Heat Load Characteristics of Sea Water Cooling Apparatus on Inshore Fishing Boat (연근해 어선용 해수냉각장치의 열부하특성)

  • The circumstance is giving the blow against fishermen with the incoming-decreasing and the difficulty of crew's supply & demand and management. In addition, the depression of the external situation like the departure of WTO system and the plan of EEZ proclaim is forcing fishery into improving their fishing condition. By this international and domestic circumstance, development of the sea water cooling apparatus for fish hold storage is demanded sincerely. First of all, we Investigated load characteristics which based on development of sea water cooling system and optimum fish hold storage. The experimental results is as follows. In creasing the speed of compressor and mass blew rate of refrigerant, the temperature of NaCl solution is low. And the load characteristics experiment on fish hold storage outlet is as fellows. As time goes by, increasing the mass flow rate of NaCl solution, temperature difference between inlet and outlet is small in a model of fish hold storage. These results provide many useful informations applicable to an actual design of sea water cooling system and optimum fish hold storage.

Acidity and Adsorption Mechanism of Solid Acid (固體酸의 酸性度와 吸着메카니즘에 관한 硏究)

  • Korean acid clays and Japanese acid clay were 1 N KCl solution and then their acidities were determined by measuring pH of the filtrates produced. And on examining the effect of neutral salt solution, such as KCl, NaCl, $BaCl_2,\;Pb(NO_3)_2\;and\;CuSO_4$, on the acidity, it was found that the effect decreased in order mentioned above and this situation proved to be in accord with Lyotrope series, i.e., the order of $K^+>Na^+>Ba^{++}>Pb^{++}>Cu^{++}.$ And after adsorbing cation, $Al_2O_3\;and\;Fe_2O_3$ which dissolved out of acid clay were measured, with the result that the amounts dissolved out were nearly proportional to acidity. This result accords with Kobayashi and Yamamoto's theory that the cause for acidity of clays is due to the fact that HCl which is formed initially by exchange of cation reacts with solid clay, and then chlorides dissolved out bring about hydrolysis, then resulting in the second formation of HCl. On measuring the rate which acid clay adsorbed dye in aqueous solution of basic and acid dyes, it was found that acid dye had no relation to acidity, and though it was not adsorbed by acid clay, the adsorption rate of basic dye was proportional to acidity. After adsorbing basic dye, pH was proportional to both acidity of solid acid and its adsorption rate of dye. Hence, it was concluded that the adsorption of basic dye was in accord with adsorption mechanism of neutral salt. This study led to find the acidic cause of solid acid and its adsorption mechanism of dye in aqueous solution.

Vibrio vulnificus Cytolysin Forms Anion-selective Pores on the CPAE Cells, a Pulmonary Endothelial Cell Line

  • Cytolysin produced by Vibrio vulnificus has been incriminated as one of the important virulence determinants in V. vulnificus infection. Ion selectivity of cytolysin-induced pores was examined in a CPAE cell, a cell line of pulmonary endothelial cell, using inside-out patch clamp techniques. In symmetrical NaCl concentration (140 mM), intracellular or extracellular application of cytolysin formed ion-permeable pores with a single channel conductance of $37.5{\pm}4.0$ pS. The pore currents were consistently maintained after washout of cytolysin. Replacement of $Na^+$ in bath solution with monovalent ions $(K^+,\;Cs^+\;or\;TEA^+)$ or with divalent ions $(Mg^{2+},\;Ca^{2+})$ did not affect the pore currents. When the NaCl concentration in bath solution was lowered from 140 to 60 and 20 mM, the reversal potential shifted from 0 to -11.8 and -28.2 mV, respectively. The relative permeability of the cytolysin pores to anions measured at $-40\;mV\;was\;Cl^-\;=\;NO_2^-\;{\geq}\;Br^-\;=\;I^-\;> \;SCN^-\;>\;acetate^-\;>\;isethionate^-\;>\;ascorbic acid^-\;>\;EDTA^{2-},$ in descending order. The cytolysin-induced pore current was blocked by $CI^-$ channel blockers or nucleotides. These results indicate that V. vulnificus cytolysin forms anion-selective pores in CPAE cells.

Short-term Preservation of Sperm in the Tiger Puffer, Takifugu rubripes (자주복(Takifugu rrbripes) 정자의 액상보존)

  • Condition for fresh storage of tiger puffer in the liquid state were investigated in several experiments. When marine fish ringer solution and 1% NaCl were used as the diluent for the short-term preservation method, spermatozoa activity index (SAI) and survival rate showed the best result among the various diluents tested. The dilution rate for the shortterm preservation of spermatozoa was suitable between 3 and 5 times with the 1% NaCi diluent. The appropriate range of temperature for the short-term preservation showed between 0 and $5^{\circ}C$. In order to keep high SAI and survival rate of spermatozoa, antibiotic addition (800 ppm neomycin) could be suggested. These results indicated that the short-term preservation method could be employed in tiger puffer spermatozoa.

