Comparison of In vitro Anti-Biofilm Activities of Natural Plant Extracts Against Environment Harmful Bacteria (천연물 성분을 이용한 환경 유해미생물의 biofilm 생성 저해능 비교에 관한 연구)

  • In this study, we investigated the in vitro anti-biofilm activities of plant extracts of chives (Allium tuberosum), garlic (Allium sativum), and radish (Raphanus sativus L.) against environment harmful bacteria (gram-positive Staphylococcus aureus and, gram-negative Salmonella typhimurium and Escherichia coli O157:H7). In the paper disc assay, garlic extracts exhibited the highest anti-biofilm activity. The Minimal Inhibitory Concentration (MIC) of all plant extracts was generally higher for gram-negative bacteria than it was for gram-positive bacteria. Gram-negative bacteria were more resistant to plant extracts. The tetrazolium dye (XTT) assay revealed that, each plant extract exhibited a different anti-biofilm activity at the MIC value depending on the pathogen involved. Among the plant extracts tested, garlic extracts (fresh juice and powder) effectively reduced the metabolic activity of the cells of food-poisoning bacteria in biofilms. These anti-biofilm activities were consistent with the results obtained through light microscopic observation. Though the garlic extract reduced biofilm formation for all pathogens tested, to elucidate whether this reduction was due to antimicrobial effects or anti-biofilm effects, we counted the colony forming units of pathogens in the presence of the garlic extract and a control antimicrobial drug. The garlic extract inhibited the E. coli O157:H7 biofilm effectively compared to the control antimicrobial drug ciprofloxacin; however, it did not inhibit S. aureus biofilm significantly compared to ciprofloxacin. In conclusion, garlic extracts could be used as natural food preservatives to prevent the growth of foodborne pathogens and elongater the shelf life of processed foods.

Effects of Natural Bioactive Substances on Hydroxyl Radical Mediated Cytotoxicity in Mouse Forebrain Cell Culture (쥐 전뇌세포 배양에 있어서 천연 생리활성물질이 하이드록실 라디칼에 의한 세포독성에 미치는 영향)

  • The biological effects of the water extracts of Rhus Verniciflua Stokes (RVS) were evaluated by protection against hydroxyl radicals. Antioxidative activities were measured using both 1,1-diphenyl-2-picrylhydrazyl (DPPH) and thiocyanate method. Also we used the Glucose oxidase (GO) 20 mU/$\textrm{m}{\ell}$ hydroxyl radical generating system in mouse forebrain cell culture. Water was used for ex-traction from RVS as a solvent which has high polarity especially. In DPPH method, the antioxidative activities of the crude water extract were stronger than any other extracts of low polar-solvents. In the antioxidative effects of mouse forebrain culture using 20 mU/$\textrm{m}{\ell}$ GO, cell viabilities were evaluated 65.6%, 68.8% at 1 $\mu\textrm{g}$. 5 $\mu\textrm{g}$ addition of crude water extracts (30 mg/$\textrm{m}{\ell}$) respectively. 10 $\mu\textrm{g}$ addition of crude water extracts had more than 86.1% cell viabilities, P<0.0l significantly, compared with the group treated with GO alone. In comparison with the antioxidative activities of several commercial antioxidants (ascorbic acid, $\alpha$-tocopherol, catalase, serum), 273 $\mu\textrm{g}$/$\textrm{m}{\ell}$ addition of crude water extracts (300 $\mu\textrm{g}$/$\textrm{m}{\ell}$) showed equivalent antioxidative effect to 25 uM ascorbic acid.

Studies on the Cytoprotective and Anti-inflammatory Effects of Isolate from Galla Rhois (오배자의 생리활성 연구)

  • Galla Rhois is a nest of parasitic bug, has been traditionally used for the treatment of the therapy of diarrhea, peptic ulcer, hemauria, etc., that showed various anti-inflammatory activity, and other biological properties. We studied the effect of Galla Rhois ethanol extract. we investigated whether compounds isolated from the ethanol extract of Galla Rhois, could modulate iNOS and COX-2 expression in RAW 264.7 macrophages stimulated with lipopolysaccharide (LPS). We found compounds that suppressed LPS-induced iNOS and COX-2 expression. Suppression of the expression of iNOS and COX-2 was in parallel with the comparable inhibition of the production of nitric oxide (NO) and prostaglandin E2 (PGE2). Our results suggest that compounds can inhibit NO and PGE2 productions through suppression of LPS-induced iNOS and COX-2 expression. Because COX-2- or iNOS-dependent mechanisms are involved in inflammation and tumor progression, our findings provide a new uncovering mechanism responsible for anti-inflammatory and antitumor effects of Galla Rhois.

