18종의 한약재 MeOH 추출물과 EtOH 추출물의 Staphylococcus aureus에 대한 항균력을 조사한 결과 하고초, 소목, 오배자 및 향유는 5mg/ml 수준에서 항균활성을 나타내었으며 지실, 금은화 및 황금 등은 10mg/ml, 오미자, 초두구 및 희렴 등은 30mg/ml 이상에서 항균성을 보였다. 최소저해농도는 MeOH추출물에서는 $1{\sim}34mg/ml$, EtOH추출물에서는 $1{\sim}46mg/ml$ 농도에서 항균활성을 보였으며 분획 층 중 EtOAc층에서 성장억제효과가 가장 높게 나타났다. 항균력이 가장 우수한 것으로 확인된 소목, 오배자 및 황금추출물의 미생물 증식억제 효과를 조사하기 위해 증식배지에 각각의 추출물을 0, 100, 300 및 500ppm의 농도로 첨가하여 균주의 증식을 조사한 결과 배양후 24시간에 소목 추출물 무첨가구의 $OD_{620}$값이 0.4인 반면 100ppm 이상의 추출물 첨가 시 $0.02{\sim}0.03$정도로 균증식이 현저히 억제되었고, 오배자 추출물 무첨가구의 $OD_{620}$값이 0.4인 반면 300ppm 이상의 추출물 첨가 시 $0.02{\sim}0.03$정도로 균증식이 현저히 억제되었으며 황금 추출물 무첨가구의 $OD_{620}$값이 0.4인 반면 300ppm 이상의 추출물 첨가 시 $0.02{\sim}0.07$ 정도로 균증식이 현저히 억제되었다. 이상의 결과로 볼 때 제대염, 부종성 피부염 및 관절염 등의 다양한 가축질병을 일으키는 Staphylococcus aureusi의 감염을 예방하거나 또는 치료할 수 있는 한약재 추출물 사료첨가제의 개발 가능성을 확인 할 수 있었다.
아세로라는 천연 비타민 C 함량이 높아 우수한 원료이지만 안정화하기가 어려워 화장품 소재로서의 연구가 거의 이루어지지 못하고 있다. 따라서 본 연구는 아세로라 추출물을 안정화 시키는 혼합 리포좀 제형을 제조하여, 안전성 시험으로 BCOP assay 및 HET-CAM assay를 통한 안점막자극시험, 인체피부 일차자극시험을 통한 피부자극시험을 평가하였다. Tyrosinase 활성저해, melanin 생성저해를 통한 미백 효과 및 procollagen type-I C-peptide 생성을 통한 주름 효과를 평가하여 기능성 화장품 소재로서의 가능성을 확인하고자 하였다. 또한, 아세로라 추출물 혼합 리포좀이 함유된 크림을 제조하여 시험대상자를 대상으로 피부주름, 피부미백에 대한 인체효능평가를 진행하였다. 아세로라 추출물 혼합 리포좀의 BCOP assay, HET-CAM assay 및 인체피부 일차자극 시험결과에서 모두 자극을 보이지 않아 피부 및 안자극에서 안전함을 확인하였다. Tyrosinase 활성저해능을 확인한 결과, 아세로라 추출물 혼합 리포좀은 1,000 ㎍/mL 농도에서 75.8%의 tyrosinase 활성 저해능을 확인하였으며, melanin 생성 저해시험에서 B16F10 세포주의 생존율에 영향을 미치지 않는 아세로라 추출물 혼합 리포좀 1,000 ㎍/mL 농도에서 46.2%의 멜라닌 생성 저해능을 확인하였다. Procollagen type-I C-peptide에서 아세로라 추출물 혼합 리포좀은 HS68 세포주의 생존율에 영향을 미치지 않는 1,000 ㎍/mL 농도에서 152.1%의 collagen 합성능을 확인하였으나, MMP-1 활성억제는 확인할 수 없었다. 아세로라 추출물 혼합 리포좀이 함유된 크림에 대한 피부주름 및 피부멜라닌 인체효력평가에서 피부주름 지표와 피부멜라닌 지표에서 통계학적으로 유의하게 개선됨을 확인하였다. 이상의 연구 결과를 바탕으로 아세로라 추출물 혼합 리포좀이 안자극과 피부자극을 나타내지 않는 안전한 천연 소재임을 확인하였으며, 피부 주름 개선 및 미백 화장품 소재로써 활용 가능성이 높을 것으로 사료된다.
