Objectives : The purpose of this study was to evaluate the significance of antibiotics in reducing postoperative infection rates and other complications following third molar surgery. Patients and methods : Two groups of patients underwent surgical extraction of third molars. The antibiotics group, n=21, received a third generation cephalosporin antibiotic for 5 days, starting from the day of surgery. The non-antibiotics group, n=26, didn't receive any antibiotics and only received analgesics to control postoperative pain. Body temperature and hematologic findings including WBC, neutrophils, lymphocytes and monocytes counts were compared between the two groups at three intervals, preoperatively, 24 hours and 7-10 days postoperatively. Pain and swelling during the follow up period were also recorded in both groups and compared in the second part of the study. Results : In the first part of the study, comparison of body temperature, CBC components (except WBCs) showed no significant difference between the two groups during the follow up period. All parameters were within the normal range at all intervals, which indicated absence of infection. In the second part, 38% of patients in the antibiotic group, compared to 54% of the non-antibiotics group, had one or more complications during the follow up period. However, three patients from the antibiotic group compared to one from the nonantibiotics group reported having a swelling of some degree. Conclusion : Based on our objective parameters (body temperature and CBC components), both groups showed no signs of infection during the follow up period. However, the results related to pain and swelling were less conclusive, probably due to small number of patients included in the study. Accordingly, we are unable to provide definite recommendations on antibiotics use in third molar surgery.
Background: NADPH oxidase (NOX) modulates cell proliferation, differentiation and immune response through generation of reactive oxygen species. Particularly, NOX2 is recently reported to be important for regulating Treg cell differentiation of CD4+ T cells. Methods: We employed ovalbumin-induced airway inflammation in wild-type and NOX2-deficient mice and analyzed tissue histopathology and cytokine profiles. Results: We investigated whether NOX2-deficiency affects T cell-mediated airway inflammation. Ovalbumin injection which activates T cell-mediated allergic response increased airway inflammation in wild-type mice, as evidenced by increased immune cell infiltration, allergic cytokine expression, and goblet cell hyperplasia in the lung. Interestingly, NOX2 knockout (KO) mice were more susceptible to allergen-induced lung inflammation compared to wild-type mice. Immune cells including neutrophils, lymphocytes, macrophages, and eosinophils were drastically infiltrated into the lung of NOX2 KO mice and mucus secretion was substantially increased in deficiency of NOX2. Furthermore, inflammatory allergic cytokines and eotaxin were significantly elevated in NOX2 KO mice, in accordance with enhanced generation of inflammatory cytokines interleukin-17 and interferon-${\gamma}$ by CD4+ T cells. Conclusion: These results indicate that NOX2 deficiency favorably produces inflammatory cytokines by T cells and thus increases the susceptibility to severe airway inflammation.
Objectives : To investigate the effect of electro-acupuncture (EA) at GB34 on hepatotoxicity in $CCl_4$-intoxicated rats. Methods : Rats were injected with $CCl_4$ and treated with acupuncture or 2 Hz electro-acupuncture (EA) at left GB34 three times a week for 10 weeks. A non-acupoint in left gluteal area was selected as a sham point. To estimate the effects of EA on hepatotoxicity in rats, body weight, liver weight and liver index were measured, and biochemical assays for serum ALT, AST, ALP and total cholesterol, and hematological analysis for RBC, WBC, PLT, hemoglobin, lymphocytes, neutrophils and monocytes, and histology analysis of liver tissue were performed. Results : 1. Lymphocyte level in blood was significantly decreased by $CCl_4$-intoxication and significantly increased by acupuncture and 2 Hz EA at left GB34. 2. Neutrophill and monocyte level in blood was increased by $CCl_4$-intoxication and significantly reduced by acupuncture and 2 Hz EA at left GB34. 3. Acupuncture and 2 Hz EA at left GB34 significantly reduced serum ALT and AST which were increased by $CCl_4$-intoxication. 4. EA at GB34 significantly reduced serum ALT and AST as compared with EA at sham point in $CCl_4$-intoxicated rat. 5. No significant difference was found between the effects of acupuncture and that of 2 Hz EA on $CCl_4$-induced liver damage in rats. Conclusions : 2 Hz EA at GB34 has hepatoprotective effects on $CCl_4$-induced liver damage in rats and the point-specificity of GB34 may be involved in these effects.
