Description of Vishniacozyma terrae sp. nov. and Dioszegia terrae sp. nov., Two Novel Basidiomycetous Yeast Species Isolated from Soil in Korea

  • Two strains, YP344 and YP579 were isolated from soil samples in Pocheon City, Gyeonggi Province, South Korea. The strains YP344 and YP579 belong to the genus Vishniacozyma and Dioszegia, respectively. The molecular phylogenetic analysis showed that the strain YP344 was closely related to Vishniacozyma peneaus. Strain YP344T differed by four nucleotide substitutions with no gap (0.70%) in the D1/D2 domain of the LSU rRNA gene and 16 nucleotide substitutions with 8 gaps (5.76%) in the ITS region. On the other hand, the strain YP579T varied from the type strain of the most closely related species, Dioszegia zsoltii var. zsoltii, by 6 nucleotide substitutions with four gaps (1.64%) in the D1/D2 domain of LSU rRNA gene and 26 nucleotide substitutions with 14 gaps (8.16%) in the ITS region. Therefore, the name Vishniacozyma terrae sp. nov. and Dioszegia terrae sp. nov. are proposed, with type strains YP344T (KCTC27988T) and YP579T (KCTC 27998T), respectively.

Alpha-Glucosidase Inhibitory Activity of Saponins Isolated from Vernonia gratiosa Hance

  • Species belonging to the Vernonia (Asteraceae), the largest genus in the tribe Vernonieae (consisting of about 1,000 species), are widely used in food and medicine. These plants are rich sources of bioactive sesquiterpene lactones and steroid saponins, likely including many as yet undiscovered chemical components. A phytochemical investigation resulted in the separation of three new stigmastane-type steroidal saponins (1 - 3), designated as vernogratiosides A-C, from whole plants of V. gratiosa. Their structures were elucidated based on infrared spectroscopy (IR), one-dimensional (1D) and two-dimensional nuclear magnetic resonance (2D NMR), high-resolution electrospray ionization mass spectrometry (HR-ESI-MS), and electronic circular dichroism analyses (ECD), as well as chemical reactivity. Molecular docking analysis of representative saponins with α-glucosidase inhibitory activity was performed. Additionally, the intended substances were tested for their ability to inhibit α-glucosidase activity in a laboratory setting. The results suggested that stigmastane-type steroidal saponins from V. gratiosa are promising candidate antidiabetic agents.

한국산 둥글넙치과(Bothidae) 어류, Bothus pantherinus의 첫기록 (First Record of Bothus pantherinus (Bothidae, Pleuronectiformes) from Korea)

  • 둥글넙치과, 별목탁가자미속에 속하는 Bothus pantherinus 1개체가 제주도 서귀포시 대정읍 모슬포항 조간대에서 2022년 8월 14일 처음 채집되었다. 본 개체는 가슴지느러미와 눈주위에 검은 점과 고리 모양의 무늬, 유안측 측선 중앙에 하나의 뚜렷한 반점을 가지는 점, 75개의 측선비늘과 7개의 모래시계 모양의 등지느러미 담기골을 가지는 점 등에서 B. pantherinus로 동정되었다. 미토콘드리아 DNA의 COI 영역 603 bp를 분석한 결과 NCBI에 등록된 B. pantherinus와 잘 일치하였다. 본 종은 인도양, 호주, 일본, 하와이 등 북위 32도에서 남위 32도 사이에 분포하는 것으로 알려져 왔으나 이번 연구를 통해 본 종의 분포역이 한국의 제주도 해역(북위 33도)까지 확장되었다. 국내에 처음 보고되는 본 종의 새로운 국명으로 "벚꽃무늬둥글넙치"를 제안한다.

  • 본 연구에서는 2018년 2~3월 제주도 남부해역에서 Bathylagidae에 속하는 Lipolagus ochotensis 자어 4개체(체장 13.4~21.3 mm)를 봉고네트로 채집하였다. L. ochotensis 자어는 몸이 길게 신장되었으며, 체고는 낮고, 눈이 돌출되며, 몸의 후반부에 흑색소포가 나 있고, 등지느러미가 몸 중앙에 위치하는 특징을 가진다. 미토콘드리아 DNA COI의 염기서열 625 bp를 분석한 결과, L. ochotensis 성어와 97.6%로 매우 가깝게 나타났다. 국내 처음 보고되는 본 종의 새로운 과명으로 "심해빙어과", 속명으로 "검은빙어속", 국명으로 "검은뺨빙어"를 각각 제안한다.

