New Sources of Resistance and Identification of DNA Marker Loci for Sheath Blight Disease Caused by Rhizoctonia solani Kuhn, in Rice

  • Sheath blight disease caused by the necrotrophic, soilborne pathogen Rhizoctonia solani Kuhn, is the global threat to rice production. Lack of reliable stable resistance sources in rice germplasm pool for sheath blight has made resistance breeding a very difficult task. In the current study, 101 rice landraces were screened against R. solani under artificial epiphytotics and identified six moderately resistant landraces, Jigguvaratiga, Honasu, Jeer Sali, Jeeraga-2, BiliKagga, and Medini Sannabatta with relative lesion height (RLH) range of 21-30%. Landrace Jigguvaratiga with consistent and better level of resistance (21% RLH) than resistant check Tetep (RLH 28%) was used to develop mapping population. DNA markers associated with ShB resistance were identified in F2 mapping population developed from Jigguvaratiga × BPT5204 (susceptible variety) using bulk segregant analysis. Among 56 parental polymorphic markers, RM5556, RM6208, and RM7 were polymorphic between the bulks. Single marker analysis indicated the significant association of ShB with RM5556 and RM6208 with phenotypic variance (R2) of 28.29 and 20.06%, respectively. Co-segregation analysis confirmed the strong association of RM5556 and RM6208 located on chromosome 8 for ShB trait. This is the first report on association of RM6208 marker for ShB resistance. In silico analysis revealed that RM6208 loci resides the stearoyl ACP desaturases protein, which is involved in defense mechanism against plant pathogens. RM5556 loci resides a protein, with unknown function. The putative candidate genes or quantitative trait locus harbouring at the marker interval of RM5556 and RM6208 can be further used to develop ShB resistant varieties using molecular breeding approaches.

  • The trials were carried out for improvement of early maturing wheat varieties, desease resistant and good quality through transfering good characters of barley to the wheat, during 1980-'81. For these purpose, Chinese Spring, a wheat variety was used as male and Manker 16 a barley variety, as female parents. EACA($\varepsilon$-Amino Caprolic Acid) known as an inhibitor of blood coagulation was injected to all barley from 14 days until heading with 1$m\ell$ per culm at the level of 500ppM. $GA_3$ solution which accelerates pollen activity was also used with 75ppM concentration after pollination for two days including check to determine the single effect of EACA. Two different methods of the cross were used. One was bud-pollination, another was post-pollination. The result indicated that the post-pollination was better than bud-pollination in terms of pollination rate, even though formation rate of normal embryo was seemingly adversed. The new plant derived from barley-wheat cross was similar in phenotype to the wheat.

Evaluation of Fiber Content According to the Cultivation Period of New Sweetpotato Varieties

  • Recently, as one of the major problems in the quality of sweetpotato, occurrence of thin and long fibrous tissues in storage root acts as a negative factor when consumers eat sweetpotato. In this study, the fiber content was compared according to the cultivation period in storage roots of 'Sodammi' and 'Hopungmi', which were newly bred and developed, and in that of 'Hogammi', which contains a lot of fibrous tissues. To isolate of fiber from storage root, the Association Official Analytical Chemists (AOAC) method was applied for quantifying fiber present in storage root of sweetpotato. The fiber contents isolated by this method is calculated by converting the weight of the storage root. The fiber content was measured every 20 days from 60 to 120 days after planting. As a result of this study, the lowest amount of fiber was 'Hopungmi' (70~140 mg/100 g), and the highest amount of fiber was observed in 'Hogammi' (115~223 mg/100 g). 'Sodammi' showed an intermediate level (104~149 mg/100 g) between the fiber content of 'Hopungmi' and 'Hogammi'. The fiber contents of 'Hopungmi' was 39% lower than that of 'Hogammi'. As the increased cultivation periods, the fiber contents showed a tendency to decrease. In the future research, the length, thickness, and fiber contents will be investigated to compare the degree of taste inhibition.

