Supplement of Conjugated Linoleic Acid Increases Neutrophil Phagocytosis in Pigs (Conjugated Linoleic Acid를 급여한 돼지의 호중구 탐식능 증강)

  • To examine the in vivo immunostimulating effect of conjugated linoleic acid (CLA) in pigs, the change of peripheral blood cells and the phagocytic response of phagocytes were evaluated. Spayed male pigs, 80 kg of average body weight, fed a diet containing either 0.5% 10t-12c CLA or 0.5% CLA mixture (mostly 9c-11t CLA and 10t-12c CLA) for 4 weeks. The change of blood cell values (PCV, WBC, differential count of WBC) and the phagocytic activities of phagocytes were evaluated on week 0, 2, 4, and 5, respectively. There were no change in the PCV values regardless of CLA supplement. The number of WBC, especially neutrophils, in pigs fed a diet with CLA was significantly increased (p<0.05 to 0.01) when compared with control pigs fed a diet without CLA. The phagocytosis of peripheral blood mononuclear cell (MNC) and peripheral blood polymorphonuclear cells (PMN) were analyzed by a flow cytometry system. There was no change in the phagocytic activity of MNC and monocyte-rich cells regardless of CLA supplement. However, the phagocytic activity of PMN composed by approximately 95% neutrophils was remarkably increased (p < 0.05 to 0.01) on week 2, 4, and 5 as compared wth control pigs. These results suggested that supplement of CLA into pigs induces the increase of neutrophil number and the enhancement of neutrophil phagocytosis.

The Efficacy of Ozonated Water Therapy on Pododermatitis of Dairy Cows (젖소 족피부염(足皮膚炎)에 대한 오존수의 치료(治療) 효과(效果))

  • This study was carried out to determine therapeutic the effect of ozonated water therapy on bovine pododermatitis. In addition, bactericidal effect of ozonated water on etiological agent of bovine pododermatitis was examined. The pathohistological examination for the pododermatitis, according to application with ozonated water and ozone ointment was investigated. Thirty healthy cattle were divided two groups(each of 15) : control group(povidone group), treatment group(ozone solution group). Various parameters were evaluated in terms of the lameness score, swelling score, lesion score, WBC count, neutrophil count, pathohistological finding, and antimicrobial action. The decrease of lameness and lesion score were shown in hoof lesions on 14 days after application of ozonated water. Significant decrease of swelling was shown in hoof lesions on 14 days 1Corresponding author after application of ozonated water(p<0.01). In hematological findings, WBC count revealed values within normal range. The number of neutrophils was slightly higher than that of normal, however, this was improved on 14 days after application of ozonated water. In pathohistological findings, recovery was rapid macroscopically and microscopically in the treatment with ozonated water on the hoof lesions and ozonated water was effective. In antimicobial action, bactericidal effect was observed in treatment with ozonated water on the hoof lesions and ozonated water was effective.

Clinical Experiences with Febrile Infants, Younger than Three Months, with Urinary Tract Infection in a Single Center for Four Years : Incidence, Pathogens and Viral Co-infections

  • Purpose: The aim of this study was to analyze the incidence and microbiological characteristics of urinary tract infection in infants aged younger three months and to compare with other infection with positive urine culture. Methods: We retrospectively reviewed the medical records of 425 infants with a tympanic temperature >$37.6^{\circ}C$, aged younger than three months, who were admitted to Cheil General Hospital in Seoul, Korea, from January 2013 to December 2016. Demographic and clinical features, laboratory findings, respiratory virus PCR and the pathogens of a urine culture were analyzed. Results: A total of 88 infants (63 males, 25 females) had urinary pathogens detected in the urine culture test. The incidence of UTI in febrile infants aged younger 3 months was 11%. The most common pathogen which causes UTI was E. coli as same as in previous studies. They were divided into a UTI group (n=48) and a non-UTI group (n=40). In comparison of both group, leukocytosis, C-reactive protein level, Absolute neutrophil count level, peak temperature is statistically significant. In both group, there were co-infections with viral pathogens in some cases, and the odd ratio of non-UTI group with viral infection was 3.28. Conclusion: The study determined the incidence and pathogen of UTI in febrile infants, aged younger three months. E. coli was responsible for the majority UTI. There were some viral co-infections in febrile infants with bacteriuria and incidence was higher in non-UTI group. WBC count, ANC count and CRP level were the differentiating factors of UTI from non-UTI group.

