Chemical Composition and Antioxidant Activity of Algerian Juniperus Phoenicea Essential Oil

  • Berries and branches essential oil of Juniperus phoenicea were obtained by electromagnetic induction heating assisted extraction and by hydrodistillation with a yield varied from ($1.2{\pm}0.3$ to $2.4{\pm}0.7%$) and from ($0.6{\pm}0.1%$ to $1.1{\pm}0.1%$), respectively. forty eight compounds were identified representing (97.2 - 99.7%) of the oil. ${\alpha}$-Pinene (40.3 - 67.8%) and ${\delta}$-3-carene (13.5 - 26.8%) were the main compounds in berries and branches essential oils. Antioxidant activity was evaluated by three means: inhibition of 2, 2-diphenyl-1-picryl hydrazyl (DPPH) free radical, reducing power and ${\beta}$-Carotene/linoleic acid bleaching. The antioxidant activity of essential oils showed $IC_{50}$ ranging from $67.6{\pm}1.02{\mu}g/mL$ to $131.5{\pm}0.8{\mu}g/mL$ for berries and from $98{\pm}1.25{\mu}g/mL$ to $166.8{\pm}0.29{\mu}g/mL$ for the branches. Berries oil show more potent antioxidant activity compared to branches. This result is supported by the three methods investigated in this work.

Radical Scavenging Activity of Grape-Seed Extracts Prepared from Different Solvents

  • Antioxidant activities of grape seeds extracted with various solvents were evaluated by measuring total phenol and flavanol contents, thiobarbituric acid reactive substances (TBARS) following lipid peroxidation, 2-deoxyribose degradation, SOD-like activity, 2,2'-azino-bis(3-ethylbenzthizaoline-6-sulfonic acid (ABTS) radical-scavenging ability, and electron-donating ability using 1,1-diphenyl-2-pycryl hydrazil (DPPH) method. Total phenol and flavanol contents of mixted-solvent extracts were higher than those of single-solvent extracts, with the mixing ratio of 17:3 (ethyl acetate: water) (EW) showed the highest contents. Antioxidant activities (%) of TBARS following phosphatidylcholine peroxidation were 14, 45, 45, 7, 4, 25, 21, 23, and 20% for ascorbic acid (AA), butylated hyroxytoluene (BHT), quercetin (Q), acetone extract (AT), ethyl acetate (EA) extract, methanol (MeOH) extract, 4:1 (EA) extract, 9:1 (EW)-extract, and 17:3 EW extract, respectively. Antioxidant activities for 2-deoxyribose degradation were 5, 80, 87, 78, 56, 73, 64, 60, and 75% in AA, BHT, Q, AT, EA, MeOH extract, 4:1 EW extract, 9:1 EW extract, and 17:3 EW extract, respectively. MeOH grape seed extract showed distinctly stronger electron-donating activity than other solvent extracts.

Antibacterial effects of Terminaliae chebula extract against major pathogens and methicillin-resistant Staphylococcus aureus (MRSA) from bovine mastitis milk (가자(Terminaliae chebula) 추출물의 젖소 유방염 주요 원인체 및 메치실린내성 황색포도상구균(MRSA)에 대한 항균효과)

  • Resistance to antibiotics is a problem all over the world, and this problem also is so extended in veterinary fields. Therefore, this study aimed to find out the antibacterial substances from natural medicinal herbs against bovine mastitis pathogens, especially methicillin-resistant Staphylococcus aureus (MRSA). Among seventy two medicinal herbs, Terminaliae chebula extract showed antimicrobial effect against MRSA isolated from mastitis milk in cow and patient in human. However, Terminaliae chebula extract didn't show antibacterial effects on various strains including other contagious and environmental pathogens related with mastitis of dairy cows.

  • Phytochemicals include plant-derived natural products that promote and improve the human metabolism and physiological activity, and there is a lot of research to find the value of the molecules is in progress. Likewise, we obtained 288 fractions of Capsicum annuum L. extract in less than 20 h using HPLC/SPE/HPLC coupling experiment through Sepbox system, an effective separation system to search for active substances in natural resources and ensure efficacy and reliability. Therefore, this experiment allowed rapid identification of biologically active molecules from the extract compared to traditional separation processes. Of the above fractions, eight fractions showed the α-glucosidase inhibitory (AGI) activity and subsequent LC-MS analysis revealed one of the active molecules as luteolin 7-O-glucoside. In addition, we proved the increase in AGI activity according to deglycosylation of flavonoid glycoside. Therefore, this study suggests that the Sepbox system can quickly separate and identify active components from plant extract, and is an effective technique for finding new active substances.

