Evaluation of time-dependent antimicrobial effect of sodium dichloroisocyanurate (NaDCC) on Enterococcus faecalis in the root canal (이염화이소시아뉼산나트륨 제재의 근관 내 사용 시 시간에 따른 E. faecalis에 대한 항균성 평가)

  • The purpose of this study was to assess the antibacterial effect of sodium dichloroisocyanurate (NaDCC), sodium hypochlorite (NaOCl), and chlorhexidine (CHX) on Enterococcus faecalis and to evaluate and to compare the time-dependant antimicrobial effect of NaDCC with NaOCl and CHX in the root canal in vitro before and after instrumentation. Extracted Human single teeth were prepared by serial instrumentation technique. The samples were autoclaved and contaminated for 3 days with E. faecalis monocultures. The teeth were then divided into 4 groups Each group was irrigated and inserted with 2% NaOCl, 2% NaDCC, 2% CHX and steri)ized saline. After 6, 12, 24, 72h, and 1 week incubation, sterilized paper point was inserted into the root canal. Paper points containing root canal contents were then placed on the agar plate. And then each root cana) was prepared with #4 and #5 GG (Gates-Glidden) drill. The debris were collected in the sterilized microtube and the plates were incubated at $37^{\circ}C$ in an increased $CO_2$ atmosphere. After 24h incubation the growth of bacteria around the paper points were measured. NaOCl and NaDCC solution shows similar antimicrobial effect for E. faecalis at 6, 12, 24, 72h and 1 week. In centrol group, irrigated with sterilized saline, no antimicrobial effect was observed. The results are in agreement with other investigators, who have shown the bactericidal property and possibility of NaDCC as a root canal irrigation solution. Thus it seems that NaDCC solutions can be clinically applied into the root canal within 1 week after dilution.

The Effect of NaCI on the Growth and Ginsenoside Production from Ginseng Hairy Root (인삼모상근의 생장과 Ginsenoside 생산에 미치는 NaCl의 영향)

  • Korean ginseng (Panax ginseng C.A. Meyer) is very difficult to obtain stable production of qualified ginseng roots because of variable stresses in soil environments. High salt concentrations in the ginseng nursery soil environment of Korea is one of important reducing factors for the stable production of quality ginseng. These studies were accomplished to identify the growth rate and production of ginsenoside from ginseng hairy root against NaCI. In the MS liquid culture, the highest contents and productivity of ginsenosides were appeared at 4 week after onset of the treatment of 0.1 M NaCI. And 0.24 M NaCI was more effective on the growth of ginseng hairy root under light condition than dark condition. Plants generally produce secondary metabolites in nature as a defense mechanism against pathogenic and insect attack. In this study, NaCI acts as a kind of stress as well as elicitor for production of ginsenosides.

Desorption and Oxidation Properties of Saturated Arsenate on LDHs (하이드로탈사이트류에 포화 흡착된 비소(V)의 탈착 및 산화 특성)

  • In order to establish the design parameters of adsorption for arsenic compounds with hydrotalcite including chlorine ion, the basic properties of adsorption and desorption as well as the oxidation of As (III) were examined in batch tests. The maximum adsorption capacities of arsenite and arsenate were 6.2 mg-As(III)/g and 103 mg-As (V)/g, respectively. Although 80.4% of maximum desorption was shown in 20% NaOH solution, 5~10% of NaOH was recommended considering operating benefits, where the proper condition of the desorption was in the range of 73% to 80%. The most suitable desorption condition was in the combination of NaCl (10~20%) and NaOH (5~10%). Within 2 minutes, As (III) was easily oxidized to As (V) with 0.0001 N KMnO4, where the maximum oxidization ratio was shown to 98.9%.

