Regulation of Neural Stem Cell Fate by Natural Products

  • Neural stem cells (NSCs) can proliferate and differentiate into multiple cell types that constitute the nervous system. NSCs can be derived from developing fetuses, embryonic stem cells, or induced pluripotent stem cells. NSCs provide a good platform to screen drugs for neurodegenerative diseases and also have potential applications in regenerative medicine. Natural products have long been used as compounds to develop new drugs. In this review, natural products that control NSC fate and induce their differentiation into neurons or glia are discussed. These phytochemicals enable promising advances to be made in the treatment of neurodegenerative diseases.

Evaluating of Validity on Peri-operative Hypothermia Management Evidence Based Guideline (저체온 관리 근거중심 가이드라인의 국내 타당성 검증)

  • The purpose of this study was to evaluate the appropriateness and applicability of a evidence-based peri-operative hypothermia management guideline developed by NCC-NSC in Korea using expert group. The expert group was composed of 180 registered nurses in surgical units and recovery room, and physicians (surgeons and anesthesiologists) who had enough experiences and knowledgeable on the management of surgical patients. Validity of guidelines were evaluated in terms of appropriateness and applicability using 9 point scale. Most of the recommendations in the guideline received high score of above 6-8 point both in appropriateness and appropriateness. However, most of the recommendations were received significantly lower score in applicability than appropriateness. This research can be used as a step to develop more acceptable hypothermia management guideline for the patients undergoing abdominal surgery in Korean. In addition, further studies which identify the barriers that inhibit applicability of recommendations should be investigated.

Applicability of CADMAS-SURF Code for the Variation of Water Level and Velocity due to Bores (CADMAS-SURF에 의한 단파의 수위 및 유속변화에 대한 예측정도의 검토)

  • This study investigates the applicability of CADMAS-SURF (Super Roller Flume for Computer Aided Design of a MArtime Structure) code basal on the Navier-Stokes solver to predict bore phenomena. The time variation of ware levels and velocities due to the bore propagation were computed for the different bore strength conditions. In order to verify the numerical results by CADMAS-SURF, laboratory experiments were also performed, using the DPIV and LDV measuring system. The numerical results were compared to the experimental data and the analytical predictions by the NSC method basal on fully nonlinear shallow-water theory by the method of characteristics. It appears that the CADMAS-SURF slightly overestimated the water-surface level measured by the laboratory experiments and its discrepancy becomes prominent as the bore strength increases. The predicted propagation speed for a bore was also slaver than that by the experiment and NSC method. However, the temporal variations in velocities were in relatively good agreement with the experimental data for all cases, except for overshooting and undershooting in the front face of a bore, which may be derived from the numerical instability. Further, CADMAS-SURF successfully simulated the decrease in the water level and velocity caused by the effects of negative waves reflected from the upstream end wall.

Flow and Strength Properties of Cement Mortar Mixed with High Range Water Reducer Containing Carboxylic Acid(II) (카르본산계 고성능 감수제를 첨가한 시멘트 모르타르의 유동 특성(II))

  • In the previous study, styrene mdleic dnhydride copolymer(SMA) as synthesized flom styrene and rnale~c dnhydr~de and further redcted with sulfuric acid to obtam water soluble SMA. In thls study, the flow dnd strcngth tests of cement mortar rmxed wth copolymers wele carried out to evaluate the capability of copolymers as high range water reducer for the con crete. It was found from flow exper~ment that the fluidity of cenient mortar rmxed wth sulfonated SMA(SSMAj was larger thdn that mxed ulth amnophenol substituted SSMA (SmSMAj. The decreasing rate of the flow of cement mortar rmxed ulth SSMA and SmSMA was significantly lower than that mixed ulth naphthalene condensate(NSC) The compressslve strength of the hardened cement mortars containing 0.5% copolymers after 28 dys curing was exarmned. 'The compressive strength of hdrdened cement mortar containing SSMA and SmSMA was mcreased up to 31% and 13%, respectively, when omp pared to the plain. As the results, the copolyniers(SSMA and SrnSMA) used in thls study are greatly expected as a good high range water reducers for the concrete.