Economical Ventilation Effectiveness to Reduce Hazardous Chemical Emissions for a Nail-Salon Worker

  • Purpose - The purpose of this study is to investigate economical ventilation effectiveness to reduce hazardous materials exposure and damage of workers by analyzing exposure amount of noxious substances under various ventilation conditions of nail salon for indoor environments. Research design, data, and methodology - This study was carried out with cooperation of Nail shop located in SeongNam city to involve an analysis of the environmental impact indoor air quality, pollutant exposure and economical cost-effectiveness in the nail workplace. The hazardous substances were PM-10(Particulate Matter-10㎛), VOCs(Volatile Organic Compounds) and Formaldehyde, which are the major materials of nail workplace. Results - PM-10 is reduced by about 60% with air cleaner, forced artificial ventilation by 32%, and natural ventilation by about 12%. TVOCs and Formaldehyde showed similar efficiency (80~100%) after natural ventilation and ventilation after 60 minutes. The removal efficiencies of VOCs and formaldehyde were similar to those of natural ventilation and mechanical ventilation system. However, in case of dust, natural ventilation was reduced by artificial ventilation system due to inflow of external dust during natural ventilation. Conclusions - If the pollution degree of outdoor air is not high, air volume is high, and natural ventilation is performed when the air conditioning and heating system is not operated. Even at the end of the work, it keeps operating for 60 minutes to remove the pollutants generated. Results of this analysis demonstrated that the worker environment can be improved by adopting institutional legislation and guidelines for ventilation.

Validation of Photo-comet Assay as a Model for the Prediction of Photocarcinogenicity

  • Recent reports on the photocarcinogenicity and photogerotoxicity of many compounds led to an increasing awareness for the need of a standard approach to test for photogenotoxicity. The comet assay has been recently validated as a sensitive and specific test system for the quantification of DNA damage. Thus, the objectives of this study are to investigate the utility of photo-comet assay for detecting photo-mutagens, and to evaluate its ability to predict rodent photo-carcinogenicity. Photo-comet assays were performed using L5178Y $Tk^{+/-}$ mouse lymphoma cells on five test substances (8-methoxypsoralen, chlorpromazine, lomefloxacin, anthracene and retinoic acid) that demonstrated positive results in photocarcinogenicity tests. For the best discrimination between the test substance-mediated DNA damage and the undesirable DNA damage caused by direct UV absorption, a UV dose-response of the cells in the absence of the test substances was firstly fnalized. Out of 5 test substances, positive comet results were obtained for chlorpromazine, lomefloxacin, anthracene and retinoic acid while 8-methoxypsoralen found negative. An investigation into the predictive value of this photo-comet assay for determining the photocarcinogenicity showed that photo-comet assay has relatively high sensitivity. Therefore, the photo-comet assay with mammalian cells seems to be a good and sensitive predictor of the photocarcinogenic potential of new substances.

The Promotive Effect of NAA, IBA and Ethychlozate on Rooting Cuttings of Certain Ornamental Plants and Some Physiological Studies. (관상식물 삽목발근에 있어서 NAA, IBA 및 Ethychlozate의 발근촉진효과와 그 생리학적연구)

