The Changes of Natural Microflora in Liver Sausage with Kimchi Powder during Storages

  • The objectives of this study were to apply the Baranyi model to predict the growth of natural microflora in liver sausage with added kimchi powder. Kimchi powder was added to the meat products at 0, 1, 2, and 3% levels. To determine and quantify the natural microflora in the meat products, total plate counts and counts of anaerobic bacteria and lactic acid bacteria were examined throughout the 28 d of storage. The obtained data were applied to the Baranyi growth model. The indices used for comparing predicted and observed data were $B_f$, $A_f$, root mean square error (RMSE), and $R^2$. Twelve predictive models were characterized by a high $R^2$ and small RMSE. The Baranyi model was useful in predicting natural microflora levels in these meat products with added kimchi powder during storage.

Biotransformation of Panax ginseng extract by rat intestinal microflora: identification and quantification of metabolites using liquid chromatography-tandem mass spectrometry

  • Background: In general, after Panax ginseng is administered orally, intestinal microbes play a crucial role in its degradation and metabolization process. Studies on the metabolism of P. ginseng by microflora are important for obtaining a better understanding of their biological effects. Methods: In vitro biotransformation of P. ginseng extract by rat intestinal microflora was investigated at $37^{\circ}C$ for 24 h, and the simultaneous determination of the metabolites and metabolic profile of P. ginseng saponins by rat intestinal microflora was achieved using LC-MS/MS. Results: A total of seven ginsenosides were detected in the P. ginseng extract, including ginsenosides Rg1, Re, Rf, Rb1, Rc, Rb2, and Rd. In the transformed P. ginseng samples, considerable amounts of deglycosylated metabolite compound K and Rh1 were detected. In addition, minimal amounts of deglycosylated metabolites (ginsenosides Rg2, F1, F2, Rg3, and protopanaxatriol-type ginsenosides) and untransformed ginsenosides Re, Rg1, and Rd were detected at 24 h. The results indicated that the primary metabolites are compound K and Rh1, and the protopanaxadiol-type ginsenosides were more easily metabolized than protopanaxatriol-type ginsenosides. Conclusion: This is the first report of the identification and quantification of the metabolism and metabolic profile of P. ginseng extract in rat intestinal microflora using LC-MS/MS. The current study provided new insights for studying the metabolism and active metabolites of P. ginseng.

미생물혼합제제 처리가 토양의 미생물상과 화학적 특성 및 시설 채소 생육에 미치는 영향 (Effect of Microorganism Mixture Application on the Microflora and the Chemical Properties of Soil and the Growth of Vegetables in Greenhouse)

  • BACKGROUND: The urgency of feeding the world's growing population while combating soil pollution, salinization and desertification requires suitable biotechnology not only to improve crop productivity but also to improve soil health through interactions of soil nutrient and soil microorganism. Interest in the utilization of microbial fertilizer has increased. A principle of nature farming is to produce abundant and healthy crops without using chemical fertilizer and pesticides, and without interrupting the natural ecosystem. Beneficial microorganisms may provide supplemental nutrients in the soil, promote crop growth, and enhance plant resistance against pathogenic microorganisms. We mixed beneficial microorganisms such as Bacillus sp. Han-5 with anti-fungal activities, Trichoderma harziaum, Trichoderma longibrachiatum with organic material degrading activity, Actinomycetes bovis with antibiotic production and Pseudomonas sp. with nitrogen fixation. This study was carried out to investigate the mixtures on the soil microflora and soil chemical properties and the effect on the growth of lettuce and cucumber under greenhouse conditions. METHODS AND RESULTS: The microbial mixtures were used with each of organic fertilizer, swine manure and organic+swine manure and compared in regard to changes in soil chemical properties, soil microflora properties and crop growth. At 50 days after the treatment of microorganism mixtures, the pH improved from 5.8 to 6.3, and the EC, $NO_3$-Na and K decreased by 52.4%, 60.5% and 29.3%, respectively. The available $P_2O_5$ and $SiO_2$ increased by 25.9% and 21.2%, respectively. Otherwise, the population density of fluorescent Pseudomonas sp. was accelerated and the growth of vegetables increased. Moreover, the population density of E. coli and Fusarium sp., decreased remarkably. The ratio of bacteria to fungi (B/F) and the ratio of Actinomycetes bovis to fungi (A/F) increased 2.3 (from 272.2 to 624.4) and 1.7 times (from 38.3 to 64), respectively. Furthermore, the growth and yield of cucumber and lettuce significantly increased by the treatment of microorganism mixtures. CONCLUSION(S): These results suggest that the treatment of microorganism mixtures improved the chemical properties and the microflora of soil and the crop growth. Therefore, it is concluded that the microorganism mixtures could be good alternative soil amendments to restore soil nutrients and soil microflora.

