Total Folate Contents of 15 Edible Plants Consumed in Korea Using Trienzyme Extraction Method (국내 소비되는 봄나물의 Trienzyme 추출법을 적용한 엽산 함량 분석)

  • Trienzyme digestion (AOAC Official Method 2004.05) procedure using protease, ${\alpha}$-amylase, and chicken pancreas conjugase was evaluated to determine its usefulness in the microbiological quantitation of total folate in foods. Folate values obtained by alkali hydrolysis (Korean Food Standards Codex) were compared to those obtained by the trienzyme method for four certified reference materials (CRM) representing diverse matrixes. Trienzyme treatment increased measurable folate from most CRM compared to levels found after alkali hydrolysis. The largest increases were observed with CRM 487 (pig liver, 5.8-fold) and CRM 121 (whole meal flour, 3.1-fold) after trienzyme digestion. Using trienzyme digestion method, total folate contents of raw and blanched edible plants were determined. Eleutherococcus senticosus ($146.9{\mu}g/100g$) showed the highest total folate content, followed by Aster glehni F. Schmidt ($142.8{\mu}g/100g$) and Ledebouriella seseloides H. Wolff ($140.4{\mu}g/100g$) on a wet weight basis. Blanching of samples resulted in an insignificant decrease in folate content for five samples and 11~63% reduction for nine samples. Our finding suggests that trienzyme digestion method is accurate for the determination of food folate in leafy vegetables.

Isolation of SYP61/OSMl that is Required for Salt Tolerance in Arabidopsis by T-DNA Tagging (애기장대에서 고염 스트레스 내성에 관여하는 OSM1/SYP61 유전자의 동정)

  • Salt stress is one of major environmental factors influencing plant growth and development. To identify salt tolerance determinants in higher plants, a large-scale screen was conducted with a bialaphos marker-based T-DNA insertional collection of Arabidopsis ecotype C24 mutants. One line for salt stress-sensitive mutant (referred to as ssm1) exhibited increased sensitivity to both ionic (NaCl) and nonionic (mannitol) osmotic stress in a root growth assay. This result suggests that ssm1 mutant is involved in ion homeostasis and osmotic compensation in plant. Molecular cloning of the genomic DNA flanking T-DNA insert of ssm1 mutant was achieved by mutant genomic DNA library screening. T-DNA insertion appeared in the first exon of an open reading frame on F3M18.7, which is the same as AtSYP61. SSM1 is SYP61/OSM1 that is a member of the SNARE superfamily of proteins required for vesicular/target membrane fusions and factor related to abiotic stress.

Mineralogical Study on the Clay Formation and Heavy Metal Speciation in the Acidified Soil Profile of the Onsan Industrial Area (온산공업지역 산성 토양 프로화일 내에서의 점토광물의 생성과 중금속 이온의 거동에 관한 광물학적 연구)

  • The present study is focused on the granite weathering and soil formation, and the heavy metal contamination in soils in the Onsan industrial area. For profile study, soil sampling was conducted on each depth and experimental analyses have been conducted on those samples. X-ray diffraction analyses show that clay minerals consist mainly of kaolin minerals, vermiculite, and minor illite. Most of kaolin minerals in the lower kiwi of the profile consist of halloysite as confirmed by formamide intercalation, but the content of halloysite decreases gradually toward the surface since it has been transformed to kaolinite in the upper part of the profile. Thermal treatment by heating at $110^{\circ}C,\;300^{\circ}C,\;and\;550^{\circ}C$ shows a diffuse and broad peak the between 10 and $14\;{\AA}$ region in X-ray diffractograms. This suggests the possible existence of the hydroxy-Al interlayerecl vermiculite. Na-citrate extraction method reaconfirms this result showing transition of $14\;{\AA}$ peak to $10\;{\AA}$ In by removing the interlayer materials and restoring the vermiculite to its original state. The occurrence of hydroxy-Al interlayered vermiculite is also supported by soil pH distribution room 3.9 In the lower part to 3.6 in the upper part of the profile. Sequential extraction experiment was conducted to investigate the states of heavy metals in soils. The experiment shows that relatively high amounts of heavy metals are concentrated in the upper part of the profile and that most of them are bound to Fe/Mn oxides and organic matters while less concentration in clay minerals. The result indicates that most of heavy metal pollutants are concentrated in the surface soil and that the low concentrations of heavy metals in clays are mainly due to the low adsorption capacities of clay minerals such as kaolin minerals and hydroxy-Al interlayered vermiculite in acidified soil condition.

