심바이오디니움으로부터 추출된 유효성분을 함유하는 신규 SUN BLOCK 물질에 관한 연구 (A Study on Nobel SUNBLOCK Materials Containing an Active Ingredient Extracted from Symbiodinium)

  • Symbiodinium is a dinoflagellate genus that coexists with coral reefs and is known to provide ultraviolet (UV) protection in nature through the synthesis of mycosporin-like amino acids (MAA). In order to develop a natural and ecofriendly sunblock for use in summer resorts, the possibility of using a Symbiodinium microbiome extract or an MAA was investigated. Two sunblocks, one containing 7% Symbiodinium extract and the other containing MAA were prepared to be tested on hairless mice and human skin. In this clinical study, the sun protection factor (SPF) and Protection factor of UV A (PFA) values of the sunblock containing either Symbiodinium extract or MAA were determined. The SPF values of the sunblock containing Symbiodinium extract and MAA were 10.43 and 10.83, respectively, and the PFA values were 3.42 and 3.39, respectively. Based on their PFA values of ≥2, the UV A protection can be graded as PA+ (low) for both the sunblocks. In addition, the UV-blocking extract of Symbiodinium has a low phototoxicity and cytotoxicity, reducing the possibility of a heavy feeling to the skin or a foreign-body sensation caused by residue build-up. The low toxicity feature of the major sunblock component will also prevent side effects, such as allergic reactions. Although Symbiodinium extract or MAA alone do not provide effectiveprotection against UV irradiation, their protection capacity can be complemented by the addition of inorganic UV-protecting compounds.

식용유지에 대한 붉나무 추출물의 항산화 효과 (Antioxidant Activity of Ethanol Extract from Rhus javanica $Linn\'{e}$ on Edible Oil)

  • 천연 항산화 물질을 분리하고자 식용 또는 한약재로 이용되는 붉나무를 75% ethyl alcohol로 항산화 성분을 추출하여 chloroform, ethyl acetate 등의 용매를 사용하여 separatory funnel, column chromatography로 분획하여 얻은 분획물의 항산화 효과를 검토하였다. 붉나무 추출물중 항산화 효과를 나타내는 유효성분은 ethyl acetate와 chloroform에 잘 용해되는 물질로, ethyl acetate로 분리한 분획물을 팜유에 600 ppm 첨가시 AI는 1.60, 돈지에서는 AI가 3.90으로 이들 용매로 분리한 분획물의 항산화 효과는 붉나무 추출 원액보다 높은 현상을 보였다. 붉나무 추출물을 팜유, 돈지 및 대두유에 농도별로 첨가하여 $60^{\circ}C$에 저장하면서 POV, AV 및 TBA가를 측정하여 이들 유지에 대한 산화안정성을 시험한 결과 Rancimat test 결과와 비슷하여 팜유에는 항산화 효과가 뚜렷하였고, 돈지에는 탁월한 결과를 보였다.

천마가 RANKL에 의해 유도된 파골세포의 분화에 미치는 효과 (Effects of Gastrodia elata Blune Water Extract on RANKL-induced Osteoclast Differentiation)

  • Impairment of balance between bone-resorbing osteoclasts and bone-forming osteoblasts result in bone disease. Especially, increased osteoclast formation and activity are responsible for bone diseases such as osteoporosis, rheumatoid arthritis, periodontal disease. Natural metabolites of plants have recently received much attention as an alternative tools for the development of novel therapeutic strategy. The aim of this study was to search the natural products to inhibit osteoclast differentiation and was to evaluate of its mechanism. Water extract of Gastrodia elata Blune significantly inhibited receptor activator of nuclear factor-κB ligand (RANKL)-induced osteoclast differentiation in bone marrow macrophages (BMMs) in a dose dependent manner. However, water extract of Gastrodia elata Blune did not affect cytotoxicity when compared with control. The mRNA expression of c-Fos, NFATc1, and TRAP induced by RANKL was inhibited by water extract of Gastrodia elata Blune treatment. Also, water extract of Gastrodia elata Blune inhibited the protein expression of c-Fos and NFATc1 expression in BMMs treated with RANKL. Water extract of Gastrodia elata Blune suppressed the phosphorylation of p38 induced by RANKL. In general, RANKL considerably inhibited the expression level of Id2 and MafB known as negative regulators of osteoclastogenesis, but RANKL did not inhibit Id2 and MafB expression in BMMs when it was co-treated with Gastrodia elata Blune. Taken together, these results suggest that Gastrodia elata Blune may be a useful drug in the treatment of bone-related disease.

