Objective: We investigated the effects of outdoor access for 1 h per day on the animal welfare (AW) of tethered cows, in terms of lying and sleeping postures, and immune function. Methods: A total of five dry cows were tethered all day indoors (tethering) for 30 days and then tethered indoors with 1 h daily outdoor access (ODA-1h) for 30 days. To analyze the effects of ODA-1h, we calculated the total duration and bout frequency per day, and bout duration of lying and sleeping postures during the last five days of each treatment period. We also analyzed the populations of T cells, B cells, and NK cells in peripheral blood mononuclear cells (PBMC) by fluorescence-activated cell sorting and determined the concanavalin A (Con A) -induced proliferation rate of T cells. Results: The mean total time per day of lying during the ODA-1h treatment was significantly shorter than that during the tethering treatment (p<0.001). The Con A-induced proliferation rate of T cells during the ODA-1h treatment was significantly higher than that during the tethering treatment (p = 0.007). The proportion of NK cells in PBMC during the ODA-1h treatment tended to be higher than that during the tethering treatment (p = 0.062). Conclusion: Although ODA-1h may decrease lying time, it increases the available space for tethered cows towards that typically found in grazing and free barn feeding systems. This increased available space promotes the expression of normal behaviors such as walking and social behaviors except lying and may also improve the immune function of tethered dry cows, thereby improving their overall welfare.

A field experiment was conducted to investigate the effect of long-term (21-year) fertilizer and compost treatments on the yield of red pepper and chemical properties in top-dong, Suwon. Six treatments were chosen for this work: No fertilization (No fert.), NPK fertilizers (NPK), NPK and compost (NPK+Compost), NP and compost (NP+Compost), NK and compost (NK+Compost), PK and compost (PK+Compost). The yield of red pepper for 21 years indicated the significant differences among the No fertilization, the PK+Compost, and other treatments. The relative yield index was 13% and 59% respectively, for the No fertilization and the PK+Compost if the average yield of red pepper for the NPK regards $20,048kg\;ha^{-1}$ as the yield index with 100%. Soil organic matter at the compost applied treatments significantly increased compared with the No fert. and the NPK. The average increase rates of soil organic matter by applying the compost ranged from 0.69 to $0.73g\;kg^{-1}\;yr^{-1}$. Available phosphate content in soil appeared the significant increase all treatments excluding the No fert. It is estimated that the available phosphate in soil was increased by $7.0mg\;kg^{-1}\;yr^{-1}$ by applying compost and $14.2mg\;kg^{-1}\;yr^{-1}$ by applying P fertilizer. Application of K fertilizer or the compost alone, the NPK, the NP+Compost, continuously caused soil K depletion whereas K fertilization plus the compost maintained at a constant level of exchangeable K. The results indicated that the addition of compost to NPK fertilizer is recommended for the maximum stable yield for red pepper and enhancement of organic matter though it is also needed for adjusting of P and K fertilization.

The immune system is a complex process within the body that protects against disease. Recently, many studies have attempted to discover immunomodulative compounds from natural sources. Pinus koraiensis (PK) cone shell is a by-product of PK. One of the major compounds of PK cone shell is dehydroabietic acid, which has bioactivity, including antiulcer and anti-inflammatory activities. Therefore, this study was performed to examine the immunomodulative effects of PK cone shell. The immunomodulatory effects of PK cone shell extracted with 20% ethanol (EtOH) in vivo were examined initially by measuring the natural killer (NK) cell activity, phagocytic activity, Th1/Th2 cytokines release, serum immunoglobulin, and T/B cell proliferation. The NK cell activity and phagocytic activity were increased significantly by a treatment with a 20% EtOH extract of PK cone shell. Th1 type cytokine and T cell proliferation increased and Th2 type cytokine, B cell proliferation and serum immunoglobulin A, G, and E decreased after a treatment with PK cone shell extract. The 20% EtOH extract of the PK cone shell normalized the unbalanced production of Th1/Th2 type cytokine. This suggests that a 20% EtOH extract of PK cone shell has great potential as a health food.

