Mineral- and Tissue-Specific Metabolic Changes in Tomato (Lycopersicon esculentum L.) Plants Grown under NPK-Starved Conditions

  • Specific metabolic network responses to mineral starvation are not well-defined. We examined a detailed broad-scale identification of metabolic responses of tomato leaf and root to N, P or K starvation. Tomato plants were grown hydroponically under optimal (5 mM N, 0.5 mM P, or 5 mM K) and starved (0.5 mM N, 0.05 mM P, or 0.5 mM K) conditions and metabolites were measured by LC-MS and GC-MS. Overall, the levels of metabolites (lipids, nucleotides, peptides and secondary metabolites) presented in this paper largely showed mineral- and tissue-specific responses. Most strikingly, G3P (glycerol-3-P), GPC (glycerol-P-choline) and choline phosphate responded differently to a type of mineral; an increase in N or K starvation and a decrease in P starvation. A dramatic increase in the levels of secondary metabolites, in particular, rutin and chlorogenate in both tomato tissues during N starvation were observed. Based on these data, it is necessary to clearly elucidate an unknown event taking place in a variety of abiotic impacts, and we are now studying to expand our knowledge on metabolic- and proteomic-responses using GS-MS and LC-MS.

Effects of Composts on the Growth, Yield and Effective Components of Turmeric (Curcuma longa L.) (울금의 생육, 수량 및 품질에 미치는 퇴비의 효과)

  • Curcuma longa L. (turmeric) is an important medicinal plant that has been historically used in herbal medicine and in the health food throughout Asia etc. Recently, the demands on rhizome of turmeric are increasing greatly by well-being boom, but there is not enough to meet the demands. To fulfill increasing demands, cultivation system strategies using the organic fertilizers are required to produce a greater amount of rhizome with good quality and yield. A field experiment was conducted to determine the effect of composts, NPK fertilizer(control, F), NPK + swine manure(SM) and NPK + fermentation manure from the wasted oriental medicine materials(OMWM), on rhizome yield and the content of bioactive components for quality. Our results showed that two compost applications can increase both rhizomes(24.1-25.9%) and curcumin(21.7-41.0%) yields, respectively, compared to F control. The content of amino acids increased significantly by SM and OMWM treatments. SM and OMWM application also increased the total phenol yields 7.8 and 8.7 g/10a compared with control 6.3 g/10a, the flavonoid yields 6.3 and 7.3 g/10a compared with control 5.3 g/10a, and also antioxidant activity 21.7 and 41%, respectively, as compared to the control. Especially, OMWM was more effective in total rhizomes yields and bioactivities and in the biosynthesis of curcumin and bioactive components than SM treatments, but the biological pathway was not clear, still. This experiment suggests that curcumin or bioactive components affected by adding SM and OMWM could increase the yields and quality of turmeric.

Economic Analysis of Growing Ginger (Zingiber officinale) Under Teak (Tectona grandis) Canopy in Southwest Nigeria

  • Multiple use forestry is capable of generating income for forest based communities through Non-Timber forest products (NTFPs) which provide food, medicine, materials for domestic use and cash income for communities adjoining forest areas in developing countries. This study evaluates the economics of producing ginger rhizomes under teak canopy in a multiple land use system during 2007 and 2008 in even aged teak plantations in Ibadan and Ife, Nigeria. Twelve $6m^2$ sample plots were randomly selected in Completely Randomized Block Design within and outside the plantation. Average ginger rhizome of (50-60 g) were planted on the slightly tilled soil. NPK 15:15:15 was applied at 180 kg/ha on a split unit dose. ANOVA, Profitability, Benefit-Cost (B/C) ratio were used to analyze data. Results showed no significant differences between sites in ginger rhizome yield, (0.089 and 0.718, ${\rho}{\leq}0.05$) in 2007 and 2008 respectively. Average yield were higher outside teak canopy in both sites and treatments, (Ibadan -40.05 g>32.9 g, Ife -67.6 g>25.2 g and Ibadan -41.3 g>31.5 g, Ife -66.8 g>25.0 g) with and without NPK respectively. NPK had no effect on yields within teak plantation, (Ibadan -31.5<32.9 g, Ife -25 g<25.2 g). Ginger rhizome production was viable financially without inorganic fertilizer during second cropping season within and outside plantation (B/C=1.02, 1.09) respectively. Ginger could be raised profitably under teak canopy, however, studies on insolation requirement of ginger under teak canopy and other tree plantations are recommended.

