Superoxide dismutase 3 protects mesenchymal stem cells through enhanced autophagy and regulation of FoxO3a trafficking

  • Therapeutic applications of mesenchymal stem cells (MSCs) are limited due to their early death within the first few days of transplantation. Therefore, to improve the efficacy of cell-based therapies, it is necessary to manipulate MSCs so that they can resist various stresses imposed by the microenvironment. Moreover, the role of superoxide dismutase 3 (SOD3) in regulating such survival under different stress conditions remain elusive. In this study, we overexpressed SOD3 in MSCs (SOD3-MSCs) and evaluated its effect under serum starvation conditions. Nutritional limitation can decrease the survival rate of transplanted MSCs and thus can reduce their efficacy during therapy. Interestingly, we found that SOD3-MSCs exhibited reduced reactive oxygen species levels and greater survival rates than normal MSCs under serum-deprived conditions. In addition, overexpression of SOD3 attenuated starvation-induced apoptosis with increased autophagy in MSCs. Moreover, we have demonstrated that SOD3 protects MSCs against the negative effects of serum deprivation via modulation of AMP-activated protein kinase/sirtulin 1, extracellular signal-regulated kinase activation, and promoted Forkhead box O3a trafficking to the nucleus. Taken together, these results demonstrate that SOD3 promotes MSCs survival and add further evidence to the concept that SOD3-MSCs may be a potential therapeutic agent with better outcomes than normal MSCs for various diseases involving oxidative stress and compromised MSCs survival during therapy.

Decrease of Thyrotropin Binding Inhibiting Immunoglobin (TBII) in Sera of Graves' Disease Patients Related with Remission Induced by Thionamide Regimens (Graves병에서 Thionamide 치료시 관해 여부에 연관한 TSH 수용체 항체의 변동에 관한 연구)

  • Serum TBII measured by radioreceptor assay using $^{125}I-bovine$ TSH and porcine thyroid well membrane was checked before, 6 months and 12 months after initiation of thionamide regimens in 63 Graves' disease patients and was related with their remission state. 1) A significant difference (p < 0.01) in pre-treatment TBII was noted between the remitted [N = 45, TBII $40.9{\pm}18.2%$ $(mean{\pm}S.D)$] and the unremitted (N = 18, TBII $64.1{\pm}15.3%$) groups. 2) After 6 months of therapy, TBII were significantly decreased in both groups (to $20.2{\pm}10.3%$ and to $45.2{\pm}16.3%$, p<0.05 for each group) 3) At 12th month, TBII activities were not significantly decreased compared to the 6th month levels in both groups. 4) 3 of the 58 patients who were initially TBII positive (over 15%) converted negative. All the 3 belonged to the remitted group. 5) No significant differences were seen in initial and posttreatment TBII levels between propylthiouracil treated (N = 36) and methimazole treated (N = 27) cases. with above mentioned results, we observed that the TBII decreased significantly with 6 months of thionamide therapy and concluded that the pretreatment measurement of serum TBII may be clinically useful in predicting the response to thionamide regimen in the treatment of Graves' disease.

Survival of Patients with Ewing's Sarcoma in Yazd-Iran

  • Background: The Ewing's sarcoma family is a group of small round cell tumors which accounts for 10-15% of all primary bone neoplasms. The aim of this study was to evaluate the survival of Ewing's sarcoma patients in our province and to determine of influencing factors. Materials and Methods: All patients with documented Ewing's sarcoma/primitive neuroectodermal tumor(PNET) family pathology were enrolled in this study during a period of eight years. For all of them local and systemic therapy were carried out. Overall and event free survival and prognostic factors were evaluated. Results: Thirty two patients were enrolled in the study. The median age was 17.5 years. Twenty (65.2%) were male and 9 (28.1%) were aged 14 years or less. Mean disease free survival was 26.8 (95%CI; 13.8-39.9) months and five year disease free survival was 26%. Mean overall survival was 38.7 months (95%CI; 25.9-50.6) and median overall survival was 24 months. Five year overall survival was 25%. From the variables evaluated, only presence of metastatic disease at presentation (p value=0. 028) and complete response (p value =0. 006) had significant relations to overall survival. Conclusions: Survival of Ewing's sarcoma in our province is disappointing. It seems to be mostly due to less effective treatment. Administration of adequate chemotherapy dosage, resection of tumor with negative margins and precise assessment of irradiation volume may prove helpful.

