Cell growth and the production of pullulan by Aureobasidium pullulan HP-2001, the UV-induced mutant of A pullulans ATCC 42023, increased with increased concentration of glucose up to 15.0% (w/v). Maximal production of pullulan in the flask scale was 27.65 g/l, when concentrations of glucose and yeast extract were 15.0 and 0.25% (w/v), respectively. Maximal conversion rate of pullulan from glucose as the carbon source was 0.37, when those of glucose and yeast extract were 5.0 and 0.15% (w/v), respectively. On the basis of total amount of pullulan, the conversion rate of pullulan from glucose, and utilization rate of glucose to cell mass and pullulan by A. pullulans HP-2001, the optimal concentrations of glucose and yeast extract for the mass production of pullulan were determined to be 10.0 and 0.25% (w/v), respectively, at which concentrations the production of pullulan and its conversion rate were 27.14 g/l and 0.27, respectively. Maximal production of pullulan with optimized concentrations of carbon and nitrogen sources by A. pullulans HP-200l in a 7-1 bioreactor was 32.12 g/l for 72 h culture, and that in a 100-1 bioreactor with the inner pressure of $0.4 kgf/cm^2$ was 36.87 g/l. Increased inner pressure of a 100-1 bioreactor resulted in a higher concentration of dissolved oxygen in the medium, which might enhance the production of pullulan by A. pullulans HP-2001.
Traditional herbal drugs of Korea, namely, Atractylodes Rhizome(蒼朮) and Atractylodes Rhizome White(白朮) which are derived from and old-grown rhizome and a newly-grown rhizome of Atractylodes japonica Koidz.(Compositae), respectively, were investigated their potential liver-protective activities against hepatotoxicity induced by $CCl_4$ intoxication in mice. Each herbal drug was extracted separately with chloroform, methanol and butanol. No extract of Atractylodes Rhizome showed a significant reduction of the duration of hypnosis produced by hexobarital after $CCl_4$ intoxication in mice. In addition, the the treatments of Atractylodes Rhizome extracts produced no decrease of aspartate aminotransferase(EC 2.6.1.1) activity in serum. However, the methanol extract of Atractylodes Rhizome White exhibited a marked protection from hepatotoxicity induced by $CCl_4$ intoxication in mice. It produced significant reductions of the duration of hypnosis and serum enzyme activity, but no other extract showed liver-protective activity against $CCl_4-induced$ hepatotoxicity.
To evaluate the radical scavenging activity of phenolic-rich fractions of Gardenia jasminoides, we first measured the levels of total polyphenols in hexane, ethyl acetate, and butanol fractions from the extract of G. jasminoides. The ethyl acetate fraction of G. jasminoides extract (GJ-EA) showed high level of phenolics, potent reducing power, and 2,2-diphenyl-1-picrylhydrazyl/2,2'-azino-bis 3-ethylbenzothiazoline-6-sulfonic acid radical scavenging effect. In addition, GJ-EA inhibited the overproduction of nitric oxide in lipopolysaccharide-activated BV-2 microglia. Furthermore, we found that GJ-EA suppressed $H_2O_2$-induced PANC-1 pancreatic cancer cell proliferation in a concentration-dependent manner and also reduced their migratory ability. These results suggest that GJ-EA may be a good source for functional foods with antioxidant and chemo-preventive activities.
