Antioxidative Effects of Flavonoids toward Modification of Human Low Density Lipoprotein (플라보노이드에 의한 사람 Low Density Lipoprotein 변형에 대한 항산화 효과)

  • The flavonoids are one of the most numberous and widespread groups of natural consituents. The low molecular weight of benzo-${\gamma}$-pyrone derivative are ubiquitous in plants and are vegetables nuts, seeds, leaves, flowers, and bark. The flavonoids constitute of a large class of compounds ubiquitous in plants containing a number of phenolic hydroxyl groups attached to ring structures conferring the antioxidant activity. Epidemiologic studies suggest that the dietary intake of antioxidants constitutes a risk factor for vasclar disease indicating that oxidation may be important in the pathogenesis of human athero-sclerosis. Elevated plasma low density lipoprotein (LDL) cholesterol concentration are associated with accelerated atherosclerosis, LDL is oxidized by smooth muscle cells resulting in several chemicals and physical changes of LDL. Oxidized LDL is responsible for cholesterol loading of macrophages foam cells formation and atherogenesis. There have been insulficient tests of the protective effects of flavonoids against LDL oxidation to make definitive statements about their structure activity relationships. How-ever hydroxylation of the flavone uncleas can appears to be advantageous because polyhydroxylated aglycone flavonoids are potent inhibitor of LDL modification. This identification may lead to new and more effective antioxidant strategies for abrogating the atherosclerotic process the leading cause of death and disability in industrialized societies.

In vitro antioxidant and free radical scavenging activities of stem extract of Euphorbia trigona Miller

  • Antioxidative and free radical scavenging properties of different stem extracts of Euphorbia trigona were evaluated and correlated with its total phenolic content. Aqueous, acetone and methanolic extracts of shade dried stem were obtained and were concentrated in vacuo. The antioxidant and free radical scavenging activities of stem extracts was determined by 2,2-Diphenyl-1-picrylhydrazyl (DPPH) scavenging assay, reducing power assay, deoxyribose degradation assay and $Fe^{2+}$ chelating assay. Total phenolic contents (TPC) were evaluated using Folin-Ciocalteu reagent. The results confirmed that the plant is a rich source of polyphenolic compounds which are invariably higher compared to other herbs. All extracts showed TPC in the range of 146.6 - 168.6 mg/g gallic acid equivalents at $300{\mu}g/ml$ of extract. Among the three extracts ME showed highest scavenging activity as evidenced by maximum scavenging of DPPH (83.2%), $OH{\bullet}$ radicals (94.81%), $Fe^{2+}$ chelating activity (88.59%) and a high reducing power 0.623 at $300{\mu}g/ml$. Our results demonstrate that Euphorbia trigona, an unexplored xerophytic plant could be potential source of natural antioxidants and phytotherapeutic agents. The plant possess invariably high amount of polyphenolic compounds with a broad spectrum of antioxidant properties and could be further used for food, feed and pharmaceutical applications.

Evaluation of the Antioxidant Properties of Pediastrum duplex and Dactylococcopsis fascicularis Microalgae

  • Interest in plant-derived food additives has increased recently, with efforts to identify natural antioxidant sources to replace synthetic antioxidants. This study evaluated the antioxidant effects of organic solvent fractions of 80% methanol extracts from Pediastrum duplex and Dactylococcopsis fascicularis microalgae. Among the solvent fractions tested, the ethyl acetate and n-hexane fractions from P duplex effectively scavenged 79.8% and 74.5% of DPPH free radicals, respectively. The chloroform fraction from D. fascicularis showed the strongest $H_2O_2$ scavenging ability (49.7%). The greatest scavenging of hydroxyl radicals (73.1%) was exhibited by the 80% methanol extract from P duplex. Aqueous residue and ethyl acetate fraction from P duplex provided the strongest nitric oxide scavenging (57.7%) and metal chelating effect (82.1%), respectively. Chloroform and ethyl acetate fractions from P duplex and n-hexane fraction from D. fascicularis exhibited significantly greater inhibition of lipid peroxidation than the commercial antioxidants. These results suggested that P duplex and D. fascicularis microalgae having potential anti oxidative compounds with various properties could be utilized in the food and pharmaceutical industries.

