Occurrence of the Root-Knot Nematode species in Vegetable Crops in Souss Region of Morocco

  • Root-knot nematodes 'Meloidogyne spp' are the most destructive group of plant parasitic nematodes causeing serious losses in vegetables crops and this damages worsened when crops grown under greenhouses conditions. In this sutdy, the distribution and characterization of root-knot nematode species collected from the Souss region of Morocco where vegetables crops intensively cultivated were determined by using both morphological and molecular tools. Out of the 110 samples collected from different greenhouses 91 (81.7%) were found to be infested with root-knot nematodes. Thirty-seven populations of root-knot nematodes were morphologically identified based on perineal patterns as well as molecularlly using species-specific primers. The obtained results indicated that Meloidogyne javanica and M. incognita were identified in 86.4% and 13.5% of the total populations, respectively. The lowest incidence of root-knot nematodes (64%) was found in Toussous, whereas the highest frequencies of 100% and 90% were detected in Taddart and Biogra, respectively. As the majority of the samples have been infested with Meloidogyne species; this indicates that there is an urgent need to provide farmers with a proper control strategy.

Biological Control of Root-Lesion Nematodes(Pratylenchus spp.) by Nematode-Trapping Fungi (선충 포식성 곰팡이를 이용한 뿌리썩이선충(Pratylenchus spp.)의 생물학적 방제)

  • For the biological control of the root-lesion nematodes, Pratylenchus spp., which damage directly and indirectly to the leaf perilla, the nematical effect of three nematode-trapping fungi, Arthrobotrys oligospora, A. conoides and A. dactyloides was evaluated in the field. Three species of Arthrobotrys were isolated from the culture soil of leaf perilla in 1998 and were observed the capture of the root-lesion nematodes, Pratylenchus spp. by adhesive hyphal networks or constricting rings on agar. At 40 days after treatment, the plant-parasitic nematodes and root-lesion nematode populations were approximately increased 3.5 fold in untreated control plot, while the nematode population in fungi treatment plots was similar to initial population. In the A. dactyloides plot, however, the population of plant-parasitic nematodes and Pratylenchus spp. was approximately reduced 65% and 53%, respectively. Thus, the fungus A. dachyloides should provide as biological agent for the control of Pratylenchus spp.

The major plant-parasitic nematodes in plastic vinyl house field (하우스시설 재배지에서 발생하는 주요 선충)

  • To know how much damages occurred by nematodes in plastic vinyl house field, soil samples were collected from strawberry cultivation areas at Buyeo and Nonsan in Chungnam and Jinju in Kyeongnam, melon cultivation area at Gocksung in Junnam and cucumber field at Gongju in Chungnam. And then, nematode samples were separated from each soil sample, and identified the kind of plant-parasitic nematodes. Plant-parasitic nematodes were separated from 52 soil samples. Among samples, Meloidogyne spp., Pratylenchus spp. and Helicotylenchus spp. were isolated from 45 (86.5%%) and 33 (63.5%) and 47 collected soil samples (90.4%), respectively. As a result of identification of plant-parasitic nematodes from regional collected soil samples, distribution of Helicotylenchus spp. was higher than any other plant-parasitic nematode. And the population of Meloidogyne spp. and Pratylenchu spp. were also higher, and a occurrence ratio of Meloidogyne spp. is higher than Pratylenchu spp. except the cucumber growing area at Gongju.

  • The entomopathogenic nematodes, Steinernema carpocapsae and Heterorhabditis bacteriophoTa, were laboratory tested for the control of rice stem borer Chilo suppressalis. When 8 cm long of two rice stems infected by rice stem borers were treated with S. carpocapsae by spray application to the concentrations of 250, 500, 1,000, or 2,000 nematodes/ml, 68.6 $\pm$ 10.0-94.9 $\pm$ 2.6 % control was obtained. When 8 cm long of rice stems infected by rice stem borers were treated with H. bacteriophoTa by spray application to the concentrations of 100, 200, 400, or 800 nematodes/ ml, 91.4 $\pm$ 0.7-100 % control was obtained. On the other hand, when 8 cm long of three rice stems were treated with H. bacteriophora by dipping application to the concentrations of 100 or 200 nematodes/ml, 46.2 $\pm$ 4.7-63.1 $\pm$ 4.7 % control was obtained. Because the moist habitat of rice stems were favorable to nematode survival and searching abilities, entomopathogenic nematode, were confirmed to be a potential biological control agents against rice stem borers.

