Characterization of Pretreatment for Barley straw by Alkaline Solutions (염기 용매를 이용한 보릿짚의 전처리 특성)

  • Lignocellulose is difficult to hydrolyze due to the presence of lignin and the technology developed for cellulose fermentation to ethanol is not yet economically viable. However, recent advances in the extremely new field of biotechnology for the ethanol production are making it possible to use of Agriculture residual biomass, e.q., Barley straw, because of their several superior aspects as Agriculture residual biomass; low lignin, high contents of carbohydrates. Barley straw consists of 39.78% cellulose (glucose), 22.56% hemicelluloses and 19.27% lignin. Pretreatment of barley straw using NaOH pretreatment solutions concentration with 2%, temperature $85^{\circ}C$ and reaction times 1 hr were investigates. $NH_4OH$ pretreatment condition was solutions concentration with 15%, temperature $60^{\circ}C$, and reaction times 24hr were investigates. Furthermore, enzymatic saccharification using cellulose at $50^{\circ}C$, pH 4.8, 180 rpm for conversion of cellulose contained in barley straw to monomeric sugar. The pretreatment of barley straw using NaOH and $NH_4OH$ can significantly improve enzymatic saccharification of barley straw by extract more lignin and increasing its accessibility to hydrolytic enzymes. The result showed NaOH pretreatment extracted yield of lignin was 24.15%. $NH_4OH$ pretreatment extracted yield of lignin was 29.09%. Shaccharification of barley straw pretreatment by NaOH for 72hr and pH 4.8 result in maximum glucose concentration 15.39g/L (58.40%) and by $NH_4OH$ for 72hr and pH 4.8 result in maximum glucose concentration 16.01g/L (64.78%).

Characteristics and Gel Properties of Gelatin from Goat Skin as Influenced by Alkaline-pretreatment Conditions

  • Characteristics and properties of gelatin from goat skin pretreated with NaOH solutions (0.50 and 0.75 M) for various times (1 to 4 days) were investigated. All gelatins contained ${\alpha}$-chains as the predominant component, followed by ${\beta}$-chain. Gelling and melting temperatures of those gelatins were $23.02^{\circ}C$ to $24.16^{\circ}C$ and $33.07^{\circ}C$ to $34.51^{\circ}C$, respectively. Gel strength of gelatins increased as NaOH concentration and pretreatment time increased (p<0.05). Pretreatment for a longer time yielded gelatin with a decrease in $L^*$-value but an increase in $b^*$-value. Pretreatment of goat skin using 0.75 M NaOH for 2 days rendered the highest yield (15.95%, wet weight basis) as well as high gel strength (222.42 g), which was higher than bovine gelatin (199.15 g). Gelatin obtained had the imino acid content of 226 residues/1,000 residues and the gelatin gel had a fine and ordered structure. Therefore, goat skin gelatin could be used as a potential replacer of commercial gelatin.

  • 본연구의 목적은DP가공에 의한 가교화를 통해 텐셀의 피브릴화를 조절할 때 DP가공 방법 및 NaOH전처리 효과를 물성, 표면형태, 역학적 성질 및 태의 변화의 관점에서 고찰하는데 있다. SEM 분석결과DP가공에 의해 피브릴 발생 정도는 감소하였다. DP가공 방법에 있어 서 WF법과 PDC법에 따른 물성의 차이는 나타나지 않았다. 역학적특성의 경우 DP가공은 DP가공 방법에 상관없이 효소처리한 직물의 인장선형성에는 큰 영 향을 주지 않았으나 인장에 너지, 굽힘 강성, 압축선형성, 압축 레질리언스, 기하학적 거칠기는 감소시켰고 인장 레질리언스, 굽힘이력, 압축에너지는 증가시켰다. 전단특성은 WF법에서는 증가한 반면, PDC법에서는 감소하여 DP가공 방법에 따른 차이를 나타내었다. WF법이 PDC법보다 더 높은 Koshi, Numeri, Fukurami 값을 보였으며, 종합태 값은 비슷하게 나타났다. NaOH 전처 리에 의해 수지부착량은 감소하였으나 감량률은 증가하였으며, DP성/물성은 더 낮게 나타났다. NaOH 전처리에 의해 인장선형성, 인장에너지, 압축 레질리언스, 전단 및 굽힘특성은 증가하였으나 인장레질리언스와 압축선형성, 압축에너지, 표면특성은 감소하였다. NaOH 전처리한 경우 Koshi는 증가하였고, Numrei와 Fukuramil는 감소하였으며, 종합태 값은 가장 낮았다. 처리한 시료들은 각각 다른 감성과 촉감을 나타냈다.