Changes of Antioxidant Effects According to Greening Period of Astragalus membranaceus var. membranaceus, Senna occidentalis, Dianthus longicalyx, and Plantago asiatica Sprout Vegetables (녹화 처리기간에 따른 황기, 석결명, 술패랭이꽃 및 질경이 새싹채소의 항산화 효과 변화)

  • The potential use of 4 plant species, Astragalus membranaceus var. membranaceus, Senna occidentalis, Dianthus longicalyx and Plantago asiatica, as new sprout vegetables with high antioxidant function was examined in the present experiments. Seeds of above plants were allowed to germinate under light condition, and seedlings were maintained under dark condition for shoot growth in length for contain period of time. Then the seedlings were put under light for photosynthesis (greening treatment) for the period of 0${\sim}$3 days. Samples were collected to analyze the changes in antioxidant levels and activity, and it was observed that antioxidant substances were affected by greening treatments, depending on plant species. In A. membranaceus, the contents of total polyphenol was highest with no greening, total flavonoids with 3 days greening, DPPH radical scavenging effects with no greening, ABTS scavenging with 1 day greening, $Fe^{2+}$ chelating effects with no greening, and inhibitory activity against linoleic acid peroxidation with 3 day greening. In S. occidentalis, highest levels of antioxidant activity and radical scavenging effects were obtained by 2 day greening, $Fe^{2+}$ chelating effects by no greening and inhibitory activity against linoleic acid peroxidation by 1 day greening. In D. longicalyx, highest levels of antioxidant activity and $Fe^{2+}$ chelating effects were obtained by 2 day greening, $Fe^{2+}$ chelating effects by no greening and inhibitory activity against linoleic acid peroxidation by 1 day greening. In D. longicalyx, highest levels antioxidant activity and $Fe^{2+}$ chelating effects were observed with 3 day greening, and highest radical scavenging effects and inhibitory activity against linoleic acid peroxidation with no greening treatment. In P. asiatica, antioxidant activity and radicals scavenging effects were highest with 2 day greening, whereas highest chelating effects was obtained with no greening and highset inhibitory activity against linoleic acid peroxidation with 3 day greening. As the length of greening treatments influenced the antioxidant levels and function in plant species tested in this experiments, different culture methods are recommended for different plant species to get maximum health benefits out of sprout vegetables.

The Nutrient Composition of Commercial Kwamegi Admixed with Functional Ingredients (기능성 소재를 첨가한 시판 과메기의 영양성분 비교)

  • Kwamegi, a traditional Korean food, is made from the flesh of Pacific saury (the fish Cololabis saira semi-dried in a cold wind off the sea, and is well known in Korea as a valuable health food. Recently, several functional materials have been developed for supplementation of Kwamegi. Here, we compared and analyzed the nutrient composition, including overall composition and mineral, vitamin, fatty acid, and amino acid levels, of several commercial Kwamegi samples prepared with addition of functional components (unsupplemented Kwamegi, Kwamegi with chitosan, and Kwamegi overlaid with gold leaf). The levels of moisture (26.4-30.8%), crude protein (29.1-32.7%), and crude ash (1.6-1.9%) did not differ greatly among samples. However, the crude lipid content of Kwamegi overlaid with gold leaf (KOGL, 32.2%) was greater than that of untreated Kwamegi (CK, 24.5%) or of Kwamegi with added chitosan (KAC, 22.9%). The levels of vitamin $B_2$ (1.8-2.0 mg/100 g) and vitamin C (6.6-6.7 mg/100 g) did not differ greatly among Kwamegi samples. However, CK had a higher vitamin A content and a greater vitamin A potency than did KAC or KOGL. The various Kwamegi samples tested contained similar levels of fatty acids and amino acids. In conclusion, no particular differences in nutrient composition were evident when commercial Kwamegi samples supplemented with functional ingredients were tested