Correlation between Quantitative Agronomical Traits of Wheat (Triticum sp.) Genetic Resources

  • Assessment of plant genetic resources is applicable when breeding improved plant caltivars. In Korea, early maturing wheat germplasm is anticipated for wheat-rice double-cropping. In this study, we assessed five agronomical traits including days of heading after sowing(HD), days of maturity after sowing(MD), culm length(CL), ear length(EL), and thousand grain weight(TW) of 500 wheat germplasms collected form 10 different countries (Mexico, the United States, Afghanistan, Ethiopia, India, Japan, China, Turkey, Pakistan and Portugal), and grown in Korea. Besides, the correlation between the agronomical traits was analyzed using XLSTAT softerware version 2019 (Addinsoft, NY, USA). The result showed wide-ranging maturity period. Among the entire population, 2 accessions (K256306/JPN/breeding line and K256328/JPN/breeding line) that matured early were identified. Furthermore, HD showed strong correlation with MD (r=0.684) and CL (r=0.610), and weak correlation with the rest two agronomical traits (EL and TW). Overall, the results of our study provides wide spectrum of prospects, and could be applicable to breed new wheat varieties with early maturity.

  • Fresh mushrooms are vulnerable to browning and tissue changes after harvest. This study monitored the external appearance, physicochemical quality indicators, and nutritional and functional components of a white beech mushroom (Hypsizygus marmoreus) variety (EG2020) newly developed in Korea during cold storage for up to 42 days. Two existing varieties of white H. marmoreus, namely H6 from Korea and HKT from Japan, were used for comparison. The mechanical texture of EG2020 was superior to H6 and HKT due to the increasing hardness of the pileus with time. Browning, in terms of the total color difference during storage, was found to be the most severe in HKT. In terms of composition, EG2020 had the highest total free sugar content, a large amount of organic acids, and higher sugar content than H6 and HKT. EG2020 also contained the largest amount of 𝛽-glucan, and its amount increased during storage. In sensory evaluation, EG2020 received higher scores than HKT in flavor, taste, appearance, and texture. Therefore, the EG2020 variety is more stable than HKT during storage and distribution.

Forecasting Fish Import Using Deep Learning: A Comprehensive Analysis of Two Different Fish Varieties in South Korea

  • Nowadays, Deep Learning (DL) technology is being used in several government departments. South Korea imports a lot of seafood. If the demand for fishery products is not accurately predicted, then there will be a shortage of fishery products and the price of the fishery product may rise sharply. So, South Korea's Ministry of Ocean and Fisheries is attempting to accurately predict seafood imports using deep learning. This paper introduces the solution for the fish import prediction in South Korea using the Long Short-Term Memory (LSTM) method. It was found that there was a huge gap between the sum of consumption and export against the sum of production especially in the case of two species that are Hairtail and Pollock. An import prediction is suggested in this research to fill the gap with some advanced Deep Learning methods. This research focuses on import prediction using Machine Learning (ML) and Deep Learning methods to predict the import amount more precisely. For the prediction, two Deep Learning methods were chosen which are Artificial Neural Network (ANN) and Long Short-Term Memory (LSTM). Moreover, the Machine Learning method was also selected for the comparison between the DL and ML. Root Mean Square Error (RMSE) was selected for the error measurement which shows the difference between the predicted and actual values. The results obtained were compared with the average RMSE scores and in terms of percentage. It was found that the LSTM has the lowest RMSE score which showed the prediction with higher accuracy. Meanwhile, ML's RMSE score was higher which shows lower accuracy in prediction. Moreover, Google Trend Search data was used as a new feature to find its impact on prediction outcomes. It was found that it had a positive impact on results as the RMSE values were lowered, increasing the accuracy of the prediction.

Effect of Varieties and Seeding Date on Over Winter and Dry Matter Yield of Italian Ryegrass in Paddy Field (논에서 이탈리안 라이그라스 품종별 파종시기가 월동 및 건물수량에 미치는 영향)

  • This experiment was carried out to investigate the effect of cultivar and seeding date on the winter survival rate, quality and DM yield of Italian ryegrass on paddy field for three years in Suwon. Seeding started from 30th Sep. 2003. at intervals of five days and finished 20th Oct. New varieties of Italian ryegrass used in this experiment were "Kospeed", "Kowinmaster" and "Hwasan 101". The winter survival average rate of 5th Oct. seeding plot was 89.8% and it decreased with delayed seeding date. The heading date of "Kospeed" was 7th~13rd May, "Kowinmaster" was 16th May, but "Hwasan 101 ho" didn't show heading until 17th May. Dry matter (DM) yields of 30th Sep. seeding plot were Kospeed 7,909kg/ha, Kowinmaster 6,398 kg/ha and Hwasan 101 ho 5,204 kg/ha. DM yield was decreased with delayed seeding date. Total digestible nutrient (TDN) yield was also decreased with delayed seeding date. Crude protein (CP) content was the highest in Kospeed. seeding plot as 18.3% and in vitro dry matter digestibility (IVDMD) was not showed significant difference among seeding dates.