Objectives : By investigating antimicrobial activity of natural extracts, identifying its usefulness as antibiotic material to oral bacteria. Methods : Antimicrobial activity tests of 25 natural extracts were implemented on 6 type strains alleged to cause dental caries and 10 clinical strains isolated and identified from dental caries. Results : Among medicinal plants, Coptis japonica, Scutellaria baicalensis, Dictamnus dasycarpus showed antimicrobial activity. Among lichens, 3 methanol extracts and 6 acetone extracts showed antimicrobial activity. Minimum Inhibitory Concentration(MIC) test of Usnea aurantiacoatra KoLRI 004184, an acetone extract of lichen with the highest antimicrobial activity, on 6 type strains involved in dental caries and 10 strains isolated from dental caries resulted as follows; Actinomyces oris 1041 $12{\mu}g/ml$, for Corynebacterium durum 3151 $13{\mu}g/ml$, for Rothia dentocariosa KCTC $3204^T$$14{\mu}g/ml$, for R. dentocariosa 911 $15{\mu}g/ml$, for R. dentocariosa 1822 $12{\mu}g/ml$, for Lacto-bacillus casei KACC $12413^T$$12{\mu}g/ml$, for L. acidophilus KACC $12419^T$$16{\mu}g/ml$, for L. rhamnosus 2421 $15{\mu}g/ml$, for Streptococcus mutans KCTC $3065^T$$15{\mu}g/ml$, for S. mutans 121 $13{\mu}g/ml$, for Streptococcus oralis 2221 $18{\mu}g/ml$, for S. salivarius KCTC 5512 $14{\mu}g/ml$, for S. salivarius 122 $12{\mu}g/ml$, for S. sanguinis KCTC $3284^T$$14{\mu}g/ml$, for S. sanguinis 912 $14{\mu}g/ml$, for Neisseria sp. KEM232 $12{\mu}g/ml$. Conclusions : Even a small amount of extract from lichen Usnea aurantiacoatra KoLRI 004184 showed very high level of antimicrobial activation against all 16 bacterial strains involved in dental caries.
To evaluate the influence of hepatic oxygen free radical systems on liver injury by topical p-phenylenediamine (PPD) application on rat skin, PPD (25 mg/16.5 $\textrm{cm}^2$) was topically applied to the abdominal region 5 times every other day and sacrificed. By PPD treatment, increasing rate of liver weight/body weight (%), serum activities of alanine aminotransferase and aspartate aminotransferase and decreasing rate of microsomal glucose-6-phosphatase activity were higher in the rats fed tungstate supplemented diet than those fed a standard diet. These findings indicate that group fed tungstate supplemented diet have more severe liver injury compared with group fed standard diet on topical PPD application. However, the activities of oxygen free radical generating enzymes such as xanthine oxidase (XO) and cytochrome P450 dependent aniline hydroxylase and those of oxygen free radical scavenging enzymes were not found to be different between these two animal groups. In the present study, a novel monitoring method to detect the generating of oxygen free radicals in liver extract was devised. Throughout this method, the oxidized PPD produced by oxygen free radicals was determined colorimetrically. The increasing rate of PPD oxidation by liver homogenate was higher in tungstate fed animals than in standard diet fed ones. Among the fractionations of liver extract, the mitochondrial and postmitochondrial fractions in the liver extract of tungstate fed animals led to a higher availability of PPD oxidation by PPD treatment compared with standard diet fed ones. In conclusion, these results suggest that an enhanced liver injury in tungstate fed animals treated with PPD may be due to oxygen free radicals produced in other systems except oxygen free radicals generating from cytosolic XO system. Especially, oxidative availability by PPD can be used for oxygen free radical detection in some tissue.

In this study, we investigate anti-allergic and anti-inflammatory effects of Leonurus sibiricus seed (LSS) extract in basophilic leukemia RBL-2H3 cells. To identify anti-allergic actions of LSS, the degranulation was evaluated in IgE and DNP-BSA stimulated RBL-2H3 cells. At the concentration of $100{\mu}g/ml$ of methanol (MeOH) extract and Methylene chloride (MC) and Ethyl acetate (EtOAc) fractions, the degranulation was significantly inhibited 16.7%, 16.7% and 27.9% respectively. And then, to assess anti-inflammatory effects of LSS, IL-4 and IL-13 mRNA level were detected in PMA/ionomycin (PI)-induced RBL-2H3 cells and cell proliferation and IL-4 mRNA level in isolated splenocytes from Balb/c mice. LSS MeOH extract and MC and EtOAc fractions significantly decreased the level of IL-4 and IL-13 mRNA in PI-induced RBL-2H3 cells and showed inhibitory effects on cell proliferation and expression of IL-4 mRNA level in mouse splenocytes. Taken together, these results suggest that LSS has potential anti-allergic and anti-inflammatory effects and EtOAc fraction is the most effective in regulating immune responses.

