한국 재래산양에 대한 L. monocytogenes균의 감염원을 조사하기 위하여 총 459개의 재래산양 분변, 비즙, 장기, 사료 및 토양재료로부터 균분리를 시도하였으며, 분리균의 항균성 물질에 대한 감수성 및 인공 감염시의 L. monocytogenes의 침입장기를 조사하였던 결과는 다음과 같다. L. monocytogenes는 분변에서만 분리되었으며 (분리율 8.2%) 분변이 본균의 주요 감염원으로 인정되었다. 가검재료로부터 L. monocytogenes의 분리방법으로는 유제화 재료를 혈액배지로써 37에서 24~48시간 동안 호기배양함이 우수하였다. 분리된 L. monocytogenes 8주 중 100%의 균주가 amikacin, colistin, tanamycin, neomycin에 내성을 나타내었으며, clinmycin, gentamicin, lincomycin, methicillin 및 refampin에 87.5%, ampicillin및 oles-ndomycin에 75.0%, Chloramphenico에 62.5%, tetracycline에 50.0% 및 cephalot hin에 37.5%의 균주가 내성이었다. Nitrofurantoin에는 전 균주가 감수성을 나타내었다. L. monocytogenes를 재래산양에 경구 및 정맥내로 접종하였을때 패혈증이 인정되었으며 대부분의 실질장기가 이 균의 침입부위인 것으로 관찰되었다.
The present study was conducted to investigate the incidence of Pasteurella multocida infection in cheju swine from March 1994 to December 1994 isolated organisms were identified by the biochemical properties, cellulose serological type and antibiotic susceptibilities. Pasteurella multocida was isolated from the Lungs of 96 pigs with pneumonia(51 %) among 188 slaughtered pigs. The majority of p multocida isolates were identical to those of the standard strains. On the classification of the capsular type of the isolated p multocida it consist of the 88 isolates of type A(91.6%) 2 isolates of type D (2%) and un classified 6 types(6.2%). The majority of the 96 isolates of p multocida highly susceptible to the antibiotics including ampicillin(Am), cephalotin(Ce), erythromycin(Em), gentamycin(Gm), kanamycin(Km), lincomycin(Lm), neomycin(Nm), penicillin(Pc), streptomycin(Sm), solfametoxazol/trimethoprim(Sxt) and tetracycline(Te)
Capacitated goat spermatozoa generated diacyl glycerol (DAG) when suspended in Krebs-Ringer bicarbonate medium and induced by progesterone or $Ca^{2+}$ ionophore A23187. We have added Sn-1-oleoyl-2-acetyl glycerol externally, to study the effect of DAG in goat sperm acrosomal exocytosis. Addition of neomycin abolished the DAG generating capacity of progesterone in a dose dependent manner, suggesting the involvement of a phosphoinositidase C activated phospholipase C system in the process. The level of increase in phosphatidic acid was considerably low and was produced well after the DAG generation thereby suggesting the involvement of a DAG kinase which phosphorylates DAG to produce PA. The inhibition of progesterone mediated effect by inhibitors of $GABA_A/Cl^{-}$ channel and $Ca^{2+}$ channels further supports the evidence that the events of binding of agonist to the receptor(s), opening of $Ca^{2+}$ channels and the activation of phospholipase C are reconciled to perform the function of acrosome reaction in capacitated goat spermatozoa.
Marker proteins of genetically-modified organisms (GMO) and their antibodies were prepared and characterized as major components of an analytical system. We selected two GMO markers, neomycin phosphotransferase II and 5- enolpyruvylshikimate-3-phosphate synthase, and produced them from E. coli employing genetic recombination technology. After purification, their structural conformation and binding affinities to the respective antibodies were characterized. The results showed that the recombinant proteins were identical with commercially obtained reference proteins. We further used them as immunogens to raise polyclonal antibodies capable of discriminating GMO containing protein from non-GMO. Well-characterized marker proteins and antibodies will be valuable as immunoreagents in constructing analytical systems such as biosensors and biochips to measure quantities of GMO.
Isolates of 70 Proteus mirabilis were tested against 10 different antibiotics by a disc diffusion method as recommended by National Committee for Clinical Laboratory Standards (NCCLS). The isolates were resistant in order of tetracycline(100.0%), enrofloxacin(95.7%), ampicillin(74.3%), choramphenicol(62.9%), cephalothin(58.6%), streptomycin(50%), kanamycin(47.2%), neomycin(35.8%), gentamicin(15.7%), and amikacin(2.9%). In the antibiotic resistant patterns, 37 kinds of multiple resistance patterns of P mirabilis isolates were detected. The highest resistant pattern was ampicillin-cephalothin-chloramphenicol-enrofloxacin-tetracycline(11.6%).
Two hundred and twenty-seven strains of Escherichia coli isolated from 25 hens (12 hens received tetracycline neomycin and sulfadimethoxine, and 13 hens not received antibiotics) were studied for the drug resistance and distribution of R factors. About 74 per cent of E. coli strains isolated from hens of a herd received antibiotics were resistance to tetracycline (TC) streptomycin (SM), chloramphenicol (CM), kanamycin (KM), ampicillin (AP) and sulfisomidine (Su), alone or in combination thereof, but only a hen among a herd not received antibiotics excreted E. coli resistant to TC and SM. Among resistant strains, about 7% were found to be resistant to TC and SM, whereas 93% were resistant to three or more antibiotics. The most common pattern was the quadruple resistant to SM, TC, KM and Su (28.7%), and followed by triple ones to SM, TC and Su (25.3%), and SM, TC and KM (24.7%). About 84% of resistant strains carried R factors which were transferable to the recipient by conjugation.
