm-1 and 1100 cm-1 , however, in addition to these absorbances, the spectrum from HCI eluant of Meju exhibited a strong absorbance at 1750cm , indicating the carbonic acid or carbonate ester.

An Antifungal Antibiotic Purified from Bacillus megaterium KL39, a Biocontrol Agent of Red-Pepper Phytophthora-Blight Disease

  • Bacillus megaterium KL39, an antibiotic-producing plant growth promoting rhizobacterium (PGPR), was selected from soil. The antifungal antibiotic, denoted KL39, was purified from culture filtrate by column chromatography using Dion HP-20, Silica gel, Sephadex LH-20, and prep-HPLC. Thin layer chromatography, employing the solvent system of ethanol:ammonia:water=8:1:1, showed the $R_{f}$. value of 0.32. The antibiotic KL39 showed a negative reaction with ninhydrin solution, positive with iodine vapor, and also positive with Ehrlich reagent. It was soluble in methanol, ethanol, butanol, and acetonitrile, but insoluble in chloroform, toluene, hexane, ethyl ether, or acetone. Its UV spectrum had the maximum absorption at 208 nm. Amino acid composition, FAB-mass, $^{1}H-NMR,\;^{13}C-NMR$, and atomic analyses showed that the antibiotic KL39 (MW=1,071) has a structure very similar to iturin E. The antibiotic KL39 has a broad antifungal spectrum against a variety of plant pathogenic fungi including Rhizoctonia solani, Pyricularia oryzae, Monilinia froeticola, Botrytis cinenea, Altenaria kikuchiana, Fusarium oxysporum, and F. solani. An MIC value of $10\;{\mu}g/ml$ was determined for Phytophthora capsici. Macromolecular incorporation studies with P. capsici using radioactive [$^{3}H-adenine$] as the precursor, indicated that the antibiotic KL39 strongly inhibits the DNA biosynthesis of the fungal cell. Microscopic observation of the antifungal action showed abnormal hyphal swelling of P. capsici. The purified antibiotic KL39 was very effective for the biocontrol of in vivo Phytophthora-blight disease of pepper.

전통된장으로부터 Angiotensin Converting Enzyme 저해물질의 분리 (Isolation of Angiotensin Converting Enzyme Inhibitor from Doenjang)

  • 전통된장으로부터 안지오텐신전환효소(angiotensin converting enzyme; ACE)를 저해하는 물질을 추출하여 그 구조를 밝혀냈다. ACE 저해물질을 열수추출한 다음 gel permeation chromatography (GPC)를 통하여 ACE 저해작용이 큰 두 개의 큰 획분을 받았다. 앞획분은 90%와 70%의 ACE 저해효과를 나타내었으나 단일물질로 분리되지 않아 계속하여 역상 HPLC를 통하여 순수 분리를 하였다. 그러나 앞획분은 순수분리되지 않아 결국 2차원 전기영동/TLC를 통하여 분리한 결과 분자량이 759.63인 아미노기를 갖고 있는 비펩타이드 물질임이 밝혀졌다. 뒷획분은 다른 조건의 HPLC(reverse column과 $NH_2$, column)를 이용하여 순수분리에 성공하였다. 이중 ACE 저해효과가 큰 물질은 분자량 271.33인 dipeptide인 arginine-proline임을 밝혀냈다. 이물질의 ACE $IC_{50}$$92\;{\mu}M$이었다. 본 연구 결과는 대부분 ACE 저해물질이 3개 내지 7개 등의 긴 펩타이드임을 감안할 때, 짧은 dipeptide로 ACE 저해펩타이드가 한국의 전통된장에서 생산할 수 있음을 보여주고 있다.

  • Total carbohydrates, amino acids and peptide-like substances in two kinds of Korean lager beers have been analyzed by the calorimetric method of Dreywood's anthrone reagent and Fowden's ninhydrin reagent. The samples were fractionated with column of ion-exchange resin. The experimental results are as follows; 1. Amounts of non-hydrolyzed carbohydrates in the part of column processed is 1. 82% and 1. 96 % (the value was measured by Bertrand's method). But the amounts of those measured by Dreywood's anthrone method are 5.57% and 4.25%, this values are much more than those of Bertrand's method. 2. It can be estimated the amounts of gum and dextrin are 3.75% and 3.30% in both two beers, by comparison of samples with the above mentioned two method. 3. The amounts of carbohydrates by anthrone reagent in acid-hydrolyzed beers are much increased than those of non-hydrolyzed, so it is suggested the presence of polymer carbohydrates which couldn't be detected by Bertrand's method. 4. Total amounts of amino acids are 0.015% and 0.025% (as glycine) in non-hydrolyzed beers measured by ninhydrine color reaction method, on the other hand the amount of amino acids in acid hydrolyzed beers are 0.06% and 0.056%, this is much more than those of nonhydrolysis. The different amounts means that of peptide-like substances. 5. It is necessary to determine the constituent of amino acid for the better taste of beer, and also it is desirable to check the role of carbohydrates in the course of fermentation, mashing and on lager beer for effective utilization of carbohydrate materials to eliminate the losses.

