First Discovery of the Lichen-Feeding Moth Bacotia sakabei (Lepidoptera: Psychidae) from Korea

  • The family Psychidae is a small group consisting of 241 genera and 1,350 species in the world. The genus Bacotia, belonging to the family Psychidae, what was established by Tutt based on the type species, Fumea sepium Speyer and Speyer, 1846. In this study, we aimed to record lichen-feeding bagworms in Korea. The genus Bacotia Tutt is reported for the first time from Korea with one newly recorded species, Bacotia sakabei Seino, 1981. All available information, including the collecting localities, host plants, illustrations of adults and genitalia are presented. A DNA barcode for precise identification of the species is also described.

  • 전남 백운산 지역의 논실 및 한재 지점에서 2015년 5월부터 8월까지 깔데기트랩(Lindgren funnel trap)을 이용하여 배줄벌레과(Biphyllidae) 곤충을 채집하였다. 그 결과 배줄벌레속(Biphyllus) 7종이 채집되었고, 그 중 3종인 둥근배줄벌레(B. aequalis (Reitter)), 담갈무늬배줄벌레(B. flexuosus (Reitter)), 애배줄벌레(B. kuzurius Sasaji)가 국내에서 처음 기록되는 종으로 확인되었으며, 이들을 동정하기 위한 검색표와 진단형질들을 도식하였다.

  • 각진장미흰깍지벌레, 참식나무흰깍지벌레와 후박나무굴깍지벌레로부터 채집된 Encarsia explorata (Silvestri) [후박굴깍지좀벌 (신칭)] 및 담배가루이로부터 채집된 Encarsia lutea (Masi) [가루이좀벌 (신칭)] 을 국내분포로 처음 보고한다. 또한 이들 종들의 검색표, 식별형질, 사진자료, 분포 및 기주 정보도 함께 제공하였다.

소아의 치아우식 부위별 우점 세균 분리 및 동정 (Isolation and identification of the abundant bacteria in dental caries in children)

  • Objectives: The study aimed to isolate the abundant bacteria in dental caries in children and to investigate the bacterial species involved in addition to those that have been previously reported. Methods: The specimens were collected from the supragingival plaques of each dental caries area, pit and fissure caries, deep dentinal caries, smooth surface caries, and dental caries, and from healthy subjects in the control group. Bacteria were cultured from these specimens, DNA was extracted from the isolated bacteria, and the 16S rRNA gene sequences were analyzed and identified. Results: Based on the results of the 16S rRNA gene sequence analysis for the 90 strains of dominant bacteria from the 45 specimens, 5, 7, 8, 7, and 13 species were identified from the supragingival plaques from healthy teeth, pit and fissure caries, deep dentinal caries, smooth surface caries, and dental caries, respectively. In healthy teeth, Actinomyces naeslundii dominated. Corynebacterium durum, Ralstonia pickettii, and Streptococcus intermedius showed equal distribution. The dominant bacterial species in dental caries, S. sanguinis, showed the greatest difference in prevalence in pit and fissure caries. In deep dentinal caries, S. mutans and Lactobacillus rhamnosus were dominant; in smooth surface caries, S. mutans and S. sanguinis were dominant; and in the supragingival plaques of dental caries, S. sanguinis and S. mutans were dominant. Conclusions: The bacterial species isolated from dental caries encompassed four phyla, eight genera, and 22 species. In addition, the SS1-2 strain, belonging to the genus Neisseria, was identified as a new species from among the isolated strains.

  • 한국 미기록종인 고창고랭이(Schoenoplectiella ${\times}$uzenensis (Ohwi ex T. Koyama) Hayasaka)를 전라북도 고창군 고수면의 저수지에서 발견하였다. 고창고랭이는 짧게 기는 지하경이 있고 소수가 1-3개가 달리는데 반하여, 송이고랭이(S. triangulata)는 지하경이 아주 짧고 줄기가 빽빽이 모여나며 소수가 5-20개가 달리고, 제주올챙이골(S. lineolata)은 지하경이 길게 발달하고 소수가 1개가 달려 뚜렷이 구별된다. 이 미기록종의 국명은 고창군에 자라기 때문에 '고창고랭이'로 하였고, 고창고랭이에 대한 기재, 도해, 칼라도판 및 근연종 사이의 차이점을 검색표와 표로 제시하였다.

Discovery of Two New Talaromyces Species from Crop Field Soil in Korea

  • Two new fungal species of the genus Talaromyces, Talaromyces purpurogenus and Talaromyces trachyspermus from the Trichocomaceae family, were recovered during an investigation of fungal communities in soil collected from the Gangwon-do and Jeollanam-do provinces of Korea. These two species have not been previously officially reported from Korea. In this study, detailed descriptions of internal transcribed spacer rDNA and beta-tubulin gene regions of these two fungi are presented. Morphological features of the two fungi in five agar media, potato dextrose, oatmeal, malt extract, czapek yeast extract, and yeast extract sucrose, are also reported. The species were identified on the basis of molecular and morphological analysis, and herein we present data with detailed descriptions and figures.