Estimation of Geochemical Evolution Path of Groundwaters from Crystalline Rock by Reaction Path Modeling (반응경로 모델링을 이용한 결정질암 지하수의 지구화학적 진화경로 예측)

  • The chemical compositions of groundwaters from the granite areas mainly belong to Ca-HC0$_{3}$ and Na-HC0$_{3}$type, and some of these belong to Ca-(CI+S0$_{4}$) and Na-(CI+S0$_{4}$) type. Spring waters and groundwaters from anorthosite areas belong to Ca-HC03 and Na-HC03 type, respectively. The result of reaction path modeling shows that the chemical compositions of aqueous solution reacted with granite evolve from initial Ca-CI type, via CaHC0$_{3}$ type, to Na-HC0$_{3}$ type. The result of rain water-anorthosite interaction is similar to evolution path of granite reaction and both of these results agree well with the field data. In the reaction path modeling of rain watergranite/anorthosite reaction, as a reaction is progressing, the activity of hydrogen ion decreases (pH increases). The concentrations of cations are controlled by the dissolution of rock-forming minerals and precipitation and re-dissolution of secondary minerals according to the pH. The continuous addition of granite causes the formation of secondary minerals in the following sequence; gibbsite plus hematite, Mn-oxide, kaolinite, silica, chlorite, muscovite (a proxy for illite here), calcite, laumontite, prehnite, and finally analcime. In the anorthosite reaction, the order of precipitation of secondary minerals is the same as with granite reaction except that there is no silica precipitation and paragonite precipitates instead of analcime. The silica and kaolinite are predominant minerals in the granite and anorthosite reactions, respectively. Total quantities of secondary minerals in the anorthosite reaction are more abundant than those in the granite reaction.

  • The effects of the cation-to-anion resin ratio and bed depth on ion exchange performance of mixed-bed were studied at ultralow solution concentration. Breakthrough curves were experimentally obtained for NaCI solution as functions of resin ratio and bed depth. The bed depth affects the pattern of the sodium breakthrough curve but not the chloride breakthrough curve in beds because of the selectivity difference. Resin selectivity determines the shape of breakthrough curves, Some sodium and chloride breakthrough curves crossed at a point as a function of resin ratio. The lower cation-to-anion resin ratio showed the higher effluent concentration or treated volume of the crossover point regardless of the total resin weight.

  • The effect of temperature on stress corrosion cracking o f Al-brass used in vessel heat exchanger tube was studied in 3.5% NaCI + 0.1% $NH_4OH$ solution. The SCC test using a CDT(constant displacement test) and the specimens using a SEN(single edge notched) specimens. For setting the environment similar to working environment of a heat exchanger, the specimens was immersed in solution and solution flow onto the specimens were performed. The results are as follows : The latent time of stress corrosion crack occurrence gets shorter, as the temperature gets higher. Dezincification phase showed around the crack occupy wider range, as the temperature gets higher. Zn composition falls under 4% at the dezincifiction area.

Influence of Soil Salinity on the Interaction between Tomato and Broomrape plant (Orobanche cernua)

  • Tomato seedlings (20- days old) were transplanted to infested soil with Orobanche cernua and non-infested soils. All plants were maintained under 0, 25, 50 and 75 mM NaCl soil salinity levels throughout their growing period under greenhouse conditions. Plants grown in O. cernua infested soil infIltrated with 0, 25, and 50 mM NaCl solution for salinity showed significant reduction in their growth as well as their total soluble carbohydrate and protein contents in compared with those grown in non-infested soil. However, under 75 mM NaCI salinity level all plants showed similar growth values whether they were grown in O. cernua infested or non-infested soil. Starting at the fifth and through out the eighth week after transplantation there was a significant increase in plant height in the 0, 25 and 50 mM NaCl irrigated plants over other treatments. Irrigation with 50 mM NaCl significantly reduced the emergence of O. cernua (2/plant) and the number of attachments (4.4 attachments) on roots of tomato. Furthermore, irrigation with 75 mM NaCl resulted in complete elimination of O. cernua emergence.

Process Optimization for Concentration and Stabilization of Recombinant Endoxylanase Expressed in Bacillus subtilis (Bacillus subtilis에서 발현된 재조합 Endoxylanase 농축과 안정화 공정의 최적화)

  • A strong constitutive PJH promoter from Bacillus sp. was applied to overexpress the endoxylanase gene (639 bp) in Bacillus subtilis. The expression plasmid, pJHKJ4, was designed to contain the $P_{JH}$ promoter and open reading frame of endoxylanase including its own promoter. The plasmid was introduced into B. subtilis DB431 and the resulting transformant was grown on LB glucose medium. At the end of cultivation, the endoxylanase activity in the culture supernatant reached about 140 DIm!. The enzyme in the supernatant was concentrated by ultrafiltration (MW cut-off 10 kDa and 30 kDa) and ammonium sulfate precipitation. For the concentration of the enzyme, ultrafiltration was more efficient than 70% ammonium sulfate precipitation. The stabilization of concentrated enzyme solution at $50^{\circ}C$ was examined with various stabilizers such as NaCI, glycerol, polyethylene glycol, sorbitol, and $CaCI_2$. The most effective stabilizers were found to be NaCI and $CaCI_2$.

Dosimetry of $^{222}Rn$ Component in Outdoor Natural Background Radiation (자연 방사선량 중 $^{222}Rn$기여 성분 정량 해석)

  • A study for the assessment of dose given by outdoor radon to respiratory system has been carried out by making use of radon-cups containing CR-39 plastic track detectors. Detection efficiencies were determined by irradiation of the radon-cups in a standard radon chamber of known concentration. Thus determined detection factors of CR-39 plastic track detector in bare, open cup and filtered cup geometry are found to be 0.273, 0.0813 and 0.0371 $trmm^{-2}$/(37$Bqm^{-3}{\cdot}d$), respectively, which are chemically etched in 30% NaOH solution of $70^{\circ}C$ for 220 minutes. The outdoor radon concentrations measured at Taejeon(Chungnam National University) from May 1988 to March 1989 are in the range of 27.4 - 135.8 Bq/$m^3$(0.74 - 3.67pCi/l)by open cup and 16.7 - 143.9 Bq/$m^3$(0.45 - 3.89 pCi/l) by filtered cup, which yield overall annual average value of outdoor radon concentration of $70.8Bq/m^3$(1.91 pCi/l). Corresponding effective dose equivalent rate to respiratory system of ICRP standard man is assessed to be 520 nSv /h.