과맥의 파성에 대한 생리화학적 연구 III. 파성심도에 따른 유수분화 및 발육과정에 있어서의 유이아미노산의 소장 (Studies on the Physiological Chemistry of Spring Habits in Naked Barley III. Variation of Free Amino acids during the Differentiation and Development of Young Spkie with Different Spring Habits)

  • In order to investigate the changes of free amino acids during the differentiation and development of young spike in naked barley, a typical spring grain, Wanju, and two winter grains, Sedohadaka and Nonsankwa No. 1-6 differing in their spring habits, were analyzed at different growth stages by thin layer chromatography. In all the varieties 22 ninhydrin positive components were detected at the sowing time of March 5 and 20 components in the sowing plots of March 30. In case of the latter plot, β-alanine was identified only in both Wanju and Sedohadaka, whereas pipecolic acid was detected only in Nonsankwa No. 1-6. Particularly, it is interesting that β-alanine was observed only in the case showing the normal heading independent of the varieties and sowing times. Whether these components are directly related to the physiology of spring habits in barley or not is also a question to be answered. Of the major amino acids, alanine and γ-aminobutyric acid were always detected in appreciably large spots, and serine, leucine, aspartic acid, valine and asparagine were somewhat larger. In the plot of march 30, glutamic acid was also detected in very large spot in both Wanju and Sedohadaka at the stage of spikelet differentiation and in Nonsankwa No. 1-6 at the stage of bract differentiation. Histidine, which showed the only qualitative difference among the varieties during seed germination, cannot be observed at all. Proline observed considerably large spot during seed germination was always detected but very small except that it was observed in large spot at the stage of floret differentiation in Nonsankwa No. 1-6 in the plot of March 5.

Aminophosphonic Acids 화합물의 생물학적 기능연구 (Study of Synthesis and Biological Function on Aminophosphonic Acids)

  • Iso-butyl bromide의 합성을 시작으로 D.L-1-amino-3-methylbutylphosphonic acid의 합성을, Diethyl phophite의 합성을 시작으로 D.L-1-amino-2-phenyl ethylphosphonic acid의 합성을 하였다. 합성되어진 필수 아미노산인 Isoleucine analogue phosphonic acid는 각각 0.2%, 0.4%를 식이에 첨가했고 Phenylalanine analogue phosphonic acid는 0.35%와 0.7%를 각각 첨가하였으며 Standard-1과 Standard-2군과 비교군인 Isoleucine 0.2%와 0.4% Phenylalanine 0.35%와 0.7%군으로써 총 100마리의 숫쥐로 각 7주씩 14주의 사육 실험을 하였다. 동물 사육 기간내의 뇨채취와 사육후의 각 장기의 채취로써 총 질소와 P, glycogen측정을 시도하였다. 이러한 일련의 실험결과 (1) D.L-1-amino-3-methylbuthylphosphonic acid나D.L-1-amino-2-phenylethylphosphonic acid의 유기 합성에 있어서는 다 수율이 낮고 상당히 높은 융점을 보였으며 ${\alpha}-NH_2$기가 Ninhydrin 반응에 예민했다. (2) Aminophosphonic acid의 첨가군(D.L-1-amino-3-methylbutyl phosphonic acid, D.L-1-amino-2-Phenylethylphosphonic acid)에서나 Amino acid(Isoleucine, Phenylalanine)첨가군에 있어서 모든 장기의 무게, 총체내질소 보유율, 간장내 총단백질량과 인의양, 및 뇨를 통한 인의 배설량에 각각 큰 차이를 보이지 않았다. 이로 보아서 Phenylalanine Aminophosphonic acid나 Isoleucine aminophosphonic acid가 다 보통 아미노산인 Isoleucine이나 Phenylalanine에 못지않게 체내에서 이용됨이 밝혀졌다. (3) 그러나 Phenylalanine aminophosphonic acid 첨가군이 간의 glycogen 함량에 있어서 Phenylalanine 첨가군과 Standard군과 비교해 보았을때 높았으며 이는 통계적인 유의성을 나타냈다.(<0.05) 이는 Phenylalanine Aminophosphonic Acid가 active state로써 간내에서 쉽게 이용되어서 Tricarboxylic cycle의 intermediate로 incorporate되는 경향으로 생각 할 수 있다.