Natural product substances have historically served as the most significant also be prepared by source of new leads for pharmaceutical development. They can chemical synthesis(both semisynthesis and total synthesis) and have played a important role in the field of organic chemistry by providing synthetic targets. Rcently, they have also been extended for commercial purpose to refer to medicinal products, health functional foods, dietary supplements and cosmetics from natural sources. A large number of currently prescribed drugs have been either directly derived from or inspired by natural products. However, with the advent of robotics, bioinformatics, high throughput screening(HTS), molecular biology-biotechnology, combinatorial chemistry, in silico(molecular modeling) and other methodologies, the pharmaceutical industry has largely moved away from plant derived natural products as a source for leads and prospective drug candidates. The strategy for natural prduct industry is now changing from drug approaches to health foods by identifying effective natural products as preparations. In Korea, a lot of development of natural product based drugs have been done, but very few on health functional foods. The concept of natural product based health foods is not active components as lead compounds but standardized extracts or preparation mixed with other medicinal plants. The representative material has been recently known to be a standardized ginseng extract "Ginsana G 115" developed by Swiss Pharmaton company. The purpose of this presentation is to underline how natural products research continues to make significant contributions in the domain of discovery and development of new health functional foods. It is proposed to present the development of high value added health food or health functional foods through scientific investigation on efficacy and standardization of new materials form natural products.
This study was performed to investigate the effect of Gleditsia sinensis Lam. extract on the physicochemical properties of emulsion-type pork sausages during storage at $10^{\circ}C$ for 4 wk. Treatments were as follows: (C, control; T1, sodium ascorbate 0.05%; T2, Gleditsia sinensis Lam. 0.05%; T3, Gleditsia sinensis Lam. 0.1%; T4, Gleditsia sinensis Lam. 0.2%; T5, Gleditsia sinensis Lam. 0.1% + sodium ascorbate 0.05%). The values of pH, moisture content, lightness, redness, and sensory attributes were all significantly decreased, while the yellowness, chroma, hue angle, and texture properties were increased during storage with increase of the Gleditsia sinensis Lam. extract added. In addition, the antioxidant activity and antimicrobial activity in the sausages displayed significant increases (p<0.05). Therefore, although it was concluded that the addition of Gleditsia sinensis Lam. extract is not effective for improvement of the physical properties compared to chemical additives in sausages, it could be applied to meat products as a natural preservatives.
In order to develop a natural food preservative, dried salviae miltiorrhizae radix (Salvia miltiorrhiza) was extracted with several solvents, and then antimicrobial activity was investigated. The optimum extracting condition for the antimicrobial sustance from the sample, minimal inhibitory concentration (MIC) of the extracted substance against microorganisms were also examined. Antimicrobial activity of the initial ethanol extract from the sample was the strongest compared to those of other solvent extracts such as n-hexane, acetone, butanol, methanol and water. the optimum extractingcondition for antimicrobial substance from the sample was shaking extraction for 2 hours at room temperature incase that 10 volumes of absolute ethanol was added to crushed Saliva Miltiorrhiza. The ethanol extract had strong growth inhibition activity against Gram-positive Bacteria (MIC, 3.13-50$\mu\textrm{g}$/ml) such as B. cereus, B, subtilis, L. minocytogenes, S. aureus, Sc. Mutans. Among Grampositive bacteria tested, Bacillus species was the most susceptibile to the extracted substance. The antimicrobial activity of the ethanol extract from the sample was weak to Gram -negative bacteria yeasts, for example MIC for Gram-negative bacteria and yeasts was 0.8mg/ml and 0.4-0.8mg/ml , respectively.
