Anti-inflammatory Flavonoids: Modulators of Proinflammatory Gene Expression

  • Plant flavonoids possess anti-inflammatory activity in vitro and in vivo. Although the action mechanisms are not fully understood, recent studies have clearly shown that certain flavonoids, especially flavone derivatives, express their anti-inflammatory activity at least in part by modulation of proinflammatory gene expression such as cyclooxygenase-2, inducible nitric oxide synthase and various cytokines. This review summarizes the recent findings of flavonoids modulating expression of proinflammatory molecules.

  • Cyclooxygenase(COX) metabolizes arachidonic acid to prostaglandins and thromboxanes. It has been reported that there is 'cross-talk' between COX-2 and nitric oxide synthase(NOS). Stimulation of eNOS of estrogen fed animal heart was not accompanied by the increase in COX-2 expression.(omitted)

Synthesis and Magnetic Properties of Expanded Graphite Oxide/Magnetic Nanoparticle Composite (Expanded Graphite 산화물과 자성 나노입자의 복합화와 자기적 특성)

  • The composites of expanded graphite oxide and magnetic nanoparticle (Ni and Co) were synthesized by using simple chemical method. From the raw material natural graphite, the expanded graphite was fabricated using sulfuric acid and $1^{st}$ heat treatment at $600^{\circ}C$ for 1 hour. The expanded graphite was changed to expanded graphite oxide by 2nd heat treatment at $1050^{\circ}C$ for 15 sec and chemical oxidation. The expanded graphite oxide/1-methyl-2-pyrrolidone solution reacts with the magnetic nanoparticle to form a magnetic graphite oxide composite. These graphite-based materials were characterized by x-ray diffractometer, Raman spectroscopy, transmission electron microscope, and vibration sample magnetometer. We expect that these results of this paper were become basis research of graphite oxide composite.

Ailanthoidol Derivatives and their Anti-inflammatory Effects

  • Ailanthoidol showed a strong $anti$-inflammatory effect in a previous result. Ailanthoidol derivatives were prepared for the $anti$-inflammatory test using Sonogashira coupling, iodine induced cyclization and Wittig reaction. $Anti$-inflammatory effects of the prepared ailanthoidol derivatives were examined in lipopolysaccharide (LPS)-stimulated RAW 264-7 macrophages. The results showed that some ailanthoidol derivatives inhibited significantly the production of inflammatory mediator nitric oxide.

Inhibitory Effect of a Sesquiterpene from Artemisia iwayomogi on Expression of Inducible Nitric Oxide Synthase by Suppression of I-κBα Degradation in LPS-stimulated RAW 264.7 Cells

  • A sesquiterpene was purified from Artemisia iwayomogi methanolic extract during the course of searching anti-inflammatory principle from medicinal plants. A sesquiterpene identified as armefolin inhibited the production of nitric oxide (NO) and attenuated inducible nitric oxide synthase (iNOS) protein level in lipopolysaccharide (LPS)-activated RAW 264.7 cells. Armefolin also down-regulated mRNA expressions of iNOS and pro-inflammatory cytokines, interleukin-$1{\beta}$ and interleukin-6 in LPS-activated macrophages. Moreover, armefolin suppressed the degradation of inhibitory-${\kappa}B{\alpha}$ (I-${\kappa}B{\alpha}$) in LPS-activated macrophages. These data suggest that armefolin from A. iwayomogi can suppress the LPS-induced production of NO and the expression of iNOS gene through inhibiting the degradation of I-${\kappa}B{\alpha}$. Taken together, armefolin from A. iwayomogi might be a candidate as promising anti-inflammatory agent.

Inhibitory Activity of Chinese Medicinal Plants on Nitric Oxide Synthesis in Lipopolysaccharide -Activated Macrophages

  • Nitric oxide (NO) produced in large amounts by the inducible nitric oxide synthase (iNOS) is known to be responsible for the vasodilation and hypotension observed in septic shock and inflammation. The inhibitors of iNOS, thus, may be useful candidate for the treatment of inflammatory diseases accompanied by the overproduction of NO. We prepared alcoholic extracts of Chinese medicinal plants and screened their inhibitory activity against NO production in lipopolysaccharide (LPS)-activated macrophages. Among the 80 kinds of extracts of herbal drugs, 15 extracts showed potent inhibitory activity of NO production above 80% at the concentration o$50\mu\textrm{g}/ml$. These potent extracts showed dose dependent inhibition of NO production of LPS-activated macrophages at the concentration of 50, 30,$10\mu\textrm{g}/ml$. Especially, Rhus chinensis, Senecio scandens and Wikstroemia indica showed most potent inhibition above 50% at the concentration of $10\mu\textrm{g}/ml$. These plants are promising candidates for the study of the activity-guided purification of active compounds and would be useful for the treatment of inflammatory diseases and endotoxemia accompanying the overproduction of NO.

  • In this study, a series of anion conductive organic/inorganic composite membranes with excellent ionic conductivity and chemical stability were prepared by introducing graphene oxide (GO) inorganic nanofiller into the quaternized poly(phenylen oxide (Q-PPO) polymer matrix. The fabricated organic/inorganic composite membranes showed higher ionic conductivity than the pristine membrane. In particular, Q-PPO/GO 0.7 showed the highest ionic conductivity value of 143.2 mS/cm at 90℃, which was 1.56 times higher than the pristine membrane Q-PPO (91.5 mS/cm). In addition, the organic/inorganic composite membrane showed superior dimensional stability and alkaline stability compared to the pristine membrane, and the physicochemical stability was improved as the content of inorganic fillers increased. Therefore, we suggest that the as-prepared organic/inorganic composite membranes are very promising materials for anion exchange membrane applications with high conductivity and alkaline stability.