Effect of Cisplatin on $Na^+/H^+$ Antiport in the OK Renal Epithelial Cell Line

  • Cis-diamminedichloroplatinum II (cisplatin), an effective antitumor agent, induces acute renal failure by unknown mechanisms. To investigate direct toxic effects of cisplatin in the renal proximal tubular transport system, OK cell line was selected as a cell model and $Na^+/H^+$ antiport activity was evaluated during a course of cisplatin treatment. The cells grown to confluence were treated with cisplatin for 1 hour, washed, and incubated for up to 48 hours. At appropriate intervals, cells were examined for $Na^+/H^+$ antiport activity by measuring the recovery of intracellular pH (pHi) after acid loading. Cisplatin of less than 50 ${\mu}M$ induced no significant changes in cell viability in 24 hours, but it decreased the viability markedly after 48 hours. In cells exposed to 50 ${\mu}M$ cisplatin for 24 hours, the $Na^+-dependent$ pHi recovery (i.e., $Na^+/H^+$ antiport) was drastically inhibited with no changes in the $Na^+-independent$ recovery. Kinetic analysis of the $Na^+-dependent$ pHi recovery indicated that the Vmax was reduced, but the apparent Km was not altered. The cellular $Na^+$ and $K^+$ contents determined immediately before the transport measurement appeared to be similar in the control and cisplatin group, thus, the driving force for $Na^+-coupled$ transport was not different. These results indicate that cisplatin exposure impairs the $Na^+/H^+$ antiport capacity in OK cells. It is, therefore, possible that in patients treated with a high dose of cisplatin, proximal tubular mechanism for proton secretion (hence $HCO_3^-$ reabsorption) could be attenuated, leading to a metabolic acidosis (proximal renal tubular acidosis).

Effect of NaCl on Biofilm Formation of the Isolate from Staphylococcus aureus Outbreak Linked to Ham

  • The objective of this study was to evaluate the effects of NaCl on the biofilm formations of the isolate from Staphylococcus aureus outbreaks linked to ham. The S. aureus ATCC13565 isolated from ham was exposed to NaCl concentrations of 0%, 2%, 4%, and 6% supplemented in tryptic soy broth (TSB) for 24 h at $35^{\circ}C$, followed by plating 0.1 mL of the culture on tryptic soy agar containing 0%, 2%, 4%, and 6% NaCl, respectively. After incubating at $35^{\circ}C$ for 24 h, the colonies on the plates were collected and diluted to $OD_{600}$ = 0.1. The diluents of S. aureus were incubated on a 96-well flat bottom plate containing TSB plus the appropriate NaCl concentrations, and the biofilm formation was quantified by crystal violet staining after being incubated at $35^{\circ}C$ for 9 h. Confocal laser scanning microscope (CLSM) was also used for visualizing the biofilm formation of S. aureus at NaCl concentrations of 0%, 2%, 4%, and 6%. The transcriptional analysis of biofilm-related genes, such as icaA, atl, clfA, fnbA, sarA, and rbf, was conducted by quantitative real-time PCR. Crystal violet staining and CLSM showed that the biofilm formations of S. aureus increased (p<0.05) along with the NaCl concentrations. Moreover, the expression of the icaA genes was higher at the NaCl concentrations of 4% and 6% as compared with 0% of NaCl by approximately 9-folds and 20-folds, respectively. These results indicated that the NaCl formulated in processed food may increase the biofilm formations of S. aureus by increasing the icaA gene expressions.

Hydrothermal Synthesis of Zeolite from Coal Fly Ash (석탄회를 이용한 제올라이트의 수열합성)

  • Phillipsite-type zeolite powders were synthesized from domestic coal fly ash by hydrothermal reaction with 2.0 M-3.5M NaOH solution at $80^{\circ}C{\sim}120^{\circ}C$ for 24 h. The properties of zeolite were investigated by XRD, SEM and IR spectroscopy. The effects of chemical composition of fly ash, reaction temperature and NaOH concentration on the zeolite synthesis were well appeared on IR spectra. It is found that the $TO_4$(T=Si, Al) tetrahedra have a well ordered structure as the $SiO_2$ content, synthesis temperature and NaOH concentration become high.