탁주발효에 있어서 발효미생물군의 변동에 대하여 (A study on the microflora changes during Takju brewing)

  • In order to study ecology of microorganisms during Takju brewing, microflora changes were examined fromm the start to the sixth day of Takju fermentation in 24 hours intervals. Takju made from rice, flour and dried sweet potato in a liter volume open container at the laboratory and a sanple of Takju brewing factory were studied for their microflora and their changes during fermentationl together with a sample of Kokja. Results obtained were as follows ; 1. The followings were the identified microorganisms in Kokja. The molds ; Absidia spinosa, Aspergillus parasiticus. The yeasts ; Candida melinii, Candida Solani, Hansenula anomala. The bacteria ; Luctobacillus casei, Leuconostoc mesenteroides, Bacillus subtilis, Bacillus pumilus. 2. Torulopsis inconspicua, Lactobacillus casei, Leuconotoc mesenteroides, Bacillus subtilis, Bacillus pumilus were isolated from main mash of laboratory-made Takju samples. The yeast, Torupsis inconspicua which was not present in Kokja and, probably of a contaminant yeast, dominated the yeast flora of Takju mash of rice, flour and sweet potato of labotatory brewing. The laboratory brewing lost also always showed large population of lactic acid bacteria flora. 3. None of the wild yeasts which were present in Kokja appeared in Takju mashes. The Kokja appears to be of no use as the yeast source for Takju fermentation. Also the Kokja appears to be of not so effective amylolytic and proteolytic enzyme sources considering the microflora characteristics. Probably the major role of Kokja in Takju fermentation may be to contribute in taste formation. 4. Inoculation of Sacharomyces cerevisiae into the mash to the level of $10^7$ ml at the start of fermentation greatly changed the ecological aspects eliminating conditions of rather slow rising of natural contaminant yeast populaiton and fermentation which might give rise to prosperity of lactic acid and Bacillus bacteria that would be avoidable. 5. Examination of microflora of the large factory scale Takju fermentation showed the quite similar pattern of microflora and their changes to that of the cultured yeast-inoculated laboratory batch Takju fermentation. The cultured yeast dominated as the only predominant microflora, and the lactic acid bacteria flora were completely suppressed and aerobic bacteria, greatly. Probably this may be the regular microflora pattern of normal Takju fermentation. The role of lactic acid bacteria and aerobic bacteria in Takju fermentation may not be clear yet from this experiment alone.

감마선 조사된 천연케이싱으로 제조한 소시지의 저장안정성 (Shelf Stability of Sausages Manufactured with Gamma-Irradiated Natural Casing)

  • We gamma-irradiated to natural casings from lamb and pork to improve sanitary quality and microbiological shelf stability of sausage stuffed in animal intestine-induced natural casing from lamb and pork. The microbial changes of irradiated casings and the microbial growth of the sausages with gamma-irradiated natural casings were observed. Population of total aerobic bacteria was decreased by gamma irradiation, dependent upon the irradiation dose. The number of microflora in lamb and pork casings was reduced by about 5.5 log cycle at 5 kGy. Population of total aerobic bacteria of sausages stuffed in non-irradiated lamb and pork casing was about 3.6 log CFU/g after just manufacture and was about 7 log after 10 day storage. However, population of total aerobic bacteria of sausages with 3 kGy-irradiated and 5 kGy-irradiated casings was 2.48 and 0.32 log in lamb, and 2.78 and 0.60 log in pork, respectively. Gamma irradiation to animal intestine- induced natural casings was effective and thus, can be used for enhancing shelf life of final products due to reduction of the number of contaminated microflora.

