황금추출물의 간장기능 회복능을 알아보기 위하여, $CCl_4$로 간중독을 유도한 쥐에게 황금추출물을 급여한 후, 혈액성상, 혈액 내 효소활성치 및 간장 내 전염증성 cytokines의 농도변화를 검토했다. RBC, Hb 및 PCV의 값은 처리군간에 유의한 차이를 나타내지 않았다. 백혈구 총수는 대조군과 비교하여 황금 처리군 모두가 낮은 수치를 나타내었으며, 황금추출물 첨가량이 증가함에 따라 neutrophils 및 eosinophile의 비율이 감소하고, lymphocytes 및 monocytes의 비율이 증가하였다. basophils의 구성비는 처리군 별 유의한 차이를 나타내지 않았다. Total protein 농도 및 albumin 농도는 처리군 간에 유의한 차이를 나타내지 않았다. 그러나 albumin/globulin 농도 비율은 대조군보다 황금추출물 처리군에서 높았다. GOT, GPT 및 LDH의 효소활성치들은 모두가 대조군보다 황금추출물 처리군에서 낮은 수치를 나타내었으며, 황금추출물 첨가량이 증가함에 따라 하락하는 경향을 보였다. 간장 내 IL-$1{\beta}$, IL-6 및 TNF-$\alpha$의 농도는 대조군보다 황금추출물 처리군 모두가 낮은 값을 나타내었으며, IL-10의 농도는 대조군보다 황금추출물 처리군 모두가 높은 값을 나타내었다. 이상의 결과를 종합해 보면 황금추출물은 간장기능회복에 효과를 나타내는 기능성 물질을 내재하고 있음을 시사해 준다.

The effects of dietary supplementation of selenium (Se), iodine (I), and a combination of both on the blood haematology, serum free thyroxine (FT4) and free triiodothyronine (FT3) hormones and glutathione peroxidase enzyme (GSH-Px) activity were examined on twenty four (7 to 8 months old, $22{\pm}1.17$ kg live weight) Kacang crossbred male goats. Animals were randomly assigned to four dietary treatments (6 animals in each group). Throughout 100 d of feeding trial, the animals of control group (CON) received a basal diet, while the other three groups were offered basal diet supplemented with 0.6 mg/kg diet DM Se (SS), or 0.6 mg/kg diet DM I (PI), or a combination of both Se and I, each at 0.6 mg/kg diet DM (SSPI). The haematological attributes which are haemoglobin (Hb), red blood cell (RBC), packed cell volume (PCV), mean cell volume (MCV), white blood cells (WBC), band neutrophils (B Neut), segmented neutrophils (S Neut), lymphocytes (Lymph), monocytes (Mono), eosinophils (Eosin) and basophils (Baso) were similar among the four treatment groups, while serum levels of Se and I increased significantly (p<0.05) in the supplemented groups. The combined dietary supplementation of Se and I (SSPI) significantly increased serum FT3 in the supplemented animals. Serum GSH-Px activity increased significantly in the animals of SS and SSPI groups. It is concluded that the dietary supplementation of inorganic Se and I at a level of 0.6 mg/kg DM increased serum Se and I concentration, FT3 hormone and GSH-Px activity of Kacang crossbred male goats.
Six week-old ICR mite which were divided into four groups including NC, RC, RR and RS were injected with sun ginseng (RS), red ginseng (RR) and saline (RC) intraperitonedlly as an amount of 60 mg/g body weight at 1 hour, 12 hours and 36 hours before the irradiation of high-energy X-ray and the mire were sacrificed at three and a half days after the irradiation. The RS group were significant increase in the weight of spleen (p<0.01) and the numbers of jejunal crypt cells (P<0.01), WBC (p<0.05), lymphocytes (p<0.05) and neutrophils (p<0.05) in comparison with the RC group. The RR group were significant increase in the numbers of jejunal crypt cells (p<0.001), WBC (p<0.05) and neutrophils (p<0.05) in comparison with the RC group. The RS group exhibited a more increase in the weights of spleen and thymus and the numbers of jejunal crypt cells and all items of hematological examination than the RR group. The values of ALT (alanine transaminase) and AST (aspartate transaminase) were significantly elevated (p<0.05) by radiation and they were significantly decreased (p<0.05) in the RS group to the values of the NC group. Taken together the above results, sun ginseng demonstrated a jejunal crypt survival effect, the protective effects on hepatocytes and immune and hematopoietic cells in mice irradiated with high-energy X-ray, and those radiation protective effects were a little higher in comparison with red ginseng.
