A comprehensive study was undertaken to evaluate the effect of naked neck (Na) gene on immune competence, serum biochemical parameters and carcass quality traits in three genotypes (NaNa, Nana and nana) of the naked neck chicken under a tropical climate (Southern India). Sixty day-old chicks (20 from each genotype) were selected randomly and reared under similar environmental conditions up to eight weeks of age. The cell mediated immune (CMI) response to phytohaemoagglutinin-P (PHA-P) was significantly higher ($p{\lgq}0.01$) in NaNa and Nana genotypes compared to nana birds. The humoral response as measured by antibody titre to sheep red blood cells (SRBC) was also significantly higher in NaNa. The total cholesterol, LDL and VLDL cholesterol levels were significantly ($p{\leq}0.01$) lower whereas HDL cholesterol level was significantly higher in NaNa and Nana compared to nana genotype. The presence of Na allele significantly increased the live weight and dressing yield, and decreased the feather cover and abdominal fat. The naked neck genotypes (NaNa/Nana) performed better than the normal (nana) siblings for almost all the traits studied.
This study was undertaken to determine whether Buthus exract(BTE) affects Na^+-K^+-ATPase$ activity of nervous tissues. The enzym activity was measured in synaptosomal fraction prepared from rabbit brain cortex. Na^+-K^+-ATPase$ activity was inhibited by BTE over concentration range of 0.05-0.5% in a dose-dependent manner. The enzyme activity was increased by an increase in $Na^+$ concentration from 5 to 100mM, $K^+$ concentration from 0.5 to 10mM, and $Mg^{2+}$ concentration from 0.2 to 5mM. These changes in ion concentrations did not produce any effect on the inhibitory effect of BTE on $Na^+-K^+-ATPase$ activity. An increase in ATP concentration from 0.1 to 3mM caused an increase in the enzyme activity. The inhibition of the enzyme activity by BTE were not different between two ATP concentrations. A sulfhydryl group protector DTT prevented PCMB-induced inhibition of $Na^+-K^+-ATPase$ activity, but the BTE-induced inhibition was not altered by DTT. The inhibition of enzyme activity by combination of ouabain and BTE was not different from that by Buthus alone. These results suggest that Buthus exerts inhibitory effect on $Na^+-K^+-ATPase$ activity in cerebral synaptosomes, and the action mechansim is similar to that of ouabain.
This study evaluated the dermal cytotoxicity of sodium bromate (NaBrO3) and the protective effect of Aster tataricus L. (AT) extract against NaBrO3-induced cytotoxicity in the cultured NIH3T3 fibroblasts. For this study, it was done the antioxidative effects such as electron donating (ED) activity and lipid peroxidation (LP) activity as well as cell viability. NaBrO3 significantly decreased cell viability in a dose-dependent manner and its XTT50 value was measured at a concentration of 54.4 μM in these cultures. The cytotoxicity of NaBrO3 was determined as highly-toxic by Borenfreund and Puerner's toxic criteria. The quercetin, antioxidant significantly increased cell viability against NaBrO3-induced cytotoxicity. Regarding the protective effect of Aster tataricus (AT) L. extract on NaBrO3-induced cytotoxicity, AT extract significantly increased the cell viability, the ED ability and the inhibitory ability of LP. From these findings, it suggested that the oxidative stress is involved in the cytotoxicity of NaBrO3, and AT extract effectively protected NaBrO3-induced cytotoxicity by antioxidative effects. Conclusively, the natural component like AT extract may be a putative therapeutic agent for the diminution or treatment of the cytotoxicity correlated with oxidative stress like hair dye component, NaBrO3.
The catalytic hydroxylation of several cycloalkanes in dichloromethane have been investigated using In(Ⅲ)-, Tl(Ⅲ)-porphyrin complexes as a catalyst and NaClO, $NaClO_{2}$, $H_{2}O_{2}$ as a terminal oxidant. Porphyrins were TPP and ($F_{20}$)TPP (TPP = tetraphenylporphyrin) and substrates were cyclopentane, cyclohexane, cycloheptane and cyclooctane. The substrate conversion yield was discussed according to the substituent effect and hinderance effect of metalloporphyrin and the radius effect of non-redox metal ion. The conversion yield of cycloalkane followed the order of $ C_{5} $ < $ C_{6}$ < $ C_{7}$ = $ C_{8}$. In this experimental condition $NaClO_2$ was rather efficient terminal oxidant than NaClO and $H_{2}O_{2}$.
