Inhibition Effect of Zizania latifolia on Apoptosis Induced by $H_2O_2$ in Neuro2A Cell ($H_2O_2$로 유발된 Neuro2A 신경세포고사에 대한 줄풀의 억제 효과)

  • The purpose of this study was to examine the inhibition effect of Zizania latifolia that has been used heart disease, Diabetes Mellitus and Skin disease for a long time on apoptosis induced by $H_2O_2$ in Neuro2A cell. Neuro2A cells were cultivated in RPMI(GibcoBRL) with $5\%$, FBS and treated with $H_2O_2$, and Zizania latifolia. We measured the cell viability and analyzed DNA fragmentation. Activity of PARP, Cytochrome C, caspase-9, caspase-3, p53, p21, Bax and Bcl-2 in the cell was examined by using western blot. The cell viability in Zizania latifolia treatment (60ug/ml<) decreased significantly compared with that of none treatment. (p<0.001) Zizania latifolia increased cell viability about twice as much as that being injury by $H_2O_2$. (Zizania Latifolia 20ug/ml, $H_2O_2$ 200uM, P<0.001) DNA fragmentation developed by $H_2O_2$, but was not developed in Zizania latifolia treatment. PARP, Cytochrome C, caspase-9 and caspase-3 activated all by $H_2O_2$ but were not activated in Zizania latifolia treatment.. P53, P2l and Bu activated by $H_2O_2$, and Bcl-2 got into inactivation. But the opposite results appeared in Zizania latifolia treatment. In conclusion, these results suggest that Zizania latifolia inhibit the development of DNA fragmentation and apoptosis by $H_2O_2$ and the antioxidant action of Zizania latifolia is effective. More researches about effect of Zizania latifolia are considered to need.

Protective Effects of Dodam Water Extract (Dodam) Against Rotenone-Induced Neurotoxicity in Neuro-2A Cells

  • Dodam formula (Dodam) has been used for neurodegenerative disease in Oriental medicine. Dodam is capable of protecting diverse kinds of cells from damage caused by a variety of toxic stimuli. In the present study, we investigated the underlying protective mechanism of Dodam on rotenone-induced cytotoxicity in rat neuroblastoma Neuro-2A cells. Treatment with Neuro-2A cells with rotenone caused the loss of cell viability, and condensation and fragmentation of nuclei, which was associated with the elevation of ROS level, and lipid peroxidation, the increase in Bax/Bcl-2 ratio. Rotenone induced mitochondrial dysfunction characterized by mitochondrial membrane potential loss and cytochrome-c release. These phenotypes induced by rotenone were reversed by pretreatment with Dodam. Our results suggested that major features of rotenone-induced neurotoxicity are partially mediated by mitochondrial dysfunction and oxidative stress, and that Dodam markedly protects Neuro-2A cells from oxidative injury. These data indicated that Dodam might provide a useful therapeutic strategy in treatment of the neurodegenerative diseases caused by oxidative injuries.

The Protective Effects of Seokchangpowonji-san on $H_2O_2$-Mediated Cell Death of Neuro 2A as an Alzheimer Model System (산화적 손상에 의한 Neuro 2A 치매모델에서 석창포원지산의 방어효과)

  • Objective : The water extract of Seokchangpowonji-san (SWS) has traditionally been used for treatment of dementia in oriental medicine. However, little is known about the mechanism by which the water extract of SWS rescues cells from neurodegenerative disease such as Alzheimer's disease. Methods & Results: This study was designed to investigate the protective mechanisms of SWS on $\beta-amyloid$ or $H_2O_2$-induced$ cytotoxicity in neuro 2A cells. $H_2O_2$ markedly decreased the viability of neuro 2A cells, which was characterized by apparent apoptotic features such as membrane blebbing as well as fragmentation of genomic DNA and nuclei. However, the water extract of SWS significantly reduced $H_2O_2-induced$ cell death and apoptotic characteristics through reduction of intracellular peroxide generation. Also, the. extract prevented the mitochondrial dysfunction including the disruption of mitochondria membrane permeability transition (MPT) and the modulation in expression of Bcl-2 family proteins in $H_2O_2­treated$ neuro 2A cells. Furthermore, pretreatment with SWS inhibited the activation of caspase-3, and in turn, degradation of ICAD/DFF45 was completely abolished in $H_2O_2-treated$ cells. Conclusion: Taken together, the data suggest that the protective effects of the water extract of SWS against $\beta-amyloid$ induced oxidative injuries may be achieved through modulation of mitochondrial dysfunction.

