곤드레 첨가 설기떡의 품질특성 (Quality Characteristics of Sulgidduk added with Cirsium setidens Nakai)

  • 본 연구는 생리활성 기능을 나타내는 곤드레 분말을 설기떡에 첨가하여 품질특성을 조사한 결과, 수분함량은 무첨가구(40.67%)에 비해 곤드레 분말 첨가군(36.51~38.43%)이 낮게 나타났고, 색도는 곤드레 분말 첨가량이 증가함에 따라 L값은 유의적으로 감소하였으며, a값과 b값은 유의적으로 증가하는 경향을 나타냈다(p<0.05). 곤드레 분말 첨가량이 증가할수록 경도, 탄력성, 씹힘성은 증가하나, 응집성은 감소하는 경향을 나타내었다(p<0.05). 관능검사 결과, 전반적인 기호도에서는 곤드레 분말 5% 첨가 설기떡 > 1% 첨가 설기떡 > 3% 첨가 설기떡 > 0% 첨가 설기떡 > 7% 첨가 설기떡 순으로 높게 평가되었다. 또한, 곤드레 분말 첨가량을 달리한 설기떡을 $18^{\circ}C$에서 3일간 저장하는 동안의 총균수 변화는 제조 직후 모든 첨가군의 총균수 함량은 2(logCFU/g) 이하이었고, 3일 저장하는 동안 0%(4.02), 1%(4.94), 3%(5.94), 5%(6.28), 7%(6.9.8)로 곤드레 첨가량이 증가할수록 총균수가 다소 증가하였으나, 곤드레 설기떡은 저장하는 동안 미생물의 증식을 억제하지 못하는 것으로 사료된다. 곤드레 분말 첨가 설기떡 제조시 다른 첨가군에 비해 가장 높게 평가된 곤드레 분말 5% 첨가구가 설기떡 제조의 가장 적합한 것으로 사료되며, 향후 다양한 제품에 곤드레 분말을 이용하고자 한다.

Sprague-Dawley 랫드를 이용한 미선나무주정추출물의 2주 반복 경구투여 독성평가 (Toxicity Assessment of Abeliophyllum distichum Nakai Ethanol Extract Orally Administered to Sprague-Dawley Rats for Two Consecutive Weeks)

  • Abeliophyllum distichum Nakai is a deciduous shrub of a flowering plant in Oleaceae. It is an important plant resource and consists of only one species in the entire world. A. distichum Nakai is well known an edible, medicinal herb in its habitat districts, but the toxicological evaluation for the safe use of its extract is still insufficient. The study characterized the toxicity of an Abeliophyllum distichum Nakai ethanol extract in Sprague-Dawley (SD) rats and determined the safe dosage levels in a 13 weeks toxicity study. Abeliophyllum distichum Nakai ethanol extract was orally administered once daily for 2 weeks at 0, 500, 1,000 and 2,000 mg/kg/day to male and female SD rats. while recording the clinical signs of toxicity, body weight, food intake/consumption, eye test and urine analysis. Only the total protein frequency in the urine of male SD rats (p<0.05), the right ovary of the 500 mg/kg group (p<0.01) and the right adrenal gland of the 1,000 mg/kg group (p<0.05) in the female rats showed statistically significant changes. But no toxic effects were noted from repeated-dose administration of the Abeliophyllum distichum Nakai ethanol extract in the SD rats during the observation period. The post-mortem examinations showed no test substance-mediated changes. The hematological analysis and clinical blood chemistry data demonstrated no toxic effects from repeated-dose administration of Abeliophyllum distichum Nakai ethanol extract in the SD rats during the observation period. Based on these results, this data suggests that a dose of 1,000 mg/kg/day is a highest treatment to administer when conducting a further 13 weeks toxicity study.

멜라노마 세포에서 당귀추출물의 MITF, TRP-1, TRP-2, tyrosinase mRNA 발현 억제 효과 (Inhibitory Efficacy of Angelica gigas Nakai on Microphthalmia-associated Transcription Factor (MITF), Tyrosinase Related Protein-1 (TRP-1), Tyrosinase Related Protein-2 (TRP-2), and Tyrosinase mRNA Expression in Melanoma Cells (B16F10))

  • 아시아에서 한방약초로 널리 알려진 당귀 추출물의 미백활성을 알아보기 위하여 tyrosinase 저해활성을 측정한 결과 1,000 ${\mu}g/ml$의 농도에서 70% 이상의 활성을 나타내었다. 또한 당귀 추출물에 대한 멜라노마 세포(B16F10)의 세포생존율을 확인한 결과 500 ${\mu}g/ml$의 농도에서 99% 이상의 세포생존율을 확인할 수 있었다. 미백 관련 인자인 MITF, TRP-1, TRP-2 및 tyrosinase의 mRNA 발현량을 측정한 결과 50 ${\mu}g/ml$의 농도에서 각각 85.7%, 123.9%, 68.8%, 208%로 당귀 추출물을 처리하지 않은 군보다 감소하였음을 확인할 수 있었다. 이러한 연구 결과에 따라 당귀 추출물이 melanin 합성과 관련이 있는 유전자 발현의 억제효과가 있음을 확인할 수 있었으며, 미백 화장품 소재로서의 가능성을 확인하였다.

