Development of the Gonads Derived from Hetero-Sexually Transferred Primordial Germ Cells (PGCs) between Embryos in the Chicken

  • Primordial germ cells (PGCs) of White Leghorn chicken embryos as a donor were transferred to Rhode Island Red chicken embryos as a recipient. At 48-50 h (stage 13-15) of incubation of fertilized eggs, donor PGCs, which were taken out from blood vessels of donor embryos, were injected into blood vessels of recipient embryos. Sex of the treated embryos was determined after the transfer of PGCs using remaining blood samples. In the present experiments, survival rate of the treated embryos was 33.3% for homo-sexual and 35.4% for hetero-sexual transfers of PGCs, respectively, when determined at 17 days of incubation. In this study, most of the treated embryos could not survive more than 18 days of incubation, though the reason for that was not clarified in the present work. The gonalds removed from embryos that died after 18 days of incubation and the organs from newly hatched chicks were examined for morphological and histological features. The gonads removed from the embryos with homo-sexual transfer of PGCs showed normal development in appearance. On the contrary, some (35.3%) of the embryos with hetero-sexual transfer of PGCs possessed abnormal gonads similar to ovotestis by histological observation. In cases where the gonads developed to be normal organs (64.7%) the sex of embryos was the same as recipient ones. The present results suggest that hetero-sexual transfer of the PGCs may bring about the possibility of development of the embryos bearing sexually different gametes, spermatogonia or oogonia.

The Influence of Alpha-fetoprotein on Natural Suppressor Cell Activity and Ehrlich Carcinoma Growth

  • The influence of alpha-fetoprotein (AFP) on the bone marrow (BM) natural suppressor (NS) cells of intact Ehrlich carcinoma -bearing CBA mice was studied. Bone marrow NS cells were fractionated into three fractions by isopycnic centrifugation on percoll gradients: NS1 (${\rho}$=1.080 g/ml), NS2 (${\rho}$=1.090 g/ml) and NS3 (1.100> ${\rho}$ > 1.090 g/ml). These fractions were highly different in their sensitivity to known NS cell inductors (interleukin (IL)-2, IL-3 or histamine). None of the NS fractions isolated from the intact mice spontaneously produced antiproliferative activity, however, they showed a high level of NS (antiproliferative and natural killer cell inhibitory) activity under the influence of AFP. A single injection of AFP to intact mice led to an increase of spontaneous NS activity and the inhibition of natural killer cell activity. NS activity, especially NS2, was increased in when tumor cells were subcutaneously inoculated three days after AFP injection. In the AFP-treated mice, the tumor mass at 14 days was 60% larger than that in the untreated mice. Our data confirmed that AFP is a tumor marker that can inhibit cancer immunity and plays a role in cancer pathogenesis.

  • The Tsushima Current has been known to branch out from the Kuroshio west of Kyushu and to flow north to the Korea Strait. Then, it has to flow across the isobaths and so needs some driving forces. As the forces, sea level difference between the Korea and Tsugaru Straits, Reynolds stress west of Kyushu and density differences have been suggested, In this paper, their roles have been numerically studied in the barotropic case. Model results show that the Tsushima Current is possible without any above force. The flows just follow isobaths over the East China Sea. They seem to be driven by their own dynamics without any external force. The mechanism is just like outflows from a gap. Model results also show that the flows in this area could be significantly affected by the external forces such as Reynolds Stress. Then the dynamics and flows in real ocean might be complicated. However, the barotropic study tells us that the Tsuahima Currents is basically driven by geostrophic adjustment.

