Cellulose degrading basidiomycetes yeast isolated from the gut of grasshopper in Korea (한국의 메뚜기의 장에서 분리된 Cellulose를 분해하는 담자균 효모)

  • Grasshoppers play vital role in the digestion of photosynthetically fixed carbons. With the aid of intestinal microflora, the grasshopper can degrade leaves constituents such as cellulose and hemicellulose. The purpose of this study was to examine cellulolytic yeast isolates from the gut of grasshoppers collected in Gyeonggi Province, South Korea. Among the yeast isolates, ON2, ON17 (two strains), and ON6 (one strain) showed positive cellulolytic activity in the CMC-plate assay. The sequence analyses of D1/D2 domains of the large subunit rDNA gene and the internal transcribed spacer (ITS) regions revealed that the strains ON2 and ON17 were most closely related to Papiliotrema aspenensis CBS $13867^T$ (100%, sequence similarity in D1/D2 domains; 99.4% sequence similarity in ITS) and strain ON6 related to Saitozyma flava (100% in D1/D2 domains; 99.0% in ITS). All these three yeast strains are capable of degrading cellulose; therefore, the members of endosymbiotic yeasts may produce their own enzymes for carbohydrate degradation and convert mobilized sugar monomers to volatile fatty acids. Thus, the endosymbiotic yeast strains ON2, ON17 (represents the genus Papilioterma) and ON6 (Saitozyma) belonging to the family Tremellomycetes, are unreported strains in Korea.

Bacteriocidal Effect of Chamical Sanitizers on Natural Microflora and Pathogenic Bacteria in Fish (화학적 살균소독제를 활용한 생선중 존재하는 자연균총 및 주요 식중독균 제어)

  • In this study, we evaluated bacteriocidal effect of CaO (scallop shell powder) for the reduction of microorganism in Scomber japonicus and Pseudosciaena ciean manchurica, and compared with main chemical sanitizers such as chlorine, ethanol, hydrogen peroxide. As a result, the effectiveness of CaO showed dramatic reduction rate for total aerobic bacteria, Escherichia coli and Vibrio parahaemolyticus. The reduction rates of total aerobic bacteria for Scomber japonicus and Pseudosciaena ciean manchurica showed $2.6{\times}10^3\;and\;3.2{\times}10^4$ respectively; Those of Escherichia Coli were $6.3{\times}10^3\;and\;1.2{\times}10^4$, those of Vibrio parahaemolyticus were $5.4{\times}10^4\;and\;5.6{\times}\2.6{\times}10^310^4$, respectively. According to this result, bacteriocidal effect of CaO was about $10^4$ and was similar to bacteriocidal effects of other three chemical sanitizers. According to these results, CaO can alternate the currently used chemical sanitizers due to its natural origin as well as the effectiveness for sterilization.

  • In this study, we evaluated bacteriocidal effect of CaO (scallop shell powder) for the reduction of microorganism in lettuce, and compared with main chemical sanitizers such as chlorine, ethanol, hydrogen peroxide. As a result, the effectiveness of CaO showed dramatic reduction rate for total aerobic bacteria, Escherichia coli, Bacillus cereus, Listeria monocytogenes, Staphylococcus aureus, and Salmonella Typhimurium and were $5.9{\times}10^3,\;1.3{\times}10^5,\;5.9{\times}10^3,\;2.7{\times}10^6,\;3.6{\times}10^3,\;4.5{\times}10^3\;and\;2.6{\times}10^4$, respectively. CaO did not show better disinfecting efficiency than chlorine or hydrogen peroxide which were used as sanitizer. In Bacillus cereus case, it showed $10^6$ reduction rate, and were $10^2{\sim}10^5$ times better reduction than ethanol sanitizer. According to these results, CaO can alternate the currently used chemical sanitizers due to its natural origin as well as the effectiveness for sterilization.

Characteristics of Traditional Wine-Koji and Isolation of Fungi (전통주 제조용 발효제의 특성 및 진균류의 분리)

  • Microflora and enzyme activity of traditional wine-koji were investigated. Bacteria was contained the greatest of $1.3\times10^7$ CFU/g-Koji, and its amylase and protease activities were 120.0 u/g and 36.6 u/g, respectively. 6 Kinds of yeast were isolated from the koji and identified as Hansenula alni (No 1), Hansenula canadensis (No 2), Hansenula silvicola (No.3), Hansenula califrnica (No 4), Hansenula beijerinckii (No 9) and Hansenula saturnus var. sturnus (No11). Furthermore, 14 kinds of mold were also isolated from the koji and identified as Rhizopus sp(No 1-41, 11 species) and Aspergillus sp.(No. 46, 53, 64, 3 species). Only Aspergillus sp. No 46 was showed a-amylase activity of 5.5 Unit and protease activity of Rhizopus sp. No 8 was the highest of 45.0 Unit.