  • An experiment of alum sludge application was carried out on a mountainous site, to know its effect on the growth of forage sorghum hybrid, Pioneer 93 1 (Sorghum bicolor x S. bicolor). When alum sludge with 80% water content was applied at the level of 133 ton per ha, the effect of the application on the growth of Pioneer 931 was much smaller than that of chemical fertilizer. But Eresh yield of the forage, with the sludge application, was greater than that without fertilizer when the soil is not fertile. But when the soil was comparatively fertile, the sludge application gave an adverse affect on the growth of the forage plant. The content of exchangeable K in the soil has a very close positive relation with the Eresh yield and plant height of the plant, while that of exchangeable Ca has a close negative relation with those growing factors. There was a tendency of yield decrease of the forage plant, when alum or compost was applied with NPK to the comparatively infertile soil especially for alum sludge. As a result, when soil is not fertile more attention will be necessary for applying alum sludge with chemical fertilizers. But to soil with better fertility, it is recommended to use alum sludge with chemical fertilizer to obtain more yield of the forage plant.

Effects of Rhizobium Inoculation on the changes of Ureide-N and Amide-N Concentration in Stem and Root exudate of Soybean Plant (대두근류균(大豆根瘤菌) 접종(接種)이 뿌리와 줄기 즙액중(汁液中) Amide-N 및 Ureide-N 농도(濃度)에 미치는 영향(影響))

  • A series of green house experiment was conducted to find but the effect of fertilizer application and inoculation of rhizobium on the changes of amide-N, ureide-N and $NH_4-N$ concentration in stem and root exudates of soybean plant growth. The results obtained were summarized as follows ; 1. Five strains of indigenous Rhizobium japonicum-nitrogen fixing activity($C_2H_2$-reducing activity) was more than 6.4 to 20.1 nmole/hr/tube-were identified from 37 soil samples in 22 areas of farmers field throughout country. 2. These identified 5 strains of rhizobium were obtained high nitrate reductase but low ammonium and nitrite oxidase activities. Among 5 strains of rhizobium the Rhizobium japonicum RjK-134 was applied for this green house experiment. 3. Dry matter yield was increased by the combination of inoculation of Rhizobium japonicum RjK-134 with no fertilizer and without nitrogen fertilizer application. However, dry matter yield was decreased with application of N and NPK with inoculation of rhizobium. 4. The concentrations of amide-N and ureide-N were increased in xylem sap than that of root exudate and higher concentration was obtained ar 30 days after planting than flowering stage (45 days after planting). 5. The combination of NPK application with inoculation of Rhrizobium japonicum RjK-134 enhanced the increase of amide-N and ureide-N concentration in xylem sap and root exudate. 6. High ammonium-N concentration in xylem sap and root exudate were obtained in combination with without-fertilizer under no inoculation of rhizobium and N and NPK application with inoculation of rhizobium.

  • The effects of rice-straw annual application on nitrogen fixing microbial flora in the soil of paddy fields and their biological activities were investigated. Experiments were performed in both NPK fertilizer applied soil and rice-straw applied soil of Agricultural Station in Aomori-ken, Japan. The following results were obtained. 1. The ARA by phototrophs was significantly increased in both soil plots. From the soil plot in which 300ppm-nitrogen was applied, the increase of ARA began to be seen from three weeks later. On the other hand, 33ppm-nitrogen applied soil plot and non-nitrogen applied soil plot showed the ARA increase from the beginning. The amount of ARA by non-phototrophs was only one-tenth of that by phototrophs. 2. For the first three weeks, the phototrophic bacteria (mainly Rhodopseudomonas) were predominant in both soil plots. Since then, as the ARA rapidly increased, the proliferation of blue-green algae forming heterocysts was remarkably promoted. Such effects were more distinct in the rice-straw annually applied soil plot than in the NPK fertilizer annually applied soil plot. 3. The degree of proliferation of blue-green algae depended on the amount of applied nitrogen. While Anabaena, Nostoc and Cylindrospermum were largely proliferated in the non-nitrogen applied soil plot, Cylindrospermum and Calothrix were in the 33ppm-nitrogen applied soil plot, but Calothrix tended to predominated in the 100ppm-nitrogen applied soil plot.

