우절의 페놀성 화합물의 분리 및 동정 (Phenolic Compounds from the Node of Lotus Rhizome (Nelumbo nucifera Gaertn))

  • The node of lotus rhizome (Nelumbo nucifera, Nymphaeaceae) have been used as a traditional medicine for the remedy of hemorrhage, blood stagnancy and thirstiness. To investigate phenolic compound from the node of Nelumbo nucifera, phytochemical isolation and structure elucidation were conducted. Four phenolic compounds were isolated from aqueous methanolic extract and the structure of these compounds were identised as (+)-catechin (1), (+)-gallocatechin (2), (+)-gallocatechin (4u-8)-catechin (3) and scolpoletin (4) respectively by the analysis of spectroscopic evidences and comparisions with the data of authentic samples.

Formulation and Sensory Evaluation of Granule and Tablet from Lotus Rhizome(Nelumbo nucifera Rhizome)

  • Nelumbo nucifera root(NNR) is used to clear summerheat(暑熱), bear Yang(陽) upwards and stop bleeding as mentioned in traditional Korean medicine. Also, it has been known that NNR is effective for lowering blood pressure and hyperlipidemia. The rhizome is considered to be nutritive, demulcent, diuretic and cholagogue and is used to treat piles, dyspepsia and diarrhea. An increasingly growing market for nutraceuticals and functional foods has triggered the study on natural sources for nutraceuticals, health foods and functional foods. But rhizome was inconvenient to formulate liquid dosage form(extract) by way of hot water because of its limited storage. Also the majority of the consumers have a complaint against the dosage. The purpose of this study was to develop the functional materials from NNR without side effects. We formulated the solid dosage form viz tablet and granule from the lotus root. Sensory evaluation was performed in terms of smell, taste, color and overall of lotus root and all colored forms(brown, dark brown, light green and yellow) of tablet and granule to evaluate the acceptability of the formulated tablets and granules. In sensory evaluation, among the formulated tablets and granules, light green granules obtained best score overall and yellow tablets showed the overall improved acceptability. In conclusion, lotus rhizome could be recommended as functional food. Further studies to clarify bioactive functions of Nelumbo nucifera in experimental animal model on atopic dermatitis are in progress.

  • To investigate anti-diabetic component from the node of lotus rhizome (Nelumbo nucifera; Nymphaeaceae), activity guided isolation was conducted. One amino acid was isolated from active fraction of the aqueous methanolic extract. The structure of this compound was identified as tryptophan (1) by the analysis of spectroscopic evidences and comparisons with the data of authentic samples. Tryptophan reduced the blood glucose level significantly in glucose-fed hyperglycemic mice compared with glucose-treated group and exhibited 44.3% of activity compared with tolbutamide,.

Effect of Nelumbo nucifera on Proliferation, Migration and Expression of MMP-2 and MMP-9 of rSMC, A431 and MDA-MB-231

  • Nelumbo nucifera Gaertn.(family Nymphaeaceae) has been used for summer heat syndrome as home remedy in Japan, China and Korea. Although whole plant parts are edible, root is commonly consumed. It has been reported that rhizome extract showed anti-diabetic and anti-inflammatory effects. However, in spite of usefulness for treatment of various diseases, the effect of Nelumbo nucifera rhizome(NNR) on proliferation, migration and matrix degrading enzymes-matrix metalloproteinsases(MMPs), the expression of which degrades extracellular matrix(ECM) leading to metastasis, has not been fully elucidated. We examined the effect of hot water extract of NNR on the proliferation, migration and secretion of MMP-2 and MMP-9 in rat smooth muscle cells(rSMC), epidermoid cancer cells(A431) and breast cancer cells(MDA-MB-231). The proliferation assay was carried out using MTT assay, the principle of which depends upon the conversion of MTT by mitochondrial dehydrogensases of viable cells to formazan crystals. The effect of NNR on migration of cells was examined using wound healing assay. Our results showed that there was inhibition in the proliferation, migration and expression of MMP-2 and MMP-9 in dose dependent fashion in all the cells used. Thus, we concluded that NNR could be used as traditional medicine in the treatment of various diseases where proliferation, migration and MMPs' expression plays a pathological role like in restenosis and metastasis.