자원식물 활용을 위한 서운산 식물상 연구 (Study of Flora in Mt. Seoun to use Plant Resources)

  • 서운산(574m)은 지리적으로 북위 36$^{\circ}$54'~ 36$^{\circ}$50', 동경 127$^{\circ}$ 16'~127$^{\circ}$19'사이에 위치하며 경기도와 충청북도에 걸쳐 있다. 본 연구는 2000년 3월~10월까지 9차례에 걸쳐 실시한 현지조사의 결과를 종합하여 소산 식물의 목록을 작성하였다. 조사지의 소산 식물은 90과 268속 383종 49변종 5품종 2교잡종 등 총 439종류로 확인되었다. 산림청 지정의 희귀 및 멸종위기 식물은 천마, 뻐국나리, 삼백초, 태백제비꽃 등이 조사되었으며, 특산식물은 총 12종으로 전체 소산식물의 2.7%를 차지하였다. 귀화식물은 총 24종류로 전체 소산식물의 5.5%에 해당되었다. 서운산에서 조사된 자원식물의 용도별 구분은 식용자원식물이 239종류(54.4%), 약용 자원식물이 270종류(61.5%), 관상용 자원식물이 217종류(49.4%), 목재용 자원식물이 46종류(10.5%), 사료용 자원식물이 56종류(12.8%)로 구분되었다.

Sprague-Dawley 랫드를 이용한 땃두릅나무 열수추출물 분말의 단회 경구투여 독성시험 및 4주 반복 경구투여 용량 결정 시험 (Single Oral Dose Toxicity Test and Four Weeks Repeated Oral Dose Determination Test of Oplopanax elatus (Nakai) Nakai Hydrothermal Extract Powder in Sprague-Dawley Rats)

  • This study was conducted to investigate the toxicity symptoms and approximate lethal dose (ALD) of Oplopanax elatus (Nakai) Nakai hydrothermal extract powder by single oral dose toxicity and 4 weeks of repeated oral dose determination. The Sprague-Dawley (SD) male and female rats were treated with 1,250 (low- dosage group), 2,500 (medium- dosage group) and 5,000 (high- dosage group) mg/kg. In the single oral dose toxicity test, no dead animals and toxic symptoms were observed during the experiment. And there were no related with anomalies in normal weight changes and autopsy results. In the four-week repeated oral dose determination test, no death animals and toxicity symptoms were observed during the experiment, and there were no abnormal results in weight changes, feed and negative intake measurements. Results of eye examination, urinalysis, hematological values and serum biochemical values, gross findings and absolute organ were not of singularity. These result demonstrated that no toxic symptoms were observed by the test substance Oplopanax elatus (Nakai) Nakai hydrothermal extract powder under this test condition, and the non-toxic content is determined to be 5,000 mg/kg/day.

곰취 분말 및 당귀 열수추출물의 생리활성을 활용한 기능성 냉면의 제조 (The Development of Functional Cold Buckwheat Noodles Using Biological Activities of Hot Water Extracts of Ligularia fischeri and Angelica gigas Nakai)