  • Effects of chemical treatments on in sacco and in vitro digestibility of barley straw by rumen fluid and pure cultures of cellulolytic bacteria were studied to evaluate the pretreatment and to improve the poor quality feed. Chemicals were applied by dissolving them in water equivalent to 40% of the weight of the straw (dry matter basis). Pretreatment with 5% NaOH yielded the largest increase in sacco digestion followed by pretreatment with 2% $(NH_4)_2SO_3$, 2.6% $NH_4OH$, 1.6% $NaHSO_3$ and untreated straw (control). In sacco dry matter digestibility of straw treated with NaOH and $(NH_4)_2SO_3$ continued to increase as the concentration of chemical increased (1 to 7.5%), as it was the in vitro dry matter loss by leaching. Treatment of barley straw with 5% NaOH enhanced significantly (p < 0.01) in vitro digestibility by rumen fluid, Fibrobacter suceinogenes and Ruminococcus albus though the fermentation products by cellulolytic bacteria were low, whereas the treatment with 5% $(NH_4)_2SO_3$ inhibited in vitro digestibility by F. succinogenes and R. albus together with lower fermentation products. Dry matter loss by leaching and bacterial digestion from barley straw treated with NaOH and $(NH_4)_2SO_3$ suggested the effect of pretreatment with these chemicals were based on leaching, and the cellulolytic bacteria had little to do with digestion.

  • Various mechanical and chemical pretreatment methods including alkali treatment, pre-beating, enzyme treatment and oxidation treatment have been used to reduce the production energy of the microfibrillated cellulose (MFC). Among them, alkali swelling can be helpful to reduce the energy consumption because the internal bonding between fibrils could be weakened. In this study, dimethyl sulfoxide (DMSO) was used as a cosolvent to improve alkali pretreatment efficiency and the effects of NaOH concentration during NaOH-DMSO swelling on changes in fiber characteristics of softwood bleached kraft pulp (SwBKP) were elucidated. For alkali treatment in H2O-DMSO solvents, fiber length were decreased with increasing NaOH concentration while fiber width, curl and WRV were increased. WRV began to increase at 8% NaOH solution. In addition, above 8% concentration of NaOH, crystalline structure of pulp fibers converted from cellulose II to cellulose III by DMSO cosolvent. Comparing the previous results with this study, it was shown that DMSO cosolvent could promote swelling of pulp fibers and thus reduce NaOH concentration for the maximum swelling of fibers.

Pretreatment and enzymatic saccharification process of rapeseed straw for production of bioethanol

  • This study was conducted to evaluate the yield of bio-ethanol produced by separate hydrolysis and fermentation (SHF) with the pretreated rapeseed straw (RS) using crude enzyme of Cellulomonas flavigena and Saccharomyces cereviase. Crude enzyme of C. flavigena showed enzymatic activity of 14.02 U/mL for CMC 133.40 U/mL, for xylan 15.21 U/mL, for locust gum and 15.73 U/mL for rapeseed straw at pH 5.0 and $40^{\circ}C$, respectively. The hemicellulose contents of RS was estimated to compromise 36.62% of glucan, 43.20% of XMG (xylan + mannan + galactan), and 2.73% of arabinan by HPLC analysis. The recovering ratio of rapeseed straw were investigated to remain only glucan 75.2% after 1% $H_2SO_4$ pretreatment, glucan 45.44% and XMG 32.13% after NaOH, glucan 44.75% and XMG 5.47% after $NH_4OH$, and glucan 41.29% and XMG 41.04% after hot water. Glucan in the pretreatments of RS was saccharified to glucose of 45.42 - 64.81% by crude enzyme of C. flavigena while XMG was made into to xylose + mannose + galactose of 58.46 - 78.59%. Moreover, about 52.88 - 58.06 % of bio-ethanol were obtained from four kinds of saccharified solutions by SHF using S. cerevisiae. Furthermore, NaOH pretreatment was determined to show the highest mass balance, in which 21.22 g of bio-ethanol was produced from 100 g of RS. Conclusively, the utilization of NaOH pretreatment and crude enzyme of Cellulomonas flavigena was estimated to be the best efficient saccharification process for the production of bio-ethanol with rapeseed straw by SHF.