Opposite Effects of Vitamin C and Vitamin E on the Antifungal Activity of Honokiol

  • The aim of the present study was to evaluate the effects of two well-known natural antioxidants, vitamin C (VC) and vitamin E (VE), on the antifungal activity of honokiol against Candida albicans. The broth microdilution method was employed to test the antifungal activities of honokiol with or without antioxidants in the medium against C. albicans strain. Intracellular reactive oxygen species and lipid peroxidation were determined by fluorescence staining assay. Mitochondrial dysfunction was assessed by detecting the mitochondrial DNA and the mitochondrial membrane potential. We observed that VC could significantly potentiate the antifungal activities of honokiol while VE reduced the effectiveness of honokiol against C. albicans. In addition, VC accelerated honokiol-induced mitochondrial dysfunction and inhibited glycolysis leading to a decrease in cellular ATP. However, VE could protect against mitochondrial membrane lipid peroxidation and rescue mitochondrial function after honokiol treatment. Our research provides new insight into the understanding of the action mechanism of honokiol and VC combination against C. albicans.

Antioxidant and Anti-Obesity Properties of Pectolinarin-rich Cirsium setidens Nakai Fine Powder (Pectolinarin 고함유 곤드레 미세분말의 항산화 및 항비만 활성)

  • The aim of this study was to evaluate the basic data of Cirsium setidens Nakai fine powder (FPC), which will then be used in the development of functional fooditems. We measures and evaluated the level of pectolinarin content, phenol content, flavonoids content, antioxidants and anti-obesity properties of FPC. Our results from the study showed that the pectolinarin, phenol, and flavonoids contents of FPC measured at 10.95±0.1 5mg/g, 12.92±0.18 mg gallic acid equivalent (GAE)/g and 26.47±0.77 mg rutin equivalent (RE)/g, respectively. The exhibited antioxidant activity of FPC increased significantly depending on the dosage, and additionally. FPC did not show any cytotoxicity up to the dosage level of 500 μg/mL. During adipocyte differentiation, FPC significantly inhibited ROS production and lipid accumulation, compared with the control substance. These results suggest that FPC could be considered a promising resource of natural antioxidants and could serve a variety of health-improving roles in the production of functional food ingredients.

Modulation of Cytotoxic Effects of Resveratrol by Its Anti- or Pro-oxidant Properties (Resveratrol의 항산화 및 산화촉진 활성이 세포독성에 미치는 영향)

  • Resveratrol is a polyphenolic compound frequently found in the diet, and its physiological actions have been extensively investigated. In the present study, modulation of the antioxidant and cytotoxic properties of resveratrol at different pHs by various antioxidants were investigated. To measure its antioxidant effects, resveratrol was incubated at different pHs, including 6.5, 7.4, and 8.0. Resveratrol incubated at pH 6.5 showed significantly higher DPPH radical scavenging activity, whereas resveratrol incubated at pH 8.0 did not show antioxidant effects. Resveratrol produced much higher amounts of $H_2O_2$ at pH 8.0 than 7.4. The cytotoxic effects of resveratrol on HeLa cells were significantly enhanced by several antioxidants, including superoxide dismutase, N-acetyl cysteine, glutathione, and ascorbic acid. The present results suggest that resveratrol shows anti- or pro-oxidant effects in different cellular organelles according to the pH conditions, and blocking of reactive oxygen species from resveratrol enhances its cytotoxic effects.

Studies on the Stability of Natural Pigment Extracted from Ascidian shell (멍게 껍질(Ascidian shell)로부터 추출한 천연색소의 안정성에 대한 연구)

  • In this study, Ascidian shell pigment was extracted, first using a 100.0 % ethanol solvent, proceeding with the dilution of it with DMSO (Dimethyl sulfoxide). The extracted pigment was evaluated to verify the stability. The absorbance of light have been evaluated according to pH levels and using the color-difference meter. As a result, it could be seen that absorbance and chromaticity ${\pm}a$ values were most stable at a pH level of 7.0 By keeping the sample at a pH level of 3.0, it could be observed that the absorbance and the chromaticity ${\pm}a$ values were decreased. Based on this observation, it can be deduced that the discoloration of the pigment can be prevented if kept at a neutral pH level. When antioxidants were added, the absorbance of the pigment increased, and the best effects could be seen in the ${\alpha}-tocopherol$ and glutathione samples.