  • Some biological activities such as an electron donating capacity, the contents of total polyphenol compounds and flavonoids, fibrinolytic activity and $\alpha$-glucosidase inhibitory activity have been detected in hot water extracts of Ligularia fischeri and Angelica gigas Nakai. To increase the usefulness of the functional ingredients for prevention and improvement of some metabolic disorders, ethanol-treated hot water extracts of Angelica gigas Nakai were prepared. A hot water extract of Ligularia fischeri has 92% of electron donating capacity, 39.4 mg/g of total polyphenol compounds, 24.8 mg/g of flavonoids and 29.8% of $\alpha$-glucosidase inhibitory activity, but no fibrinolytic activity. A hot water extract of Angelica gigas Nakai has 94.7% of electron donating capacity, 5.8 mg/g of total polyphenol compounds, 2.6 mg/g of flavonoids, 0.48 plasmin units of fibrinolytic activity and no $\alpha$-glucosidase inhibitory activity. However, with partial purification using cold ethanol treatment, the $\alpha$-glucosidase inhibitory activity of Angelica gigas Nakai was increased to 70.5%. Thus, we expected a more useful effect with the use of the addition of a cold ethanol-treated Angelica gigas Nakai extract. The L, b values of cold buckwheat noodles using a mixture of 0$\sim$3% of Ligularia fischeri powder and 0.5% of an ethanol-treated hot water extract of Angelica gigas Nakai were decreased with the addition of an increasing amount of Ligularia fischeri powder. Among the mechanical qualities, only adhesiveness was significantly higher in 3% Ligularia fischeri noodles. From sensory evaluation data, it was determined that these two functional ingredients did not ruin the color, texture, and overall acceptance of the cold buckwheat noodles. A higher amount of the extracts improved the quality of the product with little added cost.

Microbial Decontamination of Angelica gigas Nakai Using Electron Beam Irradiation

  • This study evaluated the use of electron beam irradiation for decontamination of the Korean medicinal herb, Angelica gigas Nakai. Herb samples were irradiated at doses of 2, 8, and 16 kGy, respectively. Populations of microorganisms in Angelica gigas Nakai decreased by 2$\sim$3 log cycles at 8 kGy irradiation. Electron beam irradiation caused negligible changes in Hunter color L, a, and b values. Sensory evaluations of Angelica gigas Nakai confirmed that irradiation caused no significant changes in the organoleptic properties of the samples. These results suggest that electron beam-irradiated herbs retain a better microbial safety and sensory qualities, compared with the non-irradiated.

칼잎용담의 Secoiridoid 배당체 (Secoiridoid Glucosides from the Root of Gentiana uchiyamana Nakai)

  • Gentiana uchiyamana Nakai is a perennial herb in Gentianaceous plants. From the root of Gentiana uchiyamana Nakai, the bitter secoiridoid glucosides were isolated and identified as gentiopicroside and swertiamarin. A high-performance liquid chromatographic method for quantitative determination of bitter component, i.e. gentiopicroside in the roots of Gentiana scabra and G. uchiyamana Nakai was developed, using a ${\mu}$-Bondapak $C_{18}$ column and methanol-water mobile phases. The content of gentiopicroside was calculated from calibration curve previously prepared using the standard, 3.55% in the roots of Gentiana scabra and 1.58% in Gentiana uchiyamana Nakai.

참당귀(Angelica gigas Nakai)잎 용매추출에 따른 생리활성 (Biological Activities of Extracts from Leaf of Angelica gigas Nakai)

  • Antioxidant properties and antioxidant activities were analyzed for water extracts and 50% and 70% ethanol extracts of the leaf of Angelica gigas Nakai. The polyphenol and flavonoid contents in water, 50%, and 70% ethanol extract of the leaf of Angelica gigas Nakai, it was found that the polyphenol contents were 18.75 mg GAE/g, 28.95 mg GAE/g, and 34.73 mg GAE/g, respectively, and flavonoid contents were respectively. The DPPH IC50 scavenging activity was 45.84 mg/mL, 36.44 mg/mL, 19.11 mg/mL, respectively, and theABTS+ radical scavenging ability (1 mg/mL) was 28.73%, 22.79%, and 12.70%, respectively. Tyrosinase inhibitory activity, 70% ethanol extract, 50% ethanol extract, and water extract 33.14%, and 4.53%, respectively. Nitrite scavenging activity, 70% ethanol extract, 50% ethanol extract, and water extract were in the order of 1 mg/mL scavenging activity, 36.43%, 34.80%, and 18.85%, respectively.

조릿대의 종합적 활용을 위한 조릿대 섬유 특성 평가 (Evaluation of Characteristics of Sasa quelpaertensis Nakai Stem for the Comprehensive Utilization)

  • Chemical composition, morphological properties and papermaking properties of Sasa quelpaertensis Nakai were investigated in order to use it comprehensively. The lignin contents of stalks and leaves were 18.8% and 15.3% and the holocellulose contents were 63.3% and 48.6% respectively. The contents of ash and the amount of water extract showed the higher value than those of wood or other bamboo species. The average fibers length and width of Sasa quelpaertensis Nakai were 780 ${\mu}m$ and 14.8 ${\mu}m$. The fibers of Sasa quelpaertensis Nakai stalk had thinner width and more slender structure than those of softwood. The handsheet made of Sasa quelpaertensis Nakai alkaline pulp showed higher in tensile strength and bulkier structure than those of handsheet made of soft wood unbleached kraft pulp.