Screening of DPPH radical scavenging and antimicrobial activity of extracts from local some native plants (국내 자생 식물추출물의 DPPH radical 소거활성 및 항균성 스크리닝)

  • This study was conducted to determine the antioxidant potentials of some available plants to source alternate antioxidants and antibiotics. The antioxidant activity was evaluated by determining the antioxidant activity reducing powers and 2,2-diphenyl-1-picrylhydrazyl (DPPH) scavenging activities of extracts from some local edible native plants. The DPPH radical scavenging activity of the extracts was found to have been 0.41-94.84%. The Elsholtziasplendens NAKAI extract ($85^{\circ}C$, ethanol, accelerated solvent extraction) showed the highest level of antioxidant activity. Eight samples of plant extracts were evaluated for their antimicrobial activities against three microflora (Bacillus subtilis, Escherichia coli, and Candida albicans) using disc diffusion assay. Two samples (Geranium krameri FR. Et SAV. and Cleyera japonica Thunb.) had much more intensive antimicrobial activities than the control. However, their DPPH free radical scavenging activity levels were only 14.50 and 13.85, respectively. It is suggested that they could be used as natural preservatives against bacterial contamination in cosmetics and foods, in place of the common synthetic preservatives currently used.

Analysis of Propionic acid Production in Joraengyi Rice Cake during Storage (조랭이떡 저장 중 천연유래 프로피온산 생성 특성 분석)

  • The objective of this study was to examine the natural origin of propionic acid in rice cakes by investigating the growth characteristics of the microflora and their production of propionic acid in the Joraengyi rice cake during storage period. The experiment was done in two stages within a period of three month: the rice cake fresh and contaminated with cocktail propionibacterium. The propionic acid production was analyzed according to the storage time and temperature by GC-FID (Gas chromatograph with flame ionization detector). During the storage of the fresh Joraengyi rice cake without alcohol at $30^{\circ}C$, about 95 mg/L of propionic acid was detected in 1st week, 330 mg/L in 4th week, 850 mg/L in 6th week, 970 mg/L in 8th week, and 1,040 mg/L in 12th week. During the storage of the Joraengyi rice cake which was contaminated with cocktail propionibacterium at $30^{\circ}C$, about 100 mg/L was detected from the rice cake with alcohol in the 1st week, 270 mg/L in 2nd week, about 470 mg/L in 4th week, and 660 mg/L in 8th week. This study demonstratesd the natural production of propionic acid during storage of the Joraengyi rice cake. To prevent the production, it is necessary to thoroughly manage hygiene and store it at refrigerated temperature or below $20^{\circ}C$.

Effects of Dietary Supplementation of Ginkgo Leaf and Pumpkin on the Growth Performance, Intestinal Microflora, Blood Biochemical Profile and Antioxidant Status in Broiler Chickens (사료 내 은행잎과 호박 첨가 급여가 육계의 성장, 소장 내 미생물 균총, 혈액 생화학 분석 및 체조직 내 항산화 작용에 미치는 영향)

  • The current study was performed to develop natural bio-active substances as additives for the production of high quality broiler chickens. A total of 120 male 3 day-old broiler chicks were randomly allocated to CON (control), GK2.5 (ginkgo leaf 2.5%), GK5.0 (ginkgo leaf 5.0%), PK2.5 (pumpkin 2.5%) and PK5.0 (pumpkin 5.0%) of five groups in cages (24 birds per group). All birds were fed corresponding diets from 3 to 35 d of age and determined growth performance and biological parameters including blood biochemical profiles, antioxidant status and intestinal microflora. During the entire feeding trial, GK5.0 and PK5.0 groups resulted in a significantly (P<0.05) higher FCR than GK2.5 and PK2.5 groups. Plasma triglyceride significantly (P<0.05) increased in GK5.0 group compared with the other groups, and the level of alanine transaminase (ALT) increased (P<0.05) in GK5.0 and PK5.0 groups compared with that in PK2.5 group. Dietary addition of ginkgo leaf and pumpkin significantly (P<0.05) increased superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) in the small intestine. Also, the addition of 2.5% ginkgo leaf significantly (P<0.05) increased the activities of SOD, GSH-Px and glutathione-S-transferase (GST) in the liver. Futhermore, muscle GST activity significantly (P<0.05) enhanced by dietary addition of ginko leaf and pumpkin. However, the level of lipid peroxidation (MDA) in the small intestine and muscle turned to be higher (P<0.05) in PK5.0 group. The colony forming units (CFU) of E. coli in intestinal digesta significantly (P<0.05) decreased in both ginko leaf and pumpkin supplemented groups compared with CON group. In conclusion, dietary addition of 2.5% ginko leaf and pumpkin as dietary sources can be applicable for the production of high quality broiler chickens.

