The Laccase Activity of Trametes versicolor during Cultivation on Acetylated Wood and 13C-CP/MAS NMR Study (아세틸화 처리 목재에 배양시킨 Trametes versicolor의 Laccase활성과 13C-CP/MAS NMR 분석)

  • For examine anti-degradation factors of acetylated wood, acetylated wood was incubated on Trametes versicolor. The laccase activity was examined in broth culture and solid fermentation that contain acetylated chips. The change of acetyl groups and chemical composition in the acetylated wood having massloss analysed by $^{13}C$-CP/MAS NMR. The laccase activity was detected in broth culture. When the T. versicolor contact to acetylated wood directly, the laccase activity was very low and couldn't maintain during test periods. Through the analysing of $^{13}C$-CP/MAS NMR, the acetylation took place carbohydrates as well as lignin and hydroxyl group of amorphous region was more easily substituted that of crystalline region The spectral analyses of $^{13}C$-CP/MAS NMR were shown that introduced acetyl bond was stable against fungal attack.

  • The 8-hydroxyquinoline derivative of silica gel has been prepared through the 5 step reaction. We carried out infrared as well as nuclear magnetic resonance spectrometric characterization of products taken from each step of reaction. IR study of bare silica gel showed free and hydrogen bonded hydroxyl. From the 1st step reaction, we observed IR bands for N-H and C-H as well as NMR peaks for three methylene carbons in APTS group. From the 2nd step, we observed IR bands for carbonyl, nitro and aromatic carbon group with NMR peaks for aliphatic, aromatic and carbonyl carbons. The reduction of $NO_2$ group to $NH_2$ group is confirmed by IR and NMR from 3rd step reaction. In the last step, the immobilization of 8-quinolinol is confirmed by disappearance of $N{\equiv}N$ IR peak observed in 4th step.

  • Stable isotope enrichment in proteins is necessary for high-resolution nuclear magnetic resonance (NMR) experiments. Although methods for 13C, 15N and 2H-enrichment in prokaryotic cells are well established, full processing and correct folding of complex protein systems require higher organisms as the expression host. In the present study, we review recent efforts to enrich stable isotopes in mammalian cells for protein NMR studies.

  • A novel flavonol trisulphate, quercetin 7-methyl ether $3,3',4'-tri-O-KSO_3$ was isolated from the fresh leaves of Tamarix amplexicaulis (Tamaricaceae) along with the known flavonol mono sulphates, quercetin $3-O-KSO_3$ and quercetin 4'-methyl ether $3-O-KSO_3$. Structures were achieved through conventional analytical methods, including electrophoretic analysis and confirmed by FAB-MS and NMR spectroscopy.

  • Purpose: Recently in Korea, there have been significant improvements in neonatal mortality rate (NMR) and infant mortality rate (IMR). This study aimed to investigate the proportion of the NMR among IMR, with the goal of discerning the influence of improved NMR on the reduction of IMR in the last 5 years in Korea. Methods: All data were from Statistics Korea. Changes in the NMR percentage among IMR and the percentage of the death by the distribution of the birth weight and gestational were investigated. Results: The total birth rate decreased, but the total number of preterm and low birth weight infants increased. These was a large decrease in NMR and IMR. The proportion of NMR among INR exceeded 50%. Early NMR was higher than late NMR. Among the total infant death, the mortality of preterm and low birth weight infants was high. Conclusion: Between 2005 and 2009, the total birth has declined in Korea, but the frequency of low birth weight infants is trending upward. The improvements in NMR and IMR, and the downtrend of the NMR percentage in IMR, are encouraging. It seems that the continued decrease of mortality of preterm and LBWI is required for better improvements NMR and IMR in Korea. This result is expected to be used for the basic data to improve the management of the newborns in Korea.

  • Novel measurement method has been performed for the noninvasive study of the swelling effect detected in hydrophilic polymers using Magnetic Resonance Microscopy. iN NMR images were acquired to measure geometric changes due to the swelling effect occurred in the polymer specimens. In addition to the geometric changes, the water ingress process was visualized noninvgsively. The measurement method performed .in the present study utilized some of NMR's valuable properties, both noninvasiveness and parameter selectivity. It is believed that the method used in the present study may be applicable to the study of biopolymers in which noninvasiveness is particularly important.

1H Solid-state NMR Methodology Study for the Quantification of Water Content of Amorphous Silica Nanoparticles Depending on Relative Humidity (상대습도에 따른 비정질 규산염 나노입자의 함수량 정량 분석을 위한 1H 고상 핵자기 공명 분광분석 방법론 연구)

  • The hydrogen in nominally anhydrous mineral is known to be associated with lattice defects, but it also can exist in the form of water and hydroxyl groups on the large surface of the nanoscale particles. In this study, we investigate the effectiveness of 1H solid-state nuclear magnetic resonance (NMR) spectroscopy as a robust experimental method to quantify the hydrogen atomic environments of amorphous silica nanoparticles with varying relative humidity. Amorphous silica nanoparticles were packed into NMR rotors in a temperature-humidity controlled glove box, then stored in different atmospheric conditions with 25% and 70% relative humidity for 2~10 days until 1H NMR experiments, and a slight difference was observed in 1H NMR spectra. These results indicate that amount of hydrous species in the sample packed in the NMR rotor is rarely changed by the external atmosphere. The amount of hydrogen atom, especially the amount of physisorbed water may vary in the range of ~10% due to the temporal and spatial inhomogeneity of relative humidity in the glove box. The quantitative analysis of 1H NMR spectra shows that the amount of hydrogen atom in amorphous silica nanoparticles linearly increases as the relative humidity increases. These results imply that the sample sealing capability of the NMR rotor is sufficient to preserve the hydrous environments of samples, and is suitable for the quantitative measurement of water content of ultrafine nominally anhydrous minerals depending on the atmospheric relative humidity. We expect that 1H solid-state NMR method is suitable to investigate systematically the effect of surface area and crystallinity on the water content of diverse nano-sized nominally anhydrous minerals with varying relative humidity.

  • This study describes a simple and rapid method for quantitative determination of $\gamma$-aminobutyric acid (GABA) using $^1H$ NMR spectroscopy from whole cell extracts of plant suspension cultures. When 9 cell lines derived from 8 species of higher plants maintained in liquid Marashige and Skoog (MS) medium supplemented with 1 mg/L 2,4-dichlorophenoxyacetic acid (2,4-D) were subjected to $^1H$ NMR, a cell line of Coriandrum sativum L. exhibited the highest level of GABA. The level reached up to 16.9 mg/dry wt when cells were cultured in MS medium supplemented with 0.5 mg/L 2,4-D after 3 weeks of incubation. The method for quantitative determination of GABA using $^1H$ NMR established in this study could be applied to high-throughput screening of various plant resources for GABA production and the cell suspension culture system of C. sativum could be further developed for commercial production of GABA.
