A novel antifungal antibiotic for azole-resistant Candida albicans was purified from the culture broth of Bacillus subtilis LAM 97-44 by butanol extraction, Diaion HP-20 and Dowex-50 adsorption chromatography, silica gel flash chromatography followed by HPLC and designated LAM-44A. LAM-44A was stable for 60 min at $100^{\circ}C$, and pH range from 2 to 10. MIC values were observed at $0.5-3.5\;{\mu}g/ml$ against various Candida albicans strains. The antibiotic showed no cytotoxicity for S180, MKN-45, P388, HeLa and 373 at the concentration of 1 mg/ml. LAM-f4A was colorless powder soluble in water, methanol, ethanol, butanol and negative to ninhydrin reaction. The antibiotic had maximum absorption at 273 nm in methanol, and melting point was $202^{\circ}C$. The molecular weight and formula were determined to be 282 and $C_{14}H_{34}O_5$ by $^1H-NMR,\;^{13}C-NMR$, IR spectrum and elemental analysis.
Bloody fingerprint is a very important evidence. In this study, we confirmed the enhancement effects of ninhydrin, leucocrystal violet (LCV), fuchsin acid, iodine and dimethylaminocinnamaldehyde (DMAC) on bloody fingerprints which were deposited on paper. Bloody fingerprint were deposited on paper sequentially and used after drying at room temperature. If a ridge of bloody fingerprint was clear, ninhydrin and LCV was the most effective but was not good for invisible ridge. Fuchsin acid reagent dyed paper surface so that the contrast of enhanced bloody fingerprint was decreased. Although bloody fingerprint was enhanced with iodine reagent, but the developed color was very weak after reaction. We thought that the enhancement effect of iodine to bloody fingerprint was negligible. Also, the enhancement effect of DMAC reagent to relatively clear bloody fingerprint was not good. However, it was very effective to faint or invisible ridge. By washing with methanol, contrast of enhanced bloody fingerprint was increased.
This study was carried out to investigate the effect of red ginseng water extract (RGWE) on trypsin activity. After extraction of fat soluble and saponin component from red ginseng powder by methyl alcohol, the residue was extracted with distilled water, and manufactured to water extract. The extract was dialyzed with different molecular cut off membrane. Trypsin activity demonstrated the highest level at the RGWE concentration of 9${\times}$10$\^$-2/% in reaction mixture, and also increased to 15% at 2.9${\times}$10$\^$-3/%. Km value was decreased and Vmax was increased in the present of red ginseng water extract. Red ginseng water extract was partially purified by dialysis, Bio-Gel P-I0 and DEAE-cellulose column chromatography. The active fraction demonstrated positive reaction to ninhydrin, DNS and folin reaction.

The cytotoxic effects of lipid extracts from soybean products were studied using K562 human leukemia cell, Yac1 mouse leukemia cell and S 180 mouse sarcoma cell. Total lipids from soybean powder, soybean curd residue and doenjang were extracted with chloroform/methanol (2 : 1) and water saturated butanol, consecutively, and fractionated into acetone supernatants (AS fraction) and acetone precipitates (AP fraction) by adding excess acetone. AS fraction of doenjang lipids showed the strongest cytotoxic effects on K562, Yac1 and S180 cancer cells, whereas each lipid fraction of soybean curd residue also showed relatively weak cytotoxic effects on cancer cells but soybean powder did not. AS and AP fractions of doenjang contained more free fatty acids than those of soybean curd residue and soybean. And when lipid fractions were digested with 0.4 N KOH/methanol, doenjang lipid fractions showed to contain some alkali-stable substances which showed positive reaction with ninhydrin solution on silica TLC separation.

Background: Effective decellularization and fixation process is critical, in order to use xenogenic valves clinically. In the present study, we decellularized bovine pericardium using sodium dodecyl sulfate (SDS) and N-lauroyl sarcosinate, treated with $\alpha$-galactosidase, and then fixed in various manners, to find out the most effective tissue preservation & fixation procedure. Material and Method: Bovine pericardium was decellularized with SDS and N-lauroyl sarcosinate, and treated with $\alpha$-galactosidase. Both groups were fixed differently, by varying glutaraldehyde (GA) or EDC (1-ethyl-3-(3-dimethyl aminopropyl)-carbodiimide)/N-hydroxysuccinamide (NHS) treatment conditions. Thereafter, physical examination, tensile strength test, thermal stability test, cytotoxicity test, pronase test, pronase-ninhydrin test, purpald test, permeability test, compliance test, H&E staining, DNA quantification, and $\alpha$-galactose staining were carried out to each groups. Result: GA fixed groups showed better physical properties and thermal stability than EDC/NHS fixed groups, EDC/NHS-GA dual fixed groups showed better physical properties and thermal stability than EDC/NHS fixed groups, and showed better thermal stability than GA fixed groups. In pronase test and pronase-ninhydrin test, GA fixed groups and EDC/NHS-GA dual fixed groups showed stronger crosslinks than EDC/NHS groups. Permeability and compliance tended to increase in EDC/NHS-GA dual fixed groups, compared to GA fixed groups. But, EDC/NHS-GA dual fixed groups had stronger tensile strength and lower cytotoxicity than GA fixed groups. Conclusion: We have verified that EDC/NHS-GA dual fixation can make effective crosslinks and lower the toxicity of GA fixation. Henceforth, we will verify if EDC/NHS-GA dual fixation can lower calcifications & tissue failure in vivo experiment.