Four kinds of neutral sodium salts with different anions, NaF, NaCl, NaBr, and NaI, were added to the dye bath to accurately understand the effect of anions on the reactive dyeing of silk with C. I. Reactive Black 5. The sodium cation towered the negative surface potential of the silk and increased the dye-uptake on fille fabric as reported previously. However, because of the discrepancy in the anions'inhibition power from cation's lowering: the surface negative potential the amount of the dye on the silk fiber was different from each other in the order of $F^->Cl^-> Br^-I^-$. The activation energy(E$_{a}$) lot the dyeing was in the order of $F^->Cl^-> Br^-I^-$ but the dye-uptake on the fabric and the activation free energy$(\Delta{G}^*)$, the real energy barrier fort the reaction, were in the order of $F^->Cl^-> Br^-I^-$ because the strength of the interaction of the anions with sodium cations was the salute as the order of the latter. In other words F$^{[-1000]}$ exerted the weakest electrostatic force on $Na^+$ and competed with the dyestuff anions least of all. The decrease in $\Delta{S}^*$ may be due to the looesly bonded activated complex of dyestuff anions, sodium cations and fiber molecules at transition state. It was clarified from the Brёnsted equation that sodium salts with different anions also had fille ionic strength effect and the specific salt effect on the reactive dyeing.g.
Absorption enhancing effect of 5 MSA-Na at the mucous memberanes of the rectum and duodenum was studied via in vitro sac test. There were no significant differences between two membranes. Effect of 5MSA-Na on the transfer of CMZ to the tissue was also studied via in situ loop method. There was no significant difference at the duodenum, compared to the control when CMZ was administered intravenously. Transfer of CMZ to the loop of the rectum was increased in the presence of 5MSA-Na compared to the control, which might be attributed to the enhanced permeability of the mucous memberane of the rectum.
In the rabbit renal artery, mechanisms of contraction by sodium depletion were investigated. The helical strips of isolated renal artery were immersed in the Tris-buffered salt solution. The contractions were recorded isometrically using a strain-gauge transducer. Na-free solution (Na was substituted by Li, choline or sucrose) produced contractions which were dependent on the nature of the Na substitutes. Na-free solution (choline) produced the contraction in ouabain-pretreated artery (Na loaded artery) even in the presence of verapamil. The amplitude of the contraction was dependent on the duration of the pretreatment with ouabain $(10\;^5M)$. Monensin potentiated the effect of ouabain on the contraction. Removal of Ca from bathing solution abolished the contraction and the substitution of Sr for Ca produced the contraction. Divalent cations such as Mg, Mn blocked the depolarization-induced contraction, while they had little effect on the Na-free contraction in Na loaded artery. These results suggest that the contraction induced by Na removal is dependent on the cellular Na content and may be caused by Ca influx via the Na-Ca exchange carrier.
In this research, we examined the effect of NaCl on the growth, energy metabolism, and proton motive force of Halomonas salina, and the effect of compatible solutes on the bacterium growing in the high salinity environment. H. salina was isolated from seawater and identified by 16srDNA sequencing. The growth of H. salina was not enhanced by the addition of external compatible solutes (choline and betaine) in the high salinity environment. The resting cells of H. salina absorbed more glucose in the presence of 2.0 M NaCl than in its absence. H. salina did not grow in the medium with either KCl, RbCl, CsCl, $Na_2SO_4$, or $NaNO_3$, in place of NaCl. The optimal concentration of NaCl for the growth of H. salina ranged from 1.4 M to 2.5 M, and the growth yield was decreased in the presence of NaCl below 1.4M and above 2.5M. The activity of isocitrate dehydrogenase, pyruvate dehydrogenase, and malate dehydrogenase of H. salina was not inhibited by NaCl in in vitro test. The proton translocation of H. salina was detected in the presence of NaCl only. These results indicate that NaCl is absolutely required for the normal growth and energy metabolism of H. salina, but the bacterial growth is not enhanced by the compatible solutes added to the growth medium.
