Polarity affects the antioxidant and antimicrobial activities of jellyfish (Acromitus hardenbergi) extracts

  • Jellyfish is an emerging aquaculture species, farmed for Oriental cuisines and nutraceutical ingredients. This study aimed to examine antioxidative and antimicrobial potentials of various fractions of the jellyfish, Acromitus hardenbergi. The bell and oral arms of the jellyfish were sequentially extracted with petroleum ether (PE), dichloromethane (DCM), chloroform (CHCl3), methanol (MeOH), and water (H2O) to extract its bioactive in an increasing polarity gradient. Test fractions were assayed for antiradical activities using electron spin resonance spectrometry, β-carotene-linoleate model and Folin-Ciocalteu assay; and antimicrobial activity against 2 Gram-negative bacteria, 4 Gram-positive bacteria and 2 fungal species using the disc diffusion assay. All fractions were also subjected to Fourier Transform Infrared (FTIR) analysis to identify types of functional groups present. It was found that the hydrophilic extracts (H2O fractions) possessed the most effective radical scavenging activity (p < 0.05) while the lipophilic extracts (PE fractions) the most active antimicrobial activity, especially against Gram-positive bacteria (p < 0.05). Total oxidation substrates content was found to be highest in the PE fractions of jellyfish bell and oral arms (p < 0.05). FTIR data showed that the H2O and MeOH fractions contains similar functional groups including -OH, -C=O, -N-H and -S=O groups, while the PE, DCM, and CHCl3 fractions, the -CH3, -COOH groups. This study showed that A. hardenbergi contains antioxidants and antimicrobials, thereby supporting the traditional claim of the jellyfish as an anti-aging and health-promoting functional food. Bioassay-guided fractionation approach serves as a critical milestone for the strategic screening, purification, and elucidation of therapeutically significant actives from jellyfish.

  • Semantic role labeling task(SRL) is to extract predicate and arguments such as agent, patient, place, time. In the previously SRL task studies, a pipeline method extracting linguistic features of sentence has been proposed, but in this method, errors of each extraction work in the pipeline affect semantic role labeling performance. Therefore, methods using End-to-End neural network model have recently been proposed. In this paper, we propose a neural network model using the Biaffine Average Attention model for SRL task. The proposed model consists of a structure that can focus on the entire sentence information regardless of the distance between the predicate in the sentence and the arguments, instead of LSTM model that uses the surrounding information for prediction of a specific token proposed in the previous studies. For evaluation, we used F1 scores to compare two models based BERT model that proposed in existing studies using F1 scores, and found that 76.21% performance was higher than comparison models.

Anti-inflammatory Effects of Ethanol Extract of Various Korean Compositae Herbs in LPS-induced RAW 264.7 Macrophages

  • Objective : This study was designed to evaluate candidate materials as anti-inflammation agent from extracts of various Korean Compositae herbs in Hwaak mountain. Among Korea medicinal herbs, Ainsliaea acerifolia (AA) belongs to the Compositae family, has been used for the treatment of rheumatic arthritis. However, AA has not been previously reported to have an anti-inflammatory effect. Therefore, we investigated the anti-inflammatory effects of AA and its underlying molecular mechanisms in lipopolysaccharide (LPS)-induced RAW 264.7 macrophages. Methods : Cell viability was determined by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay in RAW 264.7 macrophages. Nitric oxide (NO) was measured with Griess reagent and pro-inflammatory cytokines were detected by enzyme immunoassay (EIA) kits in LPS-stimulated RAW 264.7 macrophages. Protein expressions of inducible nitric oxide synthase, and cyclooxygenase-2 (COX-2) and p65 subunit of nuclear $factor-{\kappa}B$ ($NF-{\kappa}B$) were determined by Western blot analysis. Results : Among 8 extracts of Korean Compositae herbs tested, AA showed the inhibition of NO production without cytotoxicity. Consistent with the observation, AA reduced the expression levels of iNOS and COX-2 proteins in LPS-simulated RAW 264.7 macrophages in dose-dependent manner. In addition, AA inhibited the productions of $TNF-{\alpha}$ and IL-6 in LPS-simulated RAW 264.7 macrophages. However, AA did not inhibit activation of p65 $NF-{\kappa}B$ in LPS-simulated RAW 264.7 macrophages. Conclusion : These results suggest that down-regulation of iNOS, COX-2 protein expression and $TNF-{\alpha}$ and IL-6 production by AA are responsible for its anti-inflammatory effects.