Mycoplasma pneumoniae infection in patients with Kawasaki disease

  • Purpose: Kawasaki disease (KD) is the main cause of acquired heart disease in children. In addition to cardiovascular involvement, many complications have been recognized in KD. However, respiratory complications have been rarely reported. We investigated the differences in clinical characteristics, laboratory findings, radiography findings, and echocardiography findings of Mycoplasma pneumoniae infection and other types of pneumonia in KD patients. Methods: Among 358 patients with KD, 54 developed concurrent pneumonia. Among the 54 patients, 12 (22.2%) with high titers of anti-M. pneumoniae antibody (AMA) (>1:640) were grouped in the M. pneumoniae group and 42 were included in the control group. Serum AMA was measured in each patient. Clinical laboratory findings and total duration of fever were analyzed. Results: The duration of fever, serum hemoglobin, white blood cell count, platelet count, erythrocyte sedimentation rate, C-reactive protein level, albumin level, and the incidence of coronary arterial lesions showed no statistical difference in the 2 groups. Neutrophil count was significantly higher in the M. pneumoniae group than in the control group. Among various radiography findings observed in pneumonia, consolidation and pleural effusion were more frequent in the M. pneumoniae group than in the control group. On the other hand, parahilar peribronchial opacification, diffuse interstitial lesion, and normal findings prevailed in the control group. Conclusion: KD patients can have concurrent infections, especially pulmonary symptoms. The cause of KD is likely to be associated with M. pneumoniae infection. Thus, immediate treatment of M. pneumoniae infection in KD patients is very important.

Hematological and biochemical analysis of Korean indigenous cattle according to the ages (한우의 연령에 따른 혈액 및 혈청 화학치 분석)

  • Investigations for hematologic and biochemical values and the differential count of WBC for Korean indigenous cattle (KIC)is rarely performed. Therefore, when the index of complete blood counts (CBC) and biochemical analysis of KIC was requested, it had many difficulties to make the results for blood condition since the standard hematologic and biochemical values of KIC are lacking. In the present study, we have investigated the hematologic and biochemical values and the differential count of WBC for blood of total 252 KIC and 17 striped cattle (SC) as a control. As a result, the mean values of RBC and platelet of KIC were significantly decreased by age (P<0.05). The mean values of RBC, HCT, MCV and MCHC between KIC and SC of the same age (2 - 3 years) showed the statistical significance (P<0.05). Also, in the WBC of KIC, the mean values were decreased according to the age from $12.8{\times}10^3/{\mu}l$ under 1 year to $8.5{\times}10^3/{\mu}l$ over 5 years. In the differential count of WBC of KIC, it showed generally the rates of 45% lymphocyte and 35% segmented neutrophil. In the biochemical analysis, the mean values of BUN and globulin were significantly increased according to the age (P<0.05). Additionally, in comparative analysis between pregnant and non-pregnant group of KIC, the mean values of Hg and HCT in pregnant group were significantly decreased (P<0.05). In conclusion, data obtained from this study may be valuable as a standard for interpretation of the results in hematologic and biochemical analysis of KIC.

Predictive value of C-reactive protein for the diagnosis of meningitis in febrile infants under 3 months of age in the emergency department

  • Background: Fever is a common cause of pediatric consultation in the emergency department. However, identifying the source of infection in many febrile infants is challenging because of insufficient presentation of signs and symptoms. Meningitis is a critical cause of fever in infants, and its diagnosis is confirmed invasively by lumbar puncture. This study aimed to evaluate potential laboratory markers for meningitis in febrile infants. Methods: We retrospectively analyzed infants aged <3 months who visited the emergency department of our hospital between May 2012 and May 2017 because of fever of unknown etiology. Clinical information and laboratory data were evaluated. Receiver operating characteristic (ROC) curves were constructed. Results: In total, 145 febrile infants aged <3 months who underwent lumbar punctures were evaluated retrospectively. The mean C-reactive protein (CRP) level was significantly higher in the meningitis group than in the non-meningitis group, whereas the mean white blood cell count or absolute neutrophil count (ANC) did not significantly differ between groups. The area under the ROC curve (AUC) for CRP was 0.779 (95% confidence interval [CI], 0.701-0.858). The AUC for the leukocyte count was 0.455 (95% CI, 0.360-0.550) and that for ANC was 0.453 (95% CI, 0.359-0.547). The CRP cut-off value of 10 mg/L was optimal for identifying possible meningitis. Conclusion: CRP has an intrinsic predictive value for meningitis in febrile infants aged <3 months. Despite its invasiveness, a lumbar puncture may be recommended to diagnose meningitis in young, febrile infants with a CRP level >10 mg/L.

