Zanthoxylum rhetsa Stem Bark Extract Inhibits LPS-induced COX-2 and iNOS expression in RAW 264.7 Cells via the NF-${\kappa}B$ Inactivation

  • The methanol extract of Zanthoxylum rhetsa (MZRR) were evaluated for its ability to suppress the formation of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) in lipopolysaccharide (LPS)-activated RAW 264.7 macrophages. MZRR presented an inhibition of LPS-induced production of nitric oxide (NO) and prostaglandin $E_2$ ($PGE_2$) in RAW 264.7 macrophages. Western blotting and RT-PCR analyses demonstrated that MZRR significantly inhibited the protein and mRNA expressions of iNOS and COX-2 in LPS-activated macrophages in a dose-dependent manner. LPS-induced COX-2, iNOS, and nuclear factor kappa beta (NF-${\kappa}B$) activity were also decreased in the presence of MZRR. The production of tumor necrosis factor-$\alpha$ (TNF-$\alpha$), the mRNA expression levels of pro-inflammatory cytokines, including TNF-$\alpha$ and IL-$1{\beta}$, were reduced after MZRR administration in a dose dependent-manner. These results suggest that the MZRR extract involved in the inhibition of iNOS and COX-2 via the NF-${\kappa}B$ pathway, revealing a partial molecular basis for anti-inflammatory properties of the MZRR extract.

창출금련탕(蒼朮芩連湯) 추출물의 항산화 및 항염증 활성에 미치는 영향 (The anti-oxidant and anti-inflammatory Activites of Changchulgeumryeontang Extract)

  • Objectives : This study investigated the anti-oxidant and anti-inflammatory activities of Changchulgeumryeontang (CCGRT) extract. Methods : The macrophage cell line RAW 264.7 cells were used and MTT assay was performed to measure the cell viabilities at the various concentrations of CCGRT ($25-200{\mu}g/m{\ell}$). Nitric oxide (NO) and prostaglandin $E_2$ ($PGE_2$) were measured in LPS-induced RAW 264.7 cells. Expressions of iNOS, $NF-{\kappa}B$, $IL-1{\alpha}$, $IL-1{\beta}$ and IL-6 were also performed by real-time PCR. The anti-oxidant activities of CCGRT was measured by DPPH radical scavenging activity. Results : 1. there was no cytotoxicity in RAW264.7 cells treated with CCGRT compared to the control. 2. CCGRT treated group significantly inhibited NO and $PGE_2$ production compared to the LPS treated group. 3. CCGRT treated group significantly decreased mRNA expressions of iNOS, $NF-{\kappa}B$, $IL-1{\alpha}$, $IL-1{\beta}$ and IL-6 compared to the LPS treated group. 4. CCGTR was found to have high DPPH free radical scavenging ability. Conclusions : According to the above results, CCGRT may be a potentional choice for the treatment of inflammatory skin disease. Schisandrae Fructus ethanol extract attenuates particulate matter 2.5-induced inflammatory and oxidative responses by blocking the activation of the ROS-dependent NF-κB signaling pathway