In order to determine the functional benefits of $Cordyceps$$militaris$ in the immune system, we examined the immunomodulatory activities of $Cordyceps$$militaris$ in an immunocompromised C57BL/6 mice model. Mice were injected intraperitoneally with an immunosuppressive drug, cyclophosphamide, and then administered orally with 3% hydroxypropylmethylcellulose or 30, 100, and 300 mg/kg of 50% ethanol extract of $Cordyceps$$militaris$ (CM 30, CM 100, and CM 300, respectively) for 12 days. Mice treated with CM displayed significantly increased splenocyte proliferation and natural killer cell activity compared to immunosuppressed control mice (p<0.05). The spleen cells isolated from mice treated with CM also displayed increased production of Th1 cytokines, including IL-2, IL-12, IFN-${\gamma}$ and TNF-${\alpha}$, suggesting enhanced cellular immunity in response to CM. However, CM had no significant effect on the production of IL-4 and IL-10. These results indicate that $Cordyceps$$militaris$ enhances immune function by promoting immune cell proliferation and Th1 cytokine production.

Objectives and methods : To study on the anti-cancer, anti-metastasis and immune response improvement effects of Herbal-acupuncture with Sinomenii acuti Lignum infusion solution(SAL-HAS), we injected Sinomenii acuti Lignum infusion solution into Joksamni$(ST_{36})$ of C57BL/6 mouse which is corresponding to human Joksamni(ST36). We observed its effect on the number of $CD25^+/CD4^+,\;CD8^+/CD3e^+,\;CD69^+/B220^+,\;NK1.1^+/CD3e^+$ cells in mouse PBMCs(peripheral blood mononuclear cells), the number of the pulmonary colony, and the effect on MST(mean survival time) and ILS(increase in MST over control) of C57BL/6 mice implanted intravenously with B16-F10 melanoma. Results and Conclusions : 1. The spleen cells proliferation of the sample groups treated with SAL-HAS extract has increased significantly compared with that of the control group. 2. The percentage of the $CD25^+/CD4^+,\;CD8^+/CD3e^+,\;CD69^+/B220^+,\;NK1.1^+/CD3e^+$ cells in C57BL/6 mouse PBMCs of the sample groups treated with SAL-HAS has increased compared with that of the control group. 3. The pulmonary colony number of the sample groups SAL-HAS has decreased significantly compared with that of the control group. 4. MST and ILS of the sample groups SAL-HAS have increased significantly compared with those of the control group.
The anti-obesity effects of starter (Leuconostoc mesenteroides+Lactobacillus plantarum) fermented kimchi on 3T3-L1 adipocyte were studied using naturally fermented kimchi (NK), a functional kimchi (FK, NK supplemented with green tea), and FK supplemented with added starters (FKS). Oil red O staining and cellular levels of triglyceride (TG) and glycerol were used to evaluate the in vitro anti-obesity effects of these kimchis in 3T3-L1 cells. The expressions of adipogenesis/lipogenesis-related genes of peroxisome proliferator-active receptor (PPAR)-${\gamma}$, CCAAT/enhance-binding protein (C/EBP)-${\alpha}$, and fatty acid synthase (FAS) were determined by RT-PCR. Kimchis, especially FKS, markedly decreased TG levels and increased levels of intracellular glycerol and lipid lipolysis. In addition, FKS also reduced the mRNA levels of PPAR-${\gamma}$, C/EBP-${\alpha}$, and FAS, which are related to adipogenesis/lipogenesis in 3T3-L1 cells. These results suggest the anti-obesity effects of FKS were to due to enhanced lipolysis and reduced adipogenesis/lipogenesis in 3T3-L1 adipocytes.
The purposes of this study were to set up the method of the natural killer(NK) cell activity assay using the flow cytometer and to examine the characteristics and distribution of the NK cell during rat hepatocarcinogenesis. Forty five male 6 week-old specific pathogen free(SPF) Sprague-Dawley rats were randomly divided into three groups. Group I was the non-treated control and given normal diet and water. Group II was treated with diethylnitrosamine(DEN, 200mg/kg, i.p.) and partial hepatectomy. Group III was treated with DEN, partial hepatectomy and 0.05% phenobarbital sodium in water from 3 to 16 weeks. All animals were examined the morphology of the large granular lymphocyte(LGL), the LGL percent of the total lymphocytes and the LGL conjugation rate with YAC-1 cell in peripheral blood, spleen and liver. Moreover, activity of the LGL isolated from peripheral blood lymphocytes was determined using the flow cytometer. As results, LGL were observed in the peripheral blood, spleen and liver. LGL were observed the relatively faintly staining basophilic cytoplasm with granules, and eccentric, often kidney-shaped nuclei in Giemsa stain. Its size was $11{\sim}13{\mu}m$. LGL percentage of the isolated lymphocytes in peripheral blood, spleen and liver were 1.8~2.3%, 1.3~1.4% and 0.87~0.99%, respectively. LGL conjugation rate with YAC-1 cell was shown to be peripheral blood(9.3~10.3 %) > spleen(7.7~8.7%) > liver(5.6~7.0%). The activity of the LGL isolated from peripheral blood lymphocytes in Group I, II and III was 33.7%, 30.5% and 35.4%, respectively. However, all values were not significantly between groups.

