Compounds Obtained from Sida acuta with the Potential to Induce Quinone Reductase and to Inhibit 7,12-Dimethylbenz-[a]anthracene-Induced Preneoplastic Lesions in a Mouse Mammary Organ Culture Model

  • Activity-guided fractionation of the EtOAc-soluble extract of the whole plants of Sida acuta using a bioassay based on the induction of quinone reductase (OR) in cultured Hepa 1c1c7 mouse hepatoma cells, led to the isolation of ten active compounds of previously known structure, quindolinone (1), cryptolepinone (2), 11-methoxyquindoline (3), N-trans-feruloyltyramine (4), vomifoliol (5), loliolide (6), 4-ketopinoresinol (7), scopoletin (8), evofolin-A (9), and evofolin-B (10), along with five inactive compounds of known structure, ferulic acid, sinapic acid, syringic acid, ($\pm$)-syringaresinol, and vanillic acid. These isolates were identified by physical and spectral data measurement. A new derivative of quindolinone, 5,10-dimethylquindolin-11-one (1a) was synthesized and characterized spectroscopically. Of the active substances, compounds 1-3 and 1a exhibited the most potent QR activity, with observed CD (concentration required to double induction) values ranging from 0.01 to 0.12 $\mu$ g/mL. Six compounds were then evaluated in a mouse mammary organ culture assay, with cryptolepinone (2), N-trans-feruloyltyramine (4), and 5,10-dimethylquindolin-11-one (1a) found to exhibit 83.3, 75.0, and 66.7% inhibition of 7,12-dimethylbenz[a]anthracene-induced preneoplastic lesions, respectively, at a dose of 10 $\mu\textrm{g}$/mL.

  • The reductive pinacol coupling reaction of aldehydes or ketones creating a new C-C bond has been a major tool to produce 1,2-diol compounds. The reaction mechanism is known to be composed of sequential three steps (activation, coupling, and dissociation). In this work, we studied the dissociation step of half-titanocene-based catalytic systems. Cp and $Cp^*$ derivatives of the pinacolato-bridged dinuclear complex were synthesized and evaluated as possible models for intermediates from the coupling step. We monitored $^1H$-NMR spectra of the reaction between the metalla-pinacol intermediates and $D_2O$. New reaction routes of the dissociation step including oxo- and pinacolato-dibridged dinuclear complexes and oxo-bridged multinuclear complexes have been suggested.

A new derivative of phorbaketals isolated from a Marine Sponge Phorbas species

  • A new sesterterpenoid, phorbaketal derivative, was isolated from the marine sponge Phorbas species. Its planar structures was completely determined from a combination of extensive 1D and 2D NMR experiments and MS data, and also the stereochemistry on the chiral centers were established by the ROESY experiment and the comparison with the $^1H$ and $^{13}C$ chemical shifts of the known phorbaketal compounds. This compound 1 moderately showed cytotoxicity effect against hepatoma cancer HepG2 cell.

  • A new flavonol derivative, meliglabrin (1) along with three known flavonols, ternatin (2), meliternatin (3), and 5,4'-dihydroxy-3,7,3'-trimethoxyflavon (4) were isolated from the leaves of Melicope glabra (Blume) T.G. Hartley. Their structures were determined using extensive spectroscopic methods, including UV, IR, HRESIMS, 1D and 2D NMR. Compounds 1 - 4 were evaluated for their cytotoxicity against murine leukemia P-388 cells, compound 4 showed moderate activity.

A New Pyranoxanthone Inophyllin B from Calophyllum inophyllum

  • A new prenylated pyranoxanthone, inophyllin B (1), was isolated from the roots of Calophyllum inophyllum (Guttiferae). Together with this compound was also isolated the known pyranoxanthone brasilixanthone B (2) and two common triterpenes friedelin (3) and sitosterol (4). Structural elucidations of these compounds were achieved through $^1H,\;^{13}C$, DEPT, COSY, HSQC and HMBC experiments. The molecular mass was determined using MS techniques. The crude extract indicated weak toxicity to the larvae of Aedes aegypti. We report here the isolation, structural elucidation and bioassay data for Inophyllin B and brasilixanthone B.

  • N,N-Dialkylditihiocarbamate derivatives have been well known as broad-range fungicides. In this study, the triazole derivatives of ten new N,N-disubstituted dithiocarbamates (3a-j) were synthesized and their structures were identified by spectral and elemental analysis. Results of the antifungal activity studies showed that some of the compounds tested were active against M. canis, M. gypseum, and T rubrum at the concentration of 12.5 $\mu$g/mL when c1otrimazol was used as a standard.

  • Phyllanthus ussuriensis Rupr. et Maxim. (Euphorbiaceae) has long been used in folk medicine to treat kidney and urinary bladder disturbances, intestinal infections, diabetes, and hepatitis. Reported chemical constituents of this species are one flavonoid (rutin), two gallotannins (gallic acid, methyl gallate) and two ellagitannins.An investigation of the n-BuOH fraction of P. ussuriensis led to the isolation of two new megastigmane glycosides, 10-hydroxy-4,7-megastigmadiene-3-one-9-O-b-D-glucopyranoside (1), 10- hydroxy-4,6-megastigmadiene-3-one-9-O-b-D-glucopyranoside (2) and two known compounds roseoside (3) and 3-oxo-a-ionol-9-O-b-D-glucopyranoside (4). A New Monoterpene from the Flower Buds of Buddleja officinalis

  • A new monoterpene, crocusatin M (1), was isolated from the flower buds of Buddleja officinalis, together with four known monoterpenes, (6R)-hydroxy-1,1,5-trimethylcyclohex-4-enone (2), (+)-dehydrovomifoliol (3), 7-epiloliolide (4), and crocusatin D (5). Their structures were determined by an extensive analysis of 1D, 2D NMR, HRESI-MS, and CD data as well as by comparison of their spectroscopic data with those of literatures. All isolates were evaluated for inhibitory activities on LPS-induced nitric oxide production in RAW 264.7 cells. Among them, compounds 2 and 3 showed moderate inhibitory effects with $IC_{50}$ values of 63.8 and 24.4 ${\mu}g/ml$, respectively.

  • A new phenolic glycoside, ophiopojaponin D (1), together with two known compounds, was isolated from the tubers of a famous traditional Chinese herb-Ophiopogon japonicus Ker-Gawl. The spectroscopic and chemical data revealed their structures to be 3-tetradecyloxy-4­hydroxy-allylbenzene-4-O-$\alpha$-L-rhamnopyranosyl (1$\rightarrow$6)-$\beta$-D-glucopyranoside (1), 3, 4-dihy-droxy-allylbenzene-4-O-$\alpha$-L-rhamnopyranosyl (1$\rightarrow$6)-$\beta$-D-glucopyranoside (2) and L-pyro­glutamic acid (3).

  • A new indolinepeptide (3) was isolated, together with two known compounds, a cerebroside (1) and an alloxazine (2), from silkworm larvae infected with Paecilomyces sp. J300. On the basis of spectroscopic data, their structures were elucidated as (4E, 8E, 25, 2'R, 3R )-N-2'-hydroxy-hexadecanoyl-1-O-$\beta$-D-glucopyranosyl-9-methyl-4, 8-sphingadienine (1), 7,8-dimethylalloxazine (2) and 3$\beta$,5-dihydroxy-1-N-methyl-indoline-2$\beta$-carbonyl amino-D-alanyl-erythro-$\beta$-hydoxyiso-leucinyl-glycine (3).