  • BACKGROUND/OBJECTIVES: Schisandrae Fructus, the fruit of Schisandra chinensis Baill., has traditionally been used as a medicinal herb for the treatment of various diseases, and has proven its various pharmacological effects, including anti-inflammatory and antioxidant activities. In this study, we investigated the inhibitory effect of Schisandrae Fructus ethanol extract (SF) on inflammatory and oxidative stress in particulate matter 2.5 (PM2.5)-treated RAW 264.7 macrophages. MATERIALS/METHODS: To investigate the anti-inflammatory and antioxidant effects of SF in PM2.5-stimulated RAW 264.7 cells, the levels of pro-inflammatory mediator such as nitric oxide (NO) and prostaglandin E2 (PGE2), cytokines including interleukin (IL)-6 and IL-1β, and reactive oxygen species (ROS) were measured. To elucidate the mechanism underlying the effect of SF, the expression of genes involved in the generation of inflammatory factors was also investigated. We further evaluated the anti-inflammatory and antioxidant efficacy of SF against PM2.5 in the zebrafish model. RESULTS: The results indicated that SF treatment significantly inhibited the PM2.5-induced release of NO and PGE2, which was associated with decreased inducible NO synthase and cyclooxygenase-2 expression. SF also attenuated the PM2.5-induced expression of IL-6 and IL-1β, reducing their extracellular secretion. Moreover, SF suppressed the PM2.5-mediated translocation of nuclear factor-kappa B (NF-κB) from the cytosol into nuclei and the degradation of inhibitor IκB-α, indicating that SF exhibited anti-inflammatory effects by inhibiting the NF-κB signaling pathway. In addition, SF abolished PM2.5-induced generation of ROS, similar to the pretreatment of a ROS scavenger, but not by an inhibitor of NF-κB activity. Furthermore, SF showed strong protective effects against NO and ROS production in PM2.5-treated zebrafish larvae. CONCLUSIONS: Our findings suggest that SF exerts anti-inflammatory and antioxidant effects against PM2.5 through ROS-dependent down-regulating the NF-κB signaling pathway, and that SF can be a potential functional substance to prevent PM2.5-mediated inflammatory and oxidative damage.