겉보리에서 배양한 영지버섯 추출물의 항산화 및 항염증 효능 평가 (Evaluation of Anti-Oxidant and Anti-Inflammatory Activities of Ganoderma lucidum Cultured on Hulled Barley)

  • Background: Ganoderma lucidum cultured on hulled barley was investigated as a potential natural source of antioxidants and anti-inflammatory agents. Methods and Results: The yields from Ganoderma lucidum cultured on hulled barley water and ethanol extract were 17.69% and 25.77%, respectively. The antioxidant activity of Ganoderma lucidum cultured on hulled barley extracts was confirmed by various methods including assayss of 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2-azino-bis-(3-ethylbenzo thiazoline-6-sulfonic acid) (ABTS), nitrite radical scavenging, and $Fe^{3+}$ to $Fe^{2+}$ reducing power activity. The ethanol extract of Ganoderma lucidum cultured on hulled barley showed improved DPPH, ABTS and nitrite radical scavenging activity compared with the water extract. After treatment of RAW264.7 cells with Ganoderma lucidum cultured on hulled barley ethanol extracts, the cell viability compared with the control was 92.82%, even at a concentration of $3,000{\mu}g/m{\ell}$. The ethanol extract inhibited reactive oxygen species (ROS) generation in RAW264.7 cells stimulated with $H_2O_2$, even at low concentrations. In addition, the ethanol extract showed an inhibitory effects on the production of lipopolysaccharide-induced nitric oxide (NO) in RAW264.7 cells. Conclusions: This study suggests that the extract of Ganoderma lucidum cultured on hulled barley is a potential source of natural antioxidants and anti-inflammatory agents.

꼬시래기 유래 아가로즈와 블루베리 열매 추출물을 이용한 화장용 에멀젼 개발 (Development of Cosmetic Emulsion Using Blueberry Fruit Extract and Agarose from Gracilaria verrucosa)

  • The need for natural cosmetic ingredients has been increasing over the world nowadays. Agarose, a natural polymer from red seaweeds, has high hydrophilic character and a function of scaffolder. As skin moisturizer, agarose is adequate for percutaneous absorption. While, blueberry fruits extract possesses rich procyanidins and anthocyanins which show health benefits, anti-oxidant effect, anti-aging and anti-melanogenesis. Stability, sensory preference, skin trouble of the emulsion formula are important for cosmetic product development. In this study, we manufactured an emulsion formula for skin moisturizers using the two ingredients and tested emulsion stability and skin trouble. Total phenolic contents of the blueberry fruits extract were evaluated as well as tyrosinase inhibitory and collagenase inhibitory activities. $IC_{50}$ values of blueberry fruits extract for anti-tyrosinase and anti-collagenase activities were 168 and $112{\mu}g/mL$, respectively using gallic acid as a control ($64.8{\mu}g/mL$). The stability (pH and viscosity) of the formula containing 2% blueberry fruits extracts and 0.1% agarose was measured at five different temperatures (room temp., $25^{\circ}C$, $55^{\circ}C$, $45^{\circ}C$, $55^{\circ}C$) under the sun light at 2 day intervals for 12 days. There has been little pH change at the different temperatures. According to the sensory evaluation, there was no significant flavor, discoloration and physical changes of the formula at $25-65^{\circ}C$. These results suggest that emulsion formula containing blueberry extract and agarose could be used as a candidate for lotion and essence products.