Influence of Liquid Pig Manure on Rice Growth and Nutrient Movement in Paddy Soil under Different Drainage Conditions (배수조건이 다른 논에서 돈분뇨 액비시용이 벼 생육 및 양분이동에 미치는 영향)

  • This study was performed to assess the influences of liquid pig manure (LPM) on the rice growth and nutrient behavior in soil under two different drainage comditions. Soils of paddy field were Jeonbuk and Gangseo series, and drainage conditions of the soils were imperfectly drained and moderate well drained, respectively, Application of LPM was based on nitrogen, and the treatments included 110 (LPM-N100%), 165 (LPM-N150%) and $220(LPM-N200%)kg\;N\;ha^{-1}$. In the LPM-N150% treatment, rice growth and yield were similar to the control treatment of conventional chemical fertilizer application. Rice yield of moderately well drainedfield were 3-11% higher than that of imperfectly drained field. Loss of nitrogen through $NH_3$ gas after application of LPM was higher in no plowing than in rotary plowing field. Losses of $NO_3-N$, $NH_4-N$, $SO_4$, Ca and K through runoff water were increased as the amount of LPM application increased. Losses of $NO_3-N$ and K through infiltration were higher in LPM-150% and LPM-N200% treatments compared to the chemical fertilizer treatment. Contents of $NO_3-N$ in infiltration water measured from trasplanting to tillering stage was higher at the moderately well drained condition than imperfectly drained condition. Contents of $P_2O_5$ and K in soil were accumulated by application of LPM compared to the chemical fertilizer plot.

The Growth and Yield Differences in Kenaf (Hibiscus cannabinus L.) in Reclaimed Land Based on the Physical Types of Organic Materials (간척지에서 유기물 투입 형태에 따른 케나프의 생육반응)

  • To improve the soil of reclaimed land, we added organic materials at a level of 3,000 kg/10 a. As a result, the electrical conductivity (EC) value of reclaimed soil decreased by 58%, the organic material content increased from 6.7 to 16.0 g/kg, the porosity increased from 1.57 to 1.31%, the soil hardness decreased from 20.2 to 17.9 mm and the plow layer was deepened from 19.8 to 26.8 cm. After these physiochemical improvements to the reclaimed soil, the growth phase of crops was improved compared to that of non-treatment crops. The height of kenaf (Hibiscus cannabinus L.) cultivated in the reclaimed land containing organic materials was increased by 18.8%. Especially, the improvement effects of pellet type manure compost and rice straw on kenaf were more preferable than those of other organic materials. When the kenaf was cultivated in the reclaimed land containing organic materials, the yield increased. The average yield of the treatment crops was 9,218 kg/10 a, 2.1 times higher than that of non-treatment crops. The most effective treatments to increase the yields were pellet type manure compost (10,848 kg/10 a, 148% increase), rice straw (120% increase) and chopped kenaf (95% increase). To increase the physicochemical enhancements to the reclaimed land soil and most improve yields, the most effective type of organic materials was the pellet. The organic material types that maintained a better growth phase and most increased the yield were the liquid and pellet types. When we used pellet type organic material, the plant height of kenaf was increased by 41% in comparison with that of the non-treatment crops and yield was increased by more than 122%. Additionally liquid type organic material improved the yield (by 127%).