  • Cobrotoxin의 항염증 치료 기전에 대해 연구하기 위하여 성상세포에 LPS 및 SNP로 염증을 유도한 후 NF-${\kappa}B$와 DNA의 결합 능력, NF-kB Dependent Luciferase 발현, Astrocyte의 세포활성, NF-${\kappa}B$ 구성 단백질인 P50, P-$1{\kappa}B$, $1{\kappa}BB$ 및 염증(炎症)관련 유전자(遺傳子)인 Cox-2, iNOS, cPLA2의 발현과 GSH와 DTT로 sulf-hydryl기를 환원시 NF-${\kappa}B$와 DNA의 결합 능력, NF-${\kappa}B$ 구성 단백질인 P50 발현에 미치는 영향과 Cobrotoxin의 성상세포 내 유입 등을 관찰하여 다음과 같은 결론을 얻었다. 1. LPS 로 염증을 유발한 후 NF-${\kappa}B$와 DNA의 결합 능력을 관찰한 결과 Cobrotoxin 0.1${\mu}g/m{\ell}$ 처리군, Astrocyte 내에서의 Cobrotoxin 0.1, 0.5${\mu}g/m{\ell}$ 처리군에서 모두 대조군에 비하여 유의한 억제를 나타내었다. 2. LPS로 염증을 유발한 후 Astrocyte 내에서 NF-${\kappa}B$ Dependent Luciferase 발현을 살펴본 결과 Cobrotoxin 모든 처리군에서 대조군에 비하여 유의한 억제를 나타내었다. 3. SNP로 염증을 유발한 후 Cobrotoxin이 NF-${\kappa}B$ 구성 단백질인 P50, P-$1{\kappa}B$, $1{\kappa}B$ 발현에 미치는 영향을 살펴본 결과 P50와 $1{\kappa}B$는 Cobrotoxin 0.1, 0.5 및 $1{\mu}g/m{\ell}$ 모든 처리군에서 대조군에 비하여 유의한 억제를 나타내었고, P-$1{\kappa}B$는 Cobrotoxin $0.1{\mu}g/m{\ell}$ 처리군에서 대조군에 비하여 억제를, Cobrotoxin 0.5, $1{\mu}g/m{\ell}$ 처리군에서 각각 대조군에 비하여 유의한 억제를 나타내었다. 4. LPS로 염증을 유발한 후 Cobrotoxin이 NF-${\kappa}B$ 구성 단백질인 P50, P-$1{\kappa}B$, $1{\kappa}B$ 발현에 미치는 영향을 살펴본 결과 P50와 $1{\kappa}B$는 Cobrotoxin 0.5, $1{\mu}g/m{\ell}$ 처리군에서 각각 대조군에 비하여 유의한 억제를 나타내었다. 5. Astrocyte내에서 SNP로 염증을 유발한 후 GSH와 DTT로 sulf-hydryl기를 환원하여 NF-${\kappa}B$와 BNA의 결합 능력을 관찰한 결과 Cobrotoxin $0.5{\mu}g/m{\ell}$과 DTT 1mM Cobrotoxin $0.5{\mu}g/m{\ell}$과 DTT 5mM의 동시처리군은 각각 Cobrotoxin $0.5{\mu}g/m{\ell}$ 처리군에 비하여 유의한 증가를 나타내었다. 8. Astrocyte 내에서 LPS로 염증을 유발한 후 GSH와 DTT로 sulf-hydryl기를 환원하여 NF-${\kappa}B$와 DNA의 결합 능력을 관찰한 결과 cobrotoxin $0.5{\mu}g/m{\ell}$과 DTT 1mM Cobrotoxin $0.5{\mu}g/m{\ell}$과 DTT 5mM의 동시처리군과 Cobrotoxin $0.5{\mu}g/m{\ell}$과 GSH 1mM Cobrotoxin $0.5{\mu}g/m{\ell}$과 GSH 5mM의 동시처리군 모두에서 Cobrotoxin $0.5{\mu}g/m{\ell}$ 처리군에 비하여 유의한 증가를 나타내었다. 9. Astrocyte 내에서 SNP로 염증을 유발한 후 GSH와 DTT로 sulf-hydryl기 환원시 NF-${\kappa}B$ 구성 단백질인 P50 발현에 미치는 영향을 관찰한 결과 SNP, Cobrotoxin $1{\mu}g/m{\ell}$와 DTT 1 또는 5mM 동시처리군과 SNP, Cobrotoxin $1{\mu}g/m{\ell}$와 GSH 1 또는 5mM 동시처리군 모두에서 Cobrotoxin $1{\mu}g/m{\ell}$ 처리군에 비하여 발현의 유의한 증가를 나타내었다. 10. Cobrotoxin의 세포 내 유입 확인을 살펴본 결과 Astrocyte 내에서 Cobrotoxin이 세포내로 유입되는 것으로 나타났다. 이상의 결과로 보아 Cobrotoxin 처리는 성상 세포 들을 대상을 LPS 및 SNP로 유도된 NF-${\kappa}B$ 관련 염증 기전과 iNOS, COX-2, cPLA2와 같은 염증관련 유전자의 발현 및 NO, PGE2,에 유의한 변동을 나타내었고, 이들 결과는 Cobrotoxin의 항염증 효과 및 그 치료기전에 대하여 입증한 것이며, 향후 안전성 연구를 바탕으로 신경계 및 심혈관계 염증치료에 약침개발과 같은 적극적인 활용이 기대된다.

양제근(Rumex japonicus HOUTT.)분획의 항염증 효과 (Anti-inflammatory effect of Rumex japonicus HOUTT. in RAW 264.7 cells)

  • Objectives : The aim of this study was to authenticate whether fractionated extract of Rumex japonicus HOUTT. (RJ) has anti-inflammatory effects in mouse macrophage, RAW264.7 cells. Methods : Roots of RJ were extracted by methanol for 48hours. The methanol that gained was filtered and freeze dried. The methanol extract was dissolved in water and dichloromethane (DCM). After that, two layers were separated. Ethyl acetate (EA) added to the water layer and separated again. All the layers were filtered and freeze dried and the extracts were tested. Cytotoxic activity of extracts on RAW 264.7 cells was measured using MTS assay. The nitric oxide (NO) production was measured and proinflammatory cytokines and $PGE_2$ were measured by ELISA kit. The levels of inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2), I ${\kappa}$-B-${\alpha}$ and nuclear NF-${\kappa}B$ p65 expression were detected by western blot. Results : Our results indicated that DCM and EA extracts of RJ inhibited the LPS-induced NO, $PGE_2$ production and iNOS, COX-2 expression accompanied by an attenuation of TNF-${\alpha}$, IL-$1{\beta}$ and IL-6 production in RAW 264.7 cells most effectively. DCM and EA extracts also had suppression effects of LPS-induced NF-${\kappa}B$ and MAPKs activation. Conclusions : This results demonstrate that fractionated extract of RJ has anti-inflammatory effects and among the fractioned extract, dichloromethane and ethyl acetate extract have best anti-inflammatory effects.