The Influence of Mixed NaCl-KCl Salt on Sodium Intake and Urinary Excretion of Sodium and Potassium (혼합식염 (NaCl-KCl) 이용 조리시 나트륨${\cdot}$칼륨 섭취와 소변 중 나트륨${\cdot}$칼륨 배설에 관한 연구)

  • The purpose of this study was to investigate the influence of mixed NaCl-KCl salt on sodium intake and urinary excretion of sodium and potassium. In this study, 3-day food records for pre-experimental diet and 24-hr urine collected for 2-days, 6-day experimental diet food and 24-hr urine were used to evaluate the relationship between Na metabolism. In the experimental diet food, mixed NaCl-KCl salt was added. During the pre-experimental diet period, intakes of Na and K were 178.2 mEq and 56.4 mEq, respectively. The urinary excretion of Na and K in 24-hr were 139.6 mEq, 27.7 mEq, respectively and urinary Na/K ratio was 6.6. During the experimental diet period, intakes of Na and K were 130.2 mEq and 120.4 mEq, respectively. The urinary excretion of Na and K in 24-hr were 100.2 mEq, 37.1 mEq, respectively and urinary Na/K ratio was 2.8. According to this study, it is concluded that mixed NaCl-KCl salt diet decreased the intake of Na, and increased the intake of K.

Measurement of Na intake in Korean Adult Females (우리나라 성인 여성의 Na 섭취량 측정방법의 모색)

  • Dietary intakes of na include both the amount present in food materials (nondiscretionary Na intake) and the amount added during cooking and eating to increase salty taste and flavor (discretionary Na intake). In the present study, total Na intake was measured by duplicate food sample collected from each subject for 1 day, nondiscretionary Na intake was calculated by dietary intake record, optimum gustation of salt was measured of from pooled 3-day urine collection in healthy female college students, to measure Na intake more accurately and to find suitable method to estimate total Na intake. Mean values of total, discretionary and mondiscretionary Na intakes were 169.6 mEq, 46.2mEq, and 123.4mEq. respectively. Mean 24-hour urinary Na excretion was 137.9mEq., which was about 84.5% of total intake. Subjects with optimum gustation of salt equal to or higher than 0.5% had significantly higher total Na intake and urinary Na excretion compared to subjects with optimum gustation of salt lower than 5%. Total Na intake of subjects were significantly correlated with optimum gustation of salt, non-discretionary and discretionary Na intakes, and urinary Na excretion. Multiple regression analysis showed that the best estimate of total Na intake is obtained when both optimum gustation of salt and non-discredinary Na intake were used as independent variables (r=.7071). Among the equations using one independent variable, regression, equation with urinary Na excretion provides the best approximation (r=.6627) of total Na intake.

Effects of Aspirin on Nitrite Toxicity in Cultured Eel (Anguilla japonica) (양식 뱀장어 (Anguilla japonica)의 아질산 독성에 미치는 aspirin의 투여 효과)

  • The effect of aspirin on the recovery of nitrite-induced methemoglobinemia in cultured eels (Anguilla japonica) was studied. Methemoglobinemia was induced by exposing eels to nitrite (120 ㎎ NO2-N/ℓ) for 24 hr. The nitrite exposed eels were bathed in 20 ppm aspirin solution (Aspirin), 0.8 % NaCl solution (NaCl), 20 ppm aspirin plus 0.8% NaCl solution (NaCl + Aspirin) and 50% nitrite free water(control) for 24 hr to recover from nitrite toxicity. Peripheral blood was taken from the arterious bulb from all groups to analyse hematocrit value, hemoglobin concentration, and nitrite concentration of the blood. Histopathological features of gill were also observed. Aspirin and control groups were more effective than NaCl and NaCl + Aspirin groups in recovery of hematocrit value and hemoglobin concentration, methemoglobin rate and nitrite concentration. The histopathological features on the gill of aspirin group were similar to those of normal eels, but other groups showed focal hyperemia in the lamellar carpillaries, epithelial hyperplasia. These results suggested that aspirin was very effective to recover from methemoglobinemia in nitrite-induced cultured eels.
