Culture Conditions for In Vitro Maturation of Abattoir Derived Oocytes of Native Zebu Cows of Bangladesh

  • The objectives of the study were to determine an effective culture dish, culture duration and protein supplementation in medium for in vitro maturation (IVM) of oocytes of native zebu cows in Bangladesh. The ovaries of cows were collected from local slaughterhouse followed by aspiration of follicular fluid. The cumulus-oocyte-complexes (COCs) with more than 3 compact cumulus cell layers were cultured in tissue culture medium (TCM) 199 for maturation. The maturation of oocytes was determined by observing polar body under microscope. To determine an effective culture dish, 130 COCs derived from 48 ovaries in a well of 4-well dish and 102 COCs derived from 36 ovaries in drops covered with mineral oil within 35 mm petri dish were cultured for 24 hours. The rate of maturation of oocytes did not vary between 4-well dish ($51.3{\pm}15.0%$) and drops in petri dish ($52.4{\pm}11.6%$). To determine the effective culture duration, 185 COCs derived from 62 ovaries were cultured in drops for 18, 21, 24 and 27 hours. The rate of maturation of occytes ranged from $51.9{\pm}9.4%$ (18 hours) to $59.0{\pm}17.1%$ (27 hours) and the difference in maturation rate among different culture durations was not significant (P>0.05). To determine an effective protein supplementation, 63 oocytes from 19 ovaries were cultured separately in TCM 199 supplemented with either fetal bovine serum (FBS) or bovine serum albumin (BSA). The rate of maturation was significantly (P<0.01) higher in medium supplemented with FBS ($55.63{\pm}16.19%$) than that of BSA ($14.82{\pm}9.36%$). In conclusion, COCs of native zebu cows can be cultured for IVM either in 4-well culture dish or droplets in petri dish for 18 to 27 hours in medium supplemented with FBS.

Changes in the ruminal fermentation and bacterial community structure by a sudden change to a high-concentrate diet in Korean domestic ruminants

  • Objective: To investigate changes in rumen fermentation characteristics and bacterial community by a sudden change to a high concentrate diet (HC) in Korean domestic ruminants. Methods: Major Korean domestic ruminants (each of four Hanwoo cows; $545.5{\pm}33.6kg$, Holstein cows; $516.3{\pm}42.7kg$, and Korean native goats; $19.1{\pm}1.4kg$) were used in this experiment. They were housed individually and were fed ad libitum with a same TMR (800 g/kg timothy hay and 200 g/kg concentrate mix) twice daily. After two-week feeding, only the concentrate mix was offered for one week in order to induce rapid rumen acidosis. The rumen fluid was collected from each animals twice (on week 2 and week 3) at 2 h after morning feeding using an oral stomach tube. Each collected rumen fluid was analyzed for pH, volatile fatty acid (VFA), and $NH_3-N$. In addition, differences in microbial community among ruminant species and between normal and an acidosis condition were assessed using two culture-independent 16S polymerase chain reaction (PCR)-based techniques (terminal restriction fragment length polymorphism and quantitative real-time PCR). Results: The HC decreased ruminal pH and altered relative concentrations of ruminal VFA (p<0.01). Total VFA concentration increased in Holstein cows only (p<0.01). Terminal restriction fragment length polymorphism and real-time quantitative PCR analysis using culture-independent 16S PCR-based techniques, revealed rumen bacterial diversity differed by species but not by HC (p<0.01); bacterial diversity was higher in Korean native goats than that in Holstein cows. HC changed the relative populations of rumen bacterial species. Specifically, the abundance of Fibrobacter succinogenes was decreased while Lactobacillus spp. and Megasphaera elsdenii were increased (p<0.01). Conclusion: The HC altered the relative populations, but not diversity, of the ruminal bacterial community, which differed by ruminant species.

INFLUENCE OF AGE AT FIRST CALVING ON THE MILK PRODUCTION OF NATIVE AND CROSSBRED DAIRY COWS

  • The influence of age at first calving on the milk production of crossbred dairy cows produced under major dairy cattle crossbreeding projects in Pakistan was studied from the year 1974 to 1980. These animals were bred and raised at the Livestock Production Research Institute, Bahadarnagar (LPRI), Livestock Experiment Station, Karachi (LES) and the University of Agriculture, Faisalabad (UAF). Local Sahiwal (SWL) cattle were crossed with the European breeds, Holstein Friesian (HF) and Jersey (J) at LPRI and UAF. At LES, native Red Sindhi (RS) were crossed with the HF and J breeds. At LES and UAF, the crossbred progeny, thus, produced comprised of halfbreds only while at LPRI 3/4 HF, 3/4 J, 1/4 HF and 1/4 J groups were also available for this study. The average age at first calving was considerably higher among the native breeds. At LES, the RS calved for the first time at an age of approximately 56 months. Corresponding values for SWLs at LPRI and UAF were 44 and 64 months, respectively. The 1/2 HF and 1/2 J crossbreds calved first at the age of approximately 25, 26, 34, 36, 37 and 38 months at LPRI, LES and UAF respectively. For 3/4 HF, 3/4 K, 1/4 HF and 1/4 J groups of LPRI the average age at first calving was 29, 26, 34 and 33 months. Considerable differences in age at first calving between the crossbreds and native breeds were observed. Due to early maturity, the former attained the peak level of milk production in third lactation whereas the latter groups, because of late maturity, reached this level in their fourth lactation. This study suggested that early maturity in Zebu cattle (Box indicus) could be induced through crossbreeding with European cattle (Bos Taurus).