Antifungal activities of the extracts from 26 medicinal plants were investigated utilizing paper-disk diffusion method and (1,3)β-glucan synthase inhibitory assay. (1,3)β-glucan synthase is considered as valuable target in the development of antifungal agents. Among the screened extracts, the ethyl acetate extract of Equisetum arvense, the ethyl acetate extract of Polygonum aviculare, the butanol extract of Crataegus pinnatifida and the n-hexane extract of Saussurea lappa showed significant antifungal activities on Candida albacans in both disk diffusion and enzyme assays.
Cornis fructus has been traditionally used for treating allergy, asthma, hepatitis, and chronic nephritis. It major chemical constituents have been saponins, phenolic acids and loganin. The main aim of the present study was to examine the effect of methanol extract from Cornis fructus on melanogenesis. Cell were cultured in the presence of methanol extracts from Cornis fructus for 48 h, and there were estimated total melanin content as a final product and activity of tyrosinase, a key enzyme, in melanogenesis. Methanol extract from Cornis fructus increased the melanin content and tyrosinase activity in a dose-dependent manner. Particularly, it was observed that only methanol extract $200\;{\mu}g/ml$ stimulated the melanin secretion in B16/F10 melanoma cells by 152% at 48 h treatment and the activity of tyrosinase was increased by 261% in the presence of same concentration.
Reactive oxygen species(ROS) or free radicals are produced in the pathogenesis of human diseases including atherosclerosis, diabetes, cancer, and aging. Antioxidants are associated with the prevention of ROS-induced tissue and cellular damage in the various diseases. This study investigated the antioxidative activities of the methanol extract of Alisma plantagoaquatica var. orientale and its main component under conditions of radical generation using allophycocyanin and ferric-thiocyanate assay. Alisol B 23-acetate as a main component was isolated from the methanol extract of Alisma plantago-aquatica var. orientale. In results, the extract of Alisma plantago-aquatica var. orientale showed inhibitory activity on AAPH [2,2'-azobis(2-amidinopropane)dihydrochloride]-induced protein oxidation. Also, the extract of Alisma plantago-aquatica var. orientale and alisol B 23-acetate inhibited lipid peroxidation. These results indicate that Alisma plantago-aquatica var. orientale and alisol B 23-acetate show promise as therapeutic agents for various damages involving free radical reactions.
In this study, the anti-inflammatory effects of ethanol extract of Rumex crispus L. and its fractions were investigated in RAW 264.7 macrophages. To evaluate the anti-inflammatory effects of extract, we studied nitric oxide (NO), prostaglandin $E_2$ ($PGE_2$), and tumor necrosis factor-alpha ($TNF-{\alpha}$) levels in RAW 264.7 cells. The ethanol extract of R. crispus L. significantly decreased NO production and the levels of other inflammatory factors, such as PGE2 and $TNF-{\alpha}$, in lipopolysaccharide (LPS)-stimulated macrophages in a dose-dependent manner. We also assessed the inducible nitric oxide synthase (iNOS) and the cyclooxygenase 2 (COX-2) protein expression by western blot. Ethyl acetate fraction of R. crispus L. had the strongest anti-inflammatory effect. These results suggest that ethyl acetate extract of R. crispus L. might be beneficial in the treatment of chronic inflammatory diseases.
Peptidoglycan in inserts and mammals is well known to improve biological functions in the host's immune system. However, it is unclear how Peptidoglycan exerted its anti-inflammatory capacity especially in clam worm (Marphysa sanguinea). In this experiment, the anti-inflammatory and antioxidant effects of clam worm extract treated with (PCWE) peptidoglycan (Micrococcus luteus) in RAW264.7 cells were examined by measuring MDA, catalase, SOD, GSH-Px and inflammatory cytokines (nitric oxide, iNOS, interleukin-$1{\beta}$ and tumor necrosis factor-${\alpha}$). PCWE significantly increased the activities of catalase, SOD and GSH-Px and decreased the level of MDA. Interestingly, PCWE induced activities of SOD and GSH-Px more than clam worm extract without peptidoglycan (CWE). In addition, PCWE decreased NO production, iNOS, COX-2, TNF-${\alpha}$ and IL-$1{\beta}$ better than CWE. Taken together, these results indicate that PCWE has the potential as a natural antioxidant and a therapeutic for inflammation-related diseases.