  • Daphne L. (Thymelaceae) comprises about 95 species distributing worldwide from N Africa, N India, SE Asia to E Asia and the coast of the Mediterranean of Europe. In Korea, five species of this genus have been described. In this study, we included four species (D. genkwa, D. pseudomezereum, D. kiusiana, D. jejudoensis) from Korea, excluding cultivated D. odora. The morphological characters through local surveys and the re-classification of the specimens collected in the Korea National Herbarium (KH) were carried out and distribution maps for each taxon were also prepared. The major characters include habit, trichomes in winter bud, leaf, and twig, phyllotaxis, inflorescence, size of calyx lobe and trichomes in the calyx tube, etc. The distribution map showed that D. genkwa is mainly distributed in the coastal area of Hwanghaenam-do, Pyeongannam-do, Jeollabuk-do and Jeollanam-do, whereas D. pseudomezereum is distributed in the limestone zone of Gangwon-do, Jeollabuk-do, and Gyeongsangbuk-do. Similarly, D. kiusiana is mostly found in Jeollanam-do, Gyeongsangnam-do, and Jeju-do. In addition, D. jejudoensis is known to be distributed in forests of Murueng, Andeok, and Seonheul-ri in Jeju-do, but recently, new habitat is discovered in the island forest areas of Jeollanam-do. However, some of these individuals showed the characteristics of D. kiusiana, thus before come to any conclusion detailed taxonomic review of D. jejudoensis and D. kiusiana is required.

Steroid Components of Marine-Derived Fungal Strain Penicillium levitum N33.2 and Their Biological Activities

  • Genus Penicillium comprising the most important and extensively studied fungi has been well-known as a rich source of secondary metabolites. Our study aimed to analyze and investigate biological activities, including in vitro anti-cancer, anti-inflammatory and anti-diabetic properties, of metabolites from a marine-derived fungus belonging to P. levitum. The chemical compounds in the culture broth of P. levitum strain N33.2 were extracted with ethyl acetate. Followingly, chemical analysis of the extract leaded to the isolation of three ergostane-type steroid components, namely cerevisterol (1), ergosterol peroxide (2), and (3β,5α,22E)-ergosta-6,8(14),22-triene-3,5-diol (3). Among these, (3) was the most potent cytotoxic against human cancer cell lines Hep-G2, A549 and MCF-7 with IC50 values of 2.89, 18.51, and 16.47 ㎍/mL, respectively, while the compound (1) showed no significant effect against tested cancer cells. Anti-inflammatory properties of purified compounds were evaluated based on NO-production in LPS-induced murine RAW264.7 macrophages. As a result, tested compounds performed diverse inhibitory effects on NO production by the macrophages, with the most significant inhibition rate of 81.37±1.35% at 25 ㎍/mL by the compound (2). Interestingly, compounds (2) and (3) exhibited inhibitory activities against pancreatic lipase and α-glucosidase enzymes in vitro assays. Our study brought out new data concerning the chemical properties and biological activities of isolated steroids from a P. levitum fungus.

Morphology and phylogenetic relationships of two Antarctic strains within the genera Carolibrandtia and Chlorella (Chlorellaceae, Trebouxiophyceae)

  • The genera Carolibrandtia and Chlorella have been described as small green algae with spherical cell shapes that inhabit various environments. Species of these genera are often difficult to identify because of their simple morphology and high phenotypic plasticity. We investigated two small coccoid strains from Antarctica based on morphology, molecular phylogeny by two alignment methods which have been applied to previous phylogenetic studies of the genus Chlorella, and comparison of the secondary structures of nuclear small subunit (SSU) and internal transcribed spacer (ITS) rDNA sequences. Light microscopy of two strains revealed spherical cells containing chloroplasts with pyrenoids, and the morphological characteristics of the strains were nearly identical to those of other Chlorella species. However, based on the phylogenetic analyses of nuclear SSU and ITS rDNA sequences, it was determined that the Antarctic microalgal strains belonged to two genera, as the Chlorella and Carolibrandtia. In addition, the secondary structures of the SSU and ITS2 sequences were analyzed to detect compensatory base changes (CBCs) that were used to identify and describe the two strains. A unique CBC in the SSU rDNA gene was decisive for distinguishing strain CCAP 211/45. The ITS2 rDNA sequences for each strain were compared to those obtained previously from other closely related species. Following the comparison of morphological and molecular characteristics, we propose KSF0092 as a new species, Chlorella terrestris sp. nov., and the reassignment of the strain Chlorella antarctica CCAP 211/45 into Carolibrandtia antarctica comb. nov.

