  • Experiments were carried out in order to compare the natural antioxidants in the acetonitril extracts of raw and baked seeds of Korean sesame and perilla by thin layer and gas chromatography. The sample was dissolved in n-pentane and extracted with acetonitril and the acetonitril extract was separated by thin layer chromatography using silica gel. The spots were detected by spraying with 2, 6 -dichloroquinone -4-chlorimide, phosphomolybdic acid and dimethylamine as chromogenic reagents. Natural antioxidant, such as ${\delta}-tocopherol$ detected in raw and baked sesame and perilla seed oil by TLC and sesamol was detected only in raw and baked sesame seed oil by GC.

Optimization of Food Waste Fermentation for Probiotic Feed Production with Yeast Kluyveromyces marxianus

  • For the probiotic feed production, aerobic liquid fermentation of pulverized food wastes was attempted with a yeast Kluyveromyces marxianus. After grinding finely, optimal fermentation conditions of the substrate was investigated by shaking culture. The most active growth of the yeast was shown at solid content of 10%. The proper addition of urea(0.5g/l), o-phosphate(0.4g/l), molasses(4g/l), and yeast extract (1g/1) increased cell growth rate and viable cell count. For optimizing, the nutrients were all added to substrate and fermentation was carried in 2 litre jar fermenter. For the stimulation of hydrolyzing enzyme excretion, mixed culture with Aspersillus oryzae was also conducted. In 12 hours of fermentation, viable cell count of the yeast Kluyveromyces marxianus amounted to the number of 1.4 $\times$10$^{10}$ /1 in the culture medium.

Change in composition of gut microbiota by exposure of natural medicines including Glycyrrhizae Radix in mice

  • Many of researches have revealed that human intestinal microbiota is related to health. Several diseases like obesity, diabetes, and hypertension are affected by the microbiota directly and indirectly. So, interventions with food and drug have been tried to change a composition of the microbiota to better condition. However, few natural medicines have elucidated to date. To understand an influence on microbiota by plant materials including Glycyrrhizae Radix, the extract of medicines were administered to mice and the feces were collected before and after the administration. The feces were analyzed by terminal restriction fragment length polymorphism (T-RFLP). The changes in composition of mice gut microbiota were detected and analyzed. The data could be utilized to further study about biological activities of the plant medicines.

A New Antipsychotic Effective Neolignan from Firmiana simplex

  • A new neolignan, named simplidin was isolated from the n-butanol extract of stem of Firmiana simplex, together with six known compounds, scopoletin (1), syrigaresinol (2), aquillochin (3), nitidanin (4), tamarixetin 3-rhamnoside (6), and quercitrin (7). On the basis of spectral and chemical evidence, simplidin (5) was determined to be rel-(7R, 8R)-4, 5, 9, 9'-tetrahydroxy-3,3'­dimethoxy-7-O-5', 8-O-4'-neolignan. All the six compounds were also isolated for the first time from this plant.

  • The effects of monazite and fixing agents on cotton fabric dyed with Perilla frutescens var. acuta extract were investigated. The proper monazite treatment concentration, temperature and time were 10%(o.w.b.), $50^{\circ}C$ and 60minutes. By various fixing agent treatment, $FeSO_4$ showed a relatively high K/S value and the order of K/S value decreased as follows, cation surface active agents, soybean and NaCl. And the monazite and $FeSO_4$ fixing agent showed higher anion emissity than those of untreated cotton and other fixing agents. The cotton fabrics showed improved color fastness by monazite and fixing agents treatments with the exception of light fastness. And the cotton fabrics fixed with fixing agents were showed effective bacterial reduction with the exception of NaCl.

Isolation of Hepatic Drug Metabolism Inhibitors from the Rhizomes of Curcuma zedoaria

  • The methanolic extract of the Rhizome of Curcuma zedoaria exhibited a significant prolongation of hexobarbital (HB)-induced hypnosis. Through liquid chromatography of an ether soluble fraction. monitoring by bioassay, three sequiterpenes, germacrone (A), curzerenone (B) and germacrone epoxide (C) were isolated as active consituents. A single treatment (100-200 mg/kg, i.p.) of each compound showed not only a significant prolongation of HB-induced sleeping time but also a significant inhibition of aminopyrine N-demethylase activity in mice, and further exhibited a typical type I binding spectra with oxidized rat hepatic cytochrome P-450 induced by phenobarbital. All of the compounds provoked a sleep episode at a subhypnotic dose of HB, implying that they possess CNS depressant properties.

  • Two new 4-hydroxybenzyl alcohol derivatives (1 and 2) were isolated from the methanol extract obtained from fresh tubers of Gastrodia elata together with 4-hydroxybenzyl methyl ether, 4-hydroxybenzyl alcoho, bis(4-hydroxyphenyl)methane, 4-hydroxybenzaldehyde, $\beta$-sitosterol and plamitic acid. 1 and 2 were identified as 3-O-($4^1$-hydroxybenzyl)-$\beta$-sitosterol and 4-[$4^1$-($4^{11}$-hydroxybenzyloxy)benzyloxy]benzyl methyl ether, respectively, according to the spectroscopic data.

Cytotoxic Terpenes and Lignans from the Roots of Ainsliaea acerifolia

  • The chromatographic separation of the MeOH extract of the roots of Ainsliaea acerifolia (Compositae) led to the isolation of six known terpenes and two known lignans. Their structures were identified by spectroscopic methods as mokko lactone (1), betulonic acid (2), betulinic acid (3), zaluzanin C (4), $1{\beta}-hydroperoxygermacra-4(15)$, 5, 10(14)-triene (5), pluviatilol (6), (+)-syringaresinol (7), and glucozaluzanin C (8). Compounds $1{\sim}4$ and 8 showed non-specific significant cytotoxicity against five human tumor cell lines with $ED_{50}$ values ranging from $0.36{\sim}5.54{\mu}g/mL$.

  • The new assay method was developed by using young chicks instead of frogs for screening of anti-emetic compounds from natural sources. Comparing with our previous method using leopard and ranid frogs, the advantages of the new method were not only completely parallel results but also decreasing standard errors. Hinesol and eudesmol were isolated from MeOH extract of Atractylodes lancea rhizome as the anti-emetic principles.

  • Phytochemical investigation of the 80% MeOH extract from the leaves of Allium victorialis var. platyphyllum resulted in the isolation of seventeen compounds; two terpenes, three norsesquiterpenes, one furofuran lignan, and eleven phenolic derivatives. Their chemical structures were characterized by spectroscopic methods to be trans-phytol (1), phytene-1,2-diol (2), icariside B2 (3), (6S,9S)-roseoside (4), sedumoside G (5), pinoresinol-4-O-glucoside (6), 2-methoxy-2-(4'-hydroxyphenyl)ethanol (7), 2-hydroxy-2-(4'-hydroxyphenyl)ethanol (8), Benzyl ${\beta}$-D-glucopyranoside (9), methyl ferulate (10), trans-ferulic acid (11), methyl-p-hydroxycinnamate (12), glucosyl methyl ferulate (13), linocaffein (14), siringin (15), 2-(4-hydroxy-3-methoxyphenyl)-ethyl-O-${\beta}$-Dglucopyranoside (16), and pseudolaroside C (17). All compounds were isolated for the first time from this plant.
