Medium optimization for keratinase production by a local Streptomyces sp. NRC 13S under solid state fermentation

  • Thirteen different Streptomyces isolates were evaluated for their ability to produce keratinase using chicken feather as a sole carbon and nitrogen sources under solid state fermentation (SSF). Streptomyces sp. NRC 13S produced the highest keratinase activity [1,792 U/g fermented substrate (fs)]. The phenotypic characterization and analysis of 16S rDNA sequencing of the isolate were studied. Optimization of SSF medium for keratinase production by the local isolate, Streptomyces sp. NRC13S, was carried out using the one-variable-at-a-time and the statistical approaches. In the first optimization step, the effect of incubation period, initial moisture content, initial pH value of the fermentation medium, and supplementation of some agro-industrial by-products on keratinase production were evaluated. The strain produced about 2,310 U/gfs when it grew on chicken feather with moisture content of 75% (w/w), feather: fodder yeast ratio of 70:30 (w/w), and initial pH 7 using phosphate buffer after 8 days. Based on these results, the Box-Behnken design and response surface methodology were applied to find out the optimal conditions for the enzyme production. The corresponding maximal production of keratinase was about 2,569.38 U/gfs.

  • In this paper, we investigate MSAP-TMFT in TICN systems that will be future NCW-based tactical networks. Although MSAP-TMFT implements the WiBro technology, we propose to design a NRC(NAT Relay Cell) that functions as a relay station and at the same time as a base station in combat environments. NRCs support extension of communication distance, increased data rate, efficient radio resource management, and survavibility of combat networks. From simulation results, we show that NRCs improve the efficacy of radio resource management and system throughput compared to the legacy systems.

Design and Implementation of a Digital Control Unit for an Oxygenaire Servo Baby Incubator

  • This paper introduces a design and implementation of a digital control unit for an Oxygenaire Servo Baby Incubator. The control unit is designed and implemented according to international standards. The control unit is based on an AVR Atmel microcontroller unit. It is built for monitoring and control and displays the three main temperature values: set point temperature, baby skin temperature and air temperature. User friendly software is implemented. The implemented control unit was tested in the laboratory as well as in the field. The control unit is sensitive to change of $0.1^{\circ}C$. At startup, based on a unique control strategy, the incubator reaches its steady state in about 14 minutes. The system schematic diagrams are shown in the paper. Also, programs flow charts are presented. The control unit was designed and implemented based on a contract between the Electronics Research Institute (ERI) and ENGIMED Company. The authors would like to thank ERI and ENGIMED for introducing all required finance and shoring to complete this work.

Xylanase Production by Mixed Culture Using Crude Hemicellulose from Rice Straw Black Liquor and Peat Moss as an Inert Support

  • Black liquor (BL) is a by-product of rice straw pulping process. It is a low costs raw material for production value-adding proteins and enzymes, which has been paid more and more attention to reduce its environmental pollution. Mixed cultures of micelial fungi, Trichoderma reesei Northern Regional Research Laboratory (NRRL)11236, Trichoderma reesei NRRL 6165 and Aspergillus niger strains NRC 5A, NRC 7A, and NRC 9A were evaluated for their ability to produce xylanase using crude hemicellulose (CHC) prepared from BL and peat moss as an inert support under solid state fermentation (SSF). The most potent strains, A. niger NRC 9A (818.26 U/g CHC) and T. reesei NRRL 6165 ($100.9{\pm}57.14$ U/g CHC), were used in a mixed culture to enhance xylanase production by co-culturing under SSF. In the mixed culture, xylanase production ($1070.52{\pm}12.57$ U/g CHC) was nearly1.3 and 10.6-fold increases over the activities attained in their monocultures, A. niger NRC 9A and T. reesei NRRL 6165, respectively. Optimization of the culture parameters of the mixed culture SSF process, concentration of ammonium sulfate and corn steep liquor, CHC/peat moss ratio, inoculum size and ratios of the two strains, initial pH value, initial moisture content and incubation time, exhibited a significant increase ($2414.98{\pm}84.02$ U/g CHC) in xylanase production than before optimization.

  • In order to obtain the dynamic response behavior of the rock subjected to blasting loading, a shock-proof high sensitivity impact sensor which can measure high frequency dynamic force and strain events should be adopted. Because the impact sensors which uses quartz and piezoelectric element are costly, generally the strain measurement-based impact (SMI) sensors are applied to high speed loading devices. In this study, dynamic Brazilian tension tests of granitic rocks was conducted using the Nonex Rock Cracker (NRC) reaction driven-high speed loading device which adopts SMI sensors. The dynamic response of the granite specimens were monitored and the intermediate strain rate dependency of Brazilian tensile strengths was discussed.