The Anti-inflammatory Effects of Water Extract from Cordyceps militaris in Murine Macrophage

  • The aim of this study was to determine the in vitro anti-inflammatory effect of hot water extract from Cordyceps militaris fruiting bodies (CMWE) on lipopolysaccharide (LPS)-stimulated nitric oxide (NO) production, tumor necrosis factor-$\alpha$ (TNF-$\alpha$) and interleukin-6 (IL-6) release in RAW 264.7 cells. The treatment of macrophages with various concentrations of hot CMWE significantly reduced LPS-induced production as well as NO, TNF-$\alpha$ and IL-6 secretion in a concentration-dependent manner. These results suggest that CMWE have potent inhibitory effects on the production of these inflammatory mediators.

  • A phytochemical investigation of the ethanolic extract of Erythrina caffra flowers from an Egyptian origin yielded three C-flavonoidal glycosides; 5,7,4'-trihydroxyflavone-8-C-$\beta$-D-glucopyranoside (vitexin) (1), 5,7,4'-trihydroxyflavone-6-C-$\beta$-D-glucopyranosyl-(1 $\rightarrow$ 2)-$\beta$-D-glucopyranoside (isovitexin-2"-$\beta$-D-glucopyranoside) (2), 5, 7, 4'-trihydroxyflavone-6, 8-di-C-$\beta$-D-glucopyranoside (vicenin-2) (3) and one O-flavonoidal glycoside; kaempferol-3-O-$\beta$-D.glucopyranosyl) (1 $\rightarrow$ 2)-$\beta$-D-glucopyranoside (4). The structures of the isolated compounds (1 - 4) were elucidated using different spectral techniques (UV, 1D and 2D NMR and HRESIMS). This is the first report for the isolation of flavonoidal glycosides from Erythrina caffra. The antibacterial, antifungal, antimalarial, and antileishmanial activities of the isolates were evaluated. In addition, the cytotoxic activity of the ethanolic extract and the main fractions were tested using brine shrimp bioassay.

Characteristics and Analysis on the Refined Oil Component of Green-Tea (녹차의 정유성분에 대한 특성 및 분석)

  • This experiment extracted the natural green-tea using ethanol and obtained the refined oil component after filterated green-tea extract. This study tested the antimicrobial effect as characteristics experiment, and analyzed refined oil component with pH-meter and GC/MS. In the result of this experiment, it obtained the next conclusions. In the first result of extraction experiment, it could know that extraction ratio of refined oil component appeared about 9.0%. In the second result of characteristics experiment, it could certificate that in case of increasing the refined oil component in concentration of 100ppm and above, and according to passage of cultivation time, the number of S-aureus and E-coli in microbe decreased less and less. But in case of blank test not adding the refined oil component, the number of microbe increased more and more. In these phenomena, it could certificate that refined oil component of green-tea appeared antimicrobial effect against microbe. In the third result of instrumental analysis, refined oil component of green-tea appeared about 7.6 in 1% distilled water solution with pH-meter, and the aromatic components of benzene, bonyl acetate, campene, ${\alpha},{\beta},{\gamma}$-pinnene etcs from refined oil component of green-tea was detected with GC/MS.

  • Phytochemical studies on the whole extract of Sedum hybridum L., a Mongolian medicinal plant, has been undertaken to isolate active principles responsible for its anti-oxidative and antibacterial activities. Eighteen known compounds, i.e. (1) quercetin, (2) kaempferol, (3) herbacetin-8-O-${\beta}$-D-xylopyranoside, (4) myricetin, (5) gossypetin-8-O-${\beta}$-D-xylopyranoside, (6) gallic acid, (7) 2,4,6-tri-O-galloyl-D-glucopyranose, (8) 6-O-galloylarbutin, (9) myricetin-3-O-${\alpha}$-L-arabinofuranoside, (10) quercetin-3-O-${\alpha}$-L-arabinofuranoside, (11) caffeic acid, (12) ethylgallate, (13) (-) epigallocatechin-3-O-gallate, (14) palmitic acid, (15) stearic acid, (16) stearic acid ethyl ether, (17) ${\beta}$-sitosterol and (18) ${\beta}$-sitosteryl-O-${\beta}$-D-glucopyranose have been isolated and their molecular structures identified by spectroscopic analysis. Thirteen substances including seven flavonol components (1, 2, 3, 4, 5, 9 and 10), five gallic acid derivatives (6, 7, 8, 12 and 13) and caffeic acid (11) exhibited significant, dose-dependent, DPPH radical scavenging activity. Galloyl esters 12 and 13 were revealed to be main active principles for the antibacterial property of the extract of Sedum hybridum L.