The principal significance of the urothelial changes caused by polyomavirus activation is in an erroneous diagnosis of urothelial cancer; however, the clue to their benign nature is the smooth structureless nuclear configuration and the relative paucity of affected cells. Though virologic studies and electron microscopy are usually needed to firmly establish the diagnosis, cytology is the most readily available and rapid means of establishing a presumptive diagnosis of human polyomavirus infection. A urine specimen of a 24-year-old man with hemorrhagic cystitis beginning two months after bone marrow transplantation for acute myeloblastic leukemia(M2) was submitted for cytologic evaluation. Cytologic findings revealed a few inclusion-bearing epithelial cells intermingled with erythrocytes, neutrophils, lymphocytes, and macrophages. Most of the inclusion-bearing fells had large, round to ovoid nuclei almost completely filled with homogeneous dark, basophilic inclusion. The chromatin was clumped along the periphery and the cytoplasm was mostly degenerated. The other cells exhibited irregular inclusions attached to the nuclear membrane surrounded by an indistinct halo. These findings were consistent with polyomavirus infection.
Next to performing an expert physical examination, a blood sample submitted for a complete blood count is the most basic tool available to owners or veterinary practitioners. Reference values of complete blood count were determined at 6 different ages in 114 Thoroughbred foals during the second month of life. Hematologic results were as follows: RBC 8.2-10.5 ($10^6/{\mu}l$), Hb 10.9-13.3 (g/dl), HCT 28.2-35.2 (%), MCV 30.7-35.8 (fL), MCH 11.9-13.5 (pg), MCHC 37.9-40.5 (g/dl), RDW 24.5-25.7 (%), PLT 146.3-256.4 ($10^3/{\mu}l$), MPV 6.7-8.3(fL), total WBC 8.1-12.5 ($10^3/{\mu}l$), basophils 0.0 ($10^3/{\mu}l$), eosinophils 0.0-0.3 ($10^3/{\mu}l$), neutrophils 2.4-8.6 ($10^3/{\mu}l$), lymphocytes 1.8-2.9 ($10^3/{\mu}l$) and monocytes 0.0-1.4 ($10^3/{\mu}l$). The results of this study serve as reference ranges for Thoroughbred neonatal foals populations and can be useful for health control, regular examination and pre-sale soundness examination.
Effects of aqua-acupuncture with soshiho-tang at Kansu(BL_{18}$) and Tamsu(BL_{19}$) on the Glutamic Oxaloacetic Transaminase(GOT), Glutamic Pyruvic Transaminase(GPT), Lactic dehydrogenase(LDH) and blood pictures in rat with $CCI_4$ were determined. In the group with $CCI_4$ the activity of GOT, GPT and LDH showed a tendency to increase, however these values showed a tendency to decrease by the aqua-acupuncture with soshiho-tang throughout experimental days and these values were recovered to those of control group on the last experimental days. Counts of RBC and Hemoglobin showed a tendency to decrease in rat with $CCI_4$, however these decreased values were recovered by the aqua-acupuncture with soshiho-tang and on the last experimental days, these values were similar to those of control group. In the $CCI_4$ group, counts of WBC and the proportion of Lymphocytes showed a tendency to increase and the proportion of Neutrophils showed a tendency to decrease, however these values showed a rapid recovery by the aqua-acupuncture with soshiho-tang. In the meridian point of Kansu and Tamsu, the effects of aqua-acupuncture with soshiho-tang on recovery of liver function showed no difference. Results from this study indicate that the aqua-acupuncture with soshiho-tang can improve the liver function in rat with $CCI_4$.
