TERATOGENICITY STUDY OF RECOMBINANT GRANULOCYTE-MACROPHAGE COLONY STIMULATING FACTOR (LBD-005) IN RABBITS

  • LBD-005, a newly developed recombinant granulocyte-macrophage colony-stimulating factor, was at dose levels of 0, 20, 80 and 320ng/kg/day administered subcutaneously to pregnat New Zealand White rabbits during the organogenetic period. The dams were subjected to caesarean section on day 28 of pregnancy. Effects of test substance on dams and embryonal development of fetuses were examined. No treatment-related changes in clinical signs and necropsy findings of dams were observed in all groups. At 80 and 320 ng/kg, a significant decrease in food consumption followed by a loss in body weight was found.

  • LBD-007, a newly developed recombinant human interferon alphaA, was at dose levels of 0.3 $\times$$10^6$ , 6 $\times$ $10^6$ and 12 $\times$ $10^6$ IU/kg/day administered subcutaneously to pregnant New Zealand White rabbits during the organogenetic period. Cyclophosphamide was used as a positive control. All pregnant females were subjected to the caesarean section on day 28 of pregnancy.   • LBD-007, a newly developed recombinant human interferon ${\alpha}$A, was tested for primary eye and skin irritation in male New Zealand White rabbits. In the primary eye irritation test, 0.1ml of a solution of LBD-007 was instilled into the eye. In rinsing group, the eye was washed with water 30 seconds after instillation. No reaction was observed at the cornea, iris and conjunctivae by LBD-007. In the primary skin irritation test, LBD-007 was applied to the back of rabbits for 24 hours. Primary irritation index was "0" in test and control sites of all animals. Thus LBD-007 was evaluated as a non-irritant on the basis of the criteria of Draize et al.,(1994).

  • This study was carried out to investigate whether the use of the high endotoxin-sensitive or the low endotoxin-sensitive rabbits (New Zealand White strain) are more suited for the pyrogen test regulated by the Korean Pharmacopeia and United State Pharmacopeia. The results obtained were as follows. 1. The ferbrile responese were increased according to increased endotoxin doses used (i.e., 0.1, 1, 5, 10ng/kg) and the febrile responses reached to its maximum between 90 mins and 120 mins after the intravenous injection of endotoxin.

Teratological Study of LBD-001, a Recombinant Human Interferon $\gamma$, in Rabbits

  • LBD-001, a recombinant human interferon $\gamma$ produced by genetically engineered yeast as a host system, was intravenously administered to pregnant female rabbits (New Zealand White strain) from day 6 to 18 of gestation at dose levels of $0.35 \times 10^6$, $0. 69 \times 10^6$, and $1.38 \times 10^6$ I.U./kg/day. Hydrocortisone sodium succinate (0.3 mg/kg/day) was also given in the same way. Teratological effects of the test agents on the organogenesis of fetuses and the development of offsprings (F1 rabbits) were investigated. The results were as followings: (1) No significant changes by the treatment of LBD-001 or hydrocortisone sodium succinate were observed in the body weights, the food and water consumption, the lactating or nurshing behaviors, and the autopsy of the pregnant rabbits. (2) No significant changes in the resorption rate, the fetal organogenesis, and the normal develpoment of offsprings (F1) by the treatment of LBD-001 or hydrocortisone sodium succinate were detected. The results show that LBD-001 at the dose of $1.38 \times 10^6$ I.U./kg/day or less and hydrocortisone sodium succinate at the dose of 0.3 mg/kg/day are neither teratogenic in the organogensis of the fetuses and the development of the offsprings (F1) nor toxic to the mother rabbits.

Changes in Blood Constituents of Rabbits Subjected to Transportation under Hot, Humid Tropical Conditions

  • Unlike Europe (particularly, Italy and Spain), where a number of studies have been conducted on the stressful effects of transport on rabbit welfare, few studies have been conducted on transportation of rabbits under hot, humid tropical conditions experienced in countries like Malaysia. We studied the effects of transportation in hot humid tropical conditions of Malaysia on physiometabolic changes in New Zealand white rabbits. Eighty experimental animals were divided into two groups of 40 bucks each and transported for either 3 or 1 h. Transportation caused a significant upsurge of aspartate aminotransferase, alanine aminotransferase and creatine kinase activities (p<0.001) though did not significantly affect lactate dehydrogenase (LDH) activity (p = 0.0706). Both transportation periods caused elevation in plasma glucose levels, lactic acidosis and dehydration as evidenced through elevated packed cell volume and plasma protein concentration. It was concluded that regardless of the duration, transport of rabbits under hot humid tropical conditions, resulted in heat distress since the rabbits showed hyperglycemia, hypercalcemia, lactacidemia, lymphocytopenia, dehydration and increase in blood enzyme activities.