한국산 미기록 콩바구미 Bruchidius siliquastri (딱정벌레목: 잎벌레과)에 대한 보고 (New Record of Bruchidius siliquastri Delobel (Coleoptera, Chrysomelidae, Bruchinae) in Korea)

  • 동아시아가 원산지로 추정되며, 2007년에 신종으로 보고된 콩바구미아과(Bruchinae)에 속하는 Bruchidius siliquastri Delobel (박태기콩바 구미; 신칭)는 유럽에서 콩과(Fabaceae)인 유다박태기나무(Cercis siliquastrum L.)의 종자를 가해하는 침입종으로 간주되고 있다. 이 콩바구미는 한국에서 최근 블로거(blogger)들에 의해 정확한 동정이 이루어지지 않은 상태로 성충 사진들이 업로드(upload) 되고 있으며, 또한 저자들은 2022년 봄 순천대학교 캠퍼스에 식재된 박태기나무(C. chinensis Bunge)에서 일부 생태 및 가해를 관찰할 수 있었다. 본 연구는 한국에서 이 콩바구미의 새로운 생활사 정보를 제공하고자 한다.

Morphological and Molecular Identification of Stellantchasmus dermogenysi n. sp. (Digenea: Heterophyidae) in Thailand

  • We tried a series of morphological and molecular approaches to identify a new species of Stellantchasmus (Digenea: Heterophyidae) originating from the wrestling half-beaked fish, Dermogenys pusillus of Thailand. Adult worm samples of the new species were recovered from hamsters experimentally infected with the metacercariae from D. pusillus in Thailand. Two isolates (Thai and Korean) of Stellantchasmus falcatus were used as comparative control groups. Worm samples of 3 Stellantchasmus groups were morphologically observed and molecularly analyzed with the mitochondrial cytochrome c oxidase 1 gene. The morphological characteristics of S. dermogenysi n. sp. are similar to S. falcatus originating from brackish water fish, but minor difference was noted including the absence of the prepharynx, position of the ovary near the ceca end, smaller body size, and shorter esophageal length. A phylogenetic tree derived from neighbor-joining and maximum-likelihood methods suggests that S. dermogenysi n. sp. is separated from S. falcatus supported by high bootstrap values. The relative divergences persist between these host-specific trematodes, which we suggest should be recognized as 2 distinct species. Comparisons of S. dermogenysi n. sp. with S. falcatus isolated from mullets in Thailand and Korea indicate a genetic divergence of mitochondrial DNA of 19.4% and 21.7%, respectively. By the present study, a new species, Stellantchasmus dermogenysi n. sp. (Digenea: Heterophyidae), is proposed in Thailand based on molecular evidences, in addition to minor morphological differences between S. falcatus and the new species.

First Report of Two Plectus Species (Nematoda: Plectida) from Korea

  • The genus Plectus Bastian, 1865 represents a group of free-living freshwater nematodes belonging to the family Plectidae Örley, 1880. However, only one species has been reported thus far from Korea. Plectus aquatilis Andrássy, 1985 and Plectus cirratus Bastian, 1865 are reported for the first time from Korea, from sediments collected from the Nakdong River. Plectus aquatilis is distinguished from other Plectus species by its three longitudinal alae in the lateral field, thin and directed cephalic setae, continuous lip region (head not set-off), and rectangular shaped tail. Plectus cirratus is distinguished from other Plectus species by its large body, two longitudinal alae in the lateral field, larger stoma, and longer tail. Morphological characters and measurements of the specimens generally agree with the original descriptions of Plectus species. Here, the morphological characters (detailed morphometrics) of two species in the genus Plectus are fully redescribed and illustrated using optical microscopy. DNA barcode sequence information from the 18S rDNA gene is also provided for molecular species identification and compared with sequences from other Plectus species available on GenBank.

Identification and molecular characterization of downy mildew resistant gene candidates in maize (Zea mays subsp. Mays)

  • Downy mildew (DM), caused by several species in the Peronosclerospora and Scleropthora genera, is a major maize (Zea mays L.) disease in tropical or subtropical regions. DM is an obligate parasite species in the higher plants and spreads by oospores, wind, and mycelium in seed surface, soil, and living hosts. Owing to its geographical distribution and destructive yield reduction, DM is one of the most severe maize diseases among the maize pathogens. Positional cloning in combination with phenotyping is a general approach to identify disease resistant gene candidates in plants; however, it requires several time-consuming steps including population or fine mapping. Therefore, in the present study, we suggest a new combination strategy to improve the identification of disease resistant gene candidates. Downy mildew (DM) resistant maize was selected from five cultivars using the spreader row technique. Positional cloning and bioinformatics tools identified the DM resistant QTL marker (bnlg1702) and 47 protein coding genes annotations. Eventually, 5 DM resistant gene candidates, including bZIP34, Bak1, and Ppr, were identified by quantitative RT-PCR without fine mapping of the bnlg1702 locus. Specifically, we provided DM resistant gene candidates with our new strategy, including field selection by the spreader row technique without population preparation, the DM resistance region identification by positional cloning using bioinformatics tools, and expression level profiling by quantitative RT-PCR without fine mapping. As whole genome information is available for other crops, we propose applying our novel protocol to other crops or for other diseases with suitable adjustment.
