A study on fundamental basis of four-constitution medicine from the principle of Yeokgyeong (사상의학(四象醫學)의 역철학적(易哲學的) 기초(基礎)에 관한 연구)

  • This study searched fundamental basis of four-constitution medicine from the principle of "Yeokgyeong(易經)" that is scientific foundations of Dongmu(東武) Ijema(李濟馬)'s four-constitution medicine based on system of "Yeokgyeong(易經)" and looked into the principle of our-constitution medicine ontologically. That is to say, the translation of five phase(五行) that represented in "Hwangjenaegyeong(黃帝內經)" regulated that substance of five phase is spleen-earth(脾土). But four-constitution medicine mentions the substance as heart-earth(心土) in place of spleen-earth. Because of it's standpoint, the differences on meanings between spleen-earth and heart-earth on the substance of five-phase becomes motive to interpret scientific system of four-constitution medicine fundamental wrongly. For that reason, the research of this title is needed. The results was summarized as follows. First, in ontological view point of structure of four-constitution, five phase is substance and phenomenon, in other words it includes earth of unrevealed substance and wood, fire, metal and water of self-manifestation of existence. Second, in axiological view point, the four-constitution represent principles and contents of four virtues of human nature. And so the innate four virtues ontologically based on four-constitution of heaven. Therefore a human being is endowed innately benevolence, courtesy, justice, intelligence of four virtues. Third, the concept of greater and lesser of Eum(陰, yin) and Yang(陽, yang) in Dongmu(東武)'s four-constitution medicine is four-constitution in "Yeokgyeong(易經)". Greater principle(太極) and four-constitution is a relation of substance and phenomenon. Fourth, the origin and structure of four-constitution medicine includes the structure and principle of natural philosophical Eumyang and four-constitution, the human-centric theory and sciences of human nature and natural laws and medical experience of traditional oriental medicine and medical principle.

Ethoxy-hydroxy-benzoic Acid; A Platelet Antiaggregating Substance from Acanthopanacis Cortex (오가피로부터 혈소판 응집억제작용 물질 Ethoxy-hydroxy-benzoic Acid의 분리)

  • The BuOH fraction prepared from the methanol extract of Acanthopanacis Cortex showed inhibitory activity against ADP-induced platelet aggregation. The inhibitory activity remained in ether layer when the BuOH fraction was refluxed with 5% aq. HCl-EtOH (1 : 1 mixture) and extracted with ether. From the ether layer, ethoxy-hydroxy-benzoic acid $(m.p.\;128{\sim}130^{\circ}C)$, a platelet antiaggregating substance, was isolated.

Aburatubolactam C, a Novel Apoptosis-inducing Substance Produced by Marine Streptomyces sp. SCRC A-20

  • In the course of screening for new antitumor substances, a novel cytotoxic agent inducing apoptotic cell death was isolated from the culture broth of marine bacterial strain SCRC A-20. Strain SCRC A-20 was separated from a mollusk and was chemotaxonomically identified as a Streptomyces sp. The cytotoxic substance was purified by organic solvent extraction followed by silica gel column chromatography and preparative TLC. HRFAB-MS determined its molecular formula to be $C_{30}H_{40}N_2O_5$ (MW 508). The 1D and 2D NMR spectral data demonstrated that the substance has a novel lactam structure of a 20-membered macrocycle coupled with a unique acyl tetramine and bicyclo[3.3.0] octane, which includes three methyl groups, six olefinic protons, five carbonyl groups, a conjugated diene and a dienone. The substance, named aburatubolactam C, appeared to be cytotoxic for various continuously proliferating tumor cells of human and murine origins. The $IC_{50}$ values determined by MIT assay were in the range of 0.3 to $5.8\mug/ml$. When Jurkat T cells were treated with $3\mug/ml$. of aburatubolactam C, the apoptotic DNA fragmentation was detectable within 3 h, indicating that the cytotoxic effect of aburatubolactam C on tumor cells is attributable to the induced apoptosis.

Alterations of c-Fos mRNA Expression in Hypothalamic-Pituitary-Adrenal Axis and Various Brain Regions Induced by Intrathecal Single and Repeated Substance P Administrations in Mice

  • The effect of substance P (Sub P) injected intrathecally (I.t.) on c-fos mRNA expression in vari-ous tissues was examined in the present study. We found that a single administration of Sub P(0.5 nM) caused an increase of the c-fos mRNA level in the hypothalamic-pituitary-adrenal(HPA) axis, hippocampus, and spinal cord. The time-course study showed that c-fos mRNA level was maximal at 10 min and began to decrease 30 min after the Sub P injection in all tis-sues, and the Sub P-induced increase of the c-fos mRNA level was returned to the control level 1 h after the injection. The kinetics of the c-fos mRNA expression in mice that were repeatedly injected with Sub P (every 30 min interval up to 4 times) were different in the HPA axis, hippocampus, and spinal cord. The increased c-fos mRNA level in the hypothalamus and the spinal cord induced by I.t. injected Sub P remained at a high level. In the pituitary gland, adrenal gland, and hippocampus, the increased level of c-fos mRNA expression gradually returned to the control level during the repeated substance P injections up to 4 times. Our results suggest that spinally injected Sub P-induced pain stress increases c-fos mRNA expres-sion in the spinal cord, hippocampus, and HPA axis. In mice repeatedly injected with Sub P, the kinetics of c-fos mRNA appear to be different varied from tissue to tissue.