  • The visceral dissemination of Borrelia burgdorferi in New Zealand White rabbits was evaluated following intradermal inoculation of $1{\times}10^8$ spirochetes. We inoculated Borrelia burgdorferi B31, B garinii KW1 and B afzehlii S13, respectively, and monitored the dissemination in the experimentally infected rabbits for 28 days. In the B burgdorferi B31-challenged group, the spirochetes were completely cleared in rabbits at day 1 and visceral dissemination was not demonstrated. However, B garinii KW1 and B afzelii S13 were found to successfully disseminate in visceral organs of rabbits during the experiment period of 28 days. And experimentally infection-derived immunological responses in rabbits were identified with enzyme-linked immunosorbent assay and immunoblot analysis. Based on these results, the differences in the virulence of Lyme borrelial strains were proved in rabbit model.

Carbon Fibres for the Repair of Abdominal Wall Defects in Rabbits

  • Sixteen clinically healthy New Zealand white rabbits of either sex were divided into two equal groups I and II of 8 animals each. Under thiopental sodium (2.5%) anaesthesia a linear full thickness abdominal wall defect of 3 cm in length was created and repaired with continuous suture pattern using 3000 filaments of carbon fibres and 1~0 black braided nylon suture, ingroup I and II respectively. Increased vascularity was observed in carbon fibres (group I) and on day 30 the carbon fibres were covered by white fibrous tissue. Significantly higher (P < 0.05) values of glucose was seen on day 14 in group I, whereas, decrease in glucose value was observed in group II. Histopathologically, the carbon fiber implant induced extensive fibrous tissue (collagen fiber) reaction. Negligible inflammatory cells in the stroma indicate the host tissue tolerance to carbon fibers. Histochemically, gradually increased alkaline phosphatase activity up to day 14 in group I, suggested the proliferation of fibroblasts in early stages.

  • Previous sucessful results of neocartilage formation using tissue engineering technique in immunocompromised nude mouse xenograft model were reported. For clinical application, autogenous cell is preferrable to allogenic or xenogenic cell for circumvention of immune rejection. This study evaluates the feasibility of producing a engineered cartilage using autogenous chondrocytes. Chondrocytes were isolated from the auricular catilage of New Zealand White rabbit and seeded onto PGA polymer coated with polylactic acid in round pattern(diameter 0.7 cm, thickness 0.1 cm) at a concentration $7{\times}10^7$ chondrocytes per $cm^3$. Each Autogenous Cell-polymer constructs were implanted subcutaneously into the left side of dorsum of twelve Rabbits. Polymer templates not containg cells were implanted into the right side as a control. Fifteen rabbits were sacrificed at the following intervals: 5 rabbits at nine weeks, 7 rabbits at twelve weeksNew autogenous cartilage formation which retained the approximate dimensions of origianl round polymer template in 11 of 12 cell seeded implants. Histological examination using hematoxyline and eosin stain revealed vast majority of implants developed into mature cartilage. This study opens up the possibility of autologus cell transplant to construct autogenous cartilge.

Effects of Polyphosphate on Skin Wound Healing in Rabbits (토끼 피부창상 치유에 있어서 Polyphosphate의 효과)

  • The purpose of the present study was to investigate the effect of polyphosphate on healing of skin wound in rabbits. Four skin wounds of 8mm in diameter were induced bilaterally(4 wounds/rabbit) on the dorsolateral aspect of the trunk of 15 male New Zealand white rabbits. Ten percent polyphosphate(poly P) type 25, 10% poly P type 35, 10% poly P type 25+10% poly P type 35 and 0.85% sterile saline solution were applied on the wound area and examined grossly and histopathologically. The rate of wound contraction and histopathological changes after poly P application on the skin wound were investigated. In gross findings, the wound contraction rates of 10% poly P 25 group were significantly decreased from day 2 to day 7 after operation than those of 0.85% sterile saline solution treated group (p < 0.01). Although the hemorrhage and inflammation were observed on days 3, 6 and 9 after operation in all groups, but in 10% poly P 25 was gradually decreased. Revascularization and reepithelialization were seen 3, 6 and 9 days after operation in all groups, especially in 10% poly P 25 was formed completely on 9 days. Fibroplasia were seen 3. 6 and 9 days after operation in all groups, especially in 10% poly P 25 was compact than other groups on 9 days. The results suggested that polyphosphate, especially 10% poly P 25 would be effective on healing of skin wound in rabbits through decrease of would contraction rate, increase of revascularization, reepithelialization and fibroplasia.
