Heterogeneity of the SR-dependent Inward $Na^+-Ca^{2+}$ Exchange Current in the Heavily $Ca^{2+}-buffered$ Rat Ventricular Myocytes

  • Voltage-sensitive release mechanism was pharmacologically dissected from the $Ca^{2+}-induced\;Ca^{2+}\;release$ in the SR $Ca^{2+}$ release in the rat ventricular myocytes patch-clamped in a whole-cell mode. SR $Ca^{2+}$ release process was monitored by using forward-mode $Na^+-Ca^{2+}$ exchange after restriction of the interactions between $Ca^{2+}$ from SR and $Na^+-Ca^{2+}$ exchange within micro-domains with heavy cytosolic $Ca^{2+}$ buffering with 10 mM BAPTA. During stimulation every 10 s with a pulse roughly mimicking action potential, the initial outward current gradually turned into a huge inward current of $-12.9{\pm}0.5\;pA/pF$. From the inward current, two different inward $I_{NCX}s$ were identified. One was $10\;{\mu}M$ ryanodine-sensitive, constituting $14.2{\pm}2.3%$. It was completely blocked by $CdCl_2$ (0.1 mM and 0.5 mM) and by $Na^+-depletion$. The other was identified by 5 mM $NiCl_2$ after suppression of $I_{CaL}$ and ryanodine receptor, constituting $14.8{\pm}1.6%$. This latter was blocked by either 10 mM caffeine-induced SR $Ca^{2+}-depletion$ or 1 mM tetracaine. IV-relationships illustrated that the latter was activated until the peak in $30{\sim}35\;mV$ lower voltages than the former. Overall, it was concluded that the SR $Ca^{2+}$ release process in the rat ventricular myocytes is mediated by the voltage-sensitive release mechanism in addition to the $Ca^{2+}-induced-Ca^{2+}\;release$.

Mechanisms of Contraction Induced by Sodium Depletion in the Rabbit Renal Artery

  • In the rabbit renal artery, mechanisms of contraction by sodium depletion were investigated. The helical strips of isolated renal artery were immersed in the Tris-buffered salt solution. The contractions were recorded isometrically using a strain-gauge transducer. Na-free solution (Na was substituted by Li, choline or sucrose) produced contractions which were dependent on the nature of the Na substitutes. Na-free solution (choline) produced the contraction in ouabain-pretreated artery (Na loaded artery) even in the presence of verapamil. The amplitude of the contraction was dependent on the duration of the pretreatment with ouabain $(10\;^5M)$. Monensin potentiated the effect of ouabain on the contraction. Removal of Ca from bathing solution abolished the contraction and the substitution of Sr for Ca produced the contraction. Divalent cations such as Mg, Mn blocked the depolarization-induced contraction, while they had little effect on the Na-free contraction in Na loaded artery. These results suggest that the contraction induced by Na removal is dependent on the cellular Na content and may be caused by Ca influx via the Na-Ca exchange carrier.

[ $Ca^{2+}$ ]-dependent Long-term Inactivation of Cardiac $Na^+/Ca^{2+}$ Exchanger

  • Using BHK cells with stable expression of cardiac $Na^+/Ca^{2+}$ exchanger(BHK-NCX1), reverse mode(i.e. $Ca^{2+}$ influx mode) of NCX1 current was recorded by whole-cell patch clamp. Repeated stimulation of reverse NCX1 produced a cytosolic $Ca^{2+}$-dependent long-term inactivation of the exchanger activity. The degrees of inactivation correlated with NCX1 densities of the cells and were attenuated by reduced $Ca^{2+}$ influx via the reverse exchanger. The inactivation of NCX1 was attenuated by(i) inhibition of $Ca^{2+}$ influx with reduced extracellular $Ca^{2+}$, (ii) treatment with NCX1 blocker($Na^{2+}$), and (iii) increase of cytoplasmic $Ca^{2+}$ buffer(EGTA). In BHK-NCX1 cells transiently expressing TRPV1 channels, $Ca^{2+}$ influx elicited by capsaicin produced a marked inactivation of NCX1. We suggest that cytoplasmic $Ca^{2+}$ has a dual effect on NCX1 activities, and that allosteric $Ca^{2+}$ activation of NCX1 can be opposed by the $Ca^{2+}$-dependent long-term inactivation in intact cells.