The present study was undertaken to establish reference values for the composition blood lymphocyte populations and compare forty three Hanwoo neonatal calves (KC) with twenty one Holstein calves (HC) by blood cell count and immunophynotying. The percentages of CD2+, CD4+, CD8+, CD26+, ACT2+, MHC class, MHC class II and WC1+ T cells, B cells were determined by flow cytometry. The number of lymphocyte and monocyte in HC were higher than those of KC. However, the number of neutrophils was higher in HC than KC. The proportions of CD2+, CD4+, CD8+, MHC class, and WC1+ lymphocytes remained relatively stable during the study period, while there was a moderate increase in the relative percentage of CD26+, ACT2+, MHC class II and B cell from birth to approximately 3 weeks of age. Marked differences in the relative proportions of the lymphocyte subpopulations were noted between the individual calves. The present study shows that the T-cell subpopulations are present in peripheral blood of KC at levels comparable with HC, while the MHC class II and B cell population of KC increases significantly with age. The absolute number of WBC in KC was due to the decrease of absolute number of neutrophil rather than the increase of lymphocyte. The results indicated that KC have significantly higher number of neutrophils, and proportion of MHC class II and B cell than HC.
The study was conducted to correlate the periparturient immune status in terms of neutrophil functions and cytokine expression in peripheral blood mononuclear cell culture with impending postpartum reproductive disorders in buffaloes. Forty pregnant buffaloes were observed for occurrence of postpartum reproductive disorders (PRD), i.e., metritis, endometritis and delayed uterine involution etc., during one week prepartum to four weeks postpartum period. A representative number (n = 6) of buffaloes that did not develop any PRD were included in group I (healthy, control), while the animals which experienced PRD were assigned into group II (PRD, n = 8). The blood samples were collected at weekly interval from one week prepartum to four weeks postpartum period considering the day of calving as 'd 0'. Differential leucocytes counts, superoxide and hydrogen peroxide production activity in isolated neutrophils and the mRNA expression profile of cytokines i.e., IL-2, IL-4 and IFN-${\gamma}$ in PBMC culture were studied in all the samples. A higher total leucocytes, neutrophil and band cells count along with impaired neutrophil functions i.e., lowered level of production of superoxide and hydrogen peroxide before parturition and during early postpartum period were observed in buffaloes developing PRD. Further, a lower expression of IL-2, IFN-${\gamma}$ and IL-4 mRNA in PBMC culture was observed at calving in buffaloes that subsequently developed PRD at later postpartum. Thus, suppression in neutrophil function and cytokine expression at prepartum to early postpartum period predisposes the buffaloes to develop postpartum reproductive disorders. Hence, monitoring of neutrophils function and cytokine expression profile would be effective to predict certain reproductive disorders at late pregnancy or immediately after parturition in buffaloes. In future, this may be a novel approach for determining suitable management and therapeutic decisions for prevention of commonly occurring reproductive disorders in farm animals.
1. The sixty raised shepherd and sixty-five inhoused pet dogs in the regions of Daejon and Cheongju were subjected to investigate the TB infection by means of BCG and X-ray diagnosis. The 5 out of 65 inhoused pet and 7 out of 60 shepherd dogs were observed to be infected with TB, respectively. However, none of Mycobacterium species were detected from lung tissues of 4-slaughtered dogs showing BCG positive reaction. 2. The rats were first inoculated with 0.1ml BCG, and then 0.1ml M bovis suspended solution($1{\times}10^5$ organisms/0.2ml) 3weeks later. After 5 months, the animals were killed. The pathohistological results from both groups, TB inoculated and BCG treated groups, were observed on the surface of lung. Furthermore, the severe pathological lesion in the Iung was observed in M bovis inoculated group compared to BCG treated group. 3. The slight macrophage invasion and granuloma formation in the lung from BCG treated group were observe individually. However, it was confirmed that the lung from M bovis treated group was invaded by the macrophages and neutrophils combined with the granuloma formation. 4. When the numbers of the total cells taken from broncho-alvealar fluid in each of mouse from both groups were differentially counted, the number of total cell, neutrophils, and lymphocytes from M bovis treated group were significantly increase compared with those of BCG treated group. 5. Although there were nearly no response of the alveolar macrophages to CSF in serum obtained from control group, those from M boris treated group were significantly proliferated.
Effects of moxibution at the Gwanweon, Jungwan and Joksamlee on the blood picture in rat with transportation stress were determined. Counts of RBC showed a tendency to increase from 6 hours to 12 hours after transportation stress, however in the moxibution group, showed no changes in counts of RBC after transportation stress and the tendency of fluctuation was similiar to those of none stress group. The changes in Hb after transportation stress showed no difference among moxibution group and none moxibution group, however the changes in PCV showed a tendency to increase from 3 hours to 6 hours after transportation stress in two stress group. In the mean values of erythrocytic blood during experimental times, counts of RBC showed a high values (P<.05) in the stress only group, however in the other groups, these values showed no difference (P>.05) among treatment and the values of Hb and PCV showed no difference among 4 treatment groups. In the two stress groups, counts of WBC and Neutrophils showed a tendency to decrease after transportation stress, however the moxibution group was recovered to normal Leukocytes condition on short time compared with those of none moxibution group. In the mean values of Leukocytes during experimental times, counts of WBC and Neutrophils showed a high values and Lymphocytes showed a low values in the stress only group compared with those of other groups, however the stress group with moxibution showed no difference in Leukocytes values compared with those of normal condition group. Monocytes, Basophils and Eosinophils showed no difference among 4 treatment groups(P>.05). Results from this study indicate that the moxibution can tolerate the effects of transportation stress in rat.