대두, 홍삼, 진피로 구성된 발효 추출물의 항염증 효능에 관한 연구 (Anti-inflammatory effects of the fermentation extracts consisting of soybean, red ginseng and Citrus Unshiu Peel)

  • Objectives : Fermentation of herbs has been known to be helpful in improving the immune systems and protecting body against disease. The present study was conducted to evaluate anti-inflammatory effects of the fermentation extracts (FE) consisting of soybean, red ginseng andCitrus UnshiuPeel in lipopolysaccharide (LPS)-activated Raw264.7 cells.Methods : FE were prepared by the fermentation withBacillus Subtilisand then by extraction with ethanol (95%; prepared by the fermentation process). Cell viability was measured by MTT assay. Nitric oxide (NO) production was measured in culture media by Griess assay. The expression of nuclear factor (NF)-κB and inhibitory kappa B alpha (IκBα) was determined by Western blot.Results : LPS-induced production of NO and PGE2was dose-dependently decreased by the treatment of FE in Raw264.7 cells. These suppressive effects of FE on NO and PGE2production were related to the inhibition of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) expression. FE inhibited LPS-induced production of pro-inflammatory cytokines, TNF-α, IL-6, and IL-1βin a dose-dependent manner. Furthermore, FE inhibited the NF-κB signaling pathway through the prevention of LPS-induced degradation of IκBαin cytosol and the nuclear translocation of NF-κB.Conclusions : These findings suggest that FE could have anti-inflammatory effects on LPS-induced inflammatory responses in macrophages.

현토단(玄兎丹)의 RAW 264.7 대식 세포에서의 항염증 효과에 관한 연구 (The study of anti-inflammatory effect of Hyeonto-dan extract in RAW 264.7 macrophage)

  • Objectives : This study aimed to investigate the unknown mechanisms behind the anti- inflammatory activity of Hyeonto-dan(HT) 70% ethanol extract on LPS-stimulated RAW 264.7 cells. Methods : Cells were treated with Hyeonto-dan 1 h prior to addition of 200 ng/mL of LPS. Cell viability was measured by the MTS assay. Nitric oxide levels were determined by the Griess assay. $PGE_2$ were measured using EIA kit. Pro-inflammatory cytokine production was measured by the enzyme-linked immunosorbent assay (ELISA). The expression of COX-2, iNOS, and MAPKs was investigated by Western blot, qRT-PCR. $NF-{\kappa}B$/p65 localization and interaction of the TLR-4 receptor with LPS was examined by immunofluorescence assays. Results : Hyeonto-dan had no cytotoxicity at the measured concentration. Hyeonto-dan inhibited NO production and pro-inflammatory cytokines such as IL-6, $TNF-{\alpha}$, and PGE2 as well as the protein and mRNA expression of iNOS and COX-2. Moreover, Hyeonto-dan inhibited the interaction between LPS and TLR-4 in murine macrophages. It suppressed phosphorylation of extracellular signal-regulated kinase (ERK 1/2), c-jun N-terminal kinase (JNK 1/2) and p38. Finally, it inhibited translocation of $NF-{\kappa}B$ in response to competitive LPS. Conclusions : Based on the results of this study, Hyeonto-dan inhibited the binding of TLR-4 receptor to LPS and inhibited the phosphorylation of extracellular signaling pathway MAPKs. These inhibitory effects are thought that the amount of $NF-{\kappa}B$ delivered to the nucleus was decreased and the inflammatory reaction was prevented by decreasing the production of LPS-induced $PGE_2$, NO, IL-6 and $TNF-{\alpha}$.