The Protective Effects of Chilgi-tang on Oxidative Stress by Glucose Deprivation in Neuro 2A Cells (Glucose deprivation으로 유발된 Neuro 2A 세포의 산화적 손상에 대한 칠기탕(七氣湯)의 보호효과)

  • Objective: The water extract of Chilgi-tang (CGT) has been traditionally used in treatment of heart diseases caused by stress in Oriental Medicine. However, little is known about the mechanism by which CGT rescues neuronal cells from injury damage. Therefore, this study was designed to evaluate the protective effects of CGT on Neuro-2A cells by glucose deprivation-induced cell death. Methods: We investigated how cell death induced by glucose deprivation was associated with increased reactive oxygen species (ROS) generation. Result: The CGT treatment prior to glucose deprivation insult significantly reduced the number of cell deaths and the glucose deprivation-induced increase in ROS. Nitric oxide (NO) was also attenuated by CGT treatment. In addition, we demonstrated that the anti-cell death effect of CGT was blocked by heme oxygenase-1 (HO-1) activation. Finally, pretreatment of cells with a hemin, HO-1 inducer, reduced glucose deprivation-induced cell death. In contrast, pretreatment of cells with a ZnPP, HO-1 activity inhibitor, attenuated CGT-induced inhibition of cell death. Conclusions: These findings indicate that ROS plays an important role in glucose deprivation-induced cell death and that CGT may prevent glucose deprivation-induced cell death by inhibiting the ROS generation through HO-1 activation in Neuro-2A cells.

Adaptive Fuzzy-Neuro Controller for High Performance of Induction Motor (유도전동기의 고성능 제어를 위한 적응 퍼지-뉴로 제어기)

  • This paper is proposed adaptive fuzzy-neuro controller for high performance of induction motor drive. The design of this algorithm based on fuzzy-neural network controller that is implemented using fuzzy control and neural network. This controller uses fuzzy rule as training patterns of a neural network. Also, this controller uses the back-propagation method to adjust the weights between the neurons of neural network in order to minimize the error between the command output and actual output. A model reference adaptive scheme is proposed in which the adaptation mechanism is executed by fuzzy logic based on the error and change of nor measured between the motor speed and output of a reference model. The control performance of the adaptive fuzy-neuro controller is evaluated by analysis for various operating conditions. The results of experiment prove that the proposed control system has strong high performance and robustness to parameter variation, and steady-state accuracy and transient response.

  • This paper presents the optimal fraction of validation set to obtain a prediction accuracy of software failure count or failure time in the future by a neuro-fuzzy system. Given a fixed amount of training data, the most popular effective approach to avoiding underfitting and overfitting is early stopping, and hence getting optimal generalization. But there is unresolved practical issues : How many data do you assign to the training and validation set\ulcorner Rules of thumb abound, the solution is acquired by trial-and-error and we spend long time in this method. For the sake of optimal fraction of validation set, the variant specific fraction for the validation set be provided. It shows that minimal fraction of the validation data set is sufficient to achieve good next-step prediction. This result can be considered as a practical guideline in a prediction of software reliability by neuro-fuzzy system.

Adaptive Feedrate Neuro-Control for High Precision and High Speed Machining (고정밀 고속가공을 위한 신경망 이송속도 적응제어)

  • Finding a technique to achieve high machining precision and high productivity is an important issue for CNC machining. One of the solutions to meet better performance of machining is feedrate control. In this paper we present an adaptive feedrate neuro-control method for high precision and high speed machining. The adaptive neuro-control architecture consists of a neural network identifier(NNI) and an iterative learning control algorithm with inversion of the NNI. The NNI is an identifier for the nonlinear characteristics of feedrate and contour error, which is utilized in iterative learning for adaptive feedrate control with specified contour error tolerance. The proposed neuro-control method has been successfully evaluated for machining circular, corner and involute contours by computer simulations.

Effect of the neuroprotetion and anti-Alzheimer's disease in CT99-induced Neuro 2A cells by Ikgiansintang water extract (CT99 발현 신경 세포주에서 익기안신탕(益氣安神湯)의 신경보호 및 항치매 효과)

  • Alzheimer's disease(AD) is a geriatric dementia that is widespread in old age. In the near future AD will be the biggest problem in public health service. It has been widely believed that $A{\beta}$ peptide devided from APP causes apoptotic neurotoxicity in AD brain. However, recent evidence suggests that n99 may be an important factor causing neurotoxicity in AD. Mouse Neuro 2A cells expressed with CT99 exhibited remarkable apoptotic cell damage. We invesgated the protective effects of Ikgiansintang water extract(IGA). Findings from our experiment have shown that IGA inhibits the activities of CT99, which has neurotoxicities and apoptotic activities in cell line. In addition treatment of IGA($50{\mu}g/ml$ for 24 hours) partially prevented CT99-induced cytotoxicity in Neuro 2A cells. As the result of this study, In IGA group, the apoptosis in the nervous system is inhibited, the repair against the degerneration of Neuro 2A cells by n99 expression is promoted. Base on these findings, IGA may be beneficial for the treatment of AD.

  • In this paper, a level controller is designed with the neuro-fuzzy model based on Takagi-Sugeno fuzzy system. The fuzzy system is employed as the controller, which can be tuned by the neural network mechanism based on a gradient descent technique. The tuning mechanism will provide an optimal process input by forcing the process error to zero. The proposed controller provides the online tunable mode to adjust the consequent membership function parameters. The controller is implemented with M-file and graphic user interface (GUI) of Matlab program. The program uses MPIBM3 interface card to connect with the industrial processes In the experimentation, the proposed method is tested to vary of the process parameters, set points and load disturbance. Processes of one tank and two tanks are used to evaluate the efficiency of our controller. The results of the both processes are compared with two PID systems that are 3G25A-PIDO1-E and E5AK of OMRON. From the comparison results, our controller performance can be archived in the case of more robustness than the two PID systems.