Comparison between predicted total digestible nutrients and actual total digestible nutrients using nutrient digestibility of rice straw and timothy in ruminants

  • This study attempted to compare total digestible nutrients (TDN) calculated as Rohweder, NRC, and Waldo and Peiqiang methods and TDN measured as digestibility of in vivo appearance. Rohweder method showed that the TDN of rice straw and timothy were 54.32% and 57.79%, respectively. In NRC method, the digestibility of neutral detergent fiber (NDF) and TDN of rice straw were 50.76% and 53.15%, respectively. When NRC method was applied in Timothy, the digestibility of NDF and TDN were 51.53% and 55.22%, respectively. Waldo and Peiqiang method calculated the results through the rumen in situ test. NDF digestibility of rice straw and timothy was 44.61% and 51.82%, which was different from the results of NRC method. In addition, TDN was predicted to be 48.85% for rice straw and 55.41% for timothy. In the in vivo apparent digestibility experiment, the digestibility of NDF and TDN in rice straw was 41.10% and 44.79%, respectively. In timothy, the digestibility of NDF and TDN were measured as 51.29% and 58.18%, respectively. As a result of a series of studies, rice straw was found in Rohweder and NRC methods showed higher TDN than other methods. In this study, there was a difference in rice straw by measurement method, but there was no difference in timothy. Therefore, when evaluating the value of feed in order to provide roughage to ruminant, calculation methods must be modified and supplemented. In addition, TDN should be considered to apply several evaluation methods instead of one method.

  • 원자력발전소의 주요 기기들에 대한 가동중검사는 관련법규에 따라 철저히 수행되고 있다. 그러나 최근 선진국에서는 이에 만족하지 않고 원전 기기의 안전성을 더욱 확고히 하기 위해 기존의 가동중검사 요건을 계속 강화하고 있으며, 원전 관련 당사자들은 강화된 요건들을 충족시키기 위한 노력을 끊임없이 계속하고 있다. 이 글에서는 원전 기기 가동중검사 신규 요건들인 초음파탐상검사 시스템의 기량검증(Performance Demonstration) 요건, 비파괴검사자 및 초음파검사자 자격 인정 요건(ANSI/ASNT CP-189, Appendix VII of ASME Sec. XI), 증기발생기 전열관 와전류검사, 신호평가자 자격인정(Qualified Data Analyst : QDA), 미국규제기관(NRC)에서 발행하고 있는 NRC Bulletin, NRC information 등의 가동중검사 관련 사항들을 살펴보고 선진 외국에서는 이들 요건 및 정보에 대해 어떻게 대처하고 있는가를 알아본다. 또한 국내에서도 이들 신규 요건에 대한 대처 현황과 대처 방안을 모색한다.

GAPCON-THERMAL-2 Revision2 코드를 이용한 핵분열 생성물 방출 모델 비교 연구

  • 핵분열 생성물 방출량을 계산하는 모델들에 대한 비교 분석을 위해 GAPCON-THERMAL-2 Revision 2 (GT2R2) 코드를 이용하여 Beyer-Hann , Beyer-Hann with NRC High Burnup Correction, ANS5.4와 Modified ANS5.4 핵분열 생성물 방출 모델들을, RISO-M2-2C 핵연료봉의 실험결과와 비교하였다. Beyer-Hann 모델은 실험결과보다 낮게 예측한반면 ANS5.4 모델은 실험결과 보다 높게 예측하였다. 한편 NRC High Burnup Correction을 한 Beyer-Hann 모텔과Modified ANS5.4 모델은 실험 결과와 비슷한 방출비를 예측하였다. 이러한 결과를 확인하기 위해 국부적인 핵연료 온도와 연소도를 검토한 결과 ANS5.4 모델이 .Modified ANS5.4 모델보다 온도와 연소도에 따라 더 민감한 반응을 보이고 있으며, Beyer-Hann 모텔은 연소도 영향이 없이 각 온도 영역에서 일정하였고, Beyer-Hann with NRC High Burnup Correction 모델은 20,000MWd/MTU 연소도 이상영역에서 연소도 영향을 보이고 있다.

  • US NRC issued rulemaking of 10CFR50.55a to perform the Perservice and Inservice inspection for Reactor Vessel Head Penetration Nozzle of US Nuclaer plant. The rulemaking was required the EPRI Demonstration to verify the NDE technique performing special Ultrasonic examination. In order to meet this requirement, the UT and ECT procedures was demonstrated and the NDE personnel were qualified by EPRI. In this paper, the NDE technique and analysis method are described the Preservice inspection for the Palo Verde #1/2/3 Replacement Reactor Vessel Head Penetration Nozzle using the qualified procedures and personnel.