Objective: To observe the effects of allogeneic and autologous transfusion on cellular immunity, humoral immunity and secretion of serum inflammatory factors and perforin during the perioperative period in patients with malignant tumors. Methods: A total of 80 patients (age: 38-69 years; body weight: 40-78 kg; ASA I - II) receiving radical operation for gastro-intestinal cancer under general anesthesia were selected. All the patients were divided into four groups based on the methods of infusion and blood transfusion: blank control group (Group C), allogeneic transfusion group (group A), hemodiluted autotransfusion Group (Group H) and hemodiluted autotransfusion + allogenic transfusion Group (A+H group). Venous blood was collected when entering into the surgery room ($T_0$), immediately after surgery ($T_1$) and 24h ($T_2$), 3d ($T_3$) and 7d ($T_4$) after surgery, respectively. Moreover, flow cytometry was applied to assess changes of peripheral blood T cell subpopulations and NK cells. Enzyme linked immunosorbent assays were performed to determine levels of IL-2, IL-10, TNF-${\alpha}$ and perforin. Immune turbidimetry was employed to determine the changes in serum immunoglobulin. Results: Both CD3+ and NK cells showed a decrease at $T_1$ and $T_2$ in each group, among which, in group A, CD3+ decreased significantly at $T_2$ (P<0.05) compared with other groups, and CD3+ and NK cell reduced obviously only in group A at $T_3$ and $T_4$ (P<0.05). CD4+ cells and the ratio of D4+/CD8+ were decreased in groups A, C and A+H at $T_1$ and $T_2$ (P<0.05). No significant intra- and inter-group differences were observed in CD8+ of the four groups (P<0.05). IL-2 declined in group C at $T_1$ and $T_2$ (P<0.05) and showed a decrease in group A at each time point (P<0.05). Moreover, IL-2 decreased in group A + H only at $T_1$. No significant difference was found in each group at $T_1$ (P<0.05). More significant decrease in group ?? at $T_2$, $T_3$ and $T_4$ compared with group A (P<0.05), and there were no significant differences among other groups (P>0.05). IL-10 increased at $T_1$ and $T_2$ in each group (P<0.05), in which it had an obvious increase in group A, and increase of IL-10 occurred only in group A at $T_3$ and $T_4$ (P<0.05). TNF-${\alpha}$ level rose at $T_1$ (P<0.05), no inter- and intra-group difference was found in perforin in all groups (P<0.05). Compared with the preoperation, both IgG and IgA level decreased at $T_1$ in each group (P<0.05), and they declined only in Group A at $T_2$ and $T_3$ (P<0.05), and these parameters were back to the preoperative levels in other groups. No significant differences were observed between preoperative and postoperative IgG and IgA levels in each group at $T_4$ (P>0.05). No obvious inter- and intra-group changes were found in IgM in the four groups (P>0.05). Conclusions: Allogeneic transfusion during the perioperative period could obviously decrease the number of T cell subpopulations and NK cells and the secretion of stimulating cytokines and increase the secretion of inhibiting cytokines in patients with malignant tumors, thus causing a Th1/Th2 imbalance and transient decreasing in the content of plasma immune globulin. Autologous transfusion has little impact and may even bring about some improvement oo postoperative immune function in patients with tumors. Therefore, cancer patients should receive active autologous transfusion during the perioperative period in place of allogeneic transfusion.