  • This research offers recycling method of NaOH from mercerizing process of cotton fabric. The measuring system of NaOH concentration was designed for the mercerizing process and tested for various conditions which can be occurred in factory. The accuracy of the system was varied as the testing condition was changed. As the concentration of sulfuric acid used for titrating NaOH decreased, the accuracy of measuring system increased. The concentration of NaOH for waste water collected from mercerizing process was measured by 5.2%. As the ratio of newly mixed NaOH increased, moisture regain. water absorbency and whiteness of the specimen increased. The bending rigidity(B) and shear rigidity(G) decreased, as the ratio of newly mixed NaOH increased.

  • Preparative conditions of high-gel strength agar from Gelidium amansii have been studied, The effect of NaOH pretreatment on the quality and yields of agar extracted from Gelidium amansli was examined. The Bel strength of agar extracted from C. amansii pretreated with NaOH was higher than that of agar extracted from G. amansii non-pretreated with NaOH. The gel strength of agar extracted from G. amansii was influenced by concentration, temperature and time of pretreatment with NaOH. It was found that the proper concentration, temperature and time of NaOH pretreatment to produce high-gel strength agar was $6\%$ NaOH, $80^{\circ}C$ and 2$\~$3 hrs. The principal sugars of agar extracted from G. amansli were galactose and 3,6-anhydrogalactose.

Production of Bacterial Cellulose and Its Modification (박테리아 셀룰로오스의 생산 및 개질)

  • The bacterial celluloses are very different in its physical, chemical and morphological structures compared to wood cellulose. These fibers have many unique properties that are potentially and commercially beneficial. This study was aimed to elucidate the production of bacterial celluloses and to improve their physical properties by chemical pretreatment. Bacterial celluloses produced by static culture had gel-like pellicle structure. The pellicle thickness was increased with the increasing time, and its layer was about 1.8cm after one-month incubation. The pellicles extruded from the cells of Acetobacter had a non-crystalline structure during initial growing stages, gradually getting crystaliyzed with the incubation time elapse, and eventually fumed to the cellulose I crystals. Young's modulus of bacterial cellulose sheet was increased with increasing NaOH concentration, and resulted in the highest at 5% NaOH concentration. Similar results with NaClO3 pretreatment can be observed. Too concentrated alkali solutions induced the destruction of cellulose fibrils and changed the mechanical properties of the sheets. These alkaline pretreatment have removed non-cellulosic components(NCC) from the bacterial cellulose, and enhanced inter-abrillar bonding by direct close contact among cellulosic fibrils.

Effect of Dilute Alkali on Structural Features and Enzymatic Hydrolysis of Barley Straw (Hordeum vulgare) at Boiling Temperature with Low Residence Time

  • This work was conducted to evaluate the effect of dilute sodium hydroxide (NaOH) on barley straw at boiling temperature and fractionation of its biomass components into lignin, hemicellulose, and reducing sugars. To this end, various concentrations of NaOH (0.5% to 2%) were applied for pretreatment of barley straw at $105^{\circ}C$ for 10 min. Scanning electron microscopy (SEM), atomic force microscopy (AFM), and Fourier transform infrared (FTIR) spectroscopy studies revealed that 2% NaOH-pretreated barley straw exposed cellulose fibers on which surface granules were abolished due to comprehensive removal of lignin and hemicellulose. The X-ray diffractometer (XRD) result showed that the crystalline index was increased with increased concentration of NaOH and found a maximum 71.5% for 2% NaOH-pretreated sample. The maximum removal of lignin and hemicellulose was 84.8% and 79.5% from 2% NaOH-pretreated liquor, respectively. Reducing sugar yield was 86.5% from 2% NaOH-pretreated sample using an enzyme dose containing 20 FPU of cellulase, 40 IU of ${\beta}$-glucosidase, and 4 FXU of xylanase/g substrate. The results of this study suggest that it is possible to produce the bioethanol precursor from barley straw using 2% NaOH at boiling temperature.