Structure Study of Polycrystalline $Na_3YSi_3O_9$ and Its Substitutes Related to $Na_4CaSi_3O_9,\;Ca_3Al_2O_6$ Structure

  • The study of the $Na_3YSi_3O_9$ structure, by x-ray diffraction and infrared spectrum, showed that $Na_3YSi_3O_9$ is similar to $Na_4CaSi_3O_9$ except for its being pseudo-cubic instead of cubic. The peaks in the x-ray diffraction pattern of $Na_3YSi_3O_9$ could therefore be indexed on the basis of the $Na_4CaSi_3O_9$ cell. Also, modified $Na_3MSi_3O_9$ (M = Lu, Yb, Tm, Er, Y, Ho, Dy, Gd, Eu, and Sm) type compounds were synthesized by introducing excess sodium, decreasing M(III) concentration, and substituting small amount of phosphorus for silicon. The unit cell parameters of the composition $Na_{3.2}M_{0.7}Si_{2.9}P_{0.1}O_{8.7}$ were estimated from x-ray powder diffraction patterns using the Cohen method.

Pycnometric and Spectroscopic Studies of Red Phosphors Ca2+(1-1.5x)WO4:Eu3+x and Ca2+(1-2x)WO4:Eu3+x,Na+x

  • Red phosphors $Ca_{(1-1.5x)}Eu_xWO_4$ and $Ca_{(1-2x)}Eu^_xNa_xWO_4$ were synthesized with various concentrations x of $Eu^{3+}$ ions by using a solid-state reaction method. The crystal structure of the red phosphors were found to be a tetragonal scheelite structure with space group $I4_1/a$. X-ray diffraction (XRD) results show the (112) main diffraction peak centered at $2{\theta}=28.71^{\circ}$, and indicate that there is no basic structural deformation caused by the vacancies ${V_{Ca}}^{{\prime}{\prime}}$ or the $Eu^{3+}$ (and $Na^+$) ions in the host crystals. Densities of $Ca_{(1-1.5x)}Eu_xWO_4$ were measured on a (helium) gas pycnometer. Comparative results between the experimental and theoretical densities reveal that $Eu^{3+}$ (and $Na^+$) ions replace the $Ca^{2+}$ ions in the host $CaWO_4$. Also, the photoluminescence (PL) emission and photoluminescence excitation (PLE) spectra show the optical properties of trivalent $Eu^{3+}$ ions, not of divalent $Eu^{2+}$. Raman spectra exhibit that, without showing any difference before and after the doping of activators to the host material $CaWO_4$, all the gerade normal modes occur at the identical frequencies with the same shapes and weaker intensities after the substitution. However, the FT-IR spectra show that some of the ungerade normal modes have shifted positions and different shapes, caused by different masses of $Eu^{3+}$ ions (or $Na^+$ ions, or ${V_{Ca}}^{{\prime}{\prime}}$ vacancies) from $Ca^{2+}$.

  • Bioglass which is one of the surface active bionmaterials has a good biocompatibility but a poor mechanical strength, In the present work therefore two types of fluoride-containing bioglasses were coated on an alumina to improve mechanical strength. Crystallization of the coating layer and the hydroxyapatite formation on the bioactive glass coatings in tris-buffer solution were studied. When bioactive glass coated alumina was heat-treated Na2CaSi3O8 crystal was formed on the layer at lower temperature while wollastonite(CaSIO3) was obtained at higher temperature. Hydroxyapatite forming rate on the coating layer with Na2CaSi3O8 crystal was delayed with SiO2 contents in glass composition. However the hydroxyapatite was developed in 20minutes regardless SiO2 contents when the coating layer crystallized into wollastonite. More amount of P3+ ions were leached out of the coating layer with wollastonite than that with Na2CaSi3O8 crystal while Na+ and Ca2+ ions were leached out more easily from the Na2CaSi3O8 crystal containing coating layer.