  • A trial paste fertilizer for rice was manufactured by a developed process using major raw materials of waste liquid from glutamic acid fermentation, urea, phosphoric acid, diammomium phosphate and potassium chloride with potassium hydroxide for pH adjust and both glycerin and $HB_4O$ for enhancing and maintaining dispersion of paste. The ratios of N, $P_2O_5$ and $K_2O$ for trial products of paste fertilizer were 10:15:13 for Japonica var., and 12.5:15:13 for Indica var., and it was 6.0 in pH and 1.45g/cc in specific gravity.

  • Two trial products of monogranular compound fertilizer for red pepper under vinyl mulching cultivatin were made using the principal sources of urea, diammonium phosphate and muriate of potash in combination with fillers of treated human wastes (product I) and zeolite (Product II). A field experiment was carried out to evaluate their effects on red pepper and the results obtained were as follows. Red pepper plant growth and total red fruit yields in the trial products of one time basal application were better and higher than those of NPK split application of contrl plot. The increases of yield in the trial products might be due to steady supply of nutrients for the entire growing period. Therefore, one time basal dressing of a developed monogranular compound fertilizer for red pepper was applicable under vinyl mulching cultivation.

  • This experiment was conducted to investigate the response of the fertilizer three factors on the growth of Alopeuclus aequalis Sobol. var. amurensis(Kom.) Ohwi. The growth of Alopeuclus aequalis Sobol. var. amurensis(Kom.) Ohwi. such as plant height, tiller number and dry matter production were vigorous on the plot fertilized with phosphate(P, NP, PK, NPK). The main effect of plant height elongation and dry matter production were significant for phosphate and nitrogen, and their interaction effect was recognized, too. Tiller growth and panicle number were mainly affected by phosphate. The growth of Alopeuclus aequalis Sobol. var. amurensis(Kom.) Ohwi. depended mostly on phosphate among the fertilizer three factors. The effect of nitrogen fertilizer was very different according to phosphate fertilizer treatment.

Evaluation of Nitrogen and Phosphorus Balance in Green Manure-Rice Cropping Systems without Incorporation of Green Manure Crops (녹비작물 무 환원-벼 작부체계에서 질소와 인산수지 평가)

  • BACKGROUND: The nutrient balance in Korea during 1985-2006 had continually increased and maintained the highest levels among OECD countries. The use of green manure crops such as barley and hairy vetch is common practice for reducing chemical fertilizer application and maintaining soil fertility. However, green manure crops can also be used as a livestock feeding material which may reduce nutrient balance in the national scale. We calculated nitrogen and phosphate balance under green manure-rice cultivating system where all green manure was removed and used for feeding livestock. METHODS AND RESULTS: The barley and hairy vetch grown in pure stands or in mixtures with different sowing rates were tested for rice cultivation without chemical fertilization. The conventional fertilization (NPK) for rice cultivation was selected to compare nutrient balance with green manure-rice cultivation. Nitrogen and phosphate balance were calculated according to the surface balance method of the PARCOM guidelines. Total aboveground biomass of mixture (barley and hairy vetch) was higher compared to that of pure barley or hairy vetch. Among the mixture with barley and hairy vetch, the highest aboveground biomass was observed in B75H25 (barley 75%+hairy vetch 25%). The nitrogen and phosphate balance in the B75H25 mixture was-104 kg N/ha and-50.3 kg P/ha, respectively. CONCLUSION(S): The barley and hairy vetch mixture proved to be a very effective strategy for biomass production of green manure. The amount of nitrogen and phosphate fertilizer to be applied is estimated to be 104 kg N/ha and 50.3 kg P/ha in order to maintain soil fertility if all green manure and rice straw were removed from rice field for livestock feeding.