GABA를 담지한 자성 키토산 나노입자 제조와 약물의흡수 및 방출 연구 (The Preparation of Magnetic Chitosan Nanoparticles with GABA and Drug Adsorption-Release)

  • 약물 전달 시스템(Drug Delivery System, DDS)은 인체에 발생한 질환을 치료를 할 때 약물을 효과적으로 투약하므로써 약물성분에 의한 부작용을 최소화하고, 약물의 효능을 최대한으로 크게하기 위해 기존의 알려진 성분의 약물이나 새로운 성분의 제형을 설계하여 환자의 약물치료 과정을 최적화하는 목적을 지향하는 기술로 정의된다. 본 연구에서는 Tripolyphosphate (TPP)의 농도가 키토산과의 가교결합을 통하여 제조되는 Chitosan nanoparticles (CNPs)의 크기에 미치는 영향을 측정하여 TPP의 농도가 낮을수록 작은 크기의 입자가 형성되는 것을 확인하였다. 그리고 산화철(Fe3O4)의 양에 따른 CNPs-Fe3O4의 특성을 측정하여 Fe3O4의 양이 많을수록 자성 약물 전달체로써의 특성이 잘 나타남을 확인하였다. 닌히드린 반응(Ninhydrin test)를 통하여 저농도 구간(0.004~0.02 wt%)에서는 Y = 0.00373 exp(179.729X) - 0.0114 (R2 = 0.989), 고농도구간(0.02~0.1wt%)에서는 Y = 21.680X - 0.290 (R2 = 0.999)의 γ-aminobutyric acid (GABA)의 농도에 따른 검량선을 얻었다. 이 검량선을 사용하여 흡수를 위하여 넣어주는 GABA의 양에 따른 최대 흡수율의 관계식 Y = -136.527 exp [(-90.0862)X] + 64.724 (R2 = 0.997) 을 얻었으며, 초기에 넣어주는 GABA의 양이 약 0.04 g인 지점부터는 약 62.5%로 흡수율이 일정해 지고, 시간에 따른 GABA-Fe3O4-CNPs로부터 방출되는 GABA의 양을 측정하여 약 24 hr 이후부터 약물 방출이 종료되는 것을 확인하였다. 또한 최적의 조건에서 만들어진 GABA-Fe3O4-CNPs는 약 150 nm의 구형 입자이며, 그에 따른 입자의 특성이 잘 나타나는 것을 확인하여 약물 전달체로써 적합함을 알 수 있었다.

Chromatographic Determination of Amino Acids in Nonprotein and Protein Fraction Of Undaria Pinnatifida

  • Ion 교환수지(交換樹脂) cdumn chromatography법(法)에 의(依)해서 식용해조(食用海藻)인 '미역'의 단백성질소획분(蛋白性窒素劃分) 및 비단백성질소획분(非蛋白性窒素劃分)의 amino산함량(酸含量)을 정량(定量)하였다. 1. 단백성획분(蛋白性劃分)의 질소함량(窒素含量) (34.1mg/10g)은 비단백성획분질소(非蛋白性劃分窒素)(13i3mg/10g)의 약(約) 10배(倍)이다. 2. 양자(兩者)의 amino산조성(酸組成)은 서로 대조적(對照的)이고 그중(中) 단백성획분(蛋白性劃分)에는 필수(必須) amino산(酸)들이 고루 함유(含有)되어 있으며 그들 함유율(含有率)은 대두단백(大豆蛋白)의 경우(境遇)보다 높았다. 3. 비단백성획분(非蛋白性劃分) (80% ethanol가용(可溶))에는 상당량(相當量)의 Citrulline이 함유(含有)되어 있는것 이는 Ion 교환수지(交換樹脂) Column에서의 용리순위(溶離順位), 일차원(一次元) 및 이차원(二次元) paper chromatogram 상(上)에서의 위치(位置), 그리고 Ninhydrin 및 Ehrlich 시약(試藥)에 대(對)한 양성반응(陽性反應)에 의(依)해서 증명(證明)하였으나 그 정확(正確)한 함량(含量)은 150-cm column에서 Glycine과 중첩(重疊)되어서 용리(溶離)되는 관계(關係)로 알수 없고, 다만 이들 혼합물(混合物)의 질소량(窒素量)이 이획분전질소중(劃分全窒素中) 약(約) 30%에 상당(相當)함을 밝혔다. 4. 비단백성획분(非蛋白性劃分)에서 유리상태(遊離狀態)로 존재(存在)하는 새로운 peptide를 분리(分離)해 냈는데, 이는 염산가수분해후(鹽酸加水分解後) paper chromatography에 의(依)해서 조사(調査)한 바 Alanine, Gluatmic acid 및 Aspartic acid로 조성(組成)된 Tripeptide 임을 알았다. 5. '미역'(건조물(乾燥物))에는 약(約) 1%에 해당(該當)하는 다량(多量)의 Alanine이 함유(含有)되어 있는데 그 중(中) 약(約) 1/3량(量)이 유리상태(遊離狀態)로 존재(存在)함을 알았다.