Effect of Dietary Inuloprebiotics on Performance, Serum Immunoglobulin and Caecal Microflora in Broiler Chickens (이눌로프리바이오틱스의 사료 내 첨가가 육계의 성장능력, 혈액 면역물질 및 맹장 미생물에 미치는 영향)

  • The potential of encapsulated inuloprebiotics from domestic Jerusalem artichokes (Helianthustuberosus) as natural antibacterial growth promotor for an antibiotic replacement in broiler chickens was presently assessed through assays of growth performance, serum immunoglobulin production and influence on caecal microflora. Two hundred-forty, 1-day-old, male broilers (Ross 308) were randomly allotted to four treatments (T1-T4), with three replicate pens per treatment and 20 chicks per pen. Broiler chicks were fed a basal diet (T1: control) or basal diet plus antibiotics (T2: Chlorotetracycline, 0.10%), 300 ppm of the inuloprebiotics (T3), or 450 ppm of the inuloprebiotics (T4) for 35 days. Body weight, dressing percentage or weight of breast and thigh muscles relative to carcass weight of T3 and T4 broiler chickens was significantly (P<0.05) higher than T1 and T2 broiler chickens. The weight of abdominal fat from T3 and T4 broiler chickens were significantly (P<0.05) lower than that of T1 and T2 chickens. Serum immunoglobulins in the T3 and T4 groups were significantly (P<0.05) elevated compared to the T1 and T2 groups. The weight of immune organs, thymus and Bursa of Fabricius relative to live body weight in the T3 and T4 groups were significantly (P<0.05) higher than the T1 and T2 groups. Bifidobacteria and Lactobacillus, which are beneficial bacteria, were present in greater numbers in the caecum of T3 and T4 birds than T1 and T2 groups, whereas potentially harmful Escherichiacoli and Salmonella were present in lower numbers, with differences being significant (P<0.05). These results suggest that a diet supplemented with 300 ppm of inuloprebiotics has potential as an antibiotic replacement for organic livestock feed supplement intended to improve production of broiler chicken.

Application of a New Conjugation Method to Fish Pathogenic Bacteria Containing R Plasmid for the Analysis of Drug-Resistant Status in Aquaculture (새로운 conjugation 방법을 응용한 R plasmid 함유 어병세균의 분리와 양식장 내성균의 현황 분석)

  • To develop a new method of conjugation and to determine the distribution of R plasimds, we isolated multi-drug resistant strains from fish pathogenic bacteria in the farms of south and east seacoasts of Korea. Out of the 134 isolates examined, 10 showed resistance to chloramphenicol, tetracycline, streptomycin, ampicillin, colistin, nalidixic acid, oxolinic acid and kanamycin. One out of 10 multi-drug resistance bacteria, Vibfio damsela JE1 (V. damsela JE1), contained transferable R plasmid of chlorarnphenicol- tetracycline resistance genes and other nucleic acids encoding ampicillin and kanamycin resistance. The presence of the R plasmid was confirmed by conjugation using the chromocult medium (CC) as a selective and differential medium for transconiugants with identification based on the growth or colors of the colonies. The frequency of R plasmid transfer with filter mating method was come out much higher than that of broth mating method and appeared to be dependent upon the mating time and temperature. The optimum conditions for filter mating method were found to be 30$^{\circ}C$ and 24hrs as mating temperature and period, respectively, Moreover, donor cells with R plasmid, both isolate and standard bacteria, were shown to have an ability to transfer the plasmid against Escherichia coli K-12 HB101 (E. coli HB101) and Edwardsiella tarda (E. tarda) RE14 at fairly high frequencies, finally, we isolated 3 isolates of Sphingomonas sp., carrying R plasmid from 12 multi-drug resistant bacteria in normal microflora of the flounder (Paralichthys olivaceus) group used for the isolation of V emsela JE1 four months before. The same size and gene transfer chayateristics of R plasimds with those of V damsela JE1 confirmed that normal microflora have the reservoir activity for R plasmid in natural aquatic environment.

The Use of the Pathogen-specific Bacteriophage BCP8-2 to Develop a Rice Straw-derived Bacillus cereus-free Starter Culture (단일 박테리오파지를 이용한 볏짚 유래 Bacillus cereus free 스타터 컬쳐의 개발)

  • The purpose of this study was to develop a rice straw-derived Bacillus cereus (B. cereus)-free starter culture for traditional soybean fermented products using a B. cereus-specific bacteriophage, BCP8-2. To determine the optimal medium that supports the growth of rice straw-derived microorganisms and BCP8-2 activity, 5 different culture media were tested. The 5% ground bean (GB) medium was selected for further study. No B. cereus was detected in the BCP8-2-treated rice straw in GB medium, whereas B. cereus at a level of $10^7$ CFU/mL was recovered in the no-phage control. The total bacterial count reached approximately $10^9$ CFU/mL regardless of phage addition. When the 16S rRNA sequence-based microbial community was monitored using denaturing gradient gel electrophoresis (DGGE) and pyrosequencing, a similar microbial community was observed in the phage-treated and control samples. In conclusion, we demonstrate that phage can be used to prepare a rice straw-derived B. cereus-free starter culture with minimal effect on natural microflora.