Feasibility of Red Tide Detection Around Korean Waters Using Satellite Remote Sensing

  • The monitoring activities at the National Fisheries Research and Development Institute (NFRDI) in Korea have been extended to include all the coastal waters of Korea after the outbreak of Cochlodinium polykrikoides blooms in 1995. We used several alternative methods including climatological analysis, spectral and optical methods which may offer potential detection of the major species of red tide in Korean waters. In the climatological analysis, NOAA, SeaWiFS, OCM satellite data was chosen using the known C. polykrikoides red tide bloom data and the area was mapped by helicopter reconnaissance and ground observation. The relationship between the distribution of sea surface temperature to C. polykrikoides bloom areas was studied. The anomalies of SeaWiFS chlorophyll a imageries against the imageries of non-occurring red tide for August, 2001 showed where the C. polykrikoides occurred. The anomalies of chlorophyll a concentrations from the satellite data during red tide outbreaks showed a similar distribution of C. polykrikoides in the red tide in August, 2001. The distribution between differences in sea surface temperatures during the day and at night also showed a possibility for red tide detection. We used a corrected vegetation index (CVI) to detect floating vegetation and submerged vegetation containing algal blooms. The results of from the optical absorption of C. polykrikoides in the ultraviolet band (340 nm) showed that if we use the optical characteristics from each red tide, we will be able to establish the feasibility of red tide detection.

  • 2008 Korea-Japan Cartoon Festival is a cultural enterprise, sponsored by 8 cities of Korea and Japan. 8 cities include Pusan, Choen Nam Province, Kyung Nam Province, Cheju, Fukuoka, Saga, Nagasaki, and Yamaguchi. It was held under the sponsorship of Fukuoka, Japan, in 2007. In 2008, it was held in Pusan under the sponsorship of Korea Character Designers' Association and Pusan Becksko. Even though local autonomous entities, recently, sponsor various Festivals, 2008 Korea-Japan Cartoon Festival has so unique characteristics that it is appropriate for paper. Unique characteristics of this Festival include that it is held only two years and it is for amity of young people of East Asia. In 2008 Korea-Japan Cartoon Festival, there were cartoon contest, exhibition, seminar, and experience event. It would be the basic ideas for industries of characters, animations and games, and would broaden cultural interchange between Korea and Japan.

Plasma Soluble CD30 as a Possible Marker of Adult T-cell Leukemia in HTLV-1 Carriers: a Nested Case-Control Study

  • Elevated levels of soluble CD30 (sCD30) are linked with various T-cell neoplasms. However, the relationship between sCD30 levels and the development of adult T-cell leukemia (ATL) in human T-cell leukemia virus type 1 (HTLV-1) carriers remains to be clarified. We here investigated whether plasma sCD30 is associated with risk of ATL in a nested case-control study within a cohort of HTLV-1 carriers. We compared sCD30 levels between 11 cases (i.e., HTLV-1 carriers who later progressed to ATL) and 22 age-, sex- and institution-matched control HTLV-1 carriers (i.e., those with no progression). The sCD30 concentration at baseline was significantly higher in cases than in controls (median 65.8, range 27.2-134.5 U/mL vs. median 22.2, range 8.4-63.1 U/mL, P=0.001). In the univariate logistic regression analysis, a higher sCD30 (${\geq}30.2U/mL$) was significantly associated with ATL development (odds ratio 7.88 and the 95% confidence intervals 1.35-45.8, P = 0.02). Among cases, sCD30 concentration tended to increase at the time of diagnosis of aggressive-type ATL, but the concentration was stable in those developing the smoldering-type. This suggests that sCD30 may serve as a predictive marker for the onset of aggressive-type ATL in HTLV-1 carriers.

Age and Growth of Sea Urchin, Pseudocentrotus depressus (분홍성계(Pseudocentrotus depressus)의 연령 분석과 성장)

  • We studied age and growth of the sea urchin, Pseudocentrotus depressus, to obtain some informations regarding its sustainable production and appropriate resources reinforcement. The samples were collected at two locations (Ongpo and Bubhwan) in Jeju, Korea. Annual rings were formed from October to January, and this period was well matched with the time just prior to or during their reproduction. Two population regression lines generated by using Walford's plotting with mean radius of each age group showed significant differences in their growth rate between the two sampling locations (p<0.0l). When the regression equations were calculated using either madreporite's radius (R) and test diameter (L) or body weight (W) and test diameter (L), the results were L=23.830+ 11.735R and $W=0.0004L^3$, and no statistically significant differences were detected between the two populations (p>0.2). Based on the data of madreporite's radius and test diameters, two estimated growth equations were $L_t(mm)=72.988(1-e^{-0.412(t-0.596)}\;and\;L_t(mm)=70.195(1-e{-0.365(t-0.51l)}$ in Ongpo and Bubhwan population, respectively. Three distinct annulus groups were recognizable within the distribution of the radii of the annual rings measured from age I to age 5. The mean radii calculated from the same annulus group were all identical even though they were from different age groups.