Antiviral Activity of Hot-Water Extract and Its Ethanol Precipitate of Phellinus pini Fruiting Body (낙엽진흙버섯 자실체 유래 열수 추출물과 메탄을 침전물의 항바이러스활성)

  • The crushed fruiting body of Alaskan Porodaedalea pini (Brot.) Murrill (syn. Phellinus pini) was extracted in boiling water for 4 h with the yield of 20.5% in dry mass. This hot-water extract showed significant antiviral activity by inhibiting the plaque formation in HeLa cells by coxackievirus B3 (CVB3) and also showed highest inhibitory effect against neuraminidase activity among water extracts of various mushrooms. From the water extract, the ethanol precipitate (EP) and supernatant fraction (ES) were obtained through 75% ethanol precipitation with the yield of 43.3% and 28.3% in dry mass, respectively. Whereas ES did not show any detectable level of antiviral activity, EP showed significant dose-dependent inhibition of plaque formation by CVB3 in HeLa cells with an $EC_{50}$ (50% effective concentration) of 0.45 mg/mL. The cytotoxicity on HeLa cells by EP was relatively low with the $CC_{50}$ (50% cytotoxic concentration) of 2.25 mg/mL. EP also effectively inhibited neuraminidase activity in a dose-dependent manner showing up to 75% inhibition at 1.7 mg/mL. These results suggest that the hot-water extract and its EP of P. pini fruiting body can be a candidate for the development of a potent broad-range antiviral agent against influenza virus(Flu) as well as CVB3. The major active component of EP was shown to be a heteropolysaccharide-protein complex containing glucose as the main sugar residue with mole percentage of 79.8% and other sugars like galactose (19.2%), xylose (17%), mannose (5.8%), and fucose (4.6%) and a small portion (12.7%, in mass) of protein.

Physicochemical properties of dacquoise made with sugar or sugar replacements, tagatose, and erythritol (타가토스와 에리스리톨로 제조한 다쿠아즈의 물리화학적 특성 연구)

  • The objective of this study was to investigate the physicochemical characteristics and sensory evaluation of dacquoise made with sugar or sugar replacement (tagatose, erythritol). Sugar (S) was replaced with tagatose (T), and sugar powder was replaced with tagatose (TS), and erythritol (TE). S showed the highest sugar contents compared with TS and TE (p <0.05) but there were no significant differences with T. There were not significant differences in specific gravity and spread factor among the sweeteners. S and T tended to have higher loss factors than TS & TE. The more tagatose was increased, the more the redness value increased compared with S, and sucrose showed similar tendency for lightness (redness; TS > T > TE > S, lightness; S > T > TS > TE). TE showed good results for physical characteristics whereas T was better in the sensory test. The above results bear out that tagatose and erythritol both are suitable as alternative sweeteners for making dacquoise, but tagatose was better. Tagatose appeared to be a better replacement for sucrose in baking dacquoise, and dacquoise with 33% of the sugar replaced tagatose (T) could be suitable for use by diabetic and obese people. 95.1% tagatose (TS) maybe appropriate for diabetic patients. In conclusion, tagatose is a suitable sugar replacement for making dacquoise.

Microbiological Contamination Levels in the Processing of Korea Rice Cakes (떡류의 제조공정별 미생물학적 오염도 평가)

  • This study was conducted to evaluate microbial contamination levels of Korea traditional rice cakes such as Sirutteok, Garaetteok and Gyeongdan in the manufacturing process and environment. The microbial contamination levels such as total aerobic bacteria, fungi, coliforms, Escherichia coli, Staphylococcus aureus, Bacillus cereus and Clostridium perfringens of rice cake products were analyzed. The contamination levels of total aerobic bacteria, coliforms, fungi and B. cereus in raw materials were in the range of 2.4~4.5, ND~1.9, 1.2~2.1 and 1.0~2.1 log CFU/g, respectively. The microbial contamination levels of total aerobic bacteria, coliforms, fungi and B. cereus in manufacturing process of rice cakes were increased in the soaking and grinding steps and were decreased in steaming step. E. coli, S. aureus and C. perfringens were not detected in any manufacturing process and environment. The microbial contamination levels of raw materials and final products of rice cake were suitable for microbial safety standard in Korea. However, the manufacturing environment such as equipments and employee's sanitation were in trouble for microbial safety. The results of this study suggest that safety educatio n for personal hygiene and safetymanagement in processing environment are continuously required to assure safety in working environment and employee's individual hygiene.