The purpose of this study was to investigate the possibility of herbal medicine complex extract (NI-01), which were prepared from 6 natural materials (Cinnamomum cassia Blume, Lonicerae Flos, Paeonia suffruticosa Andrews, Arctium lappa Linne, Schzandra chinesis Bailon, Elsholtzia ciliata Hylander), as a functional material for inhibition of atopic dermatitis. anti-oxidative activity was confirmed by measuring DPPH electron donating ability and ABTS+ radical scavenging ability. Cytotoxicity and NO inhibition were measured using RAW 264.7 cells to confirm anti-inflammatory efficacy. The test substance was orally administered to the pruritus-induced ICR mice to confirm the inhibition of pruritus. The bovine cornea opacity and permeability (BCOP) assay was performed to confirm safety for irritation. NI-01 showed high antioxidant activity in DPPH and ABTS+ methods. In the anti-inflammatory effect tests with RAW 264.7 cells, NO production was inhibited at NI-01 concentrations of 50 (14.9%) and 100 (4.2%) ㎍/mL, which indicated that the anti-inflammatory effect was increased in a concentration-dependent manner. NI-01 also showed anti-itching effect after inducing of itching by compound 48/80 in ICR mice. NI-01 was proved to be a non-irritant substance in BCOP assay. The results of this study suggested that the herbal medicine combined extract (NI-01) has high antioxidant, anti-inflammatory and anti-itching effects, and safety for irritation. Therefore, herbal medicine complex extract (NI-01) is thought to be highly applicable for the inhibitory ingredients of the atopic dermatitis.

The present study was conducted to evaluate the effects of dietary mangosteen peel extract (MPE) on growth performance, serum biochemistry, jejunum morphology, and cytokine levels in growing pigs raised at a high stocking density. A total of 120 male growing pigs (43.68 ± 0.48 kg) were randomly arranged in a 2 × 2 factorial design with stocking density (high; HD, 0.55 m2/pig and normal; ND, 0.82 m2/pig) and dietary MPE (0 or 5 g/kg) as factors. Each treatment had six replicates with four or six pigs per treatment. Feed and water were provided ad libitum for 6 weeks. The HD group exhibited lower final body weight, average daily gain, and average daily feed than the ND group (p < 0.05). None of the factors affected villus height to crypt depth ratio. Dietary MPE, but not stocking density, increased IL-10 levels in the serum com-pared to the non-supplemented control diet (p < 0.05). In the microbiome analysis, alpha diversity analysis showed significant reductions in the MPE-treated group only under normal density conditions. High density stress induced gut microbiome changes and these response was differ between normal and MPE diet fed pigs. Overall, each group exhibited different major microbial composition in the gut. In conclusion, there were significant changes in the major microbial composition in response to high-density stress, and this variation was influenced by dietary treatment.

Due to COVID-19, modern people have come to prefer natural substances as anxiety due to harmful environments and various stimuli has increased. Therefore, in order to find out the appropriateness of hibiscus, which is recognized as a non-toxic plant in traditional medicine, as a natural cosmetic material, the antioxidant effects (polyphenol, flavonoid, DPPH, ABTS) of hibiscus extract were studied, and the following results were obtained. First, the total polyphenol of hibiscus was found to be 433 ㎍/mL when the HSE was 100%. Second, the total flavonoids showed high antioxidant capacity at 488 ㎍/mL in 100% of the HSE. Third, the DPPH radical scavenging ability was found to be 94.04% in the undiluted HES and 89.54% in the diluted HSE 20%. Fourth, the ABTS radical scavenging ability was 98.95% in 100% of the HSE stock solution and 94.84% in the diluted HSE 20%, respectively, showing a high scavenging ability of more than 90%. As a result of these studies, it is thought that the hibiscus extract can be used as an antioxidant raw material for natural cosmetics in the future.
MeOH extract of the leaves of Xanthium strumarium L. were found to have cytotoxic activities against five human tumor cell lines. Cytotoxicity-guided chromatographic fractionation led to the isolation of the ${\alpha}-methylene$ containing sesquiterpenes, xanthatin, 8-epi-xanthatin and 8-epi-tomentosin. 8-epi-Xanthatin was found to be far more cytotoxic than 8-epi-tomentosin, which lacks the conjugated enone moiety present in 8-epi-xanthatin.