  • The present studies were undertaken to elucidate the influence of auxins, auxin-like substance-ethychlozate ("Figaron"),and pH and sort of rooting media on rooted propagation of certainornamental woody plant cuttings, and to see possible changes in internal compositions characterizing after root-promoting treatment as the cutting stage proceeded. The experimental check-up srevealed and summarized as seen in the following;I. Effect of three different auxin treatments on rooting cuttings: 1) Promotive influence of auxin varied according to different concentration levels, hours of dipping treatment of the auxins, and kind of plants. The greatest effect was obtained for Forsythia ksreana with NAA and IBA, for Ligustrurn obtusifolium var. variegatum with NAA and ethychlozate, for Hydrangea macrophylla, Magnolia kobus, and Magnolia liliflora with NAA, lBA and ethychlozate also. The most effective level of the promotive agents was found 200mg/l for NAA, 1000mg/l for IBA, and 200mg/l for ethychlozate. For Weigela florida and Gardenia jasminoides, range of the most effective level was shown relatively wide spread. 2) NAA was more effective at its optimal level of the rooting agent than ethychiozate for Weigela florida, Viburnum awabuki, Forsythia koreana, Acer palmatum 'Nomura', Bouga invillea glabra, Elaeagnus umbellata, Prunus tomentosa, Ligustrum obtusifolium, Pyracantha coccinea, Cestrum noctu rnum, Hydrangea macrophylla, Codiaeum variegatum, Rhododen dron lateritium, and Ilex crenata var. macrophylla, and yet ethychlozate was found either as equally as effective or more so than NAA for Zebrina pendula, Hibiscus syriacus, Fatshedera lizei, Schefflera arboricola, Campsis grandiflo ra, Ixora chinensis, Euonymus japonica, and Magnolia liliflora. On the contrary, no the auxin effect was noted with Lagerstroemia indica, Trachelospermum asiaticum, and Syringa vulgaris. This probably indicates that these species are genetically different for the auxin response.II. Effect of different pH and sorts of cutting media on rooting cuttings: 1) Bougainvillea showed best in rooting for the number and dry weight at pH 6.5, more with ethychlozate than NAA, while Ligustrum did at pH 5.0 more with NAA than ethychlozate. pH 4.0 medium resulted in the best rooting for Rhododendron with NAA, more than ethychlozate. 2) Use of cutting medium with peat: perlite: vermiculite = 1:1:1 showed to give the greatest rooting percent and dry weight, apart from considering the number of roots. This apparently meant the fact that cutting medium has more to do with root growth than root differentiation. Rhododendron yet showed results with cutting media that use of peat: perlite = 2:1 mixed is more effective on rooting than using peat alone.III. Effect of auxinic treatments on rooting cuttings and change in some cutting compositions: 1) Under the climatic conditions of July having temperature $26.3\pm$$2.4^{\circ}C$for cutting bed, new roots of Magnolia started to show up generally 20 days after the cutting was made, whereas Cestrum did much earlier than that, namely 14 days after. 2) Although total carbohydrate content of Magnolia cuttings showed no marked change without auxin treatment, it did so with the treatment, especially 30 days after the start of cutting. Cestrum cuttings demonstrated a gradual in crease in total carbohydrate content as rooting took place, and the content became reduced more with auxin than with out, just about when rooting proceeded to 14 days after the start of cutting. 3) Magnolia generally showed an increase in total nitrogen content as rooting proceeded more, and Cestrum showed a decrease in total nitrogen of cuttings. The auxin treatment exhibited no pertinent relation with change in plant nitro gen when rooting is promoted with auxin treatment. 4) An abrupt drop of total sugar and reducing sugar was noticed as Magnolia rooting started, and this reduction was parti cularly outstanding with auxin treatment. Starch content also was decreased in the later stage of cutting with auxin treatment, and was rather increased without auxin. Although sugar content soon increased as cutting started with auxin treatment in the case of Cestrum, it became reduced after rooting took place. 5) Total phenol content increased with rooting, and this was especially true when rooting started. This increase was reversed somehow regardless of auxin treatment. A decrease in phenol of Magnolia was found more striking with auxin than without in the later stage of the cutting period. 6)Avena coleoptile test for auxin-like substances presented the physiologically active factor is more in easy-to-root Magnolia liliflora than hard-to-root Magnolia kobus, and the activity of auxin-like substances was much increased with auxin treatment. The increase in the growth promoting substances was markedly pronounced when rooting just started. The active growth substances decreased in the later stage of cutting, and certain inhibitory substances started appearing. Cestrum also showed physiologically similar growth promoting substances accompanying auxin-like active substances if auxin is treated, and some strong inhibitory substances seemed to appear in the later stage of cutting. 7) Mung-bean-rooting test indicated biologically that endogenous growth substances in Magnolia all promoted mung-bean rooting, and activity of the growth substances apparently stimulated mung-bean rooting with auxin more than without. Here auxin treatment seemed to give a rise to an increased activity of endogenous growth substances in cuttings. This activity was found much greater with either NAA or IBA than ethychlozate, and showed its peak of the activity when rooting first started taking place. Certain inhibitory substances for Avena coleoptile growth strongly promoted mung-bean rooting, and it was also much like in the case of Cestrum.