저장중 온도, pH, potassium sorbate를 이용한 딸기 paste의 natural microflora의 증식억제 효과 (Inhibitory Effects of Temperature, pH, and Potassium Sorbate against Natural Microflora in Strawberry Paste during Storage)

  • 시판중인 딸기의 미생물에 대한 잔류오염 정도를 파악하기 위하여 그들 잔류 미생물들을 총호기성균, 대장균군, 대장균, 유산균, 그리고 효모/곰팡이 등의 5가지로 구분하여 각각 측정하였다. 총호기성균은 약 8 log CFU/g, 대장균군은 약 2 log CFU/g, 그리고 유산균은 약 3 log CFU/g 정도씩 각각 검출되었다. 그리고 대장균과 효모/곰팡이는 검출한계(< 10 cells/g)미만에서 불검출되었다. 그리고 model system으로서 딸기 paste를 제조하여 7일 동안 저장하면서 저장온도, 보존제, 딸기 paste의 pH 등 3가지 hurdle들이 딸기에 잔류하는 총호기성균, 대장균군, 그리고 유산균의 증식억제에 미치는 영향을 파악하였다. 딸기 paste의 저장온도는 저온($4^{\circ}C$), 상온($20^{\circ}C$), 그리고 고온($37^{\circ}C$)을 선정하였고, 보존제는 0.1% potassium sorbate를 사용하였으며, 딸기 paste의 pH는 4와 7로 조정하였다. 이들 3가지 hurdle별 딸기 paste의 저장기간 중 잔류미생물에 대한 증식억제 효과는 저장온도 >pH> potassium sorbate 첨가의 순으로 나타났다. 그리고 이들 hurdle을 복합적으로 사용할 경우 딸기 paste의 pH를 4로 하여 potassium sorbate를 0.1% 첨가한 후, $4^{\circ}C$에서 저장하는 것이 가장 효율적으로 딸기 중 잔존미생물의 증식을 억제할 수 있었다.

조랭이 떡에 존재하는 자연균총 유전자 군집분석 및 천연유래 프로피온산 생성능 분석 (Genetic Analysis of Natural Microflora in the Stored Joraengyi Rice Cake and Their Capability of Propionic Acid Production)

  • 본 연구는 조랭이떡에 존재하는 미생물 군집분석과 자연균총의 프로피온산 생성능을 분석하였다. 조랭이떡의 자연균총을 유전자 분석을 통하여 군집분석을 수행해 프로피온산 생성균을 선별하였다. 선별된 프로피온산 생성균을 포도당이 첨가된 액체배지(TSB)에 배양해 온도별 생장 특성을 분석하고 가스 크로마토그래피(GC-FID)를 이용하여 프로피온산 생성량을 분석하였다. 에테르로 용매 추출하는 전 처리 방법을 확립하였고, 직선성, 검출한계, 정량한계, 회수율을 측정해 프로피온산 분석법을 검증하였다. 유통기한이 지나 미생물이 많이 자란 조랭이떡의 미생물 군집 분석 결과 총 98종의 균이 검출되었고 그 중 가장 큰 비중을 차지하는 우점종은 Lactobacills casei group으로 50.48%를 차지했고, Lactobacillus buchneri가 29.60%였다. 프로피온산 생성균은 Propionibacterium thoenii, P. cyclohexanicum, Propionibacterium_uc, P. jensenii, P. freudenreichii 등으로 나타났으며, 전체 자연균총의 약 2.4%를 차지했다. 조랭이떡 자연균총과 Lactobacillus 속은 14일 배양에서도 프로피온산을 생산하지 않았으나 P. cyclohexanicum, P. freudenreichii subsp. Shermanii, P. thoenii, P. jesenii는 4일차부터 프로피온산을 각각 263.47, 338.90, 325.43, $222.17{\mu}g/mL$를 생산하였고, 7일차에는 1,572.78, 2,496.63, 1,519.65, $2,660.41{\mu}g/mL$, 14일차에는 각각 2,462.02, 2,904.78, 2,220.64, $3,519.17{\mu}g/mL$로 프로피온산 생성량이 급격히 증가하였다. 본 연구의 결과로 살펴볼 때 전분과 포도당으로 구성된 떡류의 특성상 저장중 자연균총 미생물의 성장으로 높은 농도의 프로피온산이 천연유래로 생성될 수 있음을 알 수 있다. 본 연구는 떡류 등 전분질 식품의 프로피온산 등 천연유래 보존료 검출의 인정 및 기준규격 등 안전관리 자료로 활용될 것이다.