Evaluation on Antimicrobial Activity of Psoraleae semen Extract Controlling the Growth of Gram-Positive Bacteria

  • This study investigated bacterial growth-inhibitory effect of 69 therapeutic herbal plants extracts on 9 bacterial strains using a disc diffusion assay. Especially, the antimicrobial activity of Psoraleae semen, which showed different activity on pathogenic Gram-positive and Gram-negative bacteria, was evaluated by MIC (minimal inhibition concentration) and biofilm formation assay. The effect of Psoraleae semen extract on bacterial cell membranes was examined by measurement of protein leakage (optical density at 280 nm) and scanning electron microscope (SEM). No clear zone was formed on discs containing Gram-negative bacteria, but Gram-positive bacteria exhibited clear zones. The MICs of Psoraleae semen extract were $8{\mu}g/mL$ for Streptococcus mutans, and $16{\mu}g/mL$ for Enterococci and Staphylococcus aureus. In addition, biofilm formation was inhibited at concentration $8-16{\mu}g/mL$. Protein leakage values and SEM images revealed that cell membranes of Gram-positive bacteria were impaired following exposure to the extract. Further, the extract inhibited the growth of Listeria monocytogenes in sausages. These results indicate that Psoraleae semen extract could be utilized as a natural antimicrobial agent against Gram-positive bacteria.

Natural Iminosugar Derivatives of 1-Deoxynojirimycin Inhibit Glycosylation of Hepatitis Viral Envelope Proteins

  • A silkworm (Bombyx mori L.) extract known to contain naturally occurring iminosugars, including 1-deoxynojirimycin (1-DNJ) derived from the mulberry tree (Morus alba L.), was evaluated in surrogate HCV and HBV in vitro assays. Antiviral activity of the silkworm extract and one of its purified constituents, 1-DNJ, was demonstrated against bovine viral diarrhea virus (BVDV) and GB virus-B (GBV-B), both members of the Flaviviridae family, and against woodchuck hepatitis virus (WHV) and hepatitis B virus (HBV), both members of the Hepadnaviridae family of viruses. The silkworm extract exhibited a 1,300 fold greater antiviral effect against BVDV in comparison to purified 1-DNJ. Glycoprotein processing of BVDV envelope proteins was disrupted upon treatment with the naturally derived components. The glycosylation of the WHV envelope proteins was affected largely by treatment with the silkworm extract than with purified 1-DNJ as well. The mechanism of action for this therapy may lie in the generation of defective particles that are unable to initiate the next cycle of infection as demonstrated by inhibition of GBV-B in vitro. We postulate that the five constituent iminosugars present in the silkworm extract contribute, in a synergistic manner, toward the antiviral effects observed for the inhibition of intact maturation of hepatitis viral particles and may complement conventional therapies. These results indicate that pre-clinical testing of the natural silkworm extract with regards to the efficacy of treatment against viral hepatitis infections can be evaluated in the respective animal models, in preparation for clinical trials in humans.

병원내에서 분리된 메티실린내성 황색포도상구균에 대한 무화과잎 추출물의 항미생물효과 (Synergistic Antibacterial Activity of Fig (Ficus carica) Leaves Extract Against Clinical Isolates of Methicillin-resistant Staphylococcus aureus)