딱총새우류와 공생하는 망둑어과 한국 첫기록종, Amblyeleotris japonica (First Record of a Shrimp Goby, Amblyeleotris japonica (Gobiiformes: Gobiidae) from Korea)

  • 우리나라 제주도 남부 연안의 수심 14 m에서 채집된 1개체 (표준체장 70.5 mm)의 표본을 근거로 딱총새우류와 공생하는 망둑어과 어류 Amblyeleotris japonica를 한국 첫기록종으로 보고한다. 이 종은 몸이 다소 측편하고 새공 하단이 눈의 후연과 전새개골 하단의 중앙 아래에 위치하며, 제2등지느러미 기조수가 1극 12연조, 뒷지느러미 기조수가 1극 14연조, 종렬비늘수가 28개, 머리에서 꼬리자루까지의 몸 측면에 5개의 폭넓은 적갈색 횡대가 있는 점이 특징이다. 이 종의 속명과 국명에 대해 '새우붙이망둑속'과 '붉은동갈새우붙이망둑'을 신한국명으로 제안한다.

Description and Genomic Characteristics of Weissella fermenti sp. nov., Isolated from Kimchi

  • A Gram-positive, non-motile, and non-spore-forming lactic acid bacterium, designated as BK2T, was isolated from kimchi, a Korean traditional fermented vegetable food, and the taxonomic characteristics of strain BK2T, along with strain LMG 11983, were analyzed. Both strains optimally grew at 30℃, pH 7.0, and 1.0% NaCl. Cells of both strains were heterofermentative and facultatively anaerobic rods, demonstrating negative reactions for catalase and oxidase. Major fatty acids (>10%) identified in both strains were C18:1 ω9c, C16:0, and summed feature 7 (comprising C19:1 ω6c and/or C19:1 ω7c). The genomic DNA G+C contents of both strains were 44.7 mol%. The 16S rRNA gene sequence similarity (99.9%), average nucleotide identity (ANI; 99.9%), and digital DNA-DNA hybridization (dDDH; 99.7%) value between strains BK2T and LMG 11983 indicated that they are different strains of the same species. Strain BK2T was most closely related to Weissella confusa JCM 1093T and Weissella cibaria LMG 17699T, with 100% and 99.4% 16S rRNA gene sequence similarities, respectively. However, based on the ANI and dDDH values (92.3% and 48.1% with W. confusa, and 78.4% and 23.5% with W. cibaria), it was evident that strain BK2T represents a distinct species separate from W. confusa and W. cibaria. Based on phylogenetic, phenotypic, and chemotaxonomic features, strains BK2T and LMG 11983 represent a novel species of the genus Weissella, for which the name Weissella fermenti sp. nov. is proposed. The type of strain is BK2T (=KACC 22833T=JCM 35750T).

Antibacterial Activity of Streptomyces Strains Isolated from Different Regions of Jordan

  • Members of the genus Streptomyces produce more than 70% of antibiotics. The rise in antibiotic resistance globally enhanced the search for novel species with the ability to produce new bioactive compounds. This study was initiated to investigate different regions in Jordan for previously uncultured and rare Streptomyces species capable of producing novel antimicrobial compounds especially active against bacteria resistant to antibiotics. A total of 191 Streptomyces strains were isolated from 26 soil samples collected from different geographic regions in Jordan. Isolates were characterized based on colony and cellular morphology as well as using 16S rRNA gene sequencing. These isolates were screened for their ability to produce antibiotics by the perpendicular-cross streak method, and then tested by well diffusion method against tested pathogens. Fifty-four isolates showed potential to produce antimicrobial products especially active against resistant bacteria, 20.1% of the isolates showed inhibitory effect against Staphylococcus aureus, 16.9% against clinical MSSA strains, and 18.0% against MRSA: whereas only 4.2% against Esherichia coli, 3.2% against Klebsiella pneumonia, 2.7% against Pseudomonas aeruginosa, and 10.0% against clinical Candida albicans. Three isolates were selected for further identification due to their antibacterial activity against S. aureus, MRSA, and MSSA. These isolates were identified as follows; Streptomyces aburaviensis DSa3, Streptomyces alboniger SAb7 and Streptomyces misionensis ZAb2, based on cultural, biochemical characteristics and molecular analysis of the 16S rRNA.