Although various automated CBC analyzers with different WBC analytical principles were consequently introduced to clinical laboratory, the specific information concerning the suitability or unsuitability of aging samples is scarce. For this reason, we studied the effect of storage duration and temperature on CBC parameter in SE-9000 (SYSMEX Medical Electronics Co., Ltd., Kobe, Japan), automated CBC analyzer. We tested 32 K3-EDTA specimens with SE-9000 during 72 hours. Specimens were kept at room temperature (RT) and refrigerated and were analyzed at 0 hr, 4 hr, 8 hr, 24 hr, 48 hr and 72 hr after the collection of the specimens. The percentage changes from initial value for each parameters were calculated. Among the CBC parameters, hemoglobin, red blood cell count, mean corpuscular hemoglobin and platelets were stable for the study period at both temperatures. The mean corpuscular volume (MCV), hematocrit (Hct) and red cell distribution (RDW) increased and the mean corpuscular hemoglobin concentration (MCHC) decreased over time at room temperature. These parameters were stable when refrigerated. The leukocyte count was stable during 72hr at RT and when refrigerated. At room temperature, the relative percentages of neutrophils tend to increase, whereas those of lymphocyte and monocytes tend to decrease after 48 hours. When refrigerated, those of neutrophils and monocytes tend to increase, whereas those of lymphocytes tend to decreased over time. CBC parameters of refrigerated specimen were reliable for 72 hr for the exception of differential count from 24 hr but many CBC parameters, such as MCV, Hct, MCHC, RDW and differential count of leukocyte of blood stored at room temperature for 24 hr were unreliable.

The current observational epidemiological study analyzed blood lead levels (BLLs) in occupationally exposed workers from Riyadh region, Saudi Arabia and correlated them with the alterations in the differential cell populations of the WBC panel (lymphocytes [Lym %], mixed [Mid %] cells, and neutrophils [Neu %]). In addition, we examined the effect of confounding factors and their relation to BLLs. BLLs were estimated using the LeadCare II analyzer and hematological parameters using the ADVIA 120 analyser. An inferential analysis was conducted to detect association between the observations and the subjects' clinical characateristics. A total of 132 male subjects were included in the final analyses. Based on CDC guidelines, the subjects were categorized as Group I (BLL <10 ㎍/dL; n=118) or Group II (BLL >10 ㎍/dL; n=14) with average BLLs of 4.4 ㎍/dL and 18.1 ㎍/dL, respectively (p <0.0001). The percentages of Mid cells (p <0.0001) and neutrophils (p=0.048), were significantly altered in subjects with High BLL. A regression analysis indicated that subjects > 50 years of age had significantly higher BLLs (53.2 ㎍/dL) than younger age sub-groups (p <0.0001). Age, education, and profession were significant predictors for lead toxicity. Pb exposure is a major public health issue in Saudi Arabia and calls for further investigations on the cellular and molecular effects on hematological system.
Although there are many reports on the splenic (systemic) T cell response after Toxoptasma gondii infection, little information is available regarding the local T cell responses of peritoneal exudate cells (PEC) and gut intraepithelial Iymphocytes (IEL) following peroral infection with bradyzoites. Mice were infected with 40 cysts of the 76K strain of T. gondii, and then sacrificed at days 0, 1, 4, 7 and 10 postinfection (PI). The cellular composition and T cell responses of PEC and IEL were analyzed. The total number of PEC and IEL per mouse increased after infection, but the ratio of increase was higher in IEL. Lymphocytes were the major component of both PEC and IEL. The relative percentages of PEC macrophages and neutrophils/eosinophils increased signiflcantly at day 1 and 4 PI, whereas those of IEL did not change significantly. The percentage of PEC NK1.1 and ${\gamma\delta}T$ cells peaked at day 4 PI (p < 0.0001), and CD4 and $CD8{\alpha}T$ cells increased continuously after infection. The percentages of IEL $CD8{\alpha}$ and ${\gamma\delta}T$ cells decreased slightly at first, and then increased. CD4 and NK1.1 T cells of IEL did not change significantly after infection. $IFN-{\gamma}-producing$ PEC NK1.1 T cells increased significantly from day 1 PI, but the other T cell subsets produced $IFN-{\gamma}$ abundantly thereafter. The proportion of IEL $IFN-{\gamma}-producing$$CD8{\alpha}$ and ${\gamma\delta}T$ cells increased significantly after infection, while IEL NK1.1 T cells had similar $IFN-{\gamma}$ production patterns. Taken together, CD4 T cells were the major phenotype and the important $IFN-{\gamma}$ producing T cell subsets in PEC after oral infection with T. gondii whereas $CD8{\alpha}T$ cells had these roles in IEL. These results suggest that PEC and IEL comprise different cell differentials and T cell responses, and according to infection route these factors may contribute to the different cellular immune responses.
