Objective of this study was to examine the effect of sodium nitroprusside (SNP), a nitric oxide (NO) donor on steroid synthesis, growth and apoptosis of buffalo granulosa cells (GCs) in vitro. Follicular fluid of antral follicles (3-5 mm diameter) was aspirated and GCs were cultured in 0 (control), $10^{-3}$, $10^{-5}$, $10^{-7}$, $10^{-9}\;M$ of SNP for 48 h. To evaluate whether this effect was reversible, GCs were cultured in presence of $10^{-5}\;M$ SNP+1.0 mM $N^{\omega}$-nitro-L-arginine methyl ester (L-NAME) a NO synthase (NOS) inhibitor or hemoglobin (Hb, $1.0{\mu}g$) as NO scavenger. Nitrate/nitrite concentration was evaluated by Griess method, progesterone and estradiol concentrations by RIA and apoptosis by TUNEL assay. SNP ($10^{-3}$, $10^{-5}$, $10^{-7}\;M$) significantly (p<0.05) inhibited estradiol and progesterone synthesis, growth, disorganized GCs aggregates and induced apoptosis in a dose dependent manner. However, $10^{-9}\;M$ SNP induced the progesterone synthesis and stimulated GCs to develop into a uniform monolayer. Combination of SNP $10^{-5}$ M+L-NAME strengthened the inhibitory effect while, SNP+Hb together reversed these inhibitory effects. In conclusion, SNP at greater concentrations ($10^{-3}$, $10^{-5}$ and $10^{-7}\;M$) has a cytotoxic effect and it may lead to cell death whereas, at a lower concentration ($10^{-9}\;M$) induced progesterone synthesis and growth of GCs. These findings have important implications that NOS derived NO are involved at physiological level during growth and development of buffalo GCs which regulates the steroidogenesis, growth and apoptosis.

Abies nephrolepis(Pinaceae) extracts were tested for determined immune system regulating activity based on antiinflammatory activity, antioxidant activity and anti-proliferative effect on HeLa cell line. The A. nephrolepis extracts increased dose-dependently anti-proliferation of HeLa cell line. The DM fraction of the extracts having anti-proliferatative effects of HeLa cell line was fractionalized four subfractions($D1{\sim}D4$). Inflammation-induced NO production was inhibited by D2 and D4 in LPS-activated RAW264.7 macrophages. And also, this fractions showed antioxidant activity examined by DPPH radical scavenging effects. These results suggest that the potential use of DM fraction of A. nephrolepis in chemoprevention and regulation overproduction of NO on pathogenic conditions. The mechanism of the inflammatory effects, however, must be evaluated through various parameters in the induction cascade of NO production.
The effect of Saccharomycopsis fibuligera fermentation on the antioxidant and anti-inflammatory activity of Kerria japonica (K. japonica) extracts was studied. First, the antioxidant activity of the fermented extract was measured using the 2,2-diphenyl1-picrylhydrazyl (DPPH) and 2,2'-azinobis (3-ethylbenzthiazoline 6-sulfonic acid (ABTS) methods. Also, the quantification of polyphenols and flavonoids, which are representative components with antioxidant activity, was performed. The results of the DPPH and ABTS assays showed an increase in the antioxidant activity by 14.39% and 21.74%, respectively, due to fermentation. The polyphenol concentration increased by 10.5%, and the flavonoid concentration increased by 100.0%. In the cell experiment, a cytotoxicity test and a nitric oxide (NO) production inhibitory test were performed using RAW 264.7 cells. Both the control group and the fermentation group showed no cytotoxicity. In the NO production inhibition experiment, the fermentation group showed a 6.85% higher inhibition of NO production compared to the control group. When the inhibitory effects of the extracts on inflammatory cytokine production were assessed, the fermentation group showed 12.4% and 23.5% higher inhibition of interleukin (IL)-1β and IL-6 production, respectively, compared to the control group. In conclusion, due to its potential for inhibiting NO and inflammatory cytokine production, fermented K. japonica extracts could be considered a source of anti-inflammatory compounds.

Various kinds of antibiotics are generally believed to have inhibitory effects on the antibody response. However, as polymyxin B which belongs to the cyclic polypeptide group of antibiotic was found to have some enhancing effects on the antibody response of rabbits to enterobacterial common antigen(CA) under specified conditions, experiments were carried out on this problem with the following results. 1. When mixture of polymyxin B and CA derived from Salmonella typhimurium(STM) was treated 30 minutes at $37^{\circ}C$ and injected three times into rabbits by intravenous route, the antibody response to CA was weaker than rabbits injected CA only. 2. Mixture of polymyxin B and CA showed a marked antibody production when injected into rabbits primed with small amounts of heat-extracted antigen of STM, while the injection of CA alone showed low titers of response. 3. Mixture of polymyxin B and heat-extracted CA-containing antigen of Escherichia coli 014 also showed a increased antibody production than CA alone in rabbits primed with antigen of STM. 4. The effect of polymyxin B appeared in different ways. This antibiotic did not enhance the CA antibody response in rabbits primed with small amounts of E. coli 0111 and 055, but enhance in rabbits primed with Shigella flexneri. 5. No enhancing effect on the antibody response was observed by polymyxin B in rabbits primed with CA. 6. No enhancing effect on the antibody response was also noted in rabbits primed with STM antigen in case polymyxin B and CA were administered simultaneously but in veins of different places. 7. Bacitracin did not enhance the CA antibody response in primed rabbits with STM antigen, but neomycin slightly enhance the response. 8. Lipopolysaccharide showed no priming effect on the CA antibody response, and no enhancement of the CA antibody response in rabbits printed with STM. 9. The priming effect of STM antigen against CA antibody response was very weak as compared with the effect of CA derived from STM antigen.