Many studies have reported that bleomycin, anti-cancer drug, induces pulmonary fibrosis as a side effect. However, few investigations have focused on the dose-response effects of bleomycin on pulmonary fibrosis. Therefore, in the present study, we investigated the effects of different doses of bleomycin in male mice. ICR mice were given 3 consecutive doses of bleomycin: 1, 2, or 4 mg/kg in bleomycin-treated (BT) groups and saline only in vehicle control (VC) groups. The animals were sacrificed at 7 and 24 days postinstillation. The severity of pulmonary fibrosis was evaluated according to inflammatory cell count and lactate dehydrogenase (LDH) activity in the broncho alveolar lavage fluid (BALF), and lung tissues were histologically evaluated after hematoxylin and eosin (H&E), and Masson's trichrome staining. BT groups exhibited changed cellular profiles in BAL fluid compared to the VC group, which had an increased number of total cells, neutrophils, and lymphocytes and a modest increase in the number of macrophages at 7 days post-bleomycin instillation. Moreover, BT groups showed a dose-dependent increase in LDH levels and inflammatory cell counts. However, at 24 days after treatment, collagen deposition, interstitial thickening, and granulomatous lesions were observed in the alveolar spaces in addition to a decrease in inflammatory cells. These results indicate that pulmonary fibrosis induced by 4 mg/kg bleomycin was more severe than that induced by 1 or 2 mg/kg. These data will be utilized in experimental animal models and as basic data to evaluate therapeutic candidates through non-invasive monitoring using the pulmonary fibrosis mouse model established in this study.
Background: Lung cancer (LC) is still the primary cause of cancer deaths worldwide, and late diagnosis is a major obstacle to improving lung cancer outcomes. Recently, elevated preoperative or pretreatment neutrophil to lymphocyte ratio (NLR), platelet to lymphocyte ratio (PLR) and mean platelet volume (MPV) detected in peripheral blood were identified as independent prognostic factors associated with poor survival with various cancers, including colon cancer, esophageal cancer, gastric cancer and breast cancer. Objective: The aim of this study was to examine whether MPV, NLR and PLR could be useful inflammatory markers to differentiate lung cancer patients from healthy controls. An investigation was also made of the relationship between these markers and other prognostic factors and histopathological subgroups. Materials and Methods: Retrospectively eighty-one lung cancer patients and 81 age-sexes matched healthy subjects included into the study. Patients with hypertension, hematological and renal disease, heart failure, chronic infection, hepatic disorder and other cancer were excluded from the study. The preoperative or pretreatment blood count data was obtained from the recorded computerized database. Results: NLR and PLR values were significantly higher in the LC patients compared to the healthy subjects.( NLR: 4.42 vs 2.45 p=0.001, PLR: 245.1 vs 148.2 p=0.002) MPV values were similar in both groups (7.7 vs 7.8). No statistically significant relationship was determined between these markers (MPV, NLR and PLR) and histopathological subgroups and TNM stages. Conclusions: NLR and PLR can be useful biomarkers in LC patients before treatment. Larger prospective studies are required to confirm these findings.
To investigate the alleviative effects of Peonia japonica water extract(PJWE) on inflammation and skin barrier damage, both the irradiation of UVB and the application of squalene monohydroperoxide (Sq-OOH) to the backs of hairless mice were performed for 4 weeks. And at the same time experimental materials were applied topically. The skin erythema indices for the positive control (PC, 0.01% retinoic acid) and experimental (E, PJWE) groups were lower than that of the control (C) group. Whereas both the lipid and water capacities for the PC and E groups were higher than those of the C group. Epidermis and dermis of the C group were remarkably thickened in comparison with the PC and E groups. Relatively much less number of inflammatory cells, including lymphocytes, neutrophils and macrophages were found in dermis of the PC and E groups compared with the C group. Lipid lamellae of the C group were broken severely showing an irregular arrangement and lipid content was much reduced. Whereas those of the PC and E groups were almost intact with a regular arrangement, which were similar to that of the N group. Taken the results all together, it was confirmed that PJWE could be effective natural herbal material for the alleviation of inflammation and skin barrier damage in hairless mice skin which were induced by UVB and Sq-OOH.
The purpose of this study is to determine the changes of cytokines and immune cells in blood in castrated Hanwoo. The cytokine production and the number of immune cells were determined by collecting blood from jugular vein before castration and on 1, 7, and 28 days then after. Results of the hematological test showed that the number of leukocyte tend to increase after castration, and it significantly decreased on day 7 and day 28 (P<0.05). Lymphocytes decreased significantly on day 1 and 7 (P<0.05), and then recovered as in neutrophils on day 28. The levels of serum testosterone, TNF-a, IL-6, and anti-inflammatory IL-10 significantly decreased (P<0.05) after castration. There was also a decrease in IL-2, $IFN-{\gamma}$, and IL-4 but showed no significant difference when compared to intact ones. These results suggest that testosterone-deficiency does not affect the number of immune cells in blood, but has a close relationship with cytokine production.