Evaluation of Anti-Oxidant and Anti-Inflammatory Activities of Ganoderma lucidum Cultured on Hulled Barley (겉보리에서 배양한 영지버섯 추출물의 항산화 및 항염증 효능 평가)

  • Background: Ganoderma lucidum cultured on hulled barley was investigated as a potential natural source of antioxidants and anti-inflammatory agents. Methods and Results: The yields from Ganoderma lucidum cultured on hulled barley water and ethanol extract were 17.69% and 25.77%, respectively. The antioxidant activity of Ganoderma lucidum cultured on hulled barley extracts was confirmed by various methods including assayss of 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2-azino-bis-(3-ethylbenzo thiazoline-6-sulfonic acid) (ABTS), nitrite radical scavenging, and $Fe^{3+}$ to $Fe^{2+}$ reducing power activity. The ethanol extract of Ganoderma lucidum cultured on hulled barley showed improved DPPH, ABTS and nitrite radical scavenging activity compared with the water extract. After treatment of RAW264.7 cells with Ganoderma lucidum cultured on hulled barley ethanol extracts, the cell viability compared with the control was 92.82%, even at a concentration of $3,000{\mu}g/m{\ell}$. The ethanol extract inhibited reactive oxygen species (ROS) generation in RAW264.7 cells stimulated with $H_2O_2$, even at low concentrations. In addition, the ethanol extract showed an inhibitory effects on the production of lipopolysaccharide-induced nitric oxide (NO) in RAW264.7 cells. Conclusions: This study suggests that the extract of Ganoderma lucidum cultured on hulled barley is a potential source of natural antioxidants and anti-inflammatory agents.

Antimicrobial and Antioxidant Activity of Hot Water and Ethanol Extracts of Ricinus communis L. Leaves and Fruits (피마자(Ricinus communis L.) 잎과 열매의 열수(熱水) 및 에탄올 추출물의 항균 활성과 항산화 효능)

  • RFW and RFE of castor fruit selected as part of the development of natural antimicrobials and antioxidants yielded 15.8% and 18.4% respectively. In the results of measuring the antimicrobial activity through paper disc method, the antimicrobial activity of castor fruits in ethanol extracts appeared. Especially, the activity was excellent in P. aeruginosa and S. aureus, and antimicrobial activity of C. was 1.5mm up to 16 hours. However, the proliferation of C. was observed again after 24 hours. In the MIC experiment results of RFE, S. aureus and P. aeruginosa showed 96% and 93% of antimicrobial activity, respectively. The DPPH radical scavenging activity of RLW and castor leaf ethanol extract showed $1.8{\pm}0.6%$ and $2.1{\pm}0.7%$ free radical scavenging activity at $1000({\mu}g/m{\ell})$. This study is expected to provide basic data for the development of antimicrobial agents and antioxidants using natural products.

Antioxidative Effects of Chitosan Meat Sausage (축육 소시지에 첨가한 키토산의 항산화 효과)

  • A large quantity of fat is added in processing of emulsion sausage. It is bring about deterioration and toxic substance by oxidation. Antioxidants are generally used as a protection material of oxidation for a storage and preservation of foods. In terms of stability of foods and health of human, development of a high effective antioxidants in a natural is required. Chitosan which is made from chitin by processing of deacetylase, has various function of antibiosis, antimutation and antioxidation and so on. We studied about the antioxidation of chitosan using to emulsion sausage. As a results, antioxidative effects of chitosan were increased with th large milecular weight and the higher concentration. M.W. 30,000 and M.W. 120,000 of chitosan have more 20% of antioxidation effect in emulsion sausage. Because chitosan have not 100% of antioxidation effect, we concluded that it has synergy effect by using with other natural material which has an effect of antioxidation.