Screening of High-Palatability Rice Resources and Assessment of Eating Quality Traits of Korean Landraces and Weedy Rice Germplasms (우리나라 재래벼와 잡초벼의 식미 특성 평가 및 고식미 우수자원 탐색)

  • The eating quality of rice is one of the main concerns of rice breeding programs in many countries, especially in japonica rice cultivation areas. To select new resources with high eating quality from Korean native japonica rice, we evaluated a total of 76 varieties, including 47 native rice resources (26 landraces + 21 weedy rice) of Korea. In this study, all eating quality traits varied widely among the native resources, and some of the native resources revealed a high evaluation score in the palatability, expected eating quality, and physicochemical traits among the tested whole-plant materials. From the results, we selected two landraces (Sangdo and Waebyeo) and three weedy rice varieties (Hoengseongaengmi3, Namjejuaengmi6, and Wandoaengmi6) as promising resources for improvement of rice eating quality. Specifically, Wandoaengmi6 presented potential as a key breeding material for improving the eating quality of Korean rice cultivars, having the best evaluation results in palatability score (PS 0.83) from the sensory test and glossiness value (GV 81.8) from the Toyo taste meter of cooked rice. Given the urgent need to overcome the constraint of the narrow genetic background of Korean japonica rice, the results could be a practical solution for exploring new opportunities for improving rice eating quality through the expansion of genetic resources.

A New Early-Heading and High-Yielding Forage Rye Variety, "Olgreen" (극조숙 청예다수성 호밀 신품종 "올그린")

  • "Olgreen" (Scale cereal L.), a new rye variety was developed by National Institute of Crop Science(NICS), RDA. It was released in 2008 because of its high biomass yield potential and early maturity. It was developed from an open pollination within 10 rye varieties or lines including "Chochun" in 1995. The line 'SR95POP-S1-1072-1-2-4' was selected for its early maturity and excellent agronomic appearance, and placed in yield trials for two years from 2004 to 2005. It was designated "Homil 38" and placed in regional yield trials at the four locations around Korea from 2006 to 2008, from which the name "Olgreen" was given. Over three years, "Olgreen" averaged 8.88 ton ha-1 of forage yield (based on dry matter) harvested at late April and superior to other varieties with an increase of 10% more than the check variety "Olhomil", and 9% more than the introduced rye "Koolgrazer". Heading date of "Olgreen" was April 20 which was 3 days earlier than that of "Olhomil". It would be recommended as an early rye variety for forage or green manure in South Korea.

Characterization of a new commercial strain 'Goni' by intra-specific hyphal anastomosis in Pleurotus ostreatus (계통간 교잡에 의한 백색느타리 품종 '고니'의 육성 및 그 특성)

  • To develop new white variety of Oyster mushroom, all white varieties which have been collected and kept in the lab were revived and screened their cultural characteristics. 84 intra-specific Oyster mushroom hybrids between the white-colored mutants Suhan and Wonhyeong were developed using hyphal anastomosis technique in 2007. The Po2007-63 ($2842-7{\times}0205-7$) was shown the best cultural characteristics, selected to be a new variety and named as 'Goni'. The new commercial strain, 'Goni' has white pilei and grows well under spring and autumn conditions in Korea. The fruiting bodies of 'Goni' are of an excellent quality in that not only the stipe is thick and long but also the pileus is small and hard. The optimum temperatures for mycelial growth and fruiting body development were $25-30^{\circ}C$ and $10-16^{\circ}C$, respectively. Time period required for the initiation of the first fruiting body is about 3 to 5 days depending on the temperatures. The shape of fruiting body is thin funnel shape. Fruiting body production per bottle was about $91{\pm}13$ g which is almost 97% quantity compared to that of other variety 'Miso'. Relatively low temperature incubation ($11^{\circ}C$) resulted in the development of better quality of 'Goni' mushrooms. When two different media including potato dextrose medium and mushroom complete medium were compared, the growth of mushroom were much faster in mushroom complete medium at $20-25^{\circ}C$, but not at $25^{\circ}C$. Similar results were observed with other variety 'Miso'. Analysis of the genetic characteristics of the new commercial strain 'Goni' showed a major DNA profile as that of the parental Suhan when primer URP 1 was used, but different to 'Miso' that was used as a control. When screens were performed with primer URP 2, DNA patterns were similar both to that of the parents and 'Miso'. This new variety of the white Oyster mushroom has a clean and fresh image that corresponds well to "health food". We therefore expect that this new strain will satisfy the consumers demand for variety and excellent mushrooms.