First Record of Paramphistomes Fischoederius cobboldi and Paramphistomum epiclitum Detected in Bovine Rumen from a Local Market of Savannakhet Province, Lao PDR

  • In the present study, we report on the occurrence of paramphistomes, Fischoederius cobboldi and Paramphistomum epiclitum, in Lao PDR with the basis of molecular data. Parasite materials were collected from bovines bred in Ban Lahanam area, Savannakhet Province, Lao PDR at Lahanam public market. Morphological observations indicated 2 different species of paramphistomes. The mitochondrial gene cox1 of the specimens was successfully amplified by PCR and DNA sequencing was carried out for diagnosis of 11 specimens. Pairwise alignment of cox1 sequences were performed and confirmed F. cobboldi and P. epiclitum infecting bovines in Laos. Although there were many limiting points, as the small number of worm samples, and the restricted access of the animal host materials, we confirmed for the first time that 2 species of paramphistomes, F. cobboldi and P. epiclitum, are distributed in Lao PDR. More studies are needed to confirm the paramphistome species present in Savannakhet and its hosts to clear the natural history of these parasites of ruminants in the region and measure the impact of this parasite infection in the life and health of the local people.

Initial Report for the Radiation Effects Research Foundation F1 Mail Survey

  • To study the full health effects of parental radiation exposure on the children of the atomic bomb survivors, the Radiation Effects Research Foundation developed a cohort of 76,814 children born to atomic bomb survivors (F1 generation) to assess cancer incidence and mortality from common adult diseases. In analyzing radiation-associated health information, it is important to be able to adjust for sociodemographic and lifestyle variations that may affect health. In order to gain this and other background information on the F1 cohort and to determine willingness to participate in a related clinical study, the F1 Mail Survey Questionnaire was designed with questions corresponding to relevant health, sociodemographic, and lifestyle indicators. Between the years 2000 and 2006, the survey was sent to a subset of the F1 Mortality Cohort. A total of 16,183 surveys were completed and returned: 10,980 surveys from Hiroshima residents and 5,203 from Nagasaki residents. The response rate was 65.6%, varying somewhat across parental exposure category, city, gender, and year of birth. Differences in health and lifestyle were noted in several variables on comparison across city and gender. No major differences in health, lifestyle, sociodemographics, or disease were seen across parental exposure categories, though statistically significant tests for heterogeneity and linear trend revealed some possible changes with dose. The data described herein provide a foundation for studies in the future.

Formation and Hydrogen Absorption Properties of Intermetallic Mg-Ni Compound Nanoparticles (Mg-Ni 금속 간 화합물 나노입자의 형성과 수소저장 특성)

  • Mg-Ni nanoparticles were synthesized by a physical vapor condensation method (DC arc-discharge) in a mixture of argon and hydrogen atmosphere, using compressed mixture of micro powders as the raw materials. The crystal phases, morphology, and microstructures of nanoparticles were analyzed by means of X-ray diffraction (XRD) and high-resolution transmission electron microscopy (HRTEM). It was found that the intermetallic compounds of $Mg_2Ni$ and $Mg_2Ni$ formed with existence of phases of Mg, Ni, and MgO in Mg-Ni nanoparticles. After one cycle of hydrogen absorption/desorption process (activation treatment), Mg-Ni nanoparticles exhibited excellent hydrogen absorption properties. $Mg_2Ni$ phase became the main phase by aphase transformation during the hydrogen treatments. The phenomenon of refinement of grain size in the nanoparticle was also observed after the hydrogen absorption/desorption processes, which was attributed to the effect of volume expansion/shrinkage and subsequent break of nanoparticles. Maximum hydrogen absorption contents are 1.75, 2.21 and 2.77 wt.% at 523, 573 and 623 K, respectively.