콜린 에스테라제 저해효과 보유 식물 추출물 탐색 (Screening of Plant Extracts with Cholinesterase Inhibition Activity)

  • 본 연구는 천연식물자원을 대상으로 치매의 일종인 알츠하이머병을 개선 및 치료할 수 있는 소재를 발굴하기 위해 식물추출물에 대해 콜린성 에스테라제(AChE) 저해활성을 탐색하였다. 184개의 식물추출물을 $100{\mu}g/ml$의 단일농도에서 아세틸콜린에스테라제(AChE)을 실험한 결과, 10% 이상의 AChE 저해활성을 보였던 추출물은 황벽나무(Phellaodendron amurense Rupr., 수피, 메탄올 추출물), 황벽나무(Phellaodendron amurense Rupr., 수피, 물 추출물), 연꽃(Nelumbo nucifera Gaertn., 수술/자방, 메탄올 추출물), 쪽(Persicaria tinctoria H. GROSS, 꽃, 메탄올 추출물), 중국황련(Coptis chinensis, 뿌리줄기, 메탄올 추출물), 육계(Cinnamomum cassia Blume, 수피, 에탄올 추출물) 및 잇꽃(Carthamus tinctorius L., 열매, 에탄올 추출물) 등 7개가 확인되었다. 선발된 7개의 추출물에 대해서는 25, 20, 100 및 $200{\mu}g/ml$의 최종농도에서 AChE 및 BuChE에 대한 저해활성을 검정하였다. 그 결과, AChE 저해활성에서는 연꽃(Nelumbo nucifera Gaertn.) 수술/자방의 메탄올 추출물이 $15.0{\pm}3.4%{\sim}73.5{\pm}3.6%$, 쪽(Persicaria tinctoria H. GROSS) 꽃의 메탄올 추출물이 $19.5{\pm}9.1%{\sim}63.5{\pm}5.4%$, 중국황련(Coptis chinensis) 뿌리줄기의 메탄올 추출물이 $81.6{\pm}0.4%{\sim}58.5{\pm}2.4%$, 황벽나무(Phellaodendron amurense Rupr.) 수피의 메탄올 추출물이 $69.9{\pm}1.8%{\sim}80.5{\pm}0.9%$, 그리고 황벽나무 (Phellaodendron amurense Rupr.) 수피의 물 추출물이 $54.8{\pm}0.6%{\sim}78.3{\pm}2.6%$의 값을 보였다. BuChE 저해활성에서는 연꽃(Nelumbo nucifera Gaertn.) 수술/자방의 메탄올추출물, 쪽(Persicaria tinctoria H.GROSS) 꽃의 메탄올추출물 및 중국황련(Coptis chinensis) 뿌리줄기의 메탄올추출물이 각각 $58.9{\pm}7.8{\sim}81.6{\pm}1.9%$$45.8{\pm}9.8%{\sim}72.4{\pm}2.5%$$33.1{\pm}9.9%{\sim}55.4{\pm}5.4%$의 순으로 높은 BuChE 저해활성을 나타내었다. 이러한 실험결과를 종합하여 볼때, 연꽃(Nelumbo nucifera Gaertn.)의 수술/자방, 쪽(Persicaria tinctoria H. GROSS)의 꽃, 중국황련(Coptis chinensis)의 뿌리줄기, 황벽나무(Phellaodendron amurense Rupr.)의 수피는 알츠하이머병(AD)의 개선 효과를 나타낼 수 있는 후보 소재로 사료되었다.

무안 회산지(回山池) 백련꽃의 근경생장과 개화특성 (Rhizome Growth and Flowering of White Lotus (Nelumbo nucifera) at Muan Hoesan-Lake)

  • 무안 회산지 백련꽃의 재배와 개화 조절에 필요한 자료를 얻기 위하여 2008년 4월 20일과 5월 4일에 각각 비대근경과 종자를 용기에 식재하여 비가림 재배 관리한 후 동년 9월 4일에 근경생장 유형과 개화 특성을 조사하였다. 주근경이 분지된 것은 실생의 경우 10%, 비대근경은 15%였다. 주근경의 마디 수는 개화된 것은 14개 이상이었으며, 개화가 되지 않은 것은 11마디 이하였다. 실생 번식한 것은 모두 개화되지 않았으며, 비대근경을 식재한 것은 80%가 개화되었다. 첫 번째 꽃은 7월 말과 8월 초에 피었으며, 4.0~4.4개의 꽃이 피었다. 첫 번째 꽃대는 주근경이 분지되지 않은 것의 경우 8.8번째 마디에서, 주근경이 분지된 것은 8.0번째 마디에서 출현되었다. 화경이 지상부에 출현된 후 개화까지는 16일이 소요되었다. 백색의 꽃은 높이 15.3 cm, 폭 28.2 cm 크기로 수명은 3.3일이었으며, 생체중은 29.2 g이었다. 이상의 결과로부터 무안 백련꽃의 개화를 시키기 위해서는 근경의 비대가 시작되기 이전에 주 근경의 마디를 8마디 이상 생장시켜야 함을 알 수 있었다.