Inhibitory Effects of Scrophulariae Radix on β-hexosaminidase release and cytokine production in RBL-2H3 cells (현삼(玄蔘) 추출물이 RBL-2H3 비만세포에서 β-hexosaminidase 및 cytokine 분비에 미치는 효과)

  • Objectives : Traditional medicines isolated from natural products often have positive effects in the prevention and healing of various immune disorders, such as allergy and atopic inflammation. Scrophulariae Radix (SR) been used in oriental medicine used for treatment of acute and chronic inflammatory diseases. Mast cells are known to play important roles in the initiation of allergic reactions. In this study, we investigated the effects of SR ethanol extract on inflammatory responses in IgE-stimulated RBL-2H3 mast cells. Methods : Rat basophilic leukemia RBL-2H3 cells were purchased from Korean Cell Line Bank (KCLB No. 22256). Cell viability was measured by MTT assay. Assays for ${\beta}-Hexosaminidase$ Secretion : RBL-2H3 cells were sensitized with dinitrophenyl-ImmunoglobulinE (DNP IgE). The next antigen DNP-BSA ($25ng/m{\ell}$) was added for 10 minutes and the reaction was terminated after 5 minutes in the ice bath. To determine ${\beta}-Hexosaminidase$ release, supernatants were aliquoted into 96-well plates. Samples were mixed with substrate solution and incubated for 1 h at $37^{\circ}C$. Absorbance was measured with a spectrophotometer at 405 nm. IL-4 and tumor necrosis $factor-{\alpha}$($TNF-{\alpha}$) concentrations in cell culture supernatants were measured using enzyme-linked immunosorbent assay (ELISA) kits. Results : The cytotoxicity of SRE in RBL-2H3 cells was less than 5%. SRE inhibited DNP-IgE-imduced degranulation of mast cells in RBL-2H3 cells. Also significantly decreased the levels of inflammatory cytokine, IL-4 and TNF-alpha. In this study, the SRE showed potential anti-allergic and antiinflammatory. Conclusions : These results indicate that SRE could be inhibit the allergic response through suppressing the mast cell activation.

Antioxidant Activities of Ethanol Extracts from Solidago virga-aurea var. gigantea (울릉미역취 에탄올추출물의 항산화 활성)

  • Objectives : This study aimed to analyze contents of chlorogenic acid among major ingredients and determine the optimal solvent for the antioxidant activity from Solidago virga-aurea var. gigantea(SV) aerial part. Methods : Mature aerial part of extracted SV at the first crop on June 2018 was used. SV was mixed with each at the ethanol concentrate rates of water, 10, 30, 50, 70, and 100% and extracted them for three times for eight hours at $70^{\circ}C$. The contents of chlorogenic acid and seven kinds of antioxidant activities were measured in SV extracts. Results : The highest the contents of chlorogenic acid was from 10% ethanol extracts showed. Total polyphenol and flavonoids of SV extracts were 126.16 mg/g and 105.84 mg/g, respectively. Scavenging activities of DPPH, ABTS, and hydroxyl radical significantly increased in a dose-dependent in SV extracts, while it was almost similar with control at the concentration of $500{\mu}g/mL$. $Fe^{2+}$ chelating activity significantly increase in a dose-dependent in SV extract, it was lower than control. Nitrite scavenging activity increased, dose-dependent in the sequence of pH 1.2> pH 3.0 > pH 6.0 in SV extracts, while it was almost similar with control at the concentration of $500{\mu}g/mL$, pH 1.2, and pH 3.0. Conclusions : Thus, this study found that higher contents of chlorogenic acid and excellent antioxidant activity were contained in 10% ethanol extracts in mature aerial part of SV. It is expected to be used as basic data as natural antioxidant materials.

A Study on the Manufacture and Physical Properties of Liquid Adhesive for Wood Preservation - Focusing on the Synthesis of Gelatin and Carrageenan - (목재 보존용 액상 접착제의 제조 및 물성 연구 - 젤라틴과 카라기난 합성 중심으로 -)

  • In order to improve the usability of glue, a traditional adhesive material, and the gelation process of seaweed extract carrageenan, a liquid adhesive for wood preservation was prepared by mixing the two materials. The prepared wood adhesive was mixed with a 15 wt% aqueous solution of glue, λ-carrageenan, an antibacterial agent, an antifoaming agent, and the maximum adhesive strength of 1.80 Mpa was confirmed. As a result of comparison, the adhesive strength was superior to that of 5 different types of traditional natural adhesive ingredients (aqueous solution) and 1 type of polyvinyl acetate-based adhesive.