Comparisons of Pathological Responses in Carrot to Root-knot Nematodes

  • Carrot (Dacus carota var. sativus) is one of the top-ten most economically important vegetable crops produced worldwide, and the root-knot nematodes, Meloidogyne spp., are one of the most important pests in the carrot. In Korea, M. hapla and M. incognita are presumed to be the major root-knot nematodes distributing mostly in open carrot fields and greenhouses, respectively. In our study, currently-developed and commercial carrot cultivars and the parental lines were examined for their pathological responses to M. incognita and M. hapla 7 weeks after inoculation with about 1,000 second-stage juveniles (J2) of the nematodes. All the carrot cultivars and lines showed susceptible responses to both nematodes with the gall index (GI) of 2.4-4.4, which were always higher on the carrot plants infected with M. incognita than M. hapla. Gall sizes were remarkably larger with more serious reduction of the root growths in the plants infected with M. incognita than M. hapla, suggesting the carrot lines examined in our study were more susceptible to the former than the latter. In the infection sites of the root tissues, giant cells were more extensively formed, occupying larger stellar regions with the prominent destruction of adjacent xylem vessels by M. incognita than M. hapla. All of these results suggest M. incognita affect more seriously on the carrot plants that are grown in greenhouses, compared to M. hapla that has a major distribution in open carrot fields, which would be used for determining cropping systems based on target nematode species, their damage and pathological characteristics.

Indigenous Fungivorous Nematodes Affect the Biocontrol Efficacy of Trichoderma harzianum through Reducing the Hyphal Density

  • Indigenous fungus-feeding nematodes may adversely affect the growth and activity of introduced biocontrol fungi. Alginate pellets of the biocontrol fungus Trichoderma harzianum ThzID1-M3 and sclerotia of the fungal plant pathogen Sclerotinia sclerotiorum were added to nonsterile soil at a soil water potential of -50 or -1,000 kPa. The biomass of ThzID1-M3, nematode populations, and extent of colonization of sclerotia by ThzID1-M3 were monitored over time. The presence of ThzID1-M3 increased the nematode population under both moisture regimes (p < 0.05), and fungivores comprised 69-75% of the nematode population. By day 5, the biomass of ThzID1-M3b and its colonization of sclerotia increased and were strongly correlated (R2 = 0.98), followed by a rapid reduction, under both regimes. At -50 kPa (the wetter of the two environments), fungal biomass and colonization by ThzID1-M3 were less, in the period from 5 to 20 days, while fungivores were more abundant. These results indicate that ThzID1-M3 stimulated the population growth of fungivorous nematodes, which in turn, reduced the biocontrol ability of the fungus to mycoparasitize sclerotia. However, colonization incidence reached 100% by day 5 and remained so for the experimental period under both regimes, although hyphal fragments disappeared by day 20. Our results suggest that indigenous fungivores are an important constraint for the biocontrol activity of introduced fungi, and sclerotia can provide spatial refuge for biocontrol fungi from the feeding activity of fungivorous nematodes.

  • The fatty acid compositions of entomopathogenic nematodes Steinernema carpocapsae strain produced in vitro and in vivo were examined. Nematodes cultured both in vitro and in vivo revealed similar fatty acids compositions with respect to 16, 18, 20 carbons. However, the contents of lipids were varied by culture methods. Furthermore, it was distinctive that nematodes cultured in vitro contained fatty acids with 19 carbon. In the case of symbiotic bacterium Xenorhabdus nematophilus isolated from Steinernema carpocapsae, the major lipid component was palmitic (c16:0) fatty acids.

  • These experiments were conducted to investigate the nematicidal effects of the insecticides, Mocap(O-Ethyl-S, S-dipropyl phosphorodithioate), Carbofuran(2,3-Dihydro-2,2-dimethyl benzofuranyl ethyl carbamate), Terbufos (S-tert-buthylthio methyl O,O-diethyl phosphordithioate) and their mixtures (Mocap+carbofuran, Mocap+Terbufos, Carbofuran+Terbufos) on growth of 2year-old ginseng, Panax ginseng C.A. Meyer, and the control of root-knot nematodes. There was no evidence of plant injury from insecticide treatment of ginseng, although the rate of emergence of the treated ginseng was slightly inhibited. The insecticide treatments showed no of-flavor of ginseng plant. Terbufos and Mocap provided heifer confrol of the root-knot nematodes than carbofuran alone and their mixtures. Mixtures of the insecticides showed antagonisitic effect to the root-knot nematodes.

The Community Structure and Spatial Distribution of Meiobenthos in the Kanghwa Tidal Flat, West Coast of Korea

  • The community structure of meiobenthos was studied in tidal flats of Kanghwado in November, 1997. Nematodes were the most dominant group among 18 total meiofaunal groups at most stations except for at lower tidal flats where the most abundant groups were harpacticoid copepods. Meiobenthos were the most abundant in the upper 1cm and their density decreased depending on the depth from the surface. The highest density of the meiobenthos was 6,094 inds. $10cm^{-2}$ at the upper tidal flat, while the lowest was 1,524 ind. $10cm^{-2}$ at the lower tidal flat. The density was higher in general at the upper tidal flats, but decreased at the stations toward lower tidal flat at all transect lines. At all transect lines, nematodes decreased as stations were along toward lower tidal flat whereas harpacticoids increased. The values of N/C(nematodes/benthic harpacticoids) ratio were higher at the upper tidal flats than the lower tidal flats at all transect lines.

  • Steinemematid and heterorhabditid nematodes were not effective to control the piU bug, Armadillidium vulgare although these nematodes were able to infect pill bugs. Steinernema carpocapsae Pocheon strain and S. glaseri Dongrae strain were more effective than S. carpocapsae AU strain or Heterorhabditis bacteriophora. Nematode concentration was more important factor than host density to develop infectivity.