Methanol Extracts of Stewartia koreana Inhibit Cyclooxygenase-2 (COX-2) and Inducible Nitric Oxide Synthase (iNOS) Gene Expression by Blocking NF-κB Transactivation in LPS-activated RAW 264.7 Cells

  • Cyclooxygenase-2 (COX-2) and inducible nitric oxide synthase (iNOS) are involved in various pathophysiological processes such as inflammation and carcinogenesis. In a search for inhibitors of COX-2 and iNOS production we found that extracts of Stewartia koreana strongly inhibited NO and $PGE_2$ production in LPS-treated macrophage RAW 264.7 cells. We have now shown that the mRNA and protein levels of iNOS and COX-2 are reduced by the Stewartia koreana extract (SKE). SKE inhibited expression of an NF-${\kappa}B$ reporter gene in response to LPS, and gel mobility shift assays revealed that SKE reduced NF-${\kappa}B$ DNA-binding activity. The extract also inhibited LPS-induced phosphorylation of $I{\kappa}B-{\alpha}$ and nuclear translocation of p65. Administration of the extract reduced the symptoms of arthritis in a collagen-induced arthritic mouse model. These results indicate that Stewartia extracts contain potentially useful agents for preventing and treating inflammatory diseases.

Inhibition of Lipopolysaccharide-Induced Expression of Inducible Nitric Oxide and Cyclooxygenase-2 by Aquaous of Aconitum pseudo-laeve var. erectum in RAW 264.7 Macrophages

  • Aconitum pseudo-laeve var. erectum has traditionally been used for the treatment of water retention in the body. Administration of the aqueous extract of Aconitum pseudo-laeve var. erectum has the efficiency of anti-inflammatory activity and modulates the intestinal immune system. However, the mechanism of anti-inflammatory action of Aconitum pseudo-laeve var. erectum has not been clarified yet. In the present study, the effect of Aconitum pseudo-laeve var. erectum against LPS-stimulated expressions of COX-2 and iNOS in cells of the murine RAW 264.7 macrophages was investigated using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay, reverse transcription- polymerase chain reaction (RT-PCR), PGE2 immunoassay, and NO detection. The results of the present study indicate that Aconitum pseudo-laeve var. erectum is a potent inhibitor of the LPS-induced NO and $PGE_{2}$ production by blocking iNOS and $NF{\kappa}B$ activation in RAW 264.7 macrophages. These findings suggest that Aconitum pseudo-laeve var. erectum is a potential therapeutic for the treatment of inflammatory syndrome.

Inhibitory Effect of Chan-Su on the Secretion of PGE2 and NO in LPS-stimulated BV2 Microglial Cells

  • 본 논문은 오랫동안 민간요법으로 염증치료에 사용되어오던 섬수가 lipopolysaccharide(LPS)-자극된 BV2 소교 세포의 nitric oxide(NO) 분비에 미치는 효과에 대해 연구한 내용이다. 실험 결과 섬수는 세포 생존력에 대한 영향 없이 BV2 소교 세포에서 NO 분비를 억제시켰고, nitric oxide synthase (iNOS) 단백질도 감소시켰다. 또한 섬수는 prostaglandin E2 (PGE2) 생산 및 cyclooxygenase (COX)-2 발현을 저지하였고, proinflammatory cytokines과 ${IkB-\alpha}$감소를 억제시켰다. 따라서 섬수가 $I{\kappa}B-{\alpha}$감소를 억제함으로써 NO 합성을 저해하여 항염증작용을 할 수 있다는 내용이다.