The Effects of Digital Storytelling on Mathematical Communication Apprehension in 6th graders' Mathematics Instruction (초등학교 6학년 수학수업에 적용한 디지털 스토리텔링이 수학적 의사소통불안에 미치는 효과 분석)

  • The purpose of this study was to investigate how Storytelling in Mathematics Instruction effects students' mathematical communication apprehension. In order to do this, I selected two grade six classes with no significant difference on the Communication Apprehension(CA) test. I applied normal story telling and digital story telling to each of the classes for ten weeks then analyzed the effects through the post CA test. As a result, for Normal Storytelling Class (NSC), there was no meaningful difference in the ex ante and ex post CA test results. However, for Digital Storytelling Class (DSC), there was a meaningful difference in regards to the communication apprehension subgroup. Also, between the two NSC and DSC groups' post CA results, there was a meaningful difference in mathematics lesson and subgroup factors. Consequently, these results suggest the appliance of Digital Storytelling helps lower CA in $6^{th}$ graders participation in math class and subgroup.

Identifying Adverse Events Using International Classification of Diseases, Tenth Revision Y Codes in Korea: A Cross-sectional Study

  • Objectives: The use of administrative data is an affordable alternative to conducting a difficult large-scale medical-record review to estimate the scale of adverse events. We identified adverse events from 2002 to 2013 on the national level in Korea, using International Classification of Diseases, tenth revision (ICD-10) Y codes. Methods: We used data from the National Health Insurance Service-National Sample Cohort (NHIS-NSC). We relied on medical treatment databases to extract information on ICD-10 Y codes from each participant in the NHIS-NSC. We classified adverse events in the ICD-10 Y codes into 6 types: those related to drugs, transfusions, and fluids; those related to vaccines and immunoglobulin; those related to surgery and procedures; those related to infections; those related to devices; and others. Results: Over 12 years, a total of 20 817 adverse events were identified using ICD-10 Y codes, and the estimated total adverse event rate was 0.20%. Between 2002 and 2013, the total number of such events increased by 131.3%, from 1366 in 2002 to 3159 in 2013. The total rate increased by 103.9%, from 0.17% in 2002 to 0.35% in 2013. Events related to drugs, transfusions, and fluids were the most common (19 446, 93.4%), followed by those related to surgery and procedures (1209, 5.8%) and those related to vaccines and immunoglobulin (72, 0.3%). Conclusions: Based on a comparison with the results of other studies, the total adverse event rate in this study was significantly underestimated. Improving coding practices for ICD-10 Y codes is necessary to precisely monitor the scale of adverse events in Korea.

A Study on Alternatives for Developing the Workers' Education Programme in the Port Logistics Field using a NCS Model (국가직무능력표준을 활용한 항만물류분야 재직자의 교육과정 개발 방안에 관한 연구)

  • This study is to develop a curriculum for training workers in the port logistics field. NSC is a standardized method improving the capability and knowledge scientifically and systematically in order for individual to carry out its own job and task successfully. To develop a curriculum for workers in the port logistics field, the following methodology is used. First, questionnaire is carried out. Second, a MECE method is used to draw out a proper education programme. This study can contribute to improving the job capability of employers working at the port logistics industry. Further, it can improve firms' competitive power and furthermore national competitive edge by introducing the filed-oriented training programme to port logistics industry. In the future, a developed education is to be applied and evaluated through more intensified industry, academy, research linkage.

A Study on Feedstuff Utilization of Agricultural By-Products as a Major Feed Source in Korean Native Goat (농산부산물을 이용한 한국재래산양의 사료개발에 관한 연구)