  • 무화과는 뽕나무 과에 속하며 세계에서 가장 오래된 과일 중 하나이고, 국내에서는 주로 소화장애, 염증성궤양 그리고 가려움증 등에 사용되어 왔다. 본 연구에서는 무화과 잎을 메탄올로 추출한 후 임상진료실에서 분리된 메티실린 내성 황색포도상구균(MRSA)에 대한 항균활성을 보았다. 그 결과 분리된 MRSA 1-20에서 최소억제농도 (MIC)는 2.5 mg/mL에서 20 mg/mL 농도로 측정되었으며, 최소살균농도(MBC)는 5 mg/mL에서 20 mg/mL 농도로 관찰되었다. Oxacillin이나 ampicillin과 병용투여 시 4-8배 이상의 MIC/MBC농도가 감소되었으며, FICI 값도 0.0375-0.5이하로 병용효과를 나타내었다. 더불어 메탄올추출물 단독 사용시 보다 항생제와 병용투여 시 빠른 사멸효과를 보였다. 결론적으로 무화과 잎이 메티실린 내성 황색포도상구균에 항균활성을 나타냄을 확인하였으며, 메티실린 내성 황색포도상구균 감염에 천연 치료제로서의 가능성을 확인하였다.

녹차추출물을 첨가한 초고추장의 품질향상 (Quality Improvement of Chokochujang by the Addition of Green Tea Extract)

  • 자체 제조한 초고추장에 무첨가, 주정 3% 첨가, 녹차추출물 1, 3, 5% 첨가, 주정 1%와 녹차추출물 1% 그리고 주정 1%와 녹차추출물 2% 첨가한 시료를 각각 준비한 후 3주 동안 25, $37^{\circ}C$에 저장하면서 총균수, pH, 산도, 당도, 관능검사를 수행함으로써 초고추장의 품질변화를 조사하였다. 먼저 초고추장의 우점균주를 동점하기 위하여 분리된 균주를 16S rDNA 염기서열 분석을 수행한 결과 분리균주는 Bacillus amyloliquefaciens ER282로 최종 동정되었다. 한편 녹차추출물 자체를 1, 3, 5, 10% 농도에서 항균력을 시험한 결과 높은 항균효과를 나타내었다. 항균력이 높은 녹차추출물을 추가적으로 첨가한 초고추장 시료의 저장중의 총균수 변화를 조사한 결과 $25^{\circ}C$$37^{\circ}C$ 모두 무첨가구에 비하여 균체수가 현저히 감소하였다. 하지만 pH 산도 및 당도의 변화는 모든 첨가구에서 저장 기간에 따른 유의적 차이가 없었다. 그러나 관능검사와 전체적인 기호도 평가 결과에서는 대부분의 시료는 저장기간이 증가함에 따라 품질이 낮아지는 경향을 보였으나, 3% 녹차추출물 첨가 시료는 유의적으로 높은 기호도를 나타내었다.

Protective Effect of Aster tataricus L. Extract on the Dermal Cytotoxicity Induced by Sodium Bromate, Oxidant of Hair Dye

  • This study evaluated the dermal cytotoxicity of sodium bromate (NaBrO3) and the protective effect of Aster tataricus L. (AT) extract against NaBrO3-induced cytotoxicity in the cultured NIH3T3 fibroblasts. For this study, it was done the antioxidative effects such as electron donating (ED) activity and lipid peroxidation (LP) activity as well as cell viability. NaBrO3 significantly decreased cell viability in a dose-dependent manner and its XTT50 value was measured at a concentration of 54.4 μM in these cultures. The cytotoxicity of NaBrO3 was determined as highly-toxic by Borenfreund and Puerner's toxic criteria. The quercetin, antioxidant significantly increased cell viability against NaBrO3-induced cytotoxicity. Regarding the protective effect of Aster tataricus (AT) L. extract on NaBrO3-induced cytotoxicity, AT extract significantly increased the cell viability, the ED ability and the inhibitory ability of LP. From these findings, it suggested that the oxidative stress is involved in the cytotoxicity of NaBrO3, and AT extract effectively protected NaBrO3-induced cytotoxicity by antioxidative effects. Conclusively, the natural component like AT extract may be a putative therapeutic agent for the diminution or treatment of the cytotoxicity correlated with oxidative stress like hair dye component, NaBrO3.
