The identification of $\alpha,\beta$-4,8,13-Duvatriene.1,3-diols in fresh tobacco leaves(Nicotiana Tabaccum L.) (잎담배중 $\alpha,\beta$-4,18,13-Duvatriene-1,3-diols의 분리확인)

  • 4, 8, 13-Duvatriene-1, 3-viols have been identified in the cuticular lipid of fresh Sohyang (aromatic cultivar) tobacco leaves. Their $\alpha, \beta$-isomers were directly separated by SE-54 glass capillary column and identified by mass spectrometry. Gas chromatographic analysis revealed that the duvatrienediols are major components in the culticular lipid of Nicotiana Tabacum.

Identification of Regenerable Cells in MesophyII Protoplast Cultures (엽조직에서 나출된 원형질체의 재생 가능 세포판별)

  • This study was rimed out to examine the difference in the cell vitality between mesophyII protoplast (MP) and paraveinal mesophyII protoplast (PVMP) of Nicotiana tabaccum 'Xanti', Petunia hybrida 'Blue Star' and Chrysanthemum morifolium 'Baeckwang' by using urea permeability technique. The effects of various enzyme solutions and incubation time, NAA and thidiazron on plant regeneration from isolated protoplasts were also investigated. The vibratome technique was used for protoplast isolation and urea permeability test because the fresh living, thin tissue stripes (50 ${\mu}{\textrm}{m}$ of thickness) could be obtained with minimal damage with the vibratome. For the three plants examined, the urea permeability on the tested tissue stripes was relatively higher in PVMP than in MP by about Ks = 2.0 $\times$ 10$^{-5}$ cm/sec. The treatment of an enzyme mixture of 1.5% cellulase R-10, 1% Driselase, 0.5% Macerozyme R-10, and 0.5% Pectinase for 4 to 8 h was effective on the isolation of PVMP. The highest frequency of callus formation and plant regeneration from the isolated protoplasts was obtained with NAA 2 mg/L and thidiazuron 0.01 mg/L. Furthermore, the results demonstrated that cell devision and plantlet regeneration was more frequent in the PVMP than in the MP of the same leaf or plant We, therefore, conclude that UM is an excellent experimental material for the callus formation and regeneration from isolated protoplasts.

  • Cowper chlorotic mottle virus (CCMV) is the 'controlled' quarantine virus as plant pathogenic virus that are classed as group VI (+) ssRNA virus that belongs to the genus Bromovirus and family Bromoviridae, When plants that are Phaseolus vulgaris, Clitoria ternatea, Nicotiana tabaccum, Glycine max, Vigna unguiculata and Vigna siensis, and Arachis hypogaea is imported in domestic. In this study, inspection system is implemented to analyze CCMV accurately and rapidly by developing RT-PCR, nested PCR, and gene insertion positive control. It is expected that the method developed in this study will contribute to the plant quarantine to be consistently utilized in the field.

  • Acetolactate synthase (ALS, EC 4.1.3.18) is the first common enzyme in the biosynthesis of leucine, isoleucine, and valine. It is the target enzyme for several classes of herbicides, including the sulfonylureas, the imidazolinones, the mazolopyrimidines, the pyrimidyl-oxy-benzoates, the pyrimidyl-thio-benzens, and the 4,6-dimethoxypyrimidines. An amino-terminal fragment of the sulfonylurea-resistant ALS gene (SurB) from Nicotiana tabaccum was cloned into the bacterial expression vector pGEX-2T. The resulting recombinant plasmid pGEX-ALS1 was used to transform Escherichia coli strain BL21, and the tobacco ALS was expressed in the bacteria as a protein fused with glutathione S-transferase (GST). Polyclonal antibodies against the fusion product (GST-ALS) were produced, and the sensitivity of GST-ALS with the rabbit anti-GST-ALS IgG was up to 50 ng. This antibody was used for Western blot analysis of the partially purified ALS from barley shoots. The results suggest that the polyclonal antibody produced in this study can be used to detect plant ALS.

Flooding Stress-Induced Glycine-Rich RNA-Binding Protein from Nicotiana tabacum

  • A cDNA clone for a transcript preferentially expressed during an early phase of flooding was isolated from Nicotiana tabacum. Nucleotide sequencing of the cDNA clone identified an open reading frame that has high homology to the previously reported glycine-rich RNA-binding proteins. The open reading frame consists of 157 amino acids with an N-terminal RNA-recognition motif and a C-terminal glycine-rich domain, and thus the cDNA clone was designated as Nicotiana tabaccum glycine-rich RNA-binding protein-1 (NtGRP1). Expression of NtGRP1 was upregulated under flooding stress and also increased, but at much lower levels, under conditions of cold, drought, heat, high salt content, and abscisic acid treatment. RNA homopolymer-binding assay showed that NtGRP1 binds to all the RNA homopolymers tested with a higher affinity to poly r(G) and poly r(A) than to poly r(U) and poly r(C). Nucleic acid-binding assays showed that NtGRP1 binds to ssDNA, dsDNA, and mRNA. NtGRP1 suppressed expression of the fire luciferase gene in vitro, and the suppression of luciferase gene expression could be rescued by addition of oligonucleotides. Collectively, the data suggest NtGRP1 as a negative modulator of gene expression by binding to DNA or RNA in bulk that could be advantageous for plants in a stress condition like flooding.

Purification and Properties of $\beta-1$, 4-endoglucanase from Tobacco Suspension Cultured Cells (담배 현탁배양 세포로부터 $\beta-1$,4-endoglucanase의 정제 및 성질)

  • $\beta$-1, 4-endoglucanase (EC 3, 2, 1, 4) was isolated and purified from Nicotiana tabaccum L. Var. Virginia 115 suspensin cultured cells. The molecular weight as estimated by Sephadex G-100 was about 14, 000. The optimum pH for activity was 5.4. The Km value for carboxymethyl cellulose was 0.18 mg/unit and the enzyme was quite resistant to heating. Polyamine did not affect the activity of $\beta$-1, 4-endoglucanase in vitro but the activity increased drastically when polyamines were added in the suspension culture medium. It is suggested that increase in $\beta$-1, 4-endoglucenase activity due to polyamine might be related to growth of the plant.

Studies on Ginger Mosaic Virus (생강모자이크바이러스병에 관한 연구)

  • A mosaic virus disease of ginger plant was investigated to determine its virus group on the basis of host range, physical and chemical properties, serological behavior and electron-microscopic morphology. The disease gave rise to yellowsih and dark-green mosaic on the leaves in the early stage and stunted all the leaves as well as rhizomes in the late stage. In the field about 43\% of the plants were observed to be diseased The disease was able to be artificially infected to the ginger plants by the sap and transmission as well as to 23 other species of plants which were known to be the CMV susceptible plants by the sap transmission; Chenopodium amaranticolar, Nicotiana tabaccum var. Havana, cow pea, cucumber, tomato,... etc. The dilution end point of the virus ranged $10^{-4}-10^{-5}$ and the thermal inactivation point $65-70^{\circ}C$. Serological test showed a positive reaction by a CMV antiserum. An electron microscopy of the purified virus showed that the virus particles were spherical with a diameter of $28-32m\mu$. Virus particles from the infected tissue were observed to be free or aggregated in the mesophyll tissue of artificially infected tobacco plant. The mosaic disease of ginger plants were conclusively suggested to the CMV group.