  • This study was carried out to investigate the amounts of voluntary intake, digestibility and nitrogen retention in Korean native goats (KNG) fed agricultural by-products containing rice straw (RS) and apple pomace (AP) and to obtain a basic information for establishing the feeding system of KNG. The result are as follows. 1. Among the chemical composition of experimental diets, the highest values in crude protein (CP ; 18.6%) and crude ash contents (10.7%) were observed in alfalfa hay. Those of RS+AP treatment were significantly low 6.0 and 4.8%, respecitively. Acid detergent fiber (ADF), ether extract and non-structural carbohydrate (NSC) contents were shown an adverse tendency. 2. Dry matter (DM) intakes per day in KNG fed RS+commercial diet (CD), RS+AP and RS+AP+CD were significantly lower (P<0.05) 210.3, 228.3, respectively than 358.1g in alfalfa hay. 3. DM intakes per basal weight expressed as DM g/kg of BW0.75 and DM g/kg of BW(%) were highest (P<0.05) 60.5g and 3.3%, respectively in KNG fed alfalfa hay, any other treatments showed 35.6 to 42.5g and 2.0 to 2.3%, respectively and this result was similar to those of DM intake per day. 4. Digestibilities of DM, organic matter, CP, ADF, neutral detergent fiber (NDF). crude ash and ether extract in alfalfa hay and RS+AP+CD treatment were significantly higher (P<0.05) than RS+CD treatment. Those of CP, ADF and NDF of RS+AP treatment in not significantly different with these treatments. 5. NSC digestibility was higher in alfalfa hay (66.7%) than those of other treatments (44.2~52.0% ; P<0.05). 6. Nitrogen retentions expressed as N retention(g) and N retention (%) in KNG were highest (P<0.05) 0.6g and 15.6%, respectively in RS+AP+CD treatment and RS+CD treatment was lowest (P<0.05) -06g and -21.4%, respectively.

Effects of Schisandrae Fructus 70% Ethanol Extract on Proliferation and Differentiation of Human Embryonic Neural Stem Cells (오미자 70% 에탄올 추출물의 신경줄기세포 증식과 분화에 미치는 영향)

  • Neural stem cells (NSCs), with self-renewal and neuronal differentiation capacity, are a feasible resource in cell-based therapies for various neurodegenerative diseases and neural tissue injuries. In this study, we investigated the effects of Schisandrae Fructus (SF) on proliferation and differentiation of human embryonic NSCs. Treatment with 70% ethanol extract of SF increased the viability of NSCs derived from human embryonic stem cells, which was accompanied by increased mRNA expression of cyclin D1. Whereas 70% ethanol extract of SF also decreased the mRNA expression of nestin, it increased class III ${\beta}$-tublin (Tuj-1) and MAP2 in both growth and differentiation media. Lastly, we found increased mRNA expression of BDNF in SF-treated NSCs. In conclusion, our study demonstrates for the first time that SF induced proliferation and neuronal differentiation of NSCs and increased mRNA expression of BDNF, suggesting its potential as a regulator of NSC fate in NSC-based therapy for neuronal injuries from various diseases.

  • As part of a research program to develope 3rd generation anti tumor platinum complexes, a series of platinum complexes which have 4, 5-bis-(aminomethyl)- 1, 3-dioxolane derivatives as bidenate amine ligands, represented by the general structual formula was prepared. The R$_1$ and/or R$_2$ substituents in this series of platinum complexes can be hydrogen. alkyl, of jointly formed cyclohexane. Two Xs can be a bidenate leaving ligand such as 1, 1-cyclobutanedicarboxylate, malonate, dimethylmalonate, ethylmalonate, glycolate, L-lactate, or N-methyliminodiacetate. From based on the pharmacological and toxicological studies, we have chosen SKI 2053R, cis-malonato[(4R, 5R)-4, 5-bis(aminomethyl)-2-isopropyl-1, 3-dioxolane] platinum(II) complex (NSC D644591) as a candidate for clinical evaluation. The antitumor activity of a new anti tumor platinum complex, cis-malonato [(4R, 5R)-4, 5-bis(aminomethyl)-2-isopropyl-1, 3-dioxolane] platinum(II) (SKI 2053R, NSC D644591), was compared with those of cisplatin and carboplatin using murine tumors. We evaluated three platinum complexes against L1210/CPR, a subline of L1210 leukemia resistant to cisplatin for their abilities to overcome tumor resistance to cisplatin. The in vitro cytotoxicity of SKI 2053R to L1210 cell line was 2.5-fold less potent thann that of cisplatin, and was 10-fold more cytotoxic than that of carboplatin. SKI 2053R retained similar cytotoxic effect and anti tumor activity to L1210/CPR cell line, like the cytotoxicity of SKI 2053R to L1210 cell line, while either cisplatin or carboplatin had not property to overcome the acquired cisplatin-resistance. SKI 2053R exhibited greater or comparable antitumor activity than cisplatin or carboplatin in murine tumor models.