Anodal Effects of Transcranial Direct Current Stimulation on the Excitability of Central Neuron (양극 경두개 직류 전기 자극이 중추신경원의 흥분성에 미치는 영향)

  • Purpose : This study is to examine the effects of transcranial direct current stimulation on the excitability of the central neuron. Methods : This study selected 24 suitable women in their twenties. A positive electrode of transcranial direct current stimulation was placed on the primary motor area (M1) C4 and a negative electrode was placed on the left supraobital. A stimulation of 0.04mA/$cm^2$ was applied for 20 minutes. H-reflex and V wave used diagnostic electromyography. An active electrode was placed at the muscle belly of the medial gastrocnemius muscle at a prone posture. An electrical stimulation was given to the posterior tibial nerve. Measurements were made before and after the stimulation. All data were analyzed with SPSS 12.0 and between each measuring before and after the change of the H-reflex and V wave amplitude. Results : There were no significant differences in all H wave, M wave, and V wave amplitude before and after transcranial direct current stimulation. There were no significant differences in the change of H/M ratio and V/M ratio before and after transcranial direct current stimulation. Conclusion : We know that transcranial direct current stimulation cannot have an influence on a normal grown-up person's central neuron.

A Study on the Relations among Depression, Aggression and Draw-a-Story Response Characteristics of Middle School Students (중학생의 우울 및 공격성과 DAS반응특성과의 관계연구)

  • The purpose of this study is to inspect the use possibility of DAS as a depression assessment tool for middle school students by finding out their depression tendency and DAS characteristics. For this purpose, I have found out explored the differences between depression and aggression of in middle school students in accordance with sex and grade, and also found a relationship between DAS response characteristics, and the level of depression and aggression. The subjects of this study were 600 middle school students in Pusan and Kyungnam, and the tools used in this study were Silver's Draw-a-Story and Beck's depression scale and aggression scale. The results were analyzed two-way ANOVA analysis, Duncan test, correlation analysis and crosstab analysis. The results of this study are as follows. The depression level of the 9th grade students was higher than that of 7th grade students. And the aggression level of males was higher than in females, and higher in 8th graders than in 7th graders. The emotion content scores and the self-image scores were related to depression and aggression. So when the levels of depression and aggression were high, the emotional response of DAS and the self-image response were negative. Through this study, it is suggested that DAS has the power of discrimination as a tool measuring depression state and that DAS is meaningful because it showed it can be used as a tool for estimating the level of depression state of school students in schools.

The Study on Burden of Mothers of Children with Nephrotic Syndrome (신증후군 환아 어머니의 부담감에 대한 연구)

  • This study was designed to identity the degree of burden felt by mothers of children with nephrotic syndrome. Also, relations between the subject characteristics and burden were investigated to provide basis data for their family health and nursing intervention. The study subjects were composed of 70 mothers of nephrotic syndrome patients whose children were hospitalized in 2 Pediatric wards of University Hospital in Seoul and 1 in Pusan from Mar. 2nd, 1998 to May. 30th in the same year The questionnaires were used which dealt with burden of mothers. the questionnaires for this study designed and used by researcher placed their basis in Burden Measurement Instrument developed by Montgomery et. al (1985) and the reliability of the used instrument was .78. The data analysis was done by SPSS, t-test, ANOVA and Stepwise Multiple Regression. The results of were as follows. 1. Mean score of burden of subjects was 60.82(Maximum 86, standard deviation 1.244). 2. Of the mothers characteristics, the score of burden was high in case of no religion and low income. 3. Of the patients characteristics, the score of burden ranked as high in MCNS, doing oral therapy and injection therapy at the same time, and negative perceived patients' condition. 4. The degree of burden felt by mothers of children with nephrotic syndrome was significantly predicted by the level of pt's diagnosis(11%), pt's condition(8%), economic state (6%) and pt's sex (5%), respectively. In conclusion to above study, the researcher suggests that the development of instrument for measurement of burden is in much need. Relations between burden and social support should be studied to lessen burden of mothers of children with nephrotic syndrome.

The Effect of Insole Height on Lumbosacral Angle and Body Function in Male University Students

  • Objective: The aim of this study is to investigate the effect of insole height change in the lumbosacral angle and physical functions in healthy males. Background: In order to release male's dissatisfaction with his height and to increase satisfaction with his body, using insole is generalized. There have been researches on female's body change in accordance with function of insole and heel height, whereas there are few researches on males. Method: Participants were divided into three groups. A control group had 10 participants who wore 0cm insole. Experimental group I had 10 participants who wore 2cm insole. Experimental group II had 10 participants who wore 4cm insole. All participants wore insoles during their daily lives for a trial period of 8 weeks. The results were evaluated before and after comparison, and we measured lumbosacral angle, balance (dynamic balance, agility, quickness) and lumbar pain (LBP). Results: This study showed that insole height affected lumbosacral angle and dynamic balance and pain. In particular, there were significant differences in the 4cm group among the three groups (p<.05). The 2cm group did show a significant difference in lumbosacral angle and pain (p<.05). Furthermore, no significant difference was observed within the control group. Conclusion: The 4cm insole height suggests that the increase of lumbosacral angle contributes to some changes in LBP, balance, pain and physical functions, probably leading to negative effects on variety of activities of daily life. Application: The results of wearing insoles with proper height will help to prevent musculoskeletal disorders.