In crime scene investigation, fingerprint identification is regarded to be one of the most important techniques for personal identification. However, objective and unbiased evaluation methods that would compare the fingerprints with diverse available and developing methods are currently lacking. To develop an objective and quantitative method to improve fingerprint evaluation, a preliminary study was performed to extract useful research information from the analysis with densitometric image analysis (CP Atlas 2.0) and the Automated Fingerprint Identification System (AFIS) for the developed fingerprints on porous surfaces. First, inked fingerprints obtained by varying pressure (kg.f) and pressing time (sec.) to find optimal conditions for obtaining fingerprint samples were analyzed, because they could provide fingerprints of a relatively uniform quality. The extracted number of minutiae from the analysis with AFIS was compared with the calculated areas of friction ridge peaks from the image analysis. Inked fingerprints with a pressing pressure of 1.0 kg.f for 5 seconds provided the most visually clear fingerprints, the highest number of minutiae points, and the largest average area of the peaks of the friction ridge. In addition, the images of the developed latent fingerprints on thermal paper with the iodine fuming method were analyzed. Fingerprinting condition of 1.0 kg.f/5 sec was also found to be optimal when generating highest minutiae number and the largest average area of peaks of ridges. Additionally, when the concentration of ninhydrin solution (0.5 % vs. 5 %) was used to compare the developed latent fingerprints on print paper, the best fingerprinting condition was 2.0 kg.f/5 sec and 5 % of ninhydrin concentration. It was confirmed that the larger the average area of the peaks generated by the image analysis, the higher the number of minutiae points was found. With additional tests for fingerprint evaluation using the densitometric image analysis, this method can prove to be a new quantitative and objective assessment method for fingerprint development.
Capsaicin(8-methyl-N-vanillyl-6-nonenamide) is the principal pungent component of Capsicum fruits. This work is directed to the capsaicin-hydrolyzing enzyme playing a key role in the rate limiting and critical step of capsaicin metabolism. In order to get precise information on the enzyme's subcellular location, rat liver homogenate was divided into six subcellular fractions by differential centrifugation technique: crude nuclear pellet, PNS(post nuclear supernatant) fraction, lysosomal pellet, cytosol, Tris wash fraction, micrisomes. Capsaicin-hydrolysing enzyme activity was analysed by high performance liquid chromatography(HPLC). This enzyme was found at the highest specific activity in the microsomal fraction and co-distributed with marker enzymes of the endoplasmic reticulum, NADPH-cytochrome c reductase and nucleoside diphosphatase. This is compatible with the result of ninhydrin color reaction of vanillylamine, primary metabolite of capsaicin hydrolysis, on thin layer chromatography(TLC). This enzyme is most active at pH $8.0{\sim}9.0$. Definite subcellular location of this enzyme will make it easy to proceed with further study.
An antifungal antibiotic was produced from Bacillus sp. YJ-63, and antibiotic was purified to a homogeneity by butanol extraction, Silica-gel column chromatography, Sephadex LH-20 gel filtration column chromatography and HPLC. The purified antibiotic showed one spot on Thin Layer Chromatography, which was negative to ninhydrin and positive to iodine. This finding indicated that the antibiotic was a cyclopeptide structure. UV absorption spectrum of the antibiotic in methanol was maximal at 277 nm, which corresponded to the spectrum for all of the Iturin antibiotic. The antibiotic was stable up to $121^{cdot}C$ and in the range of pH $3.O{\sim}l2.0$ with a stable of ${\alpha}-amino$ acid and ${beta}-amino$ acid, and showed high activity for fungi and yeasts,
Clinical tests revealed that the extracts of Gelidium amansii (Gelidiaceae) had a anthelmintic action and further examinations were made on the anthelmintic components of this seaweed. This anthelmintic principle is absorbed on alumina and eluted from it by alkali solution. The active principle is absorbed on activated carbon from aqueous extract and eluted from it by methanol and it is not adsorbed on Amberite IR-120(H-form). This anthelmintic effective fraction was prepared by the use of this properties. Action of the active principle of Gelidium amansii was examined pharmacologically. The active principle of Gelidium amansii was found to decrease the tensity, tonus and mobility of Eisenia foetida(Savigny) nerve muscles. The active principle of this effective fraction was submitted to paper chromatography and spots to ninhydrin were detected at Rf; 0.30-0.31(yellow), 0.26(violet), 0.2(violet), 0.14-0.13(violet), 0.9(orange) and 0.04(violet).
Acidic polysaccharide from Korean red ginseng was found to inhibit pancreatic lipase activity and cause reduction of plasma triglyceride level after oral administration of corn oil emulsion to rats. Thus acidic polysaccharide may reduce plasma triglyceride through its inhibitory action on pancreatic lipase and successive inhibition of intestinal absorption of fat due to reduction of lipolysis. In the course of this experiment, we found an unknown ninhydrin positive substance in Korean red ginseng. The unknown substance was identified to be arginyl-fructosyl glucose(Arg - Fru - Glc). Coment of this new compound was $5.37\%$ in Korean red ginseng powder. Sucrase and maltase activities in mucous layer of rat jejunum were found to be inhibited by Arg-Fru-Glc. Physiological significance of the new compound was discussed based on these experimental results.