Effect of Organic Acids on Cr(III) Oxidation by Mn-oxide

  • Two oxidation states of chromium commonly occur in natural soil/water systems, Cr(III) and Cr(VI). The oxidized form, Cr(VI), exists as the chromate ion and is more mobile and toxic than Cr(III). Therefore oxidation of Cr(III) by various Mn-oxides in natural systems is a very important environmental concern. Organic substances can inhibit the Cr(III) oxidation by binding, Cr(III) strongly and also by dissolving Mn-oxides. Most of Cr(III) oxidation studies were carried out using in vitro systems without organic substances which exist in natural soil/water systems. In this study effect of organic acids - oxalate and pyruvate - on Cr(III) oxidation by $birnessite({\delta}-MnO_2)$ was examined. The two organic acids significantly inhibited Cr(III) oxidation by birnessite. Oxalate showed more significant inhibition than pyruvate. As solution pH was lowered in the range of 3.0 to 5.0, the Cr(III) oxidation was more strongly depressed. Addition of more organic acids reduced the Cr(III) oxidation mare extensively. Different inhibition effects by the organic acids could be due to their ability of reductive dissolution of Mn-oxides and/or Cr(III) binding. Organic acids dissolved Mn-oxide during the Cr(III) oxidation by the oxide, Dissolution by oxalic acid was much greater than that by pyruvate, and the dissolution was more extensive at lower pH. Inhibition of Cr(III) oxidation was parallel to the dissolution of Mn-oxide by organic acids. Although the effect of Cr(III) binding by organic acids on Cr(III) oxidation is not known yet, Mn-oxide dissolution by organic acids could be a main reason for the inhibition of Cr(III) oxidation by Mn-oxide in presence of organic acids. Thus oxidation of Cr(III) to Cr(VI) by various Mn-oxides in natural systems could be much less than the oxidation estimated by in vitro studies with only Cr(III) and Mn-oxides.

A study on the method of setting the water quality target level for integrated environmental management (통합 환경 관리를 위한 수질 환경의 질 목표 수준 설정 방법에 관한 연구)

  • When evaluating the extent of the impact of water pollutants on the surrounding area, we would like to present the target level (proposal) of the quality of the environment, which is the standard for evaluation. We propose the environmental quality target level for substances that need to be applied domestically by investigating overseas cases operating the integrated environmental management system and the implications of domestic environmental pollutant management. The appropriateness of the environmental quality target level reviewed in this study was determined using data from the water quality measuring network, and future improvement measures were proposed. We review the available methodologies for setting quality objectives for the environment. It proposed the environmental quality target level for 21 substances that have domestic water pollutant emission standards and do not have environmental standards, and proposed future improvement measures. If it is necessary to add quality target-level items of the environment in the future, it is believed that expansion will be possible based on the methodology presented in this study.

Anthelmintic Natural Products against Clonorchis sinensis and the Analogues (간디스토마에 작용하는 천연물 및 그 유사체)

  • Among 230 species of herbal drugs screened, thirty one show the anthelmintic activities in vitro against Clonorchis sinensis, the chinese liver fluke. The active substances have been isolated and their structures identified. Some of the active substances and their derivatives have been synthesized, followed by anthelmintic activity tests. The extracts from the active drugs cause damages in organs of the adult worm in the hepatic duct of rabbit. Some fresh water fish, which function as bad hosts for the fluke, excrete defense substances against the cercaria of the fluke. The defense substances have been isolated from Cyprinus carpio and Cyprinus carpio nudus, followed by structural identification. The results are summarized as follows: l) The bark of Machilus thunbergii as well as the seed of Schizandra chinensis contain meso-dihydro-guaiaretic acid as the anthelmintic component. Among derivatives synthesized, 4-phenyl-1-((3, 4-dihydroxyphenyl)-, 4-phenyl-1-(3-hydroxy-4-methoxy phenyl)-and 4-phenyl-1-(4-hydroxy-3-methoxy phenyl)-2, 3-dimethyl butanes show considerable activities. Administration of the bark extract mainly damages the bladder of the adult worm. 2) The active substance from the roasted fruit of Prunus mume is 2-hydroxymethylfurfural. This substance is produced during the roasting process. Administration of the fruitextract causes a damage of the bladder of the adult worm. 3) The active substance from the root of Scutellaria baicalensis is 5.2'-dihydroxy-6, 7, 8, 6'-tetramethoxyflavone. 4) Beside alantolactone, a very strong anthelmintic component is contained in the root of Inula helenium. Administration of the root extract causes irreversible damage on the worm, affecting mainly the reproductive organs. 5) The cercaricidal substances from the epidermis of C. carpio and C. carpio nudus are ethyl linoleate and linoleic acid, respertively. 6) The cercaricidal substances from various kinds of fresh water fish have different $R_f$ values, implying that the defense substances are species-specific. Unexpectedly, the fish with good host function, for example Pseudorasbora parva, excrete the defense substances, too. The defense substances are possibly organ-specific in individual species; the organs essenstial for the existence of the species excrete the defense substances, allowing other parts to be invaded by the cercaria. 7) The cercaricidal fraction of Carassius carassius is detected only in the fish which have been collected during the summer time from May to September, Its secretion is not dependent on water temperature. Thus, it seems to be possible that the secretion of the defense substance would be stimulated through a contact between the fish and cercaria.