A Study on Relationship of Symptom Distress and Natural Killer Cell Cytotoxicity in Breast Cancer Patients (치료 중인 유방암 환자의 신체적 증상과 자연살해세포 활성도의 관계)

  • The purpose of this study was to identify the relationship of symptom distress and natural killer cell cytotoxicity in breast cancer patients who had been radiation therapy and/or chemotherapy after surgery. Symptom distress measured by modified Lee's(1994) physical symptom questionnaire. For measuring the natural killer cell cytotoxic activity. 8ml to 10ml blood was collected from the subjects. Mononuclear cell was isolated by centrifuge of the blood and cultured by putting $Cr^{51}$, and reacted with target cell, K562 cell. Amount of $Cr^{51}$ was measured, and %lysis was calculated. The results were as follows. 1) Symptom distress score was 42.18, which is moderate symptom distress. 2) Natural killer cell cytotoxic activities were 42.18%lysis(effector : target cell ratio=100 : 1) and 28.05%lysis(effector : target cell ratio=50 : 1). 3) Correlation coefficients of symptom distress and natural killer cell cytotoxic activity were $-.134{\sim}-.461$. Though significant correlation was not found between total score of symptom distress and natural killer cell cytotoxic activity, 3('pain' 'feel hot on radiation site' and 'difficulty in breathing') of 19 symptom distress items and natural killer cell cytotoxic activity showed significant negative correlation(p<.05). These findings suggest that 1) breast cancer patients who had been radiation therapy and/or chemotherapy after surgery have moderate symptom distress and decreased natural killer cell cytotoxic activity. 2) The symptom distress was not related to natural killer cell cytotoxic activity.

The Two Cases Report of Bee Venom Injection on Patient with Low Back Pain Maintaining after Heating-Conduction Acupuncture Therapy (전열침 시술에 호전되지 않는 요통환자에 대한 봉독약침 치료 증례보고 2례)

  • Objectives : According to previous reports, heating-conduction acupuncture treatment is very effective for sprain and strain of ligament. But there was no report about pain aggravation by heating-conduction acupuncture and relief from bee venom injection. In this article, we report two cases of pain aggravation by heating-conduction acupuncture treatment and relief from bee venom injection. Methods : We used heating-conduction acupuncture treatment on possible sprain of sacroiliac ligament diagnosed by physical examination. Outcomes were measured by Visual Analogue Scale(VAS), Range of Motion(ROM) and Belt test, Goldthwait test. Results : The patient has shown an aggravation on pain and ROM. And after we use subcutaneous bee venom injection, pain has relieved and ROM improved. Belt test, Goldthwait test also changed from positive to negative. Conclusions : In this report, we assured the effect of Bee venom injection. Further well-designed, controlled studies and more cases are needed to differentiate the indication of heating-conduction acupuncture and bee venom injection.

A Case of Idiopathic Collapsing Glomerulopathy Showing Aggravation on a Chronic Progressive Course (만성신질환의 경과 중 급성 악화를 보인 허탈성 사구체병증의 진단 1예)

  • Collapsing glomerulopathy (CG) has become an important cause of end-stage renal disease (ESRD). First delineated from other proteinuric glomerular lesions in the 1980s, CG is now recognized as a common, distinct pattern of proliferative parenchymal injury that portends a rapid loss of renal function and poor responses to empirical therapy. The first cases in the literature trace back to human-immunodeficiency-virus(HIV)-negative patients who underwent biopsy in 1979. A 45-year-old male patient complained of hematuria and proteinuria eight years ago. He showed an abrupt serum creatinine increase from 1.75 to 2.65 mg/dL in the last preceding months. Afterwards, his serum creatinine progressively increased up to 6.82 mg/dL. Moreover, his 24 h urine protein level was determined to have reached 6,171 mg/day, as opposed to 670 mg/day a year earlier. Consequently, renal biopsy was performed, and its result showed collapsing glomerulopathy, compatible with the diagnosis. He has undergone continuous ambulatory peritoneal dialysis as renal replacement therapy. Thus, it is reported herein that a patient clinically diagnosed with chronic kidney disease eight years ago showed a sudden renal-function decrease and was clinicopathologically diagnosed with collapsing glomerulopathy based on the results of his renal biopsy.
