Cultivation of Saccharomyces cerevisiae using Defatted Rice Bran Hydrolyzed in Near-critical Water as a Culture Medium (탈지미강의 아임계수 가수분해 생성물을 배지로 이용한 Saccharomyces cerevisiae의 배양)

  • The hydrolysis of defatted rice bran using near-critical water was performed, and the feasibility of consequent hydrolyzate as a growth medium was investigated by the cultivation of Saccharomyces cerevisiae. The near-critical water hydrolysis was carried out through a series of batch experiments, and the contents of total carbohydrates, disaccharides, and monosaccharides, total organic carbon (TOC), total nitrogen (TN), pH of products were measured. The growth rate of Saccharomyces cerevisiae was measured with optical density. The yield of total carbohydrates, TOC, and TN increased with temperature below $240^{\circ}C$, however, decreased above $240^{\circ}C$. The decrease of yields above $240^{\circ}C$ was caused by the formation of organic acids, and it agreed with the change of pH of products. The yield of glucose was a maximum at $200^{\circ}C$ and it decreased dramatically at higher temperature. The growth rate of Saccharomyces cerevisiae cultivated in the hydrolyzate was similar with that in the commercial medium under certain conditions. The growth rate was correlated with the content of glucose in hydrolyzate.

Optimization of TDA Recycling Process for TDI Residue using Near-critical Hydrolysis Process (근임계수 가수분해 공정을 이용한 TDI 공정 폐기물로부터 TDA 회수 공정 최적화)

  • The recycling of TDA from solid waste of TDI plant(TDI-R) by near-critical hydrolysis reaction had been studied by means of a statistical design of experiment. The main and interaction effects of process variables had been defined from the experiments in a batch reactor and the correlation equation with process variables for TDA yield had been obtained from the experiments in a continuous pilot plant. It was confirmed that the effects of reaction temperature, catalyst type and concentration, and the weight ratio of water to TDI-R(WR) on TDA yield were significant. TDA yield decreased with increases in reaction temperature and catalyst concentration, and increased with an increase in WR. As a catalyst, NaOH was more effective than $Na_2CO_3$ for TDA yield. The interaction effects between catalyst concentration and temperature, WR and temperature, catalyst type and reaction time on TDA yield had been defined as significant. Although the effect of catalyst concentration on TDA yield at $300^{\circ}C$ as subcritical water was insignificant, the TDA yield decreased with increasing catalyst concentration at $400^{\circ}C$ as supercritical water. On the other hand, the yield increased with an increase in WR at $300^{\circ}C$ but showed negligible effect with WR at $400^{\circ}C$. The optimization of process variables for TDA yield has been explored with a pilot plant for scale-up. The catalyst concentration and WR were selected as process variables with respect to economic feasibility and efficiency. The effects of process variables on TDA yield had been explored by means of central composite design. The TDA yield increased with an increase in catalyst concentration. It showed maximum value at below 2.5 of WR and then decreased with an increase in WR. However, the ratio at which the TDA yield showed a maximum value increased with increasing catalyst concentration. The correlation equation of a quadratic model with catalyst concentration and WR had been obtained by the regression analysis of experimental results in a pilot plant.

Synthesis of Surface Active Properties of Destructible Surfactants with 1,3-Dioxane (1,3-디옥산을 함유한 분해성 계면활성제의 합성의 및 계면 특성)

  • In acid-catalyzed acetal cyclization of long aliphatic aldehydes($R=n-C_7H_{15}$ ; $n-C_9H_{19}$ ; $n-C_{11}H_{23}$) with 1,1,1-tris(hydroxymethyl)propane, 2-alkyl-5-hydroxymethyl-5-ethyl-1,3-dioxanes were obtained. The final products, sodium 2-alkyl-5-(sulfonatedpropylethermethyl)-5-ethyl-1,3-propanesultion in the presence of sodium hydride. These compounds were a new group of destructible surfactants which were readily hydrolyzed and oxidized in natural water reservoirs. Physical properties of these new compounds involved some surface properties such as Krafft point(Kp), critical micelle concentration(cmc), surface tension of aqueous solutions near cmc(${\gamma}_{min}$), foaming power, emulsion power and hydrolysis properties were determined. The destructible surfactants containing 1,3-dioxane ring were synthesized to about $85{\pm}5.5%$ yield. The cmc values of the compounds by ring method were assumed to $0.5{\sim}5.0{\times}10^{-3}mol/L$ range and surface tensions at cmc were $29.5{\sim}33.0dyne/cm$ respectively at $25^{\circ}C$. The foaming power and foam stability were $170{\sim}230mm$ and $52{\sim}135mm$ respectively at $1{\times}10^{-2}mol/L$, foam was occurred rarely below $1{\times}10^{-3}mol/L$. The emulsion property of liquid paraffin was better than that of soybean oil. For hydrolysis property with ph and time, these compounds were decomposed within about 200minutes at $ph1{\sim}2$. Hopefully these compounds are expected to be a good O/W emulsifier that have decomposability in acid and may be used in the process which do not need foaming.