Midgut Microflora of Pure Mysore (Multivoltine) and N$B_4D_2$ (Bivoltine) Silkworm (Bombyx mori L.) Races During Late Larval Instars

  • The qualitative and quantitative changes of bacterial flora associated with the Pure Mysore (Multivoltine) and NB$_4$D$_2$ (Bivoltine) silkworm (Bombyx modi L.) midgut during third, fourth and fifth instars were studied. Larvae reared on mulberry leaves were dissected and their midgut bacterial populations were enumerated through serial dilution technique and after 72 hrs of incubation period at 28 $\pm$ 1$^{\circ}C$, the bacterial population was estimated. The results showed a highest mean value of 15$\times$10/ sup 6/ sup 6/ CFU/g and 28$\times$10/ sup 6/ CFU/g in Pure Mysore and NB$_4$D$_2$races, respectively, in midgut tissue of fifth instar larvae. The natural epiphytic microflora of mulberry leaves fed during the respective instars was also studied and found maximum 14$\times$10$^3$ CFU/g in leaves fed in third instars, followed by 5.3$\times$10$^3$ CFU/g and 2.1$\times$10$^3$ CFU/g in leaves fed during fourth and fifth instars, respectively. The bacterial flora colonized in midgut was found to be elaborating amylase, caseinase, gelatinase, lipase and urease enzymes. The highest percentages of isolates were amylase producers followed by protein and lipid splitters in Pure Mysore, whereas in NB$_4$D$_2$ protein splitter were dominated followed by lipase and amylase producers in NB$_4$D$_2$. The results indicate that the natural microflora may play a vital role in the digestion of ingested food materials in silkworms.

생식원료 야채의 전처리공정에서 Hypochlorous Acid의 미생물 제어 효과 (Effect of Hypochlorous Acid to Reduce Microbial Populations in Dipping Procedure of Fresh Produce as Saengshik Raw Materials)

  • 생식 원재료 전처리공정은 1차 세척-침지-2차 세척-절단공정으로 구성되며, 본 연구에서는 hypochlorous acid를 침지공정에 적용하여 미생물을 제어하고자 하였다. 전처리 단계에서 1차 세척 후에는 감자, 당근, 케일 및 신선초의 총균수는 4.7, 5.3, 5.6, 5.7 log CFU/g이 검출되었으며, 이를 물에 5분간 침지한 경우에는 각각 0.2, 0.2, 0.4, 1.1 log CFU/g를 감소시켰다. 반면 1차 세척 후 100 ppm hypochlorous acid 용액에 5분간 침지한 시료들은 0.5, 0.5, 1.3, 2.8 log CFU/g를 감소시켜 살균효과가 더 좋았으며, 구근류(감자, 당근)보다는 엽채류(케일, 신선초)에 효과적이었다. 하지만 흐르는 물로 세척이 끝난 채소류는 절단공정을 거친 후 다시 0.1~2.7 log CFU/g만큼 미생물수가 증가하였다. 따라서 감자, 당근, 케일, 신선초에 대해 절단공정을 먼저하고 나서 침지하는 공정을 한 경우, 물을 사용하였을 때는 각각 0.9, 0.6, 0.6, 0.4 log CFU/g, hypochlorous acid 용액에 침지한 후에는 각각 3.2, 2.3, 2.5, 2.6 log CFU/g를 감소시켰다. 결론적으로 1차 세척-절단-침지-2차 세척으로 공정을 개선하고 침지단계에서 hypochlorous acid 용액을 처리하는 것이 일반미생물과 유해미생물을 효과적으로 제어할 수 있는 방법으로 나타났다.

고추장 메주와 고추 품종별 고추장의 발효특성 비교 (Quality Changes of Traditional Kochujang Prepared with Different Meju and Red Pepper during Fermentation)

  • Kochujang, fermented hot pepper-soybean paste, was prepared by two common varieties of red pepper and natural fermented (NF) or pure cultured (PC) meju (soybean mass that is soaked, steamed, mashed and fermented by natural microflora or pure strain) and monitored their taste related component, enzyme activity and microflora during fermentation at $25^{\circ}C$ for 120 days for confirming possibility of kochujang fermentation control. The reducing sugars, amino type nitrogen (reference quality factor) and amino acid content in PC kochujang were 11.4%, 0.58% and 1,372.9 mg% respectively, 7.1%, 0.42% and 1,038.7 mg% in NF kochujang. It is concerned higher ${\alpha}-{\;}and{\;}{\beta}-{\;}amylase$ and, acid and neutral protease in PC kochujang during fermentation. The meju fermented by selected pure strain (A. oryzae CBU) can be applied to produced better quality of kochujang instead of natural meju.