This study describes a preliminary evaluation of the anti-inflammatory activity and anti-atopic activity of Phormium tenax leaf extracts. P. tenax leaf was extracted using 70% ethanol and then fractionated sequentially with n-hexane, methylene chloride, ethyl acetate, n-butanol. In order to effectively screen for anti-inflammatory agents, we first investigated the inhibitory effects of P. tenax leaf crude extracts and solvent fractions on production of pro-inflammatory factors[nitric oxide(NO), prostaglandin $E_2(PGE_2)$, inducible nitric oxide synthase(iNOS) and cyclooxygenase-2(COX-2)] and pro-inflammatory cytokines [tumor necrosis $factor-{\alpha}(TNF-{\alpha})$, interleukin-6(IL-6) and $interleukin-1{\beta}(IL-1{\beta})$] in lipopolysaccharide(LPS)-stimulated RAW 264.7 cells. In addition, we also evaluated of their inhibitory effect on the atopic dermatitis-like inflammatory markers such as macrophage-derived chemokine(MDC) and thymus and activation-regulated chemokine(TARC) in HaCaT cells. Among the five solvent fractions of P. tenax, methylene chloride and ethyl acetate fractions inhibited production of pro-inflammatory factors and pro-inflammatory cytokines in a dose dependent manner, respectively. These fractions were also showed inhibitory activity for MDC and TARC expression levels in $IFN-{\gamma}-stimulated$ HaCaT cells, respectively. These results suggest that P. tenax have significantly effects of anti-inflammatory activity and anti-atopic activity that might be beneficial for the topical treatment of inflammatory skin disorders.

Many traditional herbal remedies exhibit several beneficial effects including anti-inflammation. Panax notoginseng Buck F.H. Chen. is used as a therapeutic agent to stop haemorrhages and a tonic to promote health in Korean and Chinese medicine. The pharrnacokinetic profiles of the main P. notoginseng are still not accurately investigated. The exact mechanism of the anti-inflammatory action of P. notoginseng, however, has not been determined. In the present study, we examined the effect of P. notoginseng on lipopolysaccharide (LPS)-induced nitric oxide (NO) production, and inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) gene expression in RAW264.7 macrophages. The results indicated that P. notoginseng concentration-dependently inhibited LPS-induced NO production. Furthermore, P. notoginseng inhibited the expression of LPS-induced iNOS and COX-2 proteins without an appreciable cytotoxic effect on RAW264.7 cells. Berberine also inhibited LPS-induced iNOS protein as potently as P. notoginseng. This was consistent with the findings that P. notoginseng and also berberine inhibited prostaglandin E2 synthesis induced by LPS.
Effect of glucose addition to xylose medium on xylitol production was investigated by using Candida parapsilosis ATCC 21019 mutant. With increasing the ratio of glucose to xylose in total amount of 50 g/l as glucose and/or xylose, xylitol production was decreased but ethanol and glycerol production were increased. Ethanol and glycerol concentration were maxmum in 10 g/l of xylose and 40 g/l of glucose medium as 21.5 g/l and 3.6 g/l, respecti- vely. No xylitol was formed in glucose medium without xylose because xylitol could be not produced from glucose. With increasing the ratio of glucose to xylose, the activity of xylose reductase which converted xylose to xylitol were decreased. The activities of xylitol dehydrogeiiase which converted xylitol to xylulose and then cell materials were found to be constant regardless of the ratio of glucose to xylose. This results indicated that glucose addition to xylose medium on cell growth was not affected. In order to prevent the inhibitory effect of glucose on xylitol production, glucose in a fermentor was fed with low concentration and then ethanol and glycerol was critically decreased and the xylitol yield from xylose of the culture with glucose feeding was recovered the almost same as that with only 50 g/l of xylose. However, the xylitol yield from total sugars (xylose and glucose) was decreased and glucose was not contributed to xylitol production. Therefore, the fermentation at high concentration of xylose without glucose was carried out. A final xylitol concentration of 242 g/l which corresponding 80.7% of xylitol yield was obtained from 300 g/l of xylose for 273 hours.
Objective: To derive information on the efficacy of Sam-chul-kun-bi-tang (SKT), by evaluating its anti-inflammatory effect. SKT is a widely-used herbal formula in traditional Korean medicine. In man y studies, plant-derived anti-inflammatory efficacies have been investigated for their potential inhibitory effects on lipopolysaccharide (LPS)-stimulated macrophages. This study was performed to examine the anti-inflammatory effects of SKT extract on LPS-stimulated RAW 264.7 cells. Methods: The production of nitric oxide (NO), prostaglandin $(PG)E_2$, tumor necrosis factor (TNF)-$\alpha$ and interleukin (IL)-6 were examined in a macrophage cell line, RAW 264.7 cells, in the presence of SKT. RAW 264.7 cells were incubated with LPS 1 ${\mu}g/mL$ and SKT for 18 hrs. The anti-inflammatory activity of SKT was investigated by carrageenan-induced paw edema in rats. The paw volume was measured at 2 and 4 hrs following carrageenan-induced paw edema in rats. Results: SKT showed inhibitory effect on $PGE_2$, TNF-$\alpha$ and IL-6 in LPS-stimulated RAW 264.7 cells. But SKT was not inhibitory effect on NO by LPS-stimulated RAW 264.7 cells. Administration of SKT (1 g/kg) also showed a reduction in carrageenan-induced paw edema on rats. Conclusion: These results suggest that SKT has anti-inflammatory activities in both in vitro and in vivo models.