Addition of Salts and Their Mixtures for Improvement of Storage Stability of Kimchi (김치의 저장성 향상을 위한 염혼합물의 첨가)

  • A study was carried out to investigate the control effect of sodium and potassium phosphates, sodium citrate and three different salts mixtures on kimchi fermentation when they were added into half-fermented kimchi in the concentration range of $0.001{\sim}0.01\;M$. The salts mixtures added were sodium phosphates mixture(CA-A), addition of $NaNO_2$, Ca-EDTA and BHA to CA-A(CA-B) and substitution of BHA with sodium citrate in CA-B. The results showed that sodium phosphates and sodium citrate significantly inhibited the kimchi fermentation while potassium phosphate had little effect. The order of control effect was $Na_3PO_4-Na_2HPO_4-sodium\;citrate-NaH_2PO_4-K_2HPO_4-KH_2PO_4$. Among the salts mixtures, CA-A showed the most reducing effect in the fermentation rate followed by CA-C and CA-A. The mixture of CA-C could extend the time of holding pH $4.2{\sim}4.4$ by approximately 6 times at $4{\sim}25^{\circ}C$ when it was compared to control. The microbial growth study of total and Leuconostoc mesenteroides also showed a very significant decrease in their numbers.

The Removal Rates of the Constituents of Litters in the Littoral Grassland Ecosystems in the Lake Paldangho V. Ca, Mg, and Na (팔당호 연안대 초지생태계에서 낙엽 구성성분의 유실률 V. Ca, Mg, Na)

  • This investigation was carried out to reveal the removal rates of Ca, Mg and Na among the constituents of the litters in Phragmites communis Miscanthus sacchariflorus, Typha augustata and Scirpus tabernaemontani ecosystems in the lake Paldangho. The removal constants of Ca, Mg and Na were determined by the mathematical model and the time required to decay to any percentage of each constituent was calculated by using this model. The results obtained in this study can he abstracted as follows; The removal constants of Ca, Mg and Na of the litters were 0.91, 0.86 and 0.71 in Phragmites communis, 0.64, 0.62, and 0.50 in Miscanthus sacchariflorus, 0.66, 0.28, and 0.56 in Typha angustata and 0.40, 0.55 and 0.63 in Scirpus tabernaemontani, respectively. These values of each constituent in Phragmites communis are the highest among the compared species and in each species, Ca among the studied constituent is higher than the others. The half times of Ca, Mg and Na required for the removal or accumulation of the litters on the grassland floor were 0.76, 0.80 and 0.98 years in Phragmites communis, 1.08, 1.12 and 1.39 years in Miscanthus sacehariflorus 1.05, 2.47 and 1.24 years in Typha angustata and 1.73, 1.26 and 1.10 years in Scirpus tabernaemontani, respectively. Effect of Strontium on the Uptake and Distribution of Calcium and Magnesium in Sugar beet (스티론티움에 의한 사탕무의 Ca과 Mg 흡수 및 분포)

  • To investigate the relationship between the translocation and distribution the monovalent K and Na and the divalent Sr and Ca, the natrophile and calcitrophic plant sugar beet (Beta vulgaris L.) was used. Strontium uptake and distribution are concentration and growth stage dependent. The highest Ca content occurred in the treatment of 4 : 1 mM ratio of Ca to Sr, while the highest Sr content in old leaves in the presence of 1 mM Ca and 4 mM Sr. The addition of low concentration of Sr stimulates Ca-uptake. Reversely. Sr-uptake is highest in the presence of 1 mM Ca. This result may be an antagonistic effect between Ca and Sr. The ratios of Mg to Ca and Sr are satisfactorily presented by the regression analysis. The sum of Sr and Ca contents are most significant linear to the ratio of Mg to one, showing a negative correlation. This result implies that the absorption of Mg and Ca or Sr is antagonistic. In the presence of only 5 mM Sr, K and Na-uptakes increases, while Sr in the presence of Ca does not affect the change in the K and Na assimilation and their ratios. The ratios of K to Na is also not changed. A little addition of Sr could more effectively retain the chlorophyll loss while only in the presence of Sr, the chlorophyll levels are considerably reduced.