Timed Artificial Insemination or Embryo Transfer using CIDR, Estradiol Benzoate and Prostaglandin $F_{2{\alpha}}$ for the Rebreeding of Korean Native Donor Cattle

  • This study compared the pregnancy rates of Korean native donor cattle after either a timed artificial insemination (TAI) or embryo transfer (TET) following the synchronization of ovulation using a controlled internal drug release (CIDR) device together with estradiol benzoate (EB) and prostaglandin $F_{2{\alpha}}$ ($PGF_{2{\alpha}}$). Fifty five cows and 8 heifers which had been previously used for embryo production were assigned to two treatments: (1) Thirty-two cattle received a CIDR device and 2 mg EB (Day 0), 25 mg $PGF_{2{\alpha}}$ injection at the time of CIDR removal on Day 7, and 1 mg EB injection on Day 8. All of the cattle received a TAI 30 h (Day 9) after the second EB injection (TAI group). (2) Thirty-one cattle received the same hormonal treatments as in the TAI group. The cattle with corpus luteum (CL) received a TET on Day 16 using frozen-thawed embryos (TET group). Ultrasonographic observations demonstrated that the proportion of cattle with synchronized ovulation on Day 10 and the concomitant formation of new CL on Day 13 did not differ between groups (p>0.05); the overall mean rates were 65.1 and 73.0%, respectively. The conception and pregnancy rates did not differ (p>0.05) between the TAI (12.5% and 12.5%) and TET groups (13.0% and 9.7%), respectively. We conclude that the pregnancy rate following TAI or TET in Korean native donor cattle was poor, which might be due in part to a poor synchrony of ovulation and concomitant CL formation.

  • 濟州道內 6個部落 共同牧場 野草地를 對象으로 野草의 生産性과 植生比率이 家畜 增體에 미치는 效果를 究明하기 위해 1981년 5월부터 동년 11월까지 調査, 分析한 結果는 다음과 같다. 1. 乾物收量은 7月收穫보다 10月收穫에서 2培二上 높았으며 모든 牧場이 같은 趨勢였다. 2. 野草의 祖蛋白質 含量은 7월보다 10월에서 낮았으며 粗纖維는 그 반대였다. 3. 牧場別로 볼 때 1일 增體量이 높은 河源과 海安牧場이 野草의 組蛋白質 含量이 가장 높았고 增體率이 제일 낮은 吾羅牧場에서 組蛋白質 含量이 낮았다. 4. 本 野草地에 分包된 全植生中 잔디가 56.61%로서 가장 높았으며 그 다음은 고사리 16.44%, 억새 3.76% 순위였다. 科別로는 禾本科가 72.08%. 登科 1.81% 고사리과 16.49%, 기타 9.62%였다. 5. 1일 增體率이 높은 河源牧場은 禾本科와 登科의 比率이 높았고 增體率이 낮은 吾羅와 龍崗牧場은 독생이 있는 고사리 比率이 높은 것으로 나타났다.

제주지역 한우의 소 바이러스성 설사병 바이러스 감염실태 (Prevalence of bovine viral diarrhea virus from Korean native cattle farms in Jeju)

  • Bovine viral diarrhea virus (BVDV) is an RNA virus belonging to Pestivirus in the family Flaviviridae. BVDV has economic significance for the livestock industry because of its association with acute disease, fetal loss, and birth of persistently infected (PI) animals. This study aimed to investigate the BVDV infection rates in Korean native cattle farms in Jeju for further planning of a BVDV control program in the Jeju Province. BVDV antibodies and antigens were tested in 15,842 sera collected from 302 Korean native cattle herds between January 2014 and June 2017 using enzyme-linked immunosorbent assay (ELISA). Viral antigen was detected by reverse transcription-polymerase chain reaction from 60 sera that were antigen ELISA-positive. BVDV antibodies were found in 90.7% (274/302) herds and 61.1% (9,678/15,842) cows. BVDV antigens were found in 13.2% (40/302) herds and 0.4% (61/15,842) cows. The oldest animal group (> 8 years) exhibited the highest sero-positive rates (91%), while the youngest animal group (< 1 years) had the highest antigen positivity rates (0.52%). Of the 60 antigen-positive sera, BVDV types 1 and 2 were found in 36 and 12 sera, respectively. Additionally, six animals were considered to be PI as BVDV was continually detected in annual examination.