Streptomyces spp. purchased from American Type Culture Collection and Institute for Fermentation in Osaka, and donated from Northem Regional Research Laboratory, and those isolated from soil samples were assayed to isolate many plasmids harboring streptomycetes. Among these qrganisms, 5 small size-plasmid carrying organisms SNUS 8810-597A, 8810-600, 8810-754, 8811-344, and 8811-347 were characterized and their plasmids pSJ597, pSJ600, pSJ754, pSJ344, and pSJ347 were isolated in a large scale. The plasmid harboring organisms were sensitive to neomycin, kanamycin, gentamicin, and thiostreptone, but some of them showed weak or strong resistance against streptomycin, chloramphenicol, ampicillin, and tetracycline. It was confirmed that pSJ597 and pSJ600 do not carry antibiotic biosynthetic genes. pSJ600 showed a pock-forming character.
Ten isolates of Verotoxin-producing Escherichia coli (VTEC) were detected in slaughtered cattle and investigated their phenotypic characteristics and antimicrobial susceptibility. None of the isolates was positive for eae gene. Only one isolate was positive for uidA gene. Eight out of ten isolates of VTEC were originated from broker's cattle. Thus microbiological monitoring for broker farms should be performed to minimize VTEC contamination. In the antimicrobial susceptibility test, all the isolates were highly resistant to bacitracin and lincomycin whilst they are susceptible to apramycin and neomycin.
Bradyrhizobium japonicum RJB6$str^rnal^r$에 neomycin resistant gene인 Tn5을 삽입한 표지균주를 사용하여 Pseudomonas fluorescens(PS-3)와 Bacillus subtilis(BCAC-4)를 대두에 혼합접종하고, 또한 항생제 처리후 표지균주를 접종한 대두의 생장 및 근류형성율을 조사 분석한 결과를 요약하면 다음과 같다. 1. 5년이상 대두 재배토양에 표지균주인 RJB6 $str^rnal^rneo^r$을 Ps-3와 혼합접종했을 때 접종근류균의 근류형성율은 8.5%, BCAC-4와 혼합접종한 경우는 4.3%였으나, 표지균주, Ps-3라 BCAC-4의 3 균주를 혼합접종했을 때는 2.9% 에 지나지 않았다. 2. 대두를 재배하지 않은 토양에 표지균주을 Ps-3와 혼합접종했을 때 접종근류균의 근류형성은 10.3%, BCAC-4와 혼합접종한 경우은 6%였으며, 표지균주와 Ps-3, BCAC-4의 3 균주를 혼합접종 했을 때 5.0%의 근류형성율을 나타냈다. 3. 혼합접종이 대두 생육상에 미치는 영향은 일반적으로 표지균주와 Ps-3의 혼합접종 처리구에서 가장 양호했다. 4. 항생제를 처리했을 때 5년 이상 대두재배 토양에서 접종근류균의 근류형성은 12.2-25.4% 였으며, 대두를 재배하지 않은 토양에서는 23.7-43.7% 였으나, 항생제 1,000ppm 처리구 에서 43.2%로 가장 높은 근류형성율을 보였다.
This experiment was conducted on 60 Hanwoo calves comprising five feed additive groups, with 12 calves in each group, to determine the effects of additives at pre- and post-weaning on growth performance and blood profile. The groups were control, antibiotic (Neomycin 110 ppm), illite (2%), fermented green tea probiotics (FGTP, 0.5%), and mixed additives (FGTP 0.25%, illite 1% and licorice 0.1%). The calves were offered experimental pellet feeds ad libitum and after one month were supplied with imported timothy hay. They moved freely within the group and suckled their mother' milk during the pre-weaning stage (birth to 3 months) and were separated from their dam during the post-weaning stage (4-5 months). During the pre-weaning stage, the highest average daily gain (ADG) was recorded in the antibiotic- and mixed additive-fed groups followed by FGTP, control and illite groups. In the post-weaning stage, significantly higher total weight gain and ADG were recorded in both the FGTP and mixed additive groups compared to the other groups (p<0.05). Feed efficiency of mixed additive- and illite-fed calves were almost similar with antibiotic-fed calves compared to the other two groups, but the ADG was lowest in illite-fed calves during the pre-weaning stage. In contrast, post-weaning calves fed FGTP and mixed additives showed better feed efficiency. The values of hematological indices, differential leukocyte count, blood proteins and immunoglobulin among the additive-fed calves were not significantly different (p>0.05), although hemoglobin and hematocrit values were lower in FGTP compared to control, but similar in mixed additive and antibiotic groups. These results indicate no detrimental effects of feed additives on the blood profile of calves at both pre- and post-weaning age. Serum albumin in post-weaning calves of all feed additive groups were similar but significantly lower (p<0.05) than in the control group. Post-weaning, IgM was significantly lower (p<0.05) in illite-fed calves compared to other treatment groups, but there was no difference at pre-weaning. Considering all factors, the mixed feed additives and FGTP can be the replacement feed formula for antibiotic for Hanwoo beef calf production, especially when used post- weaning.