The Extract of Limonium tetragonum Protected Liver against Acute Alcohol Toxicity by Enhancing Ethanol Metabolism and Antioxidant Enzyme Activities

  • The protective effect of EtOAc fraction of Limonium tetragonum extract (EALT) against alcohol-induced hepatotoxicity was assessed following acute ethanol intoxication in Spraque-Dawley rats. EALT (200 mg/kg p.o.) was administrated once before alcohol intake (8 g/kg, p.o.). Blood ethanol concentration, and the activities of alcohol metabolic enzymes, alcohol dehydrogenase (ADH) and aldehyde dehydrogenase (ALDH) in the liver were measured. Also, the formation of malondialdehyde (MDA) and the activities of antioxidant enzymes, superoxide dismutase (SOD), glutathione peroxidase (GSH-px), catalase were determined after acute alcohol exposure. Pretreatment of rats received ethanol with EALT significantly decreased blood ethanol concentration and elevated the activities of ADH and ALDH in liver. The increased MDA level was decreased, and the reduced activities of SOD, GSH-px and catalase were markedly preserved by the treatment with EALT. This study suggests that EALT prevent hepatic injury induced by acute alcohol which is likely related to its modulation on the alcohol metabolism and antioxidant enzymes activities.

A Study on the Dyeing of Ramie Fabric Treated with Medicinal Plants II. The Natural Dyeing On Ramie Fabric Using Artemisiae argyi Herba

  • This study was performed to investigate the effect of Artemisiae argyi Herba extract on the treatment of chromaticity and colorfastness. Artemisiae argyi Herba has been used as a Korean medicine. It is effective in removing skin disorders and suppressing pain. It is also good as a fungicide and in treating pruritis. In the long history of Korea, dyeing has been applied for a means representing the grace of natural and inner esthetic consciousness of man. Vegetable dyes give us such great benefits, diversified color; but no pollution. And ramie fabric has distinctive features such as beautiful brilliance, elegance, and strong durability. So, it is regarded as a special product of Korea traditionally. These studies were carried out to treat with acetate iron, dichloride copper and alum with a mordant to ramie fabric. The ramie fabric was died with Artemisiae argyi Herba extract. The results of experiment showed as follows: First, the chromophoric degree was the highest in acetate iron but not distinction in another mordants. Seocond, the light colorfastness was the highest in non treated and dichloride copper, but alum was the lowest. Third, the discoloration was alum and dichloride copper showed first grade in washing colorfastness. Abrasion colorfastness was not significant in this test. According the previous results, Artemisiae argyi Herba has an efficiency in removing skin disorders and suppressing pain. So it is considered that Artemisiae argyi Herba can be applied effectively to theraphy of fungicide and in treating pruritis.

Natural dyeing of silk fabric with Polygonum cuspidatum (호장근을 이용한 견직물의 천연염색)

  • In this study, the dyeing of silk fabric with Polygonum cuspidatum extracts was investigated. The contents of this study are as follows. First, the proper dyeing conditions were investigated by measuring the dye uptake (K/S value) that depended on the dyeing conditions when silk fabric was dyed with Polygonum cuspidatum extract. Second, the brightness (L), hue, and chroma differences that appear after mordanting with Al, Cu and Fe were investigated by measuring the CIELAB and Munsell values. And third, the colorfastness and antibacterial property were measured. When the silk fabric was dyed with Polygonum cuspidatum extract, the proper dyeing conditions were a colorant concentration of 90% v/v, a dyeing of time 100 minutes, a dyeing temperature of $70^{\circ}C$, and a dyeing of pH 3. In mordanting methods, the dyeabilities of post-mordanting were higher than those of premordanting. The hue value displayed yellow (Y) and yellow-red (YR) in cases of pre and post mordanting. The C value decreased by the mordanting of Polygonum cuspidatum extracts. Generally the colorfastness of mordanted fabrics was improved by mordanting. The dyed fabrics showed a 90.6% of Staphylococcus aureus reduction rate, and the dyed and mordanted fabrics showed 97.1% bacteria reduction rate. The dyed and mordanted fabrics showed above 90.5% Klebsiella pneumoniae reduction rate, and Cu mordant revealed the most effective bacterial reduction.