Reproduction of Cross- and Purebred Friesian Cattle in Northern Thailand with Special Reference to Their Milk Production

  • Reproductive data, such as numbers of days to the first estrus and A.I. service postpartum, number of days to conception, number of A.I. services required for conception, interval between the first estrus and first A.I. service and the average interval of A.I. service in Thai native-Friesian crossbred and pure Friesian dairy cows, were compiled in the National Dairy Training and Applied Research Institute in Chiang Mai, Thailand. The data were analyzed statistically and the effect of milk production on these reproductive traits was investigated. The reproductive efficiency of purebred cows was obviously inferior when compared with crossbred animals, in spite of special care being given to the purebred only in order to alleviate the effect of a tropical climate and provide better feeding. However, the regression analysis between reproductive and lactational parameters revealed a definite antagonistic effect of lactation on reproduction, especially in the purebred cows, which had a larger amount of milk production and longer lactation period. If these effects of lactation were eliminated, there would be no evident difference in reproductive efficiency between purebred and crossbred cows in the conditions of this study. Among the reproductive parameters examined, the number of days to the first estrus and interval between the first estrus and first A.I. service were less affected by breed difference and the magnitude of lactation than other reasons.

Polymorphism of Bovine Lymphocyte Antigen DRB3.2 Alleles in Iranian Native Sarabi Cows

  • Sarabi cows (n = 136) from the Sarabi Breeding Station were genotyped at bovine lymphocyte antigen (BoLA)-DRB3.2 locus by a genotyping system that used the polymerase chain reaction and restriction fragment length polymorphism. Genomic DNA was extracted from whole blood samples. A two-step polymerase chain reaction was carried out in order to amplify a 284 base-pair fragment of target gene. Nested-PCR products were digested with three restriction endonuclease enzymes RsaI, BstYI and HaeIII. Digested fragments were analyzed by polyacrylamide gel electrophoresis. Twenty-six BoLA-DRB3.2 alleles were identified with frequencies ranging from 0.4 to 15.1%. Six new allele types observed in this study have not been reported previously. Identified alleles include: BoLA-DRB3.$2^*1$, $^*2$, $^*4$, $^*6$, $^*8$, $^*12$, $^*13$, $^*14$, $^*15$, $^*16$, $^*17$, $^*23$, $^*24$, $^*25$, $^*28$, $^*32$, $^*34$, $^*35$, $^*36$, $^*37$, $^*42$, $^*46$, $^*51$, $^*kba$, $^*laa$ and $^*vaa$. Their frequencies were found to be 0.4, 0.4, 0.7, 11.4, 1.1, 1.8, 2.9, 2.2, 4.4, 9.6, 1.1, 13.6, 0.4, 0.4, 1.1, 0.7, 0.4, 6.2, 2.2, 3.7, 1.1, 7.7, 1.5, 15.1, 2.6 and 7.3% respectively. The six most frequent alleles (DRB3.2 $^*6$, $^*16$, $^*23$, $^*46$, $^*kba$ and $^*vaa$) accounted for 64.7% of the alleles in the population of this herd. Numerous studies on this locus, covering different breeds, has revealed the existence of various alleles in this locus, and new investigations have introduced novel alleles. With respect to the high number of the observed alleles in this survey and the novelty of some alleles with no previous record of reporting, it is plausible to conclude that the BoLA-DRB3.2 locus is highly polymorphic in Iranian native Sarabi cows.

  • This cepseiment was carried out to cerify the effect of thawing methods and preservative temperature on the sperm motility and fertility after thawing semen with plastic straws in fresh and warm water. Sperm motility in vitro stored at room temperature after thawing were conducted by the various storage hours. A field trial after thawing semen with warmed water in straws from Cheju native cows involving 4 technicians and 800 cows first (or second) services gave the following results. The thawing methods of warmed water for one minute in semen motility were considerably higher than that in iced water during 12 hours after thawing semen, however, the sperm survival index of ice-water shwed a better results according as the time passed away, but not significant differences. Preservative temperaure at 5$^{\circ}C$ (iced water) after thawing gave significantly better results than that of thawed at 3$0^{\circ}C$ (warmed water). The N R rate to 175 inseminations with semen thawed at 15-2$0^{\circ}C$ (fresh water) was 82.8%, 80.9% for 610 inseminations thawed in warm water. Conception rate ofthe semen thawed in warm water for 10-60 secs gave no significant difference among storage hours, because the semen used to be inseminated within one hour almost, but in decreased when semen thawed at the period of one minute over.