생약추출물의 항산화 활성검색 (Screening of Crude Drugs for Antioxidative Activity)

  • Based on DPPH radical scavenging activity and lipid peroxidation inhibitory activity, the MeOH extracts of 139 crude drugs were screened in order to search for antioxidants. Among tested samples, the extracts from the seed of Nelumbo nucifera, the fruit of Terminalia chebula var. gangetia, the root of Salvia miltiorhiza, the fruit of Ziziphus jujuba var. innermis, the root bark of Paeonia moutan, the fruit of Rubus coreanus, the fruit of Zanthoxylum schinifolium, the lignum of Caesalpinia sappan, the leaf of Pinus densiflora, the rhizome of Alpinia officinarum, the fruit of Syzygium aromaticum, the ramulus and uncus of Uncaria rhynchophylla, the root bark of Lycium chinense, and the fruit of Alpinia katsumadai showed a relatively strong antioxidative activity. Furthermore, the BuOH fraction from the extract of N. nucifera showed a potent activity in each assay. The isolation of bioactive compounds has been carried out and will be reported in the next paper.

국내 자생식물 20종의 혈관이완 효능에 대한 실험연구 (Research on vasorelaxant effects of 20 Korean native plants)

  • Objectives : The objective of present study was to investigate the vasorelaxant effects of 20 Korean native plants on isolated rat thoracic aorta precontracted with phenylephrine (PE). Methods : Dried 20 plant materials were extracted 3 times with water, ethanol, or methanol for 3h in the reflux apparatus at 70 ± 5℃. Male SD rats were anesthetized by ether inhalation, and their aorta rings were isolated and placed in 10 ㎖ Krebs Henseleit (KH) buffer. While using an isolated organ-chamber technique, the aorta rings were maintained by bubbling with a gas mixture of 95% O2-5% CO2 at 37℃. Changes in isometric tension of aorta rings were recorded via isometric transducers connected to a Powerlab Data Acquisition System. Results : Among the 20 native plants, Chrysanthemum indicum L. flower, Nelumbo nucifera Gaertn. rhizome, Schisandra chinensis (Turcz.) Baill. fruit, Anthriscus sylvestris (L.) Hoffm. root, Corydalis turtschaninovii Besser tuber, Corydalis decumbens (Thunb.) Pers. tuber, and Dolichos lablab L. seed showed significant vasorelaxant effect on the contraction of aorta rings induced by PE. In contrast, Mertensia maritima subsp. asiatica Takeda whole plant, Ajuga decumbens Thunb. whole plant, Trigonotis peduncularis (Trevis.) Benth. ex Baker & S.Moore whole plant, Dioscorea quinquelobate Thunb. rhizome, Allium microdictyon Prokh aerial part, Momordica charantia L. fruit, Carthamus tinctorius L. flower, and Clematis terniflora DC. root constricted more the aorta rings precontracted by PE Conclusion : These results suggest that the possibility as useful herbal resources for the development of functional foods or medicines for hypertension treatment.

Lotus (Nelumbo nucifera) Rhizome as an Antioxidant Dietary Fiber in Cooked Sausage: Effects on Physicochemical and Sensory Characteristics

  • The objective of this study was to determine the physicochemical and sensory properties of cooked emulsion sausages containing different levels of lotus rhizome powder (0, 1, 2, and 3%, based on total weight). Lotus rhizome powder had no significant (p>0.05) impact on pH, moisture, protein, or ash content of sausage. However, fat content was slightly but significantly (p<0.05) decreased when the level of lotus rhizome powder was increased in the sausages. The addition of lotus rhizome powder to sausages at over 1% resulted in significantly (p<0.05) darker and less red color of cooked sausage compared to control. Increase in lotus rhizome level slightly improved the emulsion stability and apparent viscosity. Significant (p<0.05) reduction in cooking loss was observed when more than 1% of lotus rhizome powder was added to sausages. The textural properties of sausages were unaffected by the inclusion of lotus rhizome except for springiness and chewiness. On the manufacture day, control sausage had significantly (p<0.05) higher TBARS value than treatments. Regarding sensory characteristics, increased levels of lotus rhizome powder decreased (p<0.05) color and juiciness scores. However, cooked sausages exhibited similar overall acceptability regardless of the level of lotus rhizome powder added to sausages. Therefore, lotus rhizome powder, an antioxidant dietary fiber, could be used as an effective natural ingredient in meat products for the development of healthier and functional food.