Removal of haloacetonitrile by adsorption on thiol-functionalized mesoporous composites based on natural rubber and hexagonal mesoporous silica

  • Haloacetonitriles (HANs) are nitrogenous disinfection by-products (DBPs) that have been reported to have a higher toxicity than the other groups of DBPs. The adsorption process is mostly used to remove HANs in aqueous solutions. Functionalized composite materials tend to be effective adsorbents due to their hydrophobicity and specific adsorptive mechanism. In this study, the removal of dichloroacetonitrile (DCAN) from tap water by adsorption on thiol-functionalized mesoporous composites made from natural rubber (NR) and hexagonal mesoporous silica (HMS-SH) was investigated. Fourier-transform infrared spectroscopy (FTIR) results revealed that the thiol group of NR/HMS was covered with NR molecules. X-ray diffraction (XRD) analysis indicated an expansion of the hexagonal unit cell. Adsorption kinetic and isotherm models were used to determine the adsorption mechanisms and the experiments revealed that NR/HMS-SH had a higher DCAN adsorption capacity than powered activated carbon (PAC). NR/HMS-SH adsorption reached equilibrium after 12 hours and its adsorption kinetics fit well with a pseudo-second-order model. A linear model was found to fit well with the DCAN adsorption isotherm at a low concentration level.

Purification and Properties of HPS (Halitosis Prevention Substance) Isolated from Cumin (Cuminum cyminum L.) Seed

  • Halitosis is mainly caused by the presence of volatile sulfur-containing compounds (VSC's) produced by proteolytic periodontopathic bacteria in the oral cavity. Various mouth-rinses have been offered on the market as solutions to reduce halitosis. The aim of this study was to find a potent substance for the prevention of halitosis. The halitosis prevention substance (HPS) from cumin seed powder was purified by solvent extraction, silica gel column chromatography and preparative TLC to yield an oil phase (0.98%). Instrumental analysis such as FT-IR, $^1H$-NMR and $^{13}C$-NMR showed that HPS contained an -OH group, -HC=CH-, -COO-, and long chain acyl group. HPS was therefore determined to be 2-hydroxyethyl-${\beta}$-undecenate. HPS inhibited the growth of Fusobacterium nucleatum and Porphyromonas gingivalis, by 72.44% and 64.37% at $1{\times}10^{-2}\;M$, and by 99.85% and 91.62% at $5\;{\times}\;10^{-2}\;M$, respectively. It also inhibited the activity of L-methionine-${\alpha}$-deamino-${\gamma}$-mercaptomethane-lyase (METase), which was produced by oral microbes. Furthermore, the VSC production by oral microbes in the human mouth air decreased with increasing HPS concentration. These results suggested that HPS from cumin seed is an efficient halitosis prevention agent.

Objective evaluation of the color of tongue substance using L*a*b* color coordinates

  • The purpose of this study was to analyze whether quantitative evaluation of the color of the tongue substance using $L^*a^*b^*$ color coordinates system could minimize the problems arising from the different illuminating conditions or not. In controlled 4 different illuminating conditions (by natural light, flashlight, f-number, shutter speed),12 healthy subjects were photographed of their tongue substance through a digital camera (C-2100uz, Olympus Co.), both on the top surface and on the bottom surface of the tongue substance by two examiners, twice at 3 day intervals. Clinician evaluation was also performed grading the redness of the tongue substance in the form of 5-points scale by 6 clinicians. As a result, there was no significant difference in color differences between the color of the tongue substance and the reference red card in the 4 different illuminating conditions. Intra-rater reliability was satisfied and even though limitedly, inter-rater reliability was satisfied. Color differences were significantly correlated with the results by the clinicians, although they were applicable limitedly to specific illuminating conditions. Our results indicate that the application of the color differences in tongue diagnosis could not only evaluate the color information quantitatively, but also minimize the problems arising from the different illuminating conditions and that there was the significant difference in the visual evaluation of the red color of the tongue substance, both between the clinicians and between the illuminating conditions.

  • This study was conducted to further purify and characterize a polysaccharide fraction from whole parts of Duchesnea indica (Andr.) Focke, which was previously reported to show an antitumor activity by us. Mild acid-treatment of the fraction by 5% acetic acid yielded three subtractions (Fr. A,B and C) on a Sephadex G-25. Fr. A was composed of uronic acid, hexose, pentose and phenolic substance, Fr. C mainly phenolic substance, and Fr. B contained hexose. All three fractions exhibited colony stimulating factor activity.

Isolation and Characterization of an Antibacterial Substance from Rheum palmatum for Treatment of Bacterial Vaginosis (대황으로부터 세균성 질염 치료를 위한 항균성 물질의 분리 및 특성)

  • Rheum palmatum has traditionally been used as a preventive agent and medication against fever and infection. The aim of this study was to isolate and characterize an antibacterial substance from R. palmatum that is effective against bacterial vaginosis. A methanol extract from R. palmatum showed antibacterial activity against Lactobacillus vaginalis KC TC 3515, Chryseobacterium gleum KCTC 2904, and Sphingomonas paucimobilis KCTC 2834, which cause bacterial vaginosis. After extraction and pH control of the methanol extract from R. palmatum, we found that acidic and alkaline extracts did not show antibacterial activity. A neutral extract (50 mg/mL) displayed an inhibitory zone of 18 mm on a nutrient agar plate with C. gleum KCTC 2904. Fractions No. 11 and 12 among 41 fractions obtained by silica gel column chromatography produced inhibitory zones of 10 mm on nutrient agar plates with C. gleum KCTC 2904. $R_f0.15$ and $R_f0.17$ spots produced by TLC of fraction No. 11 showed antibacterial activity against C. gleum KCTC 2904. Isolation and purification of the peak at a retention time (Rt) of 9.427 min was achieved by HPLC of $R_f0.29spots$. The peak at Rt 9.427 min showed antibacterial activity against C. gleum KCTC 2904.