  • Complete blood cell count(CBC) is a technique that counts leukocytes for each type of blood cell being analyzed. The principle is that laser is incident to ex vivo flowing leukocytes in a microcapillary tube and scattered light occurs by laser and leukocytes. By collecting the scattered light, we can count the types of cells because different cells generate different light-scattering patterns. However, the technique has an intrinsic limitation, scattering pattern is shown in a wide range region in the resulting, which makes it difficult to accurate analyze and use fluorescent dyes. To overcome this limitation, a new design of CBC with a dual laser, which irradiates with orthogonal angles for collecting quad-scattering information was proposed. Before development, the scattering difference depending on wavelength must be investigated to only catch up to the scattered signal by angles. Some studies, which focused on simple particles, have been conducted to theoretically and experimentally investigate different scatterings by wavelength. In this study, we propose an optical system for measuring scattered light and investigate a complex particle. As a result, the green laser made strong scattering signals in both the forward and side direction: 10% and 30%, respectively.

Differences in Clinical Laboratory Data between the Elderly and the Young Adults (노인군과 청장년군 간의 정상 검사치의 비교 분석)

  • Due to the lowering of biological functions resulted from old age, the elderly is known to have many different clinical laboratory data compared with the young adults. But, in korea, such study is lacking. This research is to find the differences between the elderly and the young adults, and also to know the sexual differences, by comparing the outcomes of the clinical. laboratory data. Along with that, it is to help clinical usage of the data in the future. The age of the elderly was between 60 and 83(average age 63.8), and that of the young. controls was between 20 and 35. In both sexes, MCV, MCH, ESR, CRP, AST, ALT, ${\gamma}$GTP, ALP, BUN, total cholesterol were significantly higher in the elderly than in the controls. And lymphocyte count(%), total bilirubin, direct bilirubin, total protein, albumin, $T_3$ were significantly lower in the elderly than in the controls(P<0.05). Hemoglobin, Hct, platelet count, $T_4$ were significantly lower only in the male elderly, and eosinophil count(%), creatinine were significantly higher only in the female elderly(P<0.05). HDL cholesterol was significantly higher only in the male elderly(P<0.01). There were no significant difference between two groups regarding WBC count, segment neutrophil count(%), monocyte count(%), TSH. Many clinical laboratory data are different between the elderly and the young adults, and some clinical laboratory data also have sexual differences.

Field Case of Babesiasis in Korean Native Cattle (한우(韓牛)에서의 Babesia병(病)의 자연발생례(自然發生例)에 대하여)

  • The authors encountered a case of Babesiasis in Korean native cative cattle at Gyeongbug prefecture. After the clinicohematological observation the patient was treated a single dose of ganaseg and the following results were obtained. 1. Initial symptoms observed were high fever, anorexia, depression and cessation of rumination and these were followed by marked hemoglobinuria and mild degree of icterus. 2. The erythrocyte count decreased to $3.6{\times}10^6/mm^3$ and the infected erythrocyte rates were 9.7 percent with Babesia sp. and 0.2 percent with Theileria sp. 3. Marked increase in band neutrophil was observed. 4. The morphology of the protozoa in erythrocytes was different from that of so-called large-type Piroplasma: various forms including paired pear-shaped, single round, oval and pear-orspindle-shaped were detected, of which the former two forms were dominant. 5. A single dose of ganaseg administered intramuscularly was highly effective for the treatment of Babesiasis.

The Effect of Erythropoietin on Ischemia-Reperfusion Injury: An Experimental Study in Rat TRAM Flap Model (백서 복직근판의 허혈-재관류 손상에 대한 Erythropoietin의 영향)

  • Purpose: Erythropoietin is traditionally known to regulate erythropoiesis, but recently its protective effect against ischemia-reperfusion injury has been studied mainly in cardiovascular and neuronal systems. This study was planned to investigate the effects of recombinant human erythropoietin on ischemia-reperfusion injury in rat TRAM flap model. Methods: Superiorly based TRAM flap was elevated and ischemic insult was given for four hours. Thirty minutes before reperfusion, single dose recombinant human Erythropoietin(5000IU/kg) was injected via intraperitoneal route in the treatment group. At 24 hours postoperatively, systemic neutrophil count, tissue myeloperoxidase activity, malonyldialdehyde amount, nitric oxide content, tissue water content and histologic finding of inflammation was evaluated. On 10 days postoperatively, flap survival rate, angiogenesis and change in hematocrit level was evaluated. Results: Tissue nitric oxide level was significantly higher and myeloperoxidase activity was significantly lower in the treatment group 24 hours after reperfusion. Tissue water content was significantly lower in the treatment group. Perivascular neutrophil infiltration and intravascular adhesion was marked in the control group. Mean flap survival after ten days was 69% in the treatment group, and 47% in the control group, demonstrating a significant difference. Neovascularization in the treatment group also outnumbered the control group. No significant hematocrit rise was noted ten days after erythropoietin administration. Conclusion: Recombinant human Erythropoietin improved flap survival in ischemia-reperfusion injured rat TRAM flaps, at least partially owing to suppressed inflammation, increased nitric oxide, and enhanced angiogenesis.