Tryptophan-derived Alkaloids from Hedera rhombea Fruits and Their Butyrylcholinesterase Inhibitory Activity

  • Alzheimer's disease (AD) is the most common age-related neurodegenerative disease in industrialized countries. It is estimated that about 47 million people living with dementia and the number of cases will be tripled by 2050. However, the exact mechanism of AD is not known, and full therapy has still not been found. Various tryptophan-derived alkaloids have been reported as promising agents for the treatment of AD. In the present study, a series of tryptophan-derived alkaloids were isolated and characterized from the methanol extract of Hedera rhombea fruit. Based on the analysis of their observed and reported spectroscopic data, their structures were identified as N-[4'-hydroxy-(E)-cinnamoyl]-L-tryptophan (1), N-[3',4'-dihydroxy-(E)-cinnamoyl]-L-tryptophan (2), N-[4'-hydroxy-(E)-cinnamoyl]-L-tryptophan methyl ester (3), and N-[3',4'-dihydroxy-(E)-cinnamoyl]-L-tryptophan methyl ester (4). These compounds were screened for anti-Alzheimer activity via their inhibition of acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) enzymes in vitro. As a result, compounds 3 and 4 showed moderate BChE inhibition with IC50 values of 86.9 and 78.4 μM, respectively, compared to those of the positive control [berberine (IC50 = 11.5 μM)]. However, all four compounds did not show significant inhibition of the AChE enzyme. This is the first time, the AChE and BChE inhibitory activities of these tryptophan-derived alkaloids were investigated and reported.

Simultaneous Determination of Eight Compounds in Lysimachia christinae by HPLC-DAD

  • Lysimachia christinae Hance was commonly used in Oriental medicine for treating the hepatitis virus, cholecystitis and cholagogic efficiency. According to the previous study, it possesses high antioxidant and anti-inflammatory activity. Simultaneous determination analytical method of isolated eight compounds, cynaroside (1), 2-(3,4-dimethoxyphenyl) ethyl O-α-L-arabinopyranosyl-(1→2)-O-[6-deoxy-α-L-mannopyranosyl-(1→3)] β-D-glucopyranoside (2), stearylester ricinoleic acid (3), (E)-4-(3,4-dimethoxyphenyl) but-3-en-1-yl palmitate (4), 2-hydroxy-24-methoxy-4-tetracosenoic acid (5), 2-hydroxy-24-propoxy-4-tetracosenoic acid (6), β-sitosterol (7), and androst-16-ene-3,6-diol (8) were established by using HPLC-DAD. This HPLC analysis was detected on a Dionex C18 column (5 ㎛, 120 Å, 4.6 mm × 150 mm) at 25℃. The mobile phase consisted of 0.1% trifluoroacetic acid and acetonitrile at a flow rate of 1 mL/min. Validation of the method was assessed by linearity, precision and accuracy test. Calibration curve was good at r2 > 0.9998. Limits of detection (LOD) ranged from 0.19 to 8.18 g/ml and Limits of quantification (LOQ) ranged from 0.19 to 24.80 g/ml. The relative standard deviations (RSD) values of precision test, intra- and inter- day, were less than 0.99% and 1.0%. The accuracy test results ranged from 98.81% to 106.49% and RSD values were less than 0.95%. These results showed that the HPLC-DAD method was very reliable and accurate for the quantity analysis of eight compounds in L. christinae extract for quality control.

Document Classification Methodology Using Autoencoder-based Keywords Embedding

  • In this study, we propose a Dual Approach methodology to enhance the accuracy of document classifiers by utilizing both contextual and keyword information. Firstly, contextual information is extracted using Google's BERT, a pre-trained language model known for its outstanding performance in various natural language understanding tasks. Specifically, we employ KoBERT, a pre-trained model on the Korean corpus, to extract contextual information in the form of the CLS token. Secondly, keyword information is generated for each document by encoding the set of keywords into a single vector using an Autoencoder. We applied the proposed approach to 40,130 documents related to healthcare and medicine from the National R&D Projects database of the National Science and Technology Information Service (NTIS). The experimental results demonstrate that the proposed methodology outperforms existing methods that rely solely on document or word information in terms of accuracy for document classification.

  • We extract the symptom of panic disorder from the context awareness environment. It extracts body context information through natural movement that exists in everyday life and uses a component of panic disorder. The ontology theory can be used to provide information on the degree of symptoms of panic disorder through inference process. For the components of panic disorder to the information processing based on ontology are defined as Classes. Panic disorder index is expressed through ontology modeling so that the condition of panic disorder can be known. The derivation of panic disorder component and panic disorder index will enable context awareness based information service for panic disorder. The context information is periodically synchronized with the context awareness on based device. Panic disorder can be used to improve the lifestyle of panic disorder.
